Epidemiological investigation of porcine pseudorabies virus and its coinfection rate in Shandong Province in China from 2015 to 2018

  • Background: Pseudorabies, also known as Aujeszky's disease, is caused by the pseudorabies virus (PRV) and has been recognized as a critical disease affecting the pig industry and a wide range of animals around the world, resulting in great economic losses each year. Shandong province, one of the most vital food animal-breeding regions in China, has a very dense pig population, within which pseudorabies infections were detected in recent years. The data, however, on PRV epidemiology and coinfection rates of PRV with other major swine diseases is sparse. Objectives: This study aimed to investigate the PRV epidemiology in Shandong and analyze the current control measures. Methods: In this study, a total number of 16,457 serum samples and 1,638 tissue samples, which were collected from 362 intensive pig farms (≥ 300 sows/farm) covered all cities in Shandong, were tested by performing enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR). Results: Overall, 52.7% and 91.5% of the serum samples were positive for PRV-gE and -gB, respectively, based on ELISA results. In addition, 15.7% of the tissue samples were PCR positive for PRV. The coinfection rates of PRV with porcine circovirus type 2 (PCV2), porcine reproductive and respiratory syndrome virus, and classical swine fever virus were measured; coinfection with PCV2 was 35.0%, higher than those of the other two viruses. Macroscopic and microscopic lesions were observed in various tissues during histopathological examination. Conclusions: The results demonstrate the PRV prevalence and its coinfection rates in Shandong province and indicate that pseudorabies is endemic in pig farms in this region. This study provides epidemiological data that can be useful in the prevention and control of pseudorabies in Shandong, China.

Development of SNP Markers for Domestic Pork Traceability (국내산 돼지고기의 원산지 검증을 위한 SNP Marker Set 개발)

  • The purpose of the study was to develop an optimum SNP marker set to be utilized for domestic pork traceability. The study tested 51 SNP markers analyzed for origin of farm to be determined from genotypes of offspring and parents in pigs. With the simulation data through random mating population (PI), half sib mating population ($PI_{half-sib}$) and full sib mating population ($PI_{sibs}$), probability of identical genotypes were analyzed as $5.63{\times}10^{-33}$, $4.35{\times}10^{-15}$ and $1.32{\times}10^{-15}$, respectively. The 51 SNP markers also had 100% accuracy for parental determination. These results suggest that if the pig breeding stock is genotyped with the 51 SNP markers, the genotype information of individual offspring can be checked for farm origins by tracing parental sow and sire. Therefore, these SNP markers will be useful to trace the pork from production to consumption in pigs.

Estimation of Production Unit Loads of Livestock Manure Based on TOC (TOC 기반 가축분뇨 발생 원단위 산정)

  • Assessment of pollutant loads for livestock manure based on total organic carbon (TOC) is being required to apply TOC as an indicator in management of total maximum daily loads. In this study, TOC based unit loads of pig manure known as highly contributing to water pollution assessed. The concentration of pig manure, amount of manure production including cleaning water, and unit loads were investigated targeting 52 farms according to 4 major river basins, rearing form, farm scale, and piggery form. The manure production was highly generated in scraper type of piggery, in small scaled farm rearing sow, and in Han River basin and Nakdong River basin. The averaged manure production was 7.4 L/head/d in total river basins. Averaged concentrations were investigated as TOC 16,037 mg/L, BOD 10,559 mg/L, TN 4,145 mg/L, and TP 503 mg/L. Corresponding unit loads were assessed as TOC 117.1 g/head/d, BOD 77.1 g/head/d, TN 34.7 g/head/d, and TP 3.67 g/head/d.

Effects of dietary supplementation of a lipid-coated zinc oxide product on the fecal consistency, growth, and morphology of the intestinal mucosa of weanling pigs

  • Background: Dietary supplementation of zinc oxide (ZnO) to 2000 to 4000 mg/kg is known to be effective for the prevention and treatment of post-weaning diarrhea in the pig. Such a 'pharmacological' supplementation, however, can potentially result in environmental pollution of the heavy metal, because dietary ZnO is mostly excreted unabsorbed. Two experiments (Exp.) were performed in the present study to determine the effects of a lipid-coated ZnO supplement Shield Zn (SZ) compared with those of ZnO. Methods: In Exp. 1, a total of 240 21-day-old weanling pigs were fed a diet supplemented with 100 mg Zn/kg as ZnO (ZnO-100), ZnO-2500, SZ-100, or SZ-200 in 24 pens for 14 days on a farm with its post-weaning pigs exhibiting a low incidence of diarrhea. Exp. 2 was performed using 192 24-day-old piglets as in Exp. 1 on a different farm, which exhibited a high incidence of diarrhea. Results: In Exp. 1, fecal consistency (diarrhea) score (FCS) was less for the ZnO-2500 and SZ-200 groups than for the SZ-100 group (P < 0.05), with no difference between the SZ-100 and ZnO-100 groups. Both average daily gain (ADG) and gain:feed ratio were less for the SZ-200 group than for the ZnO-2500 group, with no difference between the ZnO-100 group and SZ-100 or SZ-200 group. The villus height (VH), crypt depth (CD), and VH:CD ratio of the intestinal mucosa were not influenced by the treatment. In Exp. 2, FCS was lowest for the ZnO-2500 group, with no difference among the other groups. However, neither the ADG nor gain:feed ratio was influenced by the treatment. Conclusion: Results suggest that physiological SZ supplementation has less beneficial effects than pharmacological ZnO for the alleviation of diarrhea irrespective of its severity and for promoting growth without influencing their integrity of the intestinal mucosal structures with little advantage over physiological ZnO in weanling pigs with a small pen size.

A Field Survey on the Odor Concentration in Piggery by the Change of the Season (돈사 내에서 계절별 악취 발생 농도 조사 연구)

  • Six pig farms were surveyed to measure the odor concentrations and characteristics of ammonia and sulfide corollary compounds in piggery. They were depended on the scale of piggery, weather conditions such as temperature, humidity, wind speed and direction, scales and types of pig breeding, and manure treatment methods. The highest ammonia concentrations in piggery were measured during the winter, since the tight sealed insulation in piggery made less amount of generated ammonia discharged from piggery. The objective of this study was to measure concentrations of odor in the piggery by season and growing, and to measure concentrations of odor at boundary area. So, we investigated the raising managements, manure managements, and methods of reducing odor according to farm scale. We found that concentration of ammonia gas in the swine fattening piggery in winter was the highest. This result is consistent with the lower ventilation rate to maintain Indoor temperature. In this result, there was no connection between farm scale and ventilating system. Concentration of ammonia gas was 1.64 ppm at one boundary area in the middle scale. $H_2S$, $CH_3SH$, $(CH_3)_2S$, and $(CH_3)_2S_2$ were below the standard of protection odor policy.

Survey and model development of the mechanization for swine farming (양돈농가의 기계화 실태분석 및 모델개발)

  • This study was carried out to survey basic information of swine farms on the machine holdings. facility type. management of manure by farm scale and operation, and then to develop the mechanization model. Manual feeding was common for sows and nursing sows. but automation feeding was normally furnished for weaners. growing pigs and castrated male pigs. Water supplies was completely automated for all of the surveyed swine farms. Fully mechanized and automated system would not be feasible and affordable for the small scale farms breeding less than 500 heads. Because the environmental control for the nursing sows and weaner was important, some swine houses were constructed with the windowless type. However, the furnished rates ranged from 22.2% to 44.4% of the surveyed nursing sow and weaner houses at the farm scales. In the future, a computerized ventilation system would be commended for the efficient use of heat energy and to maintain the desirable temperature of swine buildings. Over-investment for large scale farm and over-crowded pigpen of small farm would cause wasting construction expenses and spreading epidermic diseases Hence, the size of swine building should follow the recommended scale. The fermentation drier was recommended for the manure management. Urine could be recycled or discharged after treating by the activated sludge process.

Effect of Enrofloxacin on Post Weaning Diarrhea in Pig (돼지의 이유자돈 설사증에 대한 Enrofloxacin의 효과)

  • For the purpose of effective therapy and prevention of post weaning E. coli diarrhea, Enrofloxacin-(Baytril premix) was administered in the feed and a series of investigations were made the occurence of diarrhea, weight gain, feed conversion rate, changes In the counts of faecal E. coli and susceptibility of the isolated E. coli strains to antimicrobials. The results obtained were as follows 1. The weaned piglets which were treated with Baytril premix gained the average body weight of 3.2kg over the control group at the age of 62 days. The result indicates a marked weight gain effect which, by statiscal analysis, showed a significant difference(P<0.01). Administration of Baytril premix also resulted in the improvement of feed conversion rate and provention of post weaning diarrhea. 2. The counts of faecal E. coli in Baytri premx treated piglets showed a marked decrease from 3.14${\times}$10$\^$8/cells/g to 2${\times}$10$\^$7/cells/g at 5days post treatment and 1${\times}$10$\^$5/cells/g at 10 days post treatment. 3. 10 strains of E. coli isolated fred the experimental pig farm were highly susceptible to Bayoil, gentamicin and trimethoprim plus sulfamethoxazole.

Genetic Association of the Porcine C9 Complement Component with Hemolytic Complement Activity

  • The complement system is a part of the natural immune regulation mechanism against invading pathogens. Complement activation from three different pathways (classical, lectin, and alternative) leads to the formation of C5-convertase, an enzyme for cleavage of C5 into C5a and C5b, followed by C6, C7, C8, and C9 in membrane attack complex. The C9 is the last complement component of the terminal lytic pathway, which plays an important role in lysis of the target cells depending on its self-polymerization to form transmembrane channels. To address the association of C9 with traits related to disease resistance, the complete porcine C9 cDNA was comparatively sequenced to detect single nucleotide polymorphisms (SNPs) in pigs of the breeds Hampshire (HS), Duroc (DU), Berlin miniature pig (BMP), German Landrace (LR), Pietrain (PIE), and Muong Khuong (Vietnamese potbelly pig). Genotyping was performed in 417 $F_2$ animals of a resource population (DUMI: $DU{\times}BMP$) that were vaccinated with Mycoplasma hyopneumoniae, Aujeszky diseases virus and porcine respiratory and reproductive syndrome virus at 6, 14 and 16 weeks of age, respectively. Two SNPs were detected within the third exon. One of them has an amino acid substitution. The European porcine breeds (LR and PIE) show higher allele frequency of these SNPs than Vietnamese porcine breed (MK). Association of the substitution SNP with hemolytic complement activity indicated statistically significant differences between genotypes in the classical pathway but not in the alternative pathway. The interactions between eight time points of measurement of complement activity before and after vaccinations and genotypes were significantly different. The difference in hemolytic complement activity in the both pathways depends on genotype, kind of vaccine, age and the interaction to the other complement components. These results promote the porcine C9 (pC9) as a candidate gene to improve general animal health in the future.

  • Sawdust, peat, and sawdust+peat were used as bulking agent in the compost production process using three different origin of manure; cow, pig, and chicken. The organic content and individual N, P, K content of the final manure compost were higher when peat or peat+sawdust were used to control the moisture. The carbon to nitrogen ratio and moisture content were low when peat or peat+sawdust were employed. In the case of cow and pig manure compost produced with peat or peat+sawdust, beneficial microorganism content was also higher than that of the manure samples produced with sawdust only. These results indicate that peat can be a useful component in the production of high quality manure compost.

Isolation and cultivation of swine encephalomyocarditis virus (돼지 뇌심근염 바이러스의 분리 배양)

  • Encephalomyocarditis(EMC) virus was isolated from the mummified and stillborn pigs at a swine farm in Chonnam Province, experienced with EMC infection over the period Oct.~Dec. of 1989. In addition some cultural, serological properties of the isolates and experimental infections in the piglets were studied. The results obtained were as follows; 1. Two EMC virus strains with HA titers and CPE similar to EMC-ATCC were established in a baby hamster kidney (BHK)-21 cell line by inoculating homogenates of brain and heart of the 19 mummified or stillborn pigs and designated $K_3$ and $K_{11}$. 2. At the second BHK-21 cell line passage of the initial isolates CPE appeared after incubation for 16~18 hours, while at the fourth and fifth passage the highest titer of HA was recorded, titer of HA using rat and guinea pig erythrocytes. 3. One pig inoculated with the isolate $K_3$ showed dyspnea as clinical signs and died at the 10 days after inoculation at necropsy white necrotic foci were observed from the dead animal heart. 4. Although all the rest surviving pigs showed increases in antibody titer and body temperature of $40^{\circ}C$ above for the initial 2~4 days followed by the return to normal, there were no gross lesions when the animals were sacrificed at the 2 weeks after inoculation.
