Evaluation of Clubroot Resistance in Chinese Cabbage and Its Inheritance in the European Turnip Line 'IT033820', a New Genetic Resource

  • Clubroot caused by the protist Plasmodiophora brassicae is one of the most destructive diseases of Brassica crops. Developing Chinese cabbage cultivars with durable clubroot resistance (CR) is an important goal of breeding programs, which will require new genetic resources to be identified and introduced. In this study, we evaluated resistance to P. brassicae race 4 using 26 Chinese cabbage (B. rapa ssp. pekinensis ) cultivars compared to the clubroot-susceptible Chinese cabbage inbred line 'BP079' and the clubroot-resistant European turnip (B. rapa ssp. rapifera ) inbred line 'IT033820'. No symptoms of clubroot disease were found in 'IT033820' infected with P. brassicae race 4, whereas the Chinese cabbage cultivars exhibited disease symptoms to various degrees. The Chinese cabbage cultivars that were reported to be clubroot-susceptible were susceptible to P. brassicae race 4; however, seven of the 20 cultivars reported to be clubroot-resistant were susceptible to this race of P. brassicae to varying degrees. Resting spores of P. brassicae were abundant within the infected root tissues of 'BP079', as revealed by light microscopy and scanning electron microscopy (SEM), but they were not detected in root tissues of 'IT033820'. Although resting spores were not detected by light microscopy in root tissues of the clubroot-resistant Chinese cabbage cultivar 'Kigokoro 75', a few spores were observed by SEM. The $F_1$ hybrids from a cross between 'IT033820' and 'BP079' showed no disease symptoms, and all $BC_1P_1$ progenies from a cross between the $F_1$ hybrid and 'IT033820' exhibited a resistance phenotype. In the $BC_1P_2$ population from a cross between the $F_1$ hybrid and 'BP079', this trait segregated at a ratio of 3(R):1(S) (${\chi}^2=1.333$, p = 0.248) at a 5% significance level. Inoculated $BC_1P_2$ plants were either highly resistant or highly susceptible to the pathogen, indicating that the CR to race 4 of P. brassicae carried by 'IT033820' is dominant. In the $F_2$ population, this trait segregated at a ratio of 15(R):1(S) (${\chi}^2=0.152$, p = 0.696) at a 5% significance level, suggesting that CR in 'IT033820' is mainly controlled by two dominant genes. Therefore, 'IT033820' represents a promising genetic resource for developing durable CR breeding lines in Chinese cabbage.

Human and Animal Disease Biomarkers and Biomonitoring of Deoxynivalenol and Related Fungal Metabolites as Cereal and Feed Contaminants (곡물 및 사료오염 데옥시니발레놀 및 대사체에 의한 인축질환 연계 생체지표 및 바이오모니터링)

  • Deoxynivalenol (DON) and related trichothecene mycotoxins are extensively distributed in the cereal-based food and feed stuffs worldwide. Recent climate changes and global grain trade increased chance of exposure to more DON and related toxic metabolites in poorly managed production systems. Monitoring the biological and environmental exposures to the toxins are crucial in protecting human and animals from toxicities of the hazardous contaminants in food or feeds. Exposure biomarkers including urine DON itself are prone to shift to less harmful metabolites by intestinal microbiota and liver metabolic enzymes. De-epoxyfication of DON by gut microbes such as Eubacterium strain BBSH 797 and Eubacterium sp. DSM 11798 leads to more fecal secretion of DOM-1. By contrast, most of plant-derived DON-glucoside is also easily catabolized to free DON by gut microbes, which produces more burden to body. Phase 2 hepatic metabolism also contributes to the glucuronidation of DON, which can be useful urine biomarkers. However, chemical modification could be very typical depending on the anthropologic or genetic background, luminal bacteria, and hepatic metabolic enzyme susceptibility to the toxins in the diet. After toxin exposure, effect biomarkers are also important in estimating the linkage and mechanisms of foodborne diseases in human and animal population. Most prominent adverse effects are demonstrated in the DON-induced immunological and behavioral disorders. For instance, acutely elevated interleukin-8 from insulted gut exposed to dietaty DON is a dominant clinical biomarker in human and animals. Moreover, subchronic exposure to the toxins is associated with high levels of serum IgA, a biological mediator of IgA nephritis. In particular, anorexia monitoring using mouse models are recently developed to monitor the biological activities of DON-induced feed refusal. It is also mechanistically linked to alteration of serotoin and peptide YY, which are promising biomarkers of neurological disorders by the toxins. As animal-alternative biomonitoring, huamn enterocyte-based assay has been developed and more realistic gut mimetic models would be useful in monitoring the effect biomarkers in resposne to toxic contaminants in the future investigations.

Comparative Analysis of the Constituents of the Leaves and Roots of Rumex crispus and their Effects on the Differentiation of Human Osteoblast-like MG-63 Cells (소리쟁이 잎과 뿌리 성분 분석 및 사람 조골 유사 MG-63 세포 분화에 미치는 효과 비교)

  • Rumex crispus (curled dock), which is a perennial wild plant, has long been used as a laxative, astringent, and medicine to treat blood and skin diseases. We recently reported that the roots of R. crispus are an effective nutraceutical for bone. This study prepared ethanol extracts of the leaves and roots of R. crispus, and analyzed the major constituents using liquid chromatography and mass spectrometry. In addition, their effects on the proliferation and differentiation of human osteoblast-like MG-63 cells, such as cell viability, alkaline phosphatase (ALP) activity, collagen content, and mineralization, were compared. The chromatograms of the chemical constituents of the two extracts exhibited quite different profiles: quercetin and quercitrin were identified as major peaks in the leaf extract, whereas cinnamtannin B1 and procyanidin isomers were the major peaks for the root extract. Neither extract was cytotoxic at concentrations of < $25{\mu}g/ml$. ALP activity and collagen synthesis-which are markers of the early stage of osteogenesis-in MG-63 cells were significantly increased upon the addition of the root extract compared with the addition of the leaf extract. In contrast, the leaf extract had a more stimulatory effect on mineralization-which is marker of the late stage of osteogenesis-in MG-63 cells than did the root extract. In conclusion, extracts of both leaves and roots of R. crispus stimulated the bone-forming activity of osteoblasts; in particular, the root extract was more effective in the early stage of osteoblast differentiation, while the leaf extract was more effective in the late stage. This difference in anabolic activity may be due to differences in the constituents of the leaves and roots. The leaves and roots of R. crispus appear to complement each other as stimulators of bone formation.

Identification of a Major QTL, qSTV11SG, Associated with Resistance to Rice Stripe Virus Disease Originated from Shingwangbyeo in Rice (Oryza Sativa L.) (신광벼 유래의 벼 줄무늬잎마름병 저항성 주동 QTL qSTV11SG탐색)

  • Virus diseases often cause serious damage to rice production in Asia. The lack of information on virus resistance genes has been a major obstacle for the breeding of resistant varieties. In order to identify DNA marker associated with resistance against rice stripe virus (RSV), the quantitative trait locus (QTL) was carried out using advanced backcross population developed from a cross between RSV-resistant tongil type cultivar Shinkwang and susceptible japonica cultivar Ilpum. A RSV resistance QTL $qSTV11^{SG}$ explaining 44.2% of the phenotypic variation was identified on chromosome 11 of Tongil type rice cultivar 'Shingwang'. $qSTV11^{SG}$ was tightly linked to DNA marker RM6897. The RM6897 divided as resistance type allele and susceptible type alleles. Twenty seven resistant varieties showed the resistant-type allele and 23 susceptible varieties were susceptible-type allele to the marker of RM6897. This results and the molecular markers presented here may be useful in rice breeding for improving RSV resistance in japonica rice.

  • A new vegetable peanut cultivar "Charmwon" (Arachis hypogaea ssp. fastigiata.L.) was developed at the Yeongnam Agricultural Research Institute, NICS, in Milyang 2006. It was developed from the cross between the disease resistant cultivar "HYQ(CG)S-10" and the high-yielding cultivar "Sinkwang". "Charmwon" which has been classified as Shinpung plant type is less branch number with early flowering and ellipse-shaped large kernel. Each pod which showed very shallow constriction had two grains with red testa and 100 seed weight was 90g in the regional yield trials (RYT). This variety also showed higher free sugar and protein contents in seed quality, and had more resistant to early and late leaf spot diseases compared with check one. In the regional yield trials "Charmwon" was out-yielded than check variety by 18% with 8.93 ton/ha for fresh pod and by 4% with 3.95 ton/ha for grain.

Organic Rice (Oryza sativa L.) Production in Eco-friendly Complex using Gelatin·Chitin Microorganisms (친환경 광역단지 내 젤라틴·키틴분해미생물을 이용한 유기 벼 생산)

  • This study was carried out to investigate the economic value of organic rice production using gelatin chitin microorganisms in eco-friendly complex, Gongsan, Naju city. The soil condition of experiment paddy field was Jeonbuk series and silt loam with a slightly poor drainage. Except for the high effective silicate, the chemical characteristics of soils used were included in the optimum range of paddy soils in Korea. In growth, plant length, tiller number, ear number, and ear length were observed to be higher in conventional paddy fields than organic paddy fields. However, number of grain per panicle and grain filling ratio (%) were higher in organic paddy fields than conventional paddy fields. Incidences of diseases and insect pests were slightly higher in the organic paddy fields. Water weevil, sheath blight, rice leaf roller and rice blast were more occurred in organic paddy field. On the other hand, false smut was higher occurred in conventional paddy field. There was a significant negative correlation between rice sheath blight and rice leaf roller, and rice yield. In the milled rice quality, the quality of organically cultivated milled rices was lower by the increase of broken rice than that of conventionally cultivated milled rices. The quality and palatability of rice were higher in organic cultivation with decreasing of protein content. Net income of conventionally and organically cultivated rice was 360,000 won/10a and 610,000 won/10a, respectively. Premium net income of the organically cultivated rice was 68%.

rvH1N1 Neuraminidase Inhibitory Activities of Phenolics from Perilla frutescens (L.) and Their Contents in Cultivars and Germplasm

  • The influenza neuraminidase (NA, E.C. 3.2.1.18), an antiviral, has been the target of high pharmaceutical companies due to its essential role in viral replication cycle. Perilla frutescens (P. frutescens) is used in traditional Chinese medicine for various diseases, such as cold due to wind-cold, headache and cough. In this context, four major polyphenolic compounds including rosmarinic acid-3-O-glucoside (1), rosmarinic acid (2), luteolin (3), and apigenin (4) isolated from P. frutescens were evaluated for their inhibitory effect on recombinant virus H1N1 neuraminidase (rvH1N1 NA). Among the test compounds, rosmarinic acid and luteolin inhibited the rvH1N1 NA with an $IC_{50}$ of 46.7 and $8.4{\mu}M$, respectively. The inhibition kinetics analyzed by the Dixon plots indicated that rosmarinic acid and luteolin were noncompetitive inhibitors and that the inhibition constant, $K_I$, was established as 43.9 and $14.3{\mu}M$, respectively. In addition, 578 genetically diverse accessions and 39 cultivars of P. frutescens were analyzed using HPLC to characterize the diversity of polyphenolic composition and concentration. The individual and total compositions exhibited significant difference (P < 0.05), especially rosmarinic acid which was detected as the predominant metabolite in all accessions (58.8%) and cultivars (62.8%). Yeupsil and Sangback cultivars exhibited the highest rosmarinic acid ($3,393.5{\mu}g/g$) and luteolin ($383.3{\mu}g/g$) content respectively. YCPL177-2 with the high concentration ($889.8{\mu}g/g$) of luteolin may be used as a genetic resource for breeding elite cultivars.

Cudrania tricuspidata Fruit Extract Ameliorates Free Fatty Acid-induced Lipid Accumulation in HepG2 Cells (유리지방산으로 지방축적을 유도한 HepG2 cells 대한 꾸지뽕 열매 추출물의 개선 효과)

  • Non-alcoholic fatty liver disease (NAFLD) is a chronic liver disease associated with various metabolic syndromes, such as obesity, dyslipidemia, hypertension, and diabetes. Cudrania tricuspidata is a medicinal plant distributed widely in Asia and has been used in clinical practice to treat various diseases. The aim of this study is to determine the lipid-lowering effects of C. tricuspidata fruit extract (CTE) using a cell model induced by free fatty acids (FFAs). HepG2 cells were exposed to 1mM FFAs (palmitic acid:oleic acid = 2:1) for 24 hr to simulate the conditions of NAFLD in vitro. CTE attenuated the increases of lipid accumulation, intracellular triglyceride, and cholesterol content and inhibited 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMG-CoA reductase) activity in the HepG2 cells in a dose-dependent manner. Also, CTE inhibited the protein expression of lipogenesis-related genes, such as sterol regulatory element-binding protein-1/-2 (SREBP-1/-2), fatty acid synthase (FAS), and stearoyl CoA desaturase-1 (SCD-1) in FFAs-induced lipid accumulation in HepG2 cells. In addition, CTE-induced adenosine monophosphate-activated protein kinase (AMPK) phosphorylation in HepG2 cells. These results suggest that CTE attenuates hepatic lipid accumulation by inhibiting lipogenesis through the modulation of the AMPK signaling pathway on FFAs-induced lipogenesis in HepG2 cells and may potentially prevent NAFLD.

'Saemimyeon', a Tongil-Type Medium-Late Maturing Rice Variety with High Amylose ContentUsed for Rice Noodle Preparation (쌀면전용 고아밀로스 중생 통일형 벼 '새미면')

  • Saemimyeon, a Tongil type, medium-late maturing rice variety, is especially used for preparing rice noodles. Its high amylose content was developed to fit market demands and to be affordable for rice processing industries. One of the high yielding lines, Milyang181 (Hanareum), was used in the final three-way cross of $IR50^*2$/YR18241-B-B-115-1-1 for yield improvement and cultivation stabilization, including disease resistance. YR24235-10-1-3, a high yielding and compact plant type, was selected and named Milyang278 after yield test at NICS (RDA, Miryang) in 2010. It was subjected to regional yield test at six sites in the middle and southern plain areas of South Korea. Saemimyeon heading occurs on August 12 and is a mid-late maturing cultivar, with resistance to leaf blast, rice stripe virus, and bacterial blight (K1-K3a), but it is susceptible to major diseases and insect pest infestation. Saemimyeon showed a high amylose content of 26.7%, with a relatively low KOH digestion value of 3.5, which are key factors in rice noodles and pasta processing. In the local adaptability tests, the yield of Saemimyeon was 7.08 MT/ha-an increase of approximately 106% compared to that of Dasan. Thus, Saemimyeon is suitable for cultivation in the southern and middle plain areas of South Korea.

'Arihyang', a Strawberry Variety with Highly Firm and Large-Sized Fruit for Forcing Culture (촉성재배용 고경도 대과성 딸기 품종 '아리향')

  • A strawberry variety 'Arihyang' was derived as an artificial cross between 'Tochiotome' and 'Seolhyang' in 2014. The seedling and line selections were conducted from 2014 to 2015. Preliminary and advanced yield trials of '14-5-5,' which was the final selected line, were conducted from 2015 to 2017. 'Arihyang' is suitable for forced cultivation and has strong plant vigor, uniformly large-sized fruit, and a high yield compared to those of the check variety, 'Seolhyang' and 'Maehyang.' Especially, vitamin C was at a significant level, which was approximately 15% higher than that of 'Seolhyang.' The average number of flowers per first flower cluster was 10.5, which could reduce the labor of thinning fruit. Its fruit has a conical shape, dark red color, and glossy skin. The fruit was of good quality but has recommendations for harvest at the fully ripened stage. 'Arihyang' has intermediate resistant to phytophthora crown rot, but is susceptible to powdery mildew, gray mold, anthracnose, and fusarium wilt. It is reguired to manage major diseases and pests using optimum cultivation techniques and chemical control.
