The present study was designed to examine whether the fermented extract using the leaves of Pinus densiflora (FPD), has an ameliorative effect on the lipid metabolism in rats with hyperlipidemia. The body weight gain was attenuated by the adminstration of either 1-dose (2 ml/kg/day) or 2-dose (4 ml/kg/day) of FPD for 2 weeks compared with that of control or hyperlipidemia rats, while blood glucose, systolic blood pressure, sGOT, and sGPT were not altered. Serum total cholesterol, LDL-cholesterol, and athelosclerotic index were partially restored by the administration of 2-dose of FPD in rats with hyperlipidemia without changes of serum HDL-cholesterol, total lipid, and triglyceride concentration. The FPD also partially ameliorated renal functional parameters including plasma cleatinine and creatinine clearance which were attenuated in rats with hyperlipidemia. These results suggest that FPD has an ameliorative effect on the lipid metabolism and renal function in rats with hyperlipidemia.
Objective: This study was conducted to investigate the supplementary effect of a phytogenic blend (SPA: a mixture containing fermented Schisandra chinensis pomace, fermented Pinus densiflora needle extract, and Allium tuberosum powder in the ratio of 2:2:1) on egg production, egg quality, blood constituents, and visceral organs in laying hens. Methods: A total of 135 Hy-line brown laying hens (48-wk-old) were randomly allocated to three dietary treatments with 5 replicates of 9 hens. The control group (CON) was fed a basal diet (no exogenous SPA addition) and the experimental groups were fed the basal diet containing SPA at the level of 0.1% and 0.3% for 6 weeks. Results: The feed intake was significantly improved in SPA supplemented groups as compared with the control (p<0.05). However, egg production, daily egg mass, and feed conversion ratio were not different among the dietary treatments (p>0.05). For egg quality traits, only Haugh unit (HU) was significantly improved in SPA (0.3%) (p<0.05) as compared with other groups. However, HU was not affected during 4-wk of storage at 18℃ among the dietary treatments (p>0.05). Furthermore, SPA supplementation did not affect the blood biochemical constituents except for the phosphate content, which was significantly higher in SPA groups than the CON group (p<0.05). There were no significant differences in visceral organ characteristics and immune indicators (immunoglobulin A [IgA], IgG, and IgM) in SPA or CON groups. Conclusion: This study suggested that the supplementation of SPA may have beneficial effects on feed intake and egg quality in laying hens.
Soil condition, total number of bacteria, soil amylase activity and microbial biomass $(CO_2-C)$ were measured at soil of different forest types. And the difference of the allelopathic effect was determined between fresh leaf extract of Quercus acutissima and Pinus rigida to the bacteria isolated from soil of different forest types. 1. Total number of bacteria in Carpinus laxiflora forest soil was 4~7 times larger than that in pinus desiflora forest soil. 2. Soil amylase activity was positively correlated with total number of soil bacteria and soil organic matter content. The amylase activity at F layer was 4~5 times larger than that at H layer, and that at H layer was 2~4 times larger than that at A layer. 3. Seasonal changes of microbial biomass showed a peak in summer, and vertical distribution of microbial biomass decreased with increasing soil depth. The microbial biomass in Pinus densiflora forest soil was larger than that in Quercus serrata forest soil. 4. Fresh leaf extract of Pinus rigida and Quercus acutissima showed an acceleration or inhibition effect on the growth of soil bacteria, and that of !. acutissima inhibited larger number of soil bacterial strains than that of P. rigida. 4.2% and 25% of soil bacterial strains isolated from soil of P. rigida and Q. acutissima forests were inhibited by fresh leaf extract of P. rigida and Q. acutissima, respectively.

This study investigates whether red pine (Pinus densiflora Sieb. et Zucc.) bark extract (PBE) can alleviate diabetes and abnormal apoptosis signaling pathways in the hippocampus of streptozotocin (STZ)-induced diabetic Sprague-Dawley (SD) rats. Two dosages of PBE (15 and 30 mg/kg of body weight/day) were administered orally to STZ-induced diabetic SD rats for 20 days. Blood glucose level and body weight were measured once per week. After 20 days of oral administration of PBE, the rat hippocampus was collected, and the production of Akt, p-Akt, GSK-3β, p-GSK-3β, tau, p-tau, Bax, and Bcl-2 proteins were determined by western blot analysis. A decrease in blood glucose level and recovery of body weight were observed in PBE-treated diabetic rats. In the Akt/GSK-3β/tau signaling pathway, PBE inhibited diabetes-induced Akt inactivation, GSK-3β inactivation, and tau hyperphosphorylation. The protein production ratio of Bax/Bcl-2 was restored to the control group level. These results suggest that PBE, rich in phenolic compounds, can be used as a functional food ingredient to ameliorate neuronal apoptosis in diabetes mellitus.
The growth-inhibiting effects of Pinus densiflpora leaf-derived materials on nine human intestinal bacteria were investigated using the impregnated paper disk method, and their activities were compared with those of 13 commercially available terpenes. The biologically active constituent of the extract of P densiflora leaf was characterized as the monoterpene (1R)-(+)-$\alpha$-pinene by various spectroscopic analyses. Responses varied according to bacterial strain, chemicals, and dose. At 10 mg/disk, limonene and (1R)-(+)-$\alpha$-pinene strongly inhibited the growth of Clostridium perfringens, Staphylococcus aureus, and Escherichia coli, without adverse effects on the growth of five lactic acid-bacteria (Bifidobacterium adolescentis, B. bifidum, B. longum, Lactobacillus acidophilus, and L. casei). Little or no inhibition against seven bacteria was observed with anethole, borneol, camphor, caryophyllene, 1,8-cineole, estragole, linalool, and $\alpha$-terpineol. Structure-activity relationship revealed that (1R)-(+)-$\alpha$-pinene had more growth-inhibiting activity against C. perfringens than (1R)-(+)-$\beta$-, (1S-(-)-$\alpha$-, and (1S-(-)-$\beta$-pinenes. Furthermore, the growth-inhibition against L. casei was much more pronounced in (1R)-(+)-$\beta$- and (In-(-)-$\beta$-pinenes than (1R)-(+)-$\alpha$- and (1S)-(-)-$\alpha$-pinenes. These results indicate that the (+)-$\alpha$ form seems to be required against C. perfringens and $\beta$ form against L. casei for growth-inhibiting activity. Morphologically, most strains of C. perfringens were damaged and disappeared at 5 and 2 mg/disk of (1R)-(+)-$\alpha$-pinene. Morphological study revealed that (1R)-(+)-$\alpha$-pinene had more growth-inhibiting activity against C. perfringens than (1R)-(+)-$\beta$-, (1S)-(-)-$\alpha$-, and (1S)-(-)-$\beta$-pinenes. As naturally occurring growth-inhibiting agents, the Pinus leaf-derived materials described above could be useful preventive agents against diseases caused by harmful intestinal bacteria such as clostridia.
The constituents from the needles of the pine tree, Pinus densiflora, were purified and investigated for antithrombotic activity. The needles were initially extracted three times with 70% ethanol, and the extract was sequentially fractionated with chloroform and n-butanol. The aqueous layer formed after n-butanol fractionation was subjected to purification by medium pressure and high pressure liquid chromatography. The two neolignans, 2, 3-dihydro-7-hydroxyl-3-hydroxymethyl-2-(4'-hydroxyl -3-methoxyphenyl)-5-benzofuranpropanol-3-O-${\alpha}$-rhamnopyranoside (a neolignan glycoside) and 2, 3-dihyro-3-hydroxymethyl-7-methoxy-2-(4'-hydroxyphenyl-3'-methoxy)-5-benxofuran propanol 4'-O-${\alpha}$-rhamnopyranoside (icariside $E_4$) were identified by $^1H$ and $^{13}C$ NMR spectra. The effect of the purified compounds, the neolignan glycoside and icariside $E_4$ on thrombin inhibition were investigated by measuring thrombin clotting time in plasma. As a result, the clotting of the neolignan glycoside was delayed four times compared to that of icariside $E_4$. In addition, an analysis of the inhibition effect by changing the concentration showed that the clotting time was delayed in accordance with an increase in the concentration of the neolignan glycoside. Furthermore, we examined the interaction of thrombin and fibrinogen to clarify the action mechanism. As a result, the delay of clotting time in the response of thrombin and pure fibrinogen may indicate that neolignan glycosides inhibit the thrombin action in a direct manner, leading to the suppression of fibrin generation.

Red pine (Pinus densiflora) is widely used traditional medicine, pharmacological and nutritional values from which the phytochemical compounds are derived. The present study was aimed to examine the antibacterial effects in the absence and presence of a immature red pine cone extract against 13 microorganisms. The components in the aqueous extract from immature red pine cone were identified by GC-MS. About 1.4% of total polyphenolic compound was measured in aqueous extract collected from immature red pine cone. Also, the high concentration of ${\beta}$-phellenandrene, ${\alpha}$-pinene, limonene, bornyl acetate and aldehyde was detected in total ion chromatograms. Of total 13 microorganisms, 4 microorganisms including Pseudomonas aeruginosa, Vibrio cholera, Listeria monocytogenes, Klebsiella pneumonia were effectively killed by aqueous extract of immature red pine cone. The highest anti-bacterial effect was detected in P. aeruginosa, followed by V. cholera, L. monocytogenes and K. pneumonia. In case of P. aeruginosa, the largest diameter of inhibition zone was maintained to 1/2 solution treated cells and slightly decreased at 1/4 and 1/8 solution treated cells. Also, in test used V. cholera and L. monocytogenes, the inhibition zone was strongly formed in only 1 and 1/2 solution treated cells, while K. pneumonia showed the very small diameter of inhibition zone in all concentrations. Therefore, these results suggested that the aqueous extracts of immature red pine cone should be considered as a new and potentially important anti-bacterial substrate to effectively prevent the microbial infection and penetration.
The objective was to examine the antifungal activity of Pinus densiflora extract for the control of turfgrass fungal diseases. Antifungal activities of the various fractions of n-hexane, methylene chloride (Ch), ethyl acetate (EtOAc), and n-butanol from P. densiflora were evaluated against Rhizoctonia solani AG1-1B, R. solani AG2-2IV, Sclerotinia homoeocarpa, R. cerealis, Pythium spp., and Colletotrichum graminicola. The Ch and EtOAc fractions showed antifungal activity against Pythium sp. and C. graminicola in paper disc assay. The effective concentration to produce 50% mycelial inhibition ($EC_{50}$) using five discriminatory concentrations of pinosylvin (1) from the Ch fraction of P. densiflora was evaluated on R. solani AG1-1B, R. solani AG2-2IV, R. cerealis, and S. homoeocarpa. S. homoeocarpa showed the highest sensitivity with the lowest mean $EC_{50}$ value ($8.426{\mu}g/mL$) among the four pathogens. Among the three Rhizoctonia pathogens, R. cerealis had the highest mean $EC_{50}$ value ($99.832{\mu}g/mL$) and R. solani AG2-2IV, with the lowest sensitivity, had the lowest $EC_{50}$ value ($39.696{\mu}g/mL$). These results suggested that pinosylvin (1) from P. densiflora could be a valuable lead compound in the improvement of a novel antifungal agent.
This study was carried out to examine antimicrobial substances from medicinal plants, the ethanol extracts of 38 medicinal plants were tested for the antimicrobial activity against Staphylococcus aureus ATCC 25923 and methicillin-resistant Staphylococcus aureus ATCC 43300. The extracts of Glycyrrhiza uralensis, Sophora flavescens, Dryopteris crassirhizoma, and Pinas densiflora showed significant antimicrobial activities against both S. aureus ATCC 25923 and methicillin-resistant S. aureus ATCC 43300. The extract of Dryopteris crassirhizoma among these medical plants showed the highest antimicrobial activity. These results suggested that the extracts from Dryopteris crassirhizoma, Sophora flavescens, Pinas densiflora, and Glycyrrhiza uralensis could be the potential source of antimicrobial agents against methicillin-resistant S. aureus and S. aureus.
This study investigates the effects of Psidium guajava L. leaf (Pg), Lagerstroemia speciosa L. leaf (Ls) and mixture A (Pg, Ls, Morus indica L. leaf extract, Pinus densiflora needles extract, Acanthopanax senticosus M. root extract) on streptozotocin (STZ)-diabetes rats. For four weeks, STZ-diabetes rats were fed crystallized extracts of Pg, Ls, and mixture A. Compared to the diabetic control group, extracts of Pg, Ls, and mixture A decreased glucose levels in rats by 20%, 14% and 24% respectively. These extracts also decreased the level of total cholesterol, triglyceride and free fatty acid, compared to the diabetic control group, while effectively increasing levels of insulin and high-density lipoprotein-cholesterol. These results showed that mixture A had greater antihyperglycemic, antihyperlipidemic, and insulin-increasing effects than the Pg and Ls extracts. Mixture A also showed better restoration of damaged beta cell function compared to Pg and Ls extracts. Therefore, it was proved that mixture A provides a beneficial synergistic effect when compared with Pg and Ls extracts used individually.
