Classification Rule for Optimal Blocking for Nonregular Factorial Designs

  • In a general fractional factorial design, the n-levels of a factor are coded by the $n^{th}$ roots of the unity. Pistone and Rogantin (2007) gave a full generalization to mixed-level designs of the theory of the polynomial indicator function using this device. This article discusses the optimal blocking scheme for nonregular designs. According to hierarchical principle, the minimum aberration (MA) has been used as an important criterion for selecting blocked regular fractional factorial designs. MA criterion is mainly based on the defining contrast groups, which only exist for regular designs but not for nonregular designs. Recently, Cheng et al. (2004) adapted the generalized (G)-MA criterion discussed by Tang and Deng (1999) in studying $2^p$ optimal blocking scheme for nonregular factorial designs. The approach is based on the method of replacement by assigning $2^p$ blocks the distinct level combinations in the column with different blocks. However, when blocking level is not a power of two, we have no clue yet in any sense. As an example, suppose we experiment during 3 days for 12-run Plackett-Burman design. How can we arrange the 12-runs into the three blocks? To solve the problem, we apply G-MA criterion to nonregular mixed-level blocked scheme via the mixed-level indicator function and give an answer for the question.

한국적 무기체계의 워게임 모델 교정에 관한 연구 (Calibration of a Korean Weapon Systems Wargame Model)

  • Some of the wargame simulators currently used in the Korean Army were developed by other countries, and do not adequately reflect the Korean Peninsula terrain and weapon systems. This implies that these war game simulators need to be calibrated with respect to the input parameters for properly assessing the effectiveness of the Korean weapon systems. In this paper, AWAM, a wargame simulator, is calibrated in terms of the time-based fighting power(FP). The FP data obtained from the Korea Combat Training Center(KCTC) are used as a reference, and the differences between the AWAM and KCTC FP data are calculated at certain points in time. Then, the Taguchi robust design method is adopted using the probabilities of hitting for the K-2 rifle as controllable input parameters. Two performance characteristics are used. One is the difference between the AWAM and KCTC FP data and the other is the score derived by grouping the difference data. For each case, optimal settings of the probabilities of hitting are determined such that the mean of each characteristic is close to 0 with its dispersion being as small as possible.

Large-Scale Fermentation for the Production of Teicoplanin From a Mutant of Actinoplanes teichomyceticus

  • Mutation and its pilot-scale fermentation were conducted for the production of teicoplanin from Actinoplanes teichomyceticus. The fermentation medium was optimized by replacement and Plackett-Burman experimental design. A maximum production of 1,500 mg/l teicoplanin was obtained by pilot-scale fermentation in an optimized medium containing (g/l): 30 g maltodextrin, 5 g glucose, 5 g yeast extract, 5 g soybean meal, 0.5 g $MgSO_4{\cdot}7H_2O$, 0.1 g NaCl, 0.1 g $CaCl_2{\cdot}2H_2O$, and 50 g Diaion HP-20. The production of teicoplanin was improved 3-fold from the parental strain by mutation, media optimization, and fermentation, and laboratory-scale fermentation was successfully demonstrated in a pilot-scale fermenter for the industrial production of teicoplanin.

Statistical Optimization for Improved Production of Cyclosporin A in Solid-State Fermentation

  • This work evaluates the effect of different amino acids on production of Cyclosporin (CyA) production in solid-state fermentation that was previously optimized for different fermentation parameters by one factor at-a-time for the maximum production of CyA by Tolypocladium inflatum MTCC557. Based on the Plackett-Burman design, glycerol, ammonium sulfate, $FeCl_3$, and inoculum size were selected for further optimization by response surface methodology (RSM). After identifying effective nutrients, RSM was used to develop mathematical model equations, study responses, and establish the optimum concentrations of the key nutrients for higher CyA production. It was observed that supplementation of medium containing (% w/w) glycerol, 1.53; ammonium sulfate, 0.95; $FeCl_3$, 0.18; and inoculum size 6.4 ml/5g yielded a maximum of 7,106 mg/kg as compared with 6,480 mg CyA/kg substrate using one factor at-a-time. In the second step, the effect of amino acids on the production of CyA was studied. Addition of $_L$-valine and $_L$-leucine in combination after 20 h of fermentation resulted in maximum production of 8,166 mg/kg.

반응표면분석법을 이용한 Lactobacillus paracasei SRCM201474의 생산배지 최적화 (Application of Response Surface Methodology in Medium Optimization to Improve Lactic Acid Production by Lactobacillus paracasei SRCM201474)

  • 본 연구는 반응표면분석법을 이용하여 L(+)형 젖산 생산향상을 위한 배지조성을 확립하기 위해 수행되었다. L(+)형 젖산을 선택적으로 고생산하는 것으로 알려진 9종의 Lactobacillus paracasei 균주를 전국에서 수집한 김치 시료로부터 선별하였으며, 젖산 생산량과 glucose로부터의 전환률 분석을 통하여 젖산 생산 배지 최적화를 수행하기 위한 균주로 SRCM201474를 선발하였다. 선택된 11개의 배지 조성 중 젖산 생산에 가장 큰 영향을 미치는 요인을 분석하기 위한 방법으로 Plack-Burman design (PBD)을 설계하였으며, 통계분석을 통해 탄소원으로는 glucose, sucrose, molasses, 질소원으로는 peptone을 최종 선정하였다. 젖산 생산 배지 최적화를 위한 각 변수들의 농도 최적화를 수행하기 위한 방법으로 반응표면분석법 중 적은 실험수로도 최적값을 산출할 수 있는 hybrid design 설계 하였다. 실험 모델에 의한 L. paracasei SRCM201474 균주를 이용한 젖산 생산배지 조성과 최적 농도는 glucose 15.48 g/l, sucrose 16.73 g/l, molasses 39.09 g/l, peptone 34.91 g/l로 나타났으며, 이때의 젖산 생산량은 33.38 g/l로 예측되었다. ANOVA 분석을 통해 가정된 실험 모델의 적합성과 유의성을 확인하였으며, 최종적으로 분석된 최적배지에서의 반복실험을 통한 젖산 생산량을 측정하여 모델에 의해 예측된 젖산생산량과 동일함을 검증하였다. 본 연구를 통해 L(+)형 젖산을 선택적으로 고생산하는 균주를 선발하였으며, 배지 최적화를 수행하여 생분해성 플라스틱 생산을 위한 산업적 젖산 생산에 적용할 수 있는 연구자료로 활용될 수 있을 것으로 판단된다.

Bacillus licheniformis SCD121067 균체 생산성 증가를 위한 통계적 생산배지 및 발효조건 최적화 (Optimization of Medium and Fermentation Conditions for Mass Production of Bacillus licheniformis SCD121067 by Statistical Experimental Design)

  • In this work, mass production of Bacillus licheniformis SCD121067 through medium optimization by statistical experimental method was studied. First, galactose, yeast extract and potassium phosphate dibasic were selected as carbon, nitrogen and phosphate sources for mass production of B. licheniformis SCD121067 by using one factor at a time method. Second, according to the result of Plackett-Burman experimental design, key factors was yeast extract and $K_2HPO$. Finally, the response surface methodology was performed to obtain the optimum concentrations of two selected variables. The optimized medium composition consisted of 20 g/L galactose, 36 g/L yeast extract, 0.41 g/L $K_2HPO4$, 0.25 g/L $Na_2CO_3$, 0.4g/L $MgSO_4$ and 0.01g/L $CaCl_2$. Dry cell weight (15.4 g/L) by optimum production medium were increased 10 times, as compared to that determined with basic production medium (1.5 g/L). Fermentation conditions were examined for the mass production of B. licheniformis. The effect of temperature, agitation speed, pH and aeration rate on the mass production of B. licheniformis were also studied in a batch fermenter which was carried out in a 2.5 L bioreactor with a working volume of 1.5 L containing optimized production medium. As a result, dry cell weight of batch culture was 30.7 g/L at $42^{\circ}C$, 300 rpm, pH 8.0 and 2 vvm.

Application of Factorial Experimental Designs for Optimization of Cyclosporin A Production by Tolypocladium inflatum in Submerged Culture

  • A sequential optimization strategy based on statistical experimental designs was employed to enhance the production of cyclosporin A (CyA) by Tolypocladium inflatum DSMZ 915 in a submerged culture. A 2-level Plackett-Burman design was used to screen the bioprocess parameters significantly influencing CyA production. Among the 11 variables tested, sucrose, ammonium sulfate, and soluble starch were selected, owing to their significant positive effect on CyA production. A response surface methodology (RSM) involving a 3-level Box-Behnken design was adopted to acquire the best process conditions. Thus, a polynomial model was created to correlate the relationship between the three variables and the CyA yield, and the optimal combination of the major media constituents for cyclosporin A production, evaluated using the nonlinear optimization algorithm of EXCEL-Solver, was as follows (g/l): sucrose, 20; starch, 20; and ammonium sulfate, 10. The predicted optimum CyA yield was 113 mg/l, which was 2-fold the amount obtained with the basal medium. Experimental verification of the predicted model resulted in a CyA yield of 110 mg/l, representing 97% of the theoretically calculated yield.

초고온성 고세균 Thermococcus onnurineus의 개미산으로부터 바이오수소 생산을 위한 통계적 배지 최적화 (Statistical Optimization of Medium for Formate-driven Bio-hydrogen Production by the Hyperthermophilic Archaeon, Thermococus onnurineus)

  • Medium compositions for the hyperthermophilic archaeon, Thermococcus onnurineus NA1 was statistically optimized to enhance formate-driven hydrogen ($H_2$) production by using response surface methodology. From the Plackett-Burman design-based experiment, it was confirmed that among the minor components of medium such as KCl, $MgSO_4$, $NH_4Cl$, Cystein-HCl, trace elements, Fe-EDTA and $CaCl_2$, the trace elements were screened as the only positively effective components with respect to $H_2$ production. Subsequently, the optimal concentrations of the trace elements and the major components of a medium such as NaCl, yeast extract and sodium formate were determined from the five-level central composite design (CCD)-based experiment. The resulting quadratic model predicted the maximum $H_2$ production of 46.6 mmol/L in serum bottle and it was validated experimentally using the optimal medium initially supplemented with 26.70 g/L of NaCl, 9.81 g/L of sodium formate, 3.50 g/L of yeast extract and 4.59 mL/L of trace elements. From the duplicate batch cultivations in the fermentor using the optimized medium, the a maximum $H_2$ production rate up to 71.8 mmol/L/h could be obtained, which was a 65% enhanced value compared with that obtained using the control medium, showing the high efficiency of the optimized medium.

Enhanced (R)-2-(4-Hydroxyphenoxy)Propionic Acid Production by Beauveria bassiana: Optimization of Culture Medium and H2O2 Supplement under Static Cultivation

  • (R)-2-(4-hydroxyphenoxy)propionic acid (HPOPA) is a key intermediate for the preparation of aryloxyphenoxypropionic acid herbicides (R-isomer). In order to improve the HPOPA production from the substrate (R)-2-phenoxypropionic acid (POPA) with Beauveria bassiana CCN-A7, static cultivation and H2O2 addition were attempted and found to be conducive to the task at hand. This is the first report on HPOPA production under static cultivation and reactive oxygen species (ROS) induction. On this premise, the cultivation conditions and fermentation medium compositions were optimized. As a result, the optimal carbon source, organic nitrogen source, and inorganic nitrogen source were determined to be glucose, peptone, and ammonium sulfate, respectively. The optimal inoculum size and fermentation temperature were 13.3% and 28℃, respectively. The significant factors including glucose, peptone, and H2O2, identified based on Plackett-Burman design, were further optimized through Central Composite Design (CCD). The optimal concentrations were as follows: glucose 38.81 g/l, peptone 7.28 g/l, and H2O2 1.08 g/l/100 ml. Under the optimized conditions, HPOPA titer was improved from 9.60 g/l to 19.53 g/l, representing an increase of 2.03-fold. The results obtained in this work will provide novel strategies for improving the biosynthesis of hydroxy aromatics.

Optimization of Medium Composition for Lipopeptide Production from Bacillus subtilis N7 using Response Surface Methodology

  • The nutritional requirements for the maximum production of lipopeptides by Bacillus subtilis N7 (B. subtilis N7) were investigated and optimized using response surface methodology (RSM) under shake flask fermentation. A one-factor-at-a-time experimental setup was used to screen carbon and nitrogen sources. A Plackett-Burman design (PBD) was employed to screen the most critical variables for lipopeptides production amongst ten nutritional elements. The central composite experimental design (CCD) was finally adopted to elucidate the composition of the fermentation medium. Statistical analyses (analysis of variance, ANOVA) of the results showed that KCl, $MnSO_4$ and $FeSO_4{\cdot}6H_2O$ were important components and that their interactions were strong. Lipopeptide production was predicted to reach 709.87 mg/L after a 60 h incubation using an optimum fermentation medium composed of glucose 7.5 g/L, peanut oil 1.25 g/L, $MgSO_4$ 0.37 g/L, $KH_2PO_4$ 0.75 g/L, monosodium glutamate 6.75 g/L, yeast extract and $NH_4Cl$ (5:3 w/w) 10 g/L, KCl 0.16 g/L, $FeSO_4{\cdot}6H_2O$ 0.24 mg/L, $MnSO_4$ 0.76 mg/L, and an initial pH of 7.0. Lipopeptide production ($706.57{\pm}3.70$ mg/L) in the optimized medium confirmed the validity of the predicted model.
