A Study on the Environment-friendly Planning Elements on High-Class Houses Kuangajeong in Chosun Dynasty (관가정(觀稼亭)으로 본 조선시대 상류주택의 환경친화적 계획요소 분석)

  • This study aims to examine the traditional living space to reflect the environment-friendly design methods and principles of reestablishment. To do so, this study carried on a related literature study and field survey. The traditional living space in terms of the environmental friendliness is reflected on site selection and space composition, and utilization of natural energy and natural materials. Focused on the Kwangajeong in Yangdong Village, this study is to identify eco-friendly techniques on the traditional living space. As a result, it shows that Kwangajeong with the side of the southeast in site selection had been considered in aspect of environmental control such as sunshine and solar radiation. Also building construction techniques had been used to minimize the variation of terrain. In aspect of environmental conservation, Kwangajeong had structurally arranged the gate, an inner yard and an inner floor to allow good ventilation. In aspect of space composition, Kwangajeong with the emphasis of scenic view had utilized the methods that attract external landscape through the control of its fences. Environment-friendly techniques and control principles in traditional space had been developed by a long experience through the long periods and the traditional techniques suitable for local climate and local environment have been developed. The technical development of these techniques to resolve the global environmental and energy issues and create a more pleasant living environment of human beings might be critical.

Assessment of Environmental Radioactivity Surveillance Results around Korean Nuclear Power Utilization Facilities in 2017

  • Background: Government conducts environmental radioactivity surveillance for verification purpose around nuclear facilities based on the Nuclear Safety Law and issues a surveillance report every year. This study aims to evaluate the short and the long-term fluctuation of radionuclides detected above MDC and their origins using concentration ratios between these radionuclides. Materials and Methods: Sample media for verification surveillance are air, rainwater, groundwater, soil, and milk for terrestrial samples, and seawater, marine sediment, fish, and seaweed for marine samples. Gamma-emitting radionuclides including $^{137}Cs$, $^{90}Sr$, Pu, $^3H$, and $^{14}C$ are evaluated in these samples. Results and Discussion: According to the result of the environmental radioactivity verification surveillance in the vicinity of nuclear power facilities in 2017, the anthropogenic radionuclides were not detected in most of the environmental samples except for the detection of a trace level of $^{137}Cs$, $^{90}Sr$, Pu, and $^{131}I$ in some samples. Radioactivity concentration ratios between the anthropogenic radionuclides ($^{137}Cs/^{90}Sr$, $^{137}Cs/^{239+240}Pu$, $^{90}Sr/^{239+240}Pu$) were similar to those reported in the environmental samples, which were affected by the global fallout of the past nuclear weapon test, and Pu atomic ratios ($^{240}Pu/^{239}Pu$) in the terrestrial sample and marine sample showed significant differences due to the different input pathway and the Pu source. Radioactive iodine ($^{131}I$) was detected at the range of < $5.6-190mBq{\cdot}kg-fresh^{-1}$ in the gulfweed and sea trumpet collected from the area of Kori and Wolsong intake and discharge. A high level of $^3H$ was observed in the air (Sangbong: $0.688{\pm}0.841Bq{\cdot}m^{-3}$) and the precipitation (Meteorology Post: $199{\pm}126Bq{\cdot}L^{-1}$) samples of the Wolsong nuclear power plant (NPP). $^3H$ concentration in the precipitation and pine needle samples showed typical variation pattern with the distance and the wind direction from the stack due to the gaseous release of $^3H$ in Wolsong NPP. Conclusion: Except for the detection of a trace level of $^{137}Cs$, $^{90}Sr$, Pu, and $^{131}I$ in some samples, anthropogenic radionuclides were below MDC in most of the environmental samples. Overall, no unusual radionuclides and abnormal concentration were detected in the 2017's surveillance result for verification. This research will be available in the assessment of environment around nuclear facilities in the event of radioactive material release.

On-Farm Study of the System of Rice Intensification (SRI) of Rainfed Lowland in Southern Cambodia (캄보디아 남부 천수답지역의 벼 집약관리재배 시스템(SRI)에 대한 현장 연구)

  • The System of Rice Intensification (SRI) has been spread very quickly in southern Cambodia. To understand the motivation of farmers in adapting SRI, and its benefits, we conducted an on-farm study at Popel commune, Tramkak district, Takeo province in southern Cambodia, during the 2012 and 2013 wet season. We noticed a significant difference between SRI and conventional farmers' practices (FP) in rainfed lowland rice ecosystem. Despite of low nitrogen input, without chemical fertilizers, high grain yield was achieved in SRI 1 (6.0 t ha) and SRI2-Bottom ($7.2t\;ha^{-1}$) in 2013. SRI 1 and SRI 2 of panicle and number of panicle were high than SR 3, FP 2, and FP 3 due to early transplant. Relationships between total number of spikelet and plant nitrogen were ($r^2=0.95$) highly positive at harvest. SRI fields were, most of them achieving highly superior yield and number of panicle compared to their FP fields. The results indicated that SRI practices of planting younger seedling, with organic material and topography of paddy, lead to increased grain yield.

Identification, Enzymatic Activity, and Decay Ability of Basidiomycetous Fungi Isolated from the Decayed Bark of Mongolian Oak (Quercus mongolica Fisch. ex Ledeb.)

  • Decay fungi can decompose plant debris to recycle carbon in the ecosystem. Still, they can also be fungal pathogens, which can damage living trees and/or wood material and cause a large amount of timber loss. We isolated and identified basidiomycetous fungi from the decayed bark of Mongolian oak wrapped with sticky roll traps. The degrading enzyme activities were then tested for all fungal isolates. The decay ability of selected isolates was assessed based on the weight loss of wood discs after inoculating with culture suspension of decay fungi under the different humidity levels. A total of 46 basidiomycetous fungal isolates belonged to 12 species, and 10 genera were obtained from Jong Myo (16 isolates), Chang Kyung palace (7 isolates), Cheong Gye (10 isolates), and Gun Po (13 isolates). Gymnopus luxurians was the most dominant fungus in the present study, and this species distributed in all survey sites with 9 isolates in Jong Myo, followed by 3 isolates in Chang Kyung palace, while Cheong Gye and Gun Po had only 1 isolate each. Among 46 isolates, 44 isolates secreted at least one enzyme, while 25 isolates produced both cellulase and phenol oxidase enzymes, and 2 isolates produced neither. The assessment of decay ability by artificial inoculation indicated that the weight loss of wood discs was significantly influenced by humidity conditions when inoculated with bark decay fungi. The percent weight losses by G. luxurians inoculation in RH of 90-100% and RH of 65-75% were 4.61% and 2.45%, respectively. The weight loss caused by Abortiporus biennis were 6.67% and 0.46% in RH of 90-100% and RH of 45-55%, respectively. The humidity reduction approach should be applied for further studies to control the growth and spread of bark decay fungi on the trunks wrapped with sticky roll traps.

Sawdust Substitution in Growth Medium of Oyster Mushroom for Using Its By-product Spent Mushroom Substrates as Ruminant Feed (수확 후 배지의 가축 사료화를 위한 느타리 생육배지 톱밥 대체재료 선발 연구)

  • To replace the sawdust in the growth medium of oyster mushroom to utilize its by-product spent mushroom substrates (SMS) as feed for ruminant, we performed cultivation test using cotton seed hull pellet (CSHP), corn stalk pellet (CSP), corncob (CC), and analyzed the feed chemical properties of those SMS. As a result of cultivation test, CC and CSHP treatment took 27 days for spawn run, 4 days for primordium formation, and 3 days for development fruiting bodies, resulting in a total cultivation period of 34 days. The yield per bottle was 134 g for CC treatment, similar to 130 g for control, while CSHP treatment (112 g) and CSP treatment (68 g) were lower than that of control. The highest biological efficiency (BE) was shown in CC treatment as 80.1%, which was 11.4% higher than 68.7% of control. The SMS of CC treatment had a relatively low content of neutral detergent fiber and acid detergent fiber, and in particular, lignin content was the lowest and crude protein content was the highest among other treatments. Therefore, CC as a substitute material for sawdust was capable of stable mushroom production and excellent nutritional value as a feed for its by-products.

Adipocyte differentiation inhibition, whitening, antibacterial and antioxidant activities of extracts from Aloe vera by-product (알로에 베라 가공 부산물 추출물의 지방세포 분화억제, 미백, 항균 및 항산화 활성)

  • Aloe has been widely used as a cosmetic and medicinal plant. Until now, several effects such as antioxidant, anti-cancer, anti-diabetic, immunity and whitening of aloe gel extract have been reported, but research on aloe by-products occurring in food processing has not been actively conducted. In this study, we investigated whether the aloe by-product extract from food processing could be used as a functional biomaterial. Cytotoxicity was not seen in both the mixer and press extracts. Inhibition of 3T3-L1 adipocyte differentiation was detected only in the mixer extract and not in the press. It was confirmed that hyaluronic acid accumulation and tyrosinase inhibition increased according to the treatment concentration of the mixer extract. The antimicrobial activity of the mixer extract was observed in the Porphyromonas gingivalis strain, but not in the Streptococcus mutans strain. Antioxidant activity through DPPH and SOD analysis increased with the concentration of the mixer extract. In summary, it was confirmed that the mixer extract of aloe by-products has the effect of inhibiting adipocyte differentiation, moisturizing, whitening, and antioxidant, suggesting the possibility of using it as a functional bio-material for health drinks or beauty masks.

Atractylenoide II Isolated from Atractylodes macrocephala Inhibited Inflammatory Responses in Lipopolysaccharide-induced RAW264.7 Macrophages and BV2 Microglial Cells (백출에서 분리된 Atractylenolide II의 RAW264.7 대식세포와 BV2 미세아교세포에서의 항염증 효과)

  • Atractylodes macrocephala is a perennial herb and is a member of the Compositae family. This plant is known to contain various bioactive constituents indicating anti-inflammatory, neuroprotective, anti-oxidant, immunological enhancement, and gastroprotective effects. In this investigation, we isolated four compounds with similar chemical structures from A. macrocephala, and evaluated their anti-inflammatory effects. Among the four compounds, compound 2(atractylenolide II) showed the second-best inhibitory effect on the lipopolysaccharide(LPS)-induced production of nitric oxide in RAW264.7 macrophages and BV2 microglial cells. Compound 2 also inhibited the LPS-induced the production of prostaglandin E2(PGE2), and the expression of inducible nitric oxide synthase(iNOS) and cyclooxygenase(COX)-2 proteins in both cells. In addition, compound 2 suppressed the production of pro-inflammatory cytokines including interleukin(IL)-1β, IL-6, and tumor necrosis factor(TNF)-α. These inhibitory effects were contributed by inactivation of nuclear factor kappa B(NF-κB) and mitogen-activated protein kinases(MAPKs) pathways by treatment with compound 2. This compound did not induce the expression of heme oxygenase(HO)-1 protein indicating that the anti-inflammatory effect of compound 2 was independent with HO-1 protein. Taken together, these results suggested that atractylenolide II can be a candidate material to treat inflammatory diseases.

Analysis of Essential Oil Components using Elsholtzia splendense Nakai, a Fragrant Plant Distributed in Korea (자생 방향성 식물 꽃향유의 정유성분 분석)

  • In order to extract the essential oil contained in the Elsholtzia splendense Nakai, a fully-bloomed individual was collected and the living body was used for the experiment. Plants were divided into 0.7kg of flowers and 1.5kg of leaves + stems, and extracted by parts and used for scent pattern analysis, and extracted after adding 5kg of outpost for the analysis of essential oil components. Essential oil extraction was performed using the SDE method improved by Schultz et al. (1977), and the extraction time was limited to 1 hour after the material started boiling. The extracted essential oil component was analyzed for fragrance pattern analysis using an e-nose, and the results of analyzing the substance of the essential oil component by GC-MS are as follows. 1. As for the fragrance pattern analysis, in the case of essential oils extracted from flowers, the scent quality was the best at 10-20 minutes, and the scents extracted from stems and leaves were somewhat of poor scent quality, but the fragrance was good at 10-40 minutes. The intensity of scent was the strongest in 10-20 minutes, and the intensity of incense was high even in 30-40 minutes. The scent extracted from the stems and leaves was generally not strong, but appeared high in 10 to 20 minutes. 2. There were 40 kinds of essential oils contained in Elsholtzia splendense Nakai oil. Among them, Mequinol, Benzene, 1,2,3,4-tetramethyl, Elsholtziaketone, and Dehydroelsholtziaketone were identified.

Effect of Essential Oil from Cuscuta japonica CHOISY on Proliferation and Migration in Human Skin Keratinocyte (새삼(Cuscuta japonica CHOISY) 유래 정유의 피부 각질형성세포 증식 및 이주에 미치는 효과)

  • The migration and proliferation of keratinocytes are key events in re-epithelization, itself a major phase in the wound healing process. Cuscuta japonica Choisy (CJC) is used as a traditional medicine to improve liver, heart, and intestinal function, and its extracts are reported to have various biological properties such as whitening, anti-oxidancy, and an anti-acne effect. However, it is not yet known in particular whether or not CJC essential oil (CJCEO) affects skin regeneration. In the present study, we isolated CJCEO by solvent extraction and tested its effect on wound healing responses using normal human keratinocytes, namely HaCaT cells. We found that CJCEO induced proliferation as well as migration in HaCaT cells in a dose-dependent manner. Compared with a control group, CJCEO treatment at 250 ㎍/ml increased proliferation by 239.98±5.51% in HaCaT cells in a dose and migration by 124.86±6.06%. Moreover, the oil induced sprout outgrowth and, at 250 ㎍/ml, increased collagen synthesis by 148.56±15.47% in HaCaT cells. These results demonstrate that CJCEO may promote skin regeneration and wound healing by increasing the migration, proliferation, and collagen synthesis of HaCaT cells. We therefore suggest that CJCEO could be used as a cosmetic material.

Effect on Nitrous Oxide Emission in Applying Livestock Manure Compost for Strawberry (Fragaria × ananassa Duch.) Cultivation in Plastic Film House (딸기(Fragaria × ananassa Duch.) 시설재배에서 가축분 퇴비 시용이 아산화질소 배출에 미치는 영향)

  • This study was conducted to investigate the growth characteristics of strawberries and N2O emission by treating the compost for each type of livestock manure, which was an organic farming material, as a basal fertilization in plastic film house. Livestock manure compost, which made from cattle manure, swine manure, and poultry manure as raw materials, were applied to this experiment, treated by mixing or single on the basis of nitrogen content with the standard amount of fertilizer for strawberries. Total emission of N2O were 10.7% higher than those in poultry manure compost treatment compared to the inorganic fertilizer treatment, but 16.5~41.9% lower than those in other livestock manure compost treatment. The period of N2O emission mainly was up to the 17th day after fertilizer application, accounting for 70~87% of the total amount of discharge, and 13~30% of the total amount was emitted for 158 days later. N2O emission was decreased significantly NH4+-N content in the soil, and increased NO3--N. As compared with control, the number of leaves, leaf width and crown diameter of livestock manure compost treatments were not significantly different, leaf length of cattle+poultry, cattle+ swine, swine+poultry treatment higher, and SPAD (soil plant analysis development) values of cattle+poultry treatment highest. There was no significant difference in weight and sugar content of strawberry fruits among treatments.
