적심 및 BA 엽면 살포가 긴산꼬리풀의 생육에 미치는 영향 (Growth of Veronica longifolia L. as Affected by Pinching and Foliar Application of BA)

  • 본 연구는 국내 자생 긴산꼬리풀의 관상식물로서 활용을 위하여 적심 후 BA 엽면살포의 효과에 대해 알아보기 위하여 실시하였다. 긴산꼬리풀이 10 cm 정도 자랐을 때 적심 0, 1, 2 주 후 0, 500, 1000, 2000 mg/L의 BA를 엽면살포하여 식물의 생육과 개화 특성을 살펴보았다. 긴산꼬리풀의 분지 형성은 적심 1주 후 2000 mg/L를 처리하였을 시 가장 우수하게 나타났으며, 모든 처리구에서 왜성화 효과가 나타났다. 또한 첫 개화일수가 BA 처리구에서 지연되는 효과가 있었으며, 꽃의 길이와 화서의 길이가 감소하는 경향을 보였다. 본 연구결과를 통하여 국내 긴산꼬리풀의 적심과 BA 엽면살포가 관상식물로써의 활용 능력을 증대시킬 수 있을 것이라 판단된다.

  • 생장조절물질(生長調節物質)의 처리(處理)가 콩나물의 ethylene 발생(發生)에 미치는 효과(效果)에 관(關)하여 연구(硏究)한 결과(結果)를 요약(要約)하면 다음과 같다. 1. 콩나물의 ethylene 발생(發生)에 미치는 auxin의 처리(處理)는 뚜렷한 영향(影響)이 보이지 않았다. 2. 콩나물의 ethylene 발생(發生)에 미치는 cytokinin 및 유사물질(類似物質)의 처리효과(處理效果)는 BA가 ethylene 발생(發生)을 크게 촉진(促進)시킨 반면(反面) kinetin, zeatin, zeatin riboside는 미미(微微)하였다. 3. 콩나물의 ethylene 발생(發生)에 미치는 BA 농도별(濃度別) 처리효과(處理效果)는 농도(濃度)가 높을 수록 ethylene 발생(發生)이 증가(增加)되었다. 4. BA의 처리(處理)는 단기간내(短期間內) ethylene 발생(發生)을 증가(增加)시켜서 신장억제(伸長抑制)와 비대촉진(肥大促進) 효과(效果)를 보이나 4PU-30은 ethylene 발생(發生)을 지속적(持續的)으로 증대(增大)시키는 효과(效果)를 보였다. 5. 생장조절물질(生長調節物質)이 처리(處理)된 콩나물에서의 ethylene 발생정도(發生程度)는 BA 25ppm > IAA 25 + BA 25 ppm ${\geq}$ 4 PU-30 25 ppm > IAA 25 ppm > control 순(順)이었다. 6. 콩나물의 세근발생(細根發生)은 cytokinin 중(中) BA에 의(依)해 억제(抑制)되며, 비대촉진(肥大促進)은 ethylene에 의해 촉진(促進)되는데, BA는 ethylene 발생(發生)을 크게 촉진(促進)시켰다.

  • 생장점 배양에 의한 바이러스 무병주 생산 여부를 판단하고자 바이러스 감염실태를 조사한 결과 지황1호와 국내재래종 모두 여러 종류의 바이러스에 감염되어 있었고 주종바이러스는 potexvirus와 TMV였다. 생장점 배양시 신초형성은 kinetin 첨가 배지에서 가장 양호하였으며 2.4-D 첨가 배지에서는 캘러스로 발달하였는데 캘러스로부터 다수의 신초를 획득할 수 있었다. 기내 재분화 식물체의 투과전자현미경(TEM)과 지표식물을 이용한 바이러스 검정결과 생장점 배양 유래의 식물체들은 83.3%가 바이러스 무병주임을 확인할 수 있었다.

The Effects of Growth Regulators and Medium Strength on the Shoot and Bud Formation from the Shoot Apex of Chinese Yam (Dioscorea opposita Thunb)

  • Plantlet regeneration from the shoot apex was studied in three different genotypes of the chinese yam (Dioscorea opposita Thunb) cv. Jnagma and Danma, Dunggunma. The effects of plant growth regulators and inorganic salts concentration of the culture medium on bud induction and shoot growth were examined. The combinations of 0.2 mg/L BAP + 0.2mg/L kinetin, 0.01mg/L NAA + 0.2 mg/L kinetin and a single treatment of 0.2mg/L BAP were equally effective for bud and shoot formation from the shoot apices in the three cultivars. Auxin (2,4-D, NAA) treatment enhanced calli formation from the cultured apices. Also, the shoot apices of the cv. Dunggunma produced more callus and buds on the culture medium (MS) containing 0.05mg/L NAA and 0.5-1.0mg/L SAP. Lower salt strength of medium inhibited shoot elongation but did not have much effect on the shoot and bud induction from the shoot apices. These results will be useful to obtain disease-free plants of the Chinese yam.

Plant Regeneration Depending on Explant Type in Chrysanthemum coronarium L.

  • Plant regeneration depending on explant type was inves-tigated with cotyledon, hypocotyl, and leaf explants of garland chrysanthemum (Chrysanthemum coronarium L.) cultured on MS basal medium supplemented with various concentrations of SAP and NAA combination. Among the three different types of explants, hypocotyl explants grown on MS medium containing $1.0{\mu}M\;NAA,\;1.0{\mu}M\;BA\;and\;6{\mu}M\;AgNO_3$ produced the highest adventitious shoots (4.67 per explant). Hypocotyl explants not only produced more vigorous shoots, which regenerated aster than the cotyledon and leaf explants. An efficient root formation was observed in MS medium containing $3\%$sucrose. The concentration of NAA did not show significant effects on root formation. Results from this experiment suggested that hypocotyl explants were efficient for the regeneration of garland chrysanthemum.

Effect of plant growth regulators and antioxidants on in vitro plant regeneration and callus induction from leaf explants of purple passion fruit (Passiflora edulis Sims)

  • Purple passion fruit (Passiflora edulis Sims) is one of the introduced tropical plants, an increasing interest has arisen due to its distinctive taste and attractive flavor. It is expected that passion fruit production and planted area will increase gradually in the years ahead because of high profitability and consumer's demands of healthful ingredients. So we tried to investigate the effect of plant growth regulators and antioxidants on in vitro plant regeneration and callus induction from leaf explants of passion fruit for an establishment of optimal mass propagation system. Young leaf explants of purple passion fruit were cultured in Murashige and Skoog (MS) medium containing different growth regulators and antioxidant additives to induce the shoot organogenesis. After 8 weeks, the highest embryogenic callus formation rate was obtained in MS medium supplemented with $1mg{\cdot}L^{-1}$ 6-benzylaminopurine (BAP) and $2mg{\cdot}L^{-1}$ 2,4-dichlorophenoxyacetic acid (2,4-D), furthermore, the shoot development via organogenesis was also observed. Silver nitrate ($AgNO_3$), which was added into the medium to minimize the adverse effects of leached phenolics, was effective for reduction of medium browning and sudden explant death. In the medium supplemented with $1mg{\cdot}L^{-1}$ BAP and $1mg{\cdot}L^{-1}$ gibberellic acid ($GA_3$), shoots were most vigorously regenerated and elongated. Most shoots rooted successfully in half strength medium with $1mg{\cdot}L^{-1}$ indol-3 acetic acid (IAA), and more than 90% of plantlets survived after 4-month acclimatization period.

  • 예취한 페레니얼 라이그라스의 재생에 미치는 식물 생장 조절물질의 영향을 조사한 결과, 0.1. 0.5 mg/L의 cytokinin 처리시 가장 높은 재생력을 나타내었으며, kinetin과 2iP가 BAP보다 재생에 보다 좋은 영향을 미쳤다. 2,4-D의 경우, 0.1 mg/L에서 가장 높은 재생력을 나타내었으며, GA$_3$은 초기재생력은 왕성하였으나 예취 2주 이후의 생장이 느린 것으로 나타났다. Cytokinin과 auxin의 세포분열에 미치는 경합을 조사한 결과, kinetin과 2,4-D를 단독으로 처리했을 때보다 두 생장 조절물질을 함께 처리했을 때 더 좋은 재생력을 나타내었다. 페레니얼 라이그라스의 현탁 배양세포에서도 cytokinin 과 auxin을 단독으로 처리했을 때보다 함께 처리했을 때 세포분열이 왕성한 것으로 나타났다.

Cytokinin signaling promotes root secondary growth and bud formation in Panax ginseng

  • Background: Panax ginseng, one of the valuable perennial medicinal plants, stores numerous pharmacological substrates in its storage roots. Given its perennial growth habit, organ regeneration occurs each year, and cambium stem cell activity is necessary for secondary growth and storage root formation. Cytokinin (CK) is a phytohormone involved in the maintenance of meristematic cells for the development of storage organs; however, its physiological role in storage-root secondary growth remains unknown. Methods: Exogenous CK was repeatedly applied to P. ginseng, and morphological and histological changes were observed. RNA-seq analysis was used to elucidate the transcriptional network of CK that regulates P. ginseng growth and development. The HISTIDINE KINASE 3 (PgHK3) and RESPONSE REGULATOR 2 (PgRR2) genes were cloned in P. ginseng and functionally analyzed in Arabidopsis as a two-component system involved in CK signaling. Results: Phenotypic and histological analyses showed that CK increased cambium activity and dormant axillary bud formation in P. ginseng, thus promoting storage-root secondary growth and bud formation. The evolutionarily conserved two-component signaling pathways in P. ginseng were sufficient to restore CK signaling in the Arabidopsis ahk2/3 double mutant and rescue its growth defects. Finally, RNA-seq analysis of CK-treated P. ginseng roots revealed that plant-type cell wall biogenesis-related genes are tightly connected with mitotic cell division, cytokinesis, and auxin signaling to regulate CK-mediated P. ginseng development. Conclusion: Overall, we identified the CK signaling-related two-component systems and their physiological role in P. ginseng. This scientific information has the potential to significantly improve the field-cultivation and biotechnology-based breeding of ginseng.

  • This study was undertaken to investigate the influence of kinetin and 2, 4-dichlorophenoxyacetic acid on the rate of growth, the contents of RNA, DNA, and protein. And also the effect of plant growth regulator on isoperoxidases in callus derived from root (root-callus) and petiole (petiolecallus) was investigated. The rate of growth in petiole-callus was higher than the rootcallus at 0.1 mg/l kinetin and 1mgfl 2,4-D. At 1mgll kinetic, the rate of growth increased, but at high concentration the rate of growth decreased fast. The contents of RNA, DNA and protein also increased, but it did not coincide with the increase of the growth rate of callus. The isoperoxidases of callus grown at various amounts of 2,4-D and kinetic occurred in an almost fashion, but those of root-callus appeared different from those of petiole-callus.

In vitro propagation of Phaleonopsis hybrid 'Little gem' by culturing apical part and axillary bud of flower stalk

  • The in vitro propagation of the commercially important Phalaeonopsis hybrid 'Little gem' was achieved by culturing the apical part and axillary buds excised from flower stalks. The explants were cultured on 5 different basal media: $3.0{\cdot}L^{-1}$ Hyponex and $4.0{\cdot}L^{-1}$ peptone ($H_3P_4$) and Murashige & Skoog (MS) media were shown to be suitable for shoot regeneration. The MS medium supplemented with $5.0mg{\cdot}L^{-1}$ 6-benzylaminopurine (BA) was found to be more efficient for shoot regeneration. However, the number of shoots induced by axillary buds was higher than that induced by the apical part. Incubation of the apical part under darkness for one week, as well as of the explants in the same medium with activated charcoal (AC) $0.5g{\cdot}L^{-1}$ promoted shoot regeneration and shoot growth; similar growth was not observed with axillary buds.
