Induction of apoptosis by water extract Glycyrrhizae radix in human bladder T24 cancer cells (인체 방광암 T24 세포에서 Glycyrrhizae radix 열수추출물에 의한 apoptosis 유도)

  • Glycyrrhizae radix is one of the most frequently prescribed ingredients in Oriental medicine, and G. radix extract has been shown to exert anti-cancer effects. However, the cellular and molecular mechanisms of apoptosis by G. radix are poorly defined. In the present study, it was examined the biochemical mechanisms of apoptosis by water extract of G. radix (WEGR) in human bladder T24 cancer cells. It was found that WEGR could inhibit the cell growth of T24 cells in a dose-dependent manner, which was associated with the induction of apoptotic cell death, as evidenced by the formation of apoptotic bodies, DNA fragmentation and increased populations of annexin-V positive cells. The induction of apoptotic cell death by WEGR was connected with an up-regulation of pro-apoptotic Bax protein expression and down-regulation of anti-apoptotic Bcl-2 and Bcl-xL proteins, and inhibition of apoptosis family proteins (XIAP, cIAP-1 and cIAP-2). In addition, apoptosis-inducing concentrations of WEGR induced the activation of caspase-9, an initiator caspase of the mitochondrial-mediated intrinsic pathway, and caspase-3, accompanied by proteolytic degradation of poly (ADP-ribose)-polymerase. WEGR also induced apoptosis via a death receptor-mediated extrinsic pathway by caspase-8 activation, resulting in the down-regulation of total Bid and suggesting the existence of cross-talk between the extrinsic and intrinsic pathways. Taken together, the present results suggest that WEGR may be a potential chemotherapeutic agent for the control of human bladder cancer cells.

Baicalein induces cell death in Human Lung Carcinoma A549 Cells: Role of Apoptosis and Autophagy pathway (인체폐암 A549 세포에서 Baicalein에 의한 세포사멸 유도: Apoptosis와 Autophagy 경로의 역할)

  • Baicalein is one of the main flavonoids derived from roots of Scutellaria baicalensis Georgi, a traditional Oriental medicine. Although baicalein has high antitumor effect on several human carcinomas, the mechanism responsible for this property is not unclear. In this study, the data revealed that baicale-ininduced growth inhibition was associated with the induction of apoptosis connecting with cytochrome c release, down-regulation of anti-apoptotic Bcl-xl and increased the percentage of cells with a loss of mitochondria membrane permeabilization. Baicalein also induced the proteolytic activation of caspases and cleavage of PARP; however, blockage of caspases activation by z-VAD-fmk inhibited baicalein-induced apoptosis. In addition, baicalein enhanced the formation of autophagosomes and up-regulated LC3-II/LC3-I ratio. Interestingly, the pretreatment of bafilomycin A1 recovered baicalein-induced cell death suggesting that autophagy by baicalein roles as protective autophagy. Taken together, our results indicated that this flavonoid induces apoptosis and cell protective autophagy. These data means combination treatment with baicalein and autophagy inhibitor might be a promising anticancer drug.

Anti-inflammatory effects of DATS via suppression of cross talk between the TLR4/NF-κB and CXCL12/CXCR4 pathways in LSP-stimulated RAW 264.7 macrophages (LSP로 유도된 RAW 264.7 대식세포에서 TLR4/NF-κB와 CXCL12/CXCR4 경로 억제를 통한 DATS의 항염증 효과)

  • Diallyl trisulfide (DATS) is an organic polysulfide compound found in garlic. Although certain studies have demonstrated that DATS possesses strong anti-inflammatory activity, the underlying molecular mechanisms remain largely unresolved. In this study, we examined whether DATS exerts anti-inflammatory activity and investigated the possible mechanisms. Our results indicated that DATS significantly suppressed the lipopolysaccharide (LPS)-induced release of nitric oxide (NO) and prostaglandin E2 by inhibiting inducible NO synthase and cyclooxygenase-2 expression at the transcriptional and post-transcriptional levels in RAW 264.7 macrophages. DATS also down-regulated Toll-like receptor 4 (TLR4) and myeloid differentiation factor 88 expression, and inhibited nuclear translocation of nuclear transcription factor-kappa B (NF-${\kappa}B$) in LPS-stimulated 264.7 macrophages. Furthermore, we found that these inhibitory effects of DATS were associated with the inhibition of chemokine receptor (CXCR4) and ligand (CXCL12) expression, and reactive oxygen species generation. Overall, the present data indicated that DATS had anti-inflammatory effects on LPS-activated macrophages, possibly via inhibiting the TLR4/NF-kB and/or chemokine signaling pathways, and DATS could be a potential drug therapy for inflammation and its associated diseases.

Difference of Physiochemical Characteristics Between Citrus Bacterial Canker Pathotypes and Identification of Korean Isolates with Repetitive Sequence PCRs

  • The difference of carbon source utilization and fatty acid composition between the pathotypes of Xanthomonas strains, which causing citrus bacterial canker was compared, and the physiochemical characteristics were used to analyze relationship of the strains for the first time. The pattern of several carbon sources utilization and fatty acids composition reliably discriminated the pathotypes of Xanthomonas strains. The dendrogram which was constructed by 95 carbon source utilization profiles differentiated X. axonopodis pv. citri A, $A^*$ and $A^w$ from the other pathotypes. When the dendrogram was drawn by combined analysis of carbon source utilization pattern and fatty acid composition, X. axonopodis pv. aurantifolii B, C and X. axonopodis pv. citrumelo formed a distinct cluster. The difference of carbon source utilization and fatty acid composition could be used effectively for the identification of pathotypes of citrus bacterial canker. The physiochemical characteristics strongly indicated that the strains isolated in Korea belong to X. axonopodis pv. citri A type. The cluster analysis by the band patterns of ERIC-, BOX- and REP-PCR allowed the discrimination of the pathotypes isolated from Korea. However, the rep-PCRs could not differentiate X. axonopodis pv. citri A types from $A^*$ and $A^w$ types. The overall results of metabolic profiles and rep-PCRs strongly indicated that the Korean isolates are X. axonopodis pv. citri A type.

Phosphorus Accumulation and Utilization Efficiency in Soybean Plant under Atmospheric CO2 Enrichment

  • Soybean plants(Glycine max [L.] merr.) inoculated with Bradyrhizobium japonicum MN110 were grown in growth chambers under 400 or $800{\mu}l{\cdot}l^{-1}$ atmospheric $CO_2$ and harvested at 25, 28, 32, and 35 DAT to examine the effect of $CO_2$ enrichment on phosphorus accumulation, uptake, and utilization efficiency during vegetative growth. Phosphorus concentration in leaf was lower in high $CO_2$ plant by 47% at 25 DAT and 34% at 35 DAT than those in the control plant but phosphorus concentrations in stem, root and nodule were not affected by $CO_2$ enrichment. Total phosphorus accumulation increased 3.9-fold in high $CO_2$ plant and 3.2-fold in the control plant between 25 and 35 DAT. Elevated $CO_2$ caused a decrease in the whole plant phosphorus concentration by 35%, which was due almost entirely to a decrease in the phosphorus concentration of leaves. $CO_2$ enrichment increased phosphorus utilization efficiency in the whole plant by 70% during the experimental period. Plants exposed to high $CO_2$ had larger root systems than under ambient $CO_2$, but high $CO_2$ plants had lower P-uptake efficiency. Averaged over four harvests, plants at high $CO_2$ had 38% larger root mass that was more than offset the 20% lower efficiency of P-uptake and accounted for increased phosphorus accumulation by high $CO_2$ plant. These results suggest that the reduced phosphorus concentration in soybean plant under $CO_2$ enrichment may be an acclimation response to high $CO_2$ concentration or enhanced starch accumulation, resulting in the plants to have a lower phosphorus requirement on a unit dry weight basis or a high phosphorus utilization efficiency under these conditions.

A Basic Study on Utilization of Angelica acutiloba Kitag (Tanggui)

  • This study was conducted to determine feasibility of utilization of Angelica acutiloba. Especially, the quality characteristics of bread prepared with the addition of Angelica acutiloba powder were investigated. Sensory evaluation and spoilage test were conducted for preparation of functional breads which added with ground plant matters (leaves and stems) from Angelica acutiloba. The result showed that the functional breads had high score of overall liking as well as low spoilage rate when added with 0.5 to 1.0% ground plant matters of Angelica acutiloba. Consumer acceptability evaluation showed a significant preference when added 0.5 to 1.0% ground leaves and stems of Angelica acutiloba into breads. Functional breads which added powder of Angelica acutiloba inhibited the growth of fungi. The more addition of Angelica acutiloba powder, the higher the degrees of this inhibited. These results suggested that the shelf-lives of the breads were extended by the addition of Angelica acutiloba powder. Further studies were required for improvement of functionality and diversity of bread products using medicinal plant materials as an additive.

  • To study the possibility of agricultural utilization of the sewage sludge and the converter slag, the chemical properties of soil, the growth response of corn plant and uptake of inorganic nutrients in plant tissues were investigated by application of the composts made of the sewage sludge and converter slag. Uptake of inorganic nutrients in stem and leaf of corn plant were decreased by applications of the composts. The content of heavy metals in soil and corn plant were investigated, but the results of show that the concentrations of heavy metals are much low.

  • In this paper, we presents a systematic methodology for minimizing tardiness and maximizing resource utilization in a multi-plant supply chain. A methodology is represented to a multi-objective mathematical program model. The model offers flexible and efficient multi-plant planning and scheduling. Also, We develope a realistic and flexible planning model using the genetic algorithm to solve the model.
