시스템비계의 현장적용 확대 연구로 품질 및 안전제고 (조선 및 플랜트산업 현장을 기준으로) (Enhanced Quality and Safety by Expanding Field Application of System Scaffolding (Based on the Shipbuilding and Plant Industry Sites))

  • 연구목적: 건설 현장에서 고소 작업시 일부 사용하고 있는 시스템비계를 선박건조 및 플랜트 산업현장의 다양한 형태의 구조물에 확대 적용이 가능토록 방법을 연구하여 품질향상과 안전성을 높이는 방안을 제시하고자 한다. 연구방법: 8개 기업을 대상으로 실태조사를 실시하였으며, 문헌이나 인터넷 조사등을 분석하여 직접 현장조사와 실무자, 전문가의 의견을 반영한 방안을 수립하였다. 연구결과: 관련 특허 분석, 업체와의 FGI 등을 통한 분석하고 시공업체의 전문성 제고를 위한 개선 방안과 다양한 형태에도 적용이 가능한 자재의 연구 개발을 통한 적용 확대의 가능성을 발견하였다 결론:시스템비계는 안전측면으로 그 중요성이 높으나 전 세계적으로 관련 연구는 부진한 편이다. 이에 software적인 전문성 제고와 다양한 형태에도 적용이 가능한 사용자재의 개발연구로 품질 및 안전에 대한 요구를 충족할 수 있을 것이다.

  • 대형 석탄 화력발전소의 보일러는 안정적인 전력량 수급을 위하여, 계속적인 운전 혹은 기동 정지가 반복하면서 운전 되는 등 매우 열악한 상태에서 운전되고 있기 때문에 신뢰성을 유지하기 위한 주기적 점검과 정비는 필수적이다. 정비를 위한 기존의 비계시스템은 모든 하중이 하부로 집중되어 일부 부재에 문제 발생시 연쇄적으로 비계 시스템이 붕괴되는 문제점이 있다. 이에 따라 본 연구에서는 새로운 노내비계를 개발하여 상부에서 하중을 분산시키고, 일부 부재에 문제가 발생하더라도 연쇄 붕괴의 위험을 최소화하였다. 또한, 보일러 튜브의 구조로 인하여 직접적인 설치가 어려운 경우 노내비계의 설치를 위한 인양시스템을 개발하여 노내비계의 구조적인 안정성을 향상시키고 비계 작업자의 안전성을 확보하여 발전설비의 건전성을 확보할 수 있었다.

Current status of whole-genome sequences of Korean angiosperms

  • Owing to the rapid development of sequencing technologies, more than 1,000 plant genomes have been sequenced and released. Among them, 69 Korean plant taxa (85 genome sequences) contain at least one whole-genome sequence despite the fact that some samples were not collected in Korea. The sequencing-by-synthesis method (next-generation sequencing) and the PacBio (third-generation sequencing) method were the most commonly used in studies appearing in 65 publications. Several scaffolding methods, such as the Hi-C and 10x types, have also been used for pseudo-chromosomal assembly. The most abundant families among the 69 taxa are Rosaceae (10 taxa), Brassicaceae (7 taxa), Fabaceae (7 taxa), and Poaceae (7 taxa). Due to the rapid release of plant genomes, it is necessary to assemble the current understanding of Korean plant species not only to understand their whole genomes as our own plant resources but also to establish new tools for utilizing plant resources efficiently with various analysis pipelines, including AI-based engines.

철골용 클램프 시공방향에 따른 변형 및 성능확인 (Steel Frame Clamp Deformation and Performance Check based on Clamping Orientation)

  • 정부[3]에서는 강관비계를 재래식 비계로 규정하고 일체형 작업발판인 시스템비계를 우선 설치하도록 제도화 하고 강관비계의 사용을 최대한 지양하고 있는 추세이다. 하지만 발전소, 플랜트 현장 등 장비의 진입이 안 되거나 본 구조물의 구조가 복잡한 곳, 그리고 지면에서 비계를 쌓아 올리지 못하는 곳에서는 강관비계를 설치하여 작업할 수 밖에 없다. 고소부위의 H-beam 구조물에 강관비계를 설치하여 작업자들이 안전하게 작업을 할 수 있도록 작업공간을 형성하기 위해서는 철골용 클램프의 성능은 매우 중요한 요소이다. 본 연구에서는 철골용 클램프의 설치 방향별 부재의 변형 및 인장하중에 대해 성능시험 및 구조해석 모델링을 통해 확인한 결과 설치 방향별 성능은 안전인증 기준인 인장하중 10,000N을 만족하는 것으로 확인 되었다. 다만 성능값은 만족하지만 부착부 누름 볼트의 변형발생이 확인되었고, 부착부 본체는 변형이 미미한 것으로 확인되었으므로 철골용 클램프는 하중이 전달되는 방향으로 설치하여 부착부 본체에 하중이 제대로 전달될 수 있도록 하는 것이 보다 안전한 방법이다.

화력발전소 보일러 수퍼히트부 안전발판 개발 연구 (A Development of Platforms for Boiler of Thermal Power Plant)

  • The catastrophic collapse of the in-boiler scaffolding system in the two thermal power plants occurred in March and April 2012. After site investigation and document review, it was found that the specialized scaffolding system was imported for overhaul & maintenance and that the system did not get the safety certification at the import. In this regard, this study developed & proposed an access platform and a support for the vertical tube section of the super heat as well as a variable-length platform for the horizontal tube section, satisfying the domestic certification standards. The access platform was developed to be easy to handle by the worker with a weight of about 0.069 kN, which could reduce the risk of falling accidents and workers' musculoskeletal diseases. For the variable-length platform, it is possible to cope with various changes in length between the horizontal tubes associated with the increase of rigidity in the overlapping and the elimination of the protrusion.

Trends in Hybrid Cultured Meat Manufacturing Technology to Improve Sensory Characteristics

  • The projected growth of global meat production over the next decade is attributed to rising income levels and population expansion. One potentially more pragmatic approach to mitigating the adverse externalities associated with meat production involves implementing alterations to the production process, such as transitioning to cultured meat, hybrid cultured meat, and meat alternatives. Cultured meat (CM) is derived from animal stem cells and undergoes a growth and division process that closely resembles the natural in vivo cellular development. CM is emerging as a widely embraced substitute for traditional protein sources, with the potential to alleviate the future strain on animalderived meat production. To date, the primary emphasis of cultured meat research and production has predominantly been around the ecological advantages and ethical considerations pertaining to animal welfare. However, there exists substantial study potential in exploring consumer preferences with respect to the texture, color, cuts, and sustainable methodologies associated with cultured meat. The potential augmentation of cultured meat's acceptance could be facilitated through the advancement of a wider range of cuts to mimic real muscle fibers. This review examines the prospective commercial trends of hybrid cultured meat. Subsequently, the present state of research pertaining to the advancement of scaffolding, coloration, and muscle fiber development in hybrid cultured meat, encompassing plant-based alternatives designed to emulate authentic meat, has been deliberated. However, this discussion highlights the obstacles that have arisen in current procedures and proposes future research directions for the development of sustainable cultured meat and meat alternatives, such as plant-based meat production.

원통형 탈황설비 정비용 비계 연구개발 (Research & Development on the Scaffolding for Cylindrical Desulfurizers Facility Maintenance)

  • The desulfurizers facility is cylindrical shape. To operate properly it needs nozzles cleaned, get rid of lime adhesion and sludge, repair the wear and corrosion of facility regularly. For this purpose, workers shall access the ceiling or vertical wall at high place. Ordinary scaffoldings such as steel pipe scaffolding or system scaffolding have been using so that workers can access them. With these ordinary scaffoldings, openings around cylindrical wall are inevitable which make workers can expose always to the risk of falling. The purpose of this study is to develop customized scaffolding to minimize the openings to prevent workers form falling during maintenance it. It consists of a hexagonal central tower and six trapezoidal outer towers. And the bracing among the towers have connected each other for self-standing and for maintaining the structure of towers. Span decks, the circumference footstools, steps, etc. are laid on each floor. The safety is reviewed by structural analysis and performance test. With this study, openings each floor of this scaffold are removed. The gap between the cylindrical wall and the edge of the work stage is approximately 100 mm. Therefore, we expect that workers can work safely and efficiently.

Virtual Design and Construction (VDC)-Aided System for Logistics Monitoring: Supply Chains in Liquefied Natural Gas (LNG) Plant Construction

  • Many conventional management methods have emphasized the minimization of required resources along the supply chain. Accordingly, this paper presents a proposed method called the Virtual Design and Construction (VDC)-aided system. It is based on object-oriented resource control, in order to accomplish a feed-forward control monitoring supply chain logistics. The system is supported by two main parts: (1) IT-based Technologies; and (2) VDC Models. They enable the system to convey proactive information from the detection technology to its linked visualization. The paper includes a field study as the system's pre-test: the Scaffolding Works in a LNG Mega Project. The study demonstrates a system of real-time productivity monitoring by use of the RFIDbased Mobile Information Hub. The on-line 'productivity dashboard' provides an opportunity to display the continuing processes for each work-package. This research project offers the observed opportunities created by the developed system. Future work will entail research experiments aimed towards system validation.

Effect of Multiple Copies of Cohesins on Cellulase and Hemicellulase Activities of Clostridium cellulovorans Mini-cellulosomes

  • Cellulosomes in Clostridium cellulovorans are assembled by the interaction between the repeated cohesin domains of a scaffolding protein (CbpA) and the dockerin domain of enzyme components. In this study, we determined the synergistic effects on cellulosic and hemicellulosic substrates by three different recombinant mini-cellulosomes containing either endoglucanase EngB or endoxylanase XynA bound to mini-CbpA with one cohesin domain (mini-CbpAl), two cohesins (mini-CbpA12), or four cohesins (mini-CbpAl234). The assembly of EngB or XynA with mini-CbpA increased the activity against carboxymethyl cellulose, acid-swollen cellulose, Avicel, xylan, and com fiber 1.1-1.8-fold compared with that for the corresponding enzyme alone. A most distinct improvement was shown with com fiber, a natural substrate containing xylan, arabinan, and cellulose. However, there was little difference in activity between the three different mini-cellulosomes when the cellulosomal enzyme concentration was held constant regardless of the copy number of cohesins in the cellulosome. A synergistic effect was observed when the enzyme concentration was increased to be proportional to the number of cohesins in the mini-cellulosome. The highest degree of synergy was observed with mini-CbpAl234 (1.8-fold) and then mini-CbpAl2 (1.3-fold), and the lowest synergy was observed with mini-CbpAl (1.2-fold) when Avicel was used as the substrate. As the copy number of cohesin was increased, there was more synergy. These results indicate that the clustering effect (physical enzyme proximity) of the enzyme within the mini-cellulosome is one of the important factors for efficient degradation of plant cell walls.

Apoptosis of Kinetin Riboside in Colorectal Cancer Cells Occurs by Promoting β-Catenin Degradation

  • The Wnt/β-catenin pathway plays essential roles in regulating various cellular behaviors, including proliferation, survival, and differentiation [1-3]. The intracellular β-catenin level, which is regulated by a proteasomal degradation pathway, is critical to Wnt/β-catenin pathway control [4]. Normally, casein kinase 1 (CK1) and glycogen synthase kinase-3β (GSK-3β), which form a complex with the scaffolding protein Axin and the tumor suppressor protein adenomatous polyposis coli (APC), phosphorylate β-catenin at Ser45, Thr41, Ser37, and Ser33 [5, 6]. Phosphorylated β-catenin is ubiquitinated by the β-transducin repeat-containing protein (β-TrCP), an F-box E3 ubiquitin ligase complex, and ubiquitinated β-catenin is degraded via a proteasome pathway [7, 8]. Colorectal cancer is a significant cause of cancer-related deaths worldwide. Abnormal up-regulation of the Wnt/β-catenin pathway is a major pathological event in intestinal epithelial cells during human colorectal cancer oncogenesis [9]. Genetic mutations in the APC gene are observed in familial adenomatous polyposis coli (FAP) and sporadic colorectal cancers [10]. In addition, mutations in the N-terminal phosphorylation motif of the β-catenin gene were found in patients with colorectal cancer [11]. These mutations cause β-catenin to accumulate in the nucleus, where it forms complexes with transcription factors of the T-cell factor/lymphocyte enhancer factor (TCF/LEF) family to stimulate the expression of β-catenin responsive genes, such as c-Myc and cyclin D1, which leads to colorectal tumorigenesis [12-14]. Therefore, downregulating β-catenin response transcription (CRT) is a potential strategy for preventing and treating colorectal cancer. Plant cytokinins are N6-substituted purine derivatives; they promote cell division in plants and regulate developmental pathways. Natural cytokinins are classified as isoprenoid (isopentenyladenine, zeatin, and dihydrozeatin), aromatic (benzyladenine, topolin, and methoxytopolin), or furfural (kinetin and kinetin riboside), depending on their structure [15, 16]. Kinetin riboside was identified in coconut water and is a naturally produced cytokinin that induces apoptosis and exhibits antiproliferative activity in several human cancer cell lines [17]. However, little attention has been paid to kinetin riboside's mode of action. In this study, we show that kinetin riboside exerts its cytotoxic activity against colon cancer cells by suppressing the Wnt/β-catenin pathway and promoting intracellular β-catenin degradation.
