소태나무(Picrasma quassioides (D.Don) Benn.)의 성분분석과 생리활성 평가 (Studies for Component Analysis and Biological Evaluation in Picrasma quassioides (D.Don) Benn. Extracts)

  • This study investigates the chemical components and biological activities in Picrasma quassioides (D.Don) Benn. extracts for the purpose of developing natural pharmaceutical products. The proximate components in Picrasma quassioides (D.Don) Benn. extracts were 81.56% of carbohydrate, 5.80% of crude fat, 7.75% of crude protein, and 2.07% of crude ash. Calcium was the most predominant mineral and followed phosphorus, potassium, sodium, and magnesium, in a decreasing order. The content of the unsaturated fatty acids, such as oleic acid, linoleic acid, and linolenic acid, was find to be higher than those of saturated fatty acids. Water and methanol extract exhibited antioxidative activity with $IC_{50}$ of $128{\mu}g/ml$ and $144{\mu}g/ml$, respectively, suggesting that the extracts are the major biological component in Picrasma quassioides (D.Don) Benn. extracts. The ${\alpha}-glucosidase$ inhibitory activity showed that methanol and water extract were the high inhibitory activity. Considering these results, water extracts of Picrasma quassioides (D/Don) Benn. showed the possibility that they can be used as a new material for natural pharmaceutical products.

Antioxidant and Anticancer Activity of Fractions from Picrasma quassioides (D. Don) Benn. Methanolic Extract

  • The potential antioxidant and anticancer activities of Hexane, EtOAc (Ethyl acetate), BuOH (n-Buthanol) and water fractions from methanolic (MeOH) extract of Picrasma quassioides (D. Don) Benn. were evaluated in vitro. Tested fractions showed strong antioxidant activity, especially EtOAc fraction had the highest activity ($IC_{50}\;=\;114.01\;{\mu}g/mL$), containing high total phenolics and total flavonoids contents, showed $67.59\;Tan\;{\mu}g/mg$ and $64.95\;Que\;{\mu}g/mg$ respectively. Anticancer activity of these fractions was tested by MTT assay on HT-29 (the human colon carcinoma cells) cell line. BuOH fraction not only showed very high anticancer activity, but also had no cytotoxic effect on 293 (the human normal kidney cells) cell line. Considering these results, we used BuOH fraction of MeOH crude extract from P. quassioides (D.Don) Benn. to do assessment of apoptosis by flow cytometry and the mRNA expression levels of widely established apoptotic-related genes on HT-29 cell line. All the experiments showed that BuOH fraction can induce apoptosis on HT-29 cell line strongly. Taken together, methanolic extract of P. quassioides has potential for antioxidant and anticancer activities products.

Isolation and Biological Activities of an Alkaloid Compound (3-methylcanthin-5, 6-dione) from Picrasma quassiodes (D. Don) Benn.

  • An alkaloid, 3-methylcanthin-5, 6-dione, was isolated from the stem of Picrasma quassioides (D. Don) Benn. and characterized by comprehensive analyses of its 1D and 2D NMR spectra. It was also evaluated for its cytotoxic activity in vitro against three human cancer cell lines (MDA-MB-231, HT-29 and NCI-N87), using MTT assays. We found that 3-methylcanthin-5, 6-dione exhibited significant anti-inflammatory activity via inhibiting NO production induced in LPS-stimulated murine macrophage RAW264.7 cells. The antioxidant activity of 3-methylcanthin-5, 6-dione was measured by DPPH free radical scavenging assays, hydroxyl radical scavenging assays and reducing power assays. Our results showed that 3-methylcanthin-5, 6-dione has significant biological activities.

소태나무 잎 및 편백나무 추출물의 항산화 효과 (Antioxidant Effects of Picrasma quassioides and Chamaecyparis obtusa (S. et Z.) ENDL Extracts)

  • 실험에서는 소태나무, 편백의 유용자원으로의 이용가능성을 알아보기 위해, 소태나무 잎과 편백 목질부를 분쇄한 후 추출물을 제조하여 이들 각각의 총 폴리페놀 및 플라보노이드의 함량을 측정 후 항산화제로 널리 알려진 BHA와의 비교측정으로 항산화 활성을 검색해 보았다. 먼저 소태나무 잎, 편백나무의 메탄올 추출물에 존재하는 총 폴리페놀 및 플라보노이드 함량을 측정한 결과, 소태나무 잎 추출물의 총 폴리페놀 함량은 $367.52{\mu}g/mg$, 편백나무 추출물의 총 폴리페놀 함량은 $138.3{\mu}g/mg$으로 편백나무 보다는 소태나무 잎에서 높은 폴리페놀 함량을 보였다. 총 플라보노이드 함량 역시 소태나무 잎과 편백나무에서 각각 46.41 및 $8.12{\mu}g/mg$으로 소태나무 잎에서 플라보노이드 함량이 높게 나타났다. 각 시료의 DPPH 소거 활성을 농도별로 측정한 결과, $10{\mu}g/ml$의 농도에서 소태나무 잎이 97%, $200{\mu}g/ml$의 농도에서 편백나무가 90%의 소거능을 보였고, BHA ($5{\mu}g/ml$)에서 81% 정도의 항산화능을 보였다. 또한 $ABTS+^{\bullet}$ 소거활성을 Trolox와 비교한 결과, $ABTS+^{\bullet}$ 소거활성법에서 표준물질로 사용되는 Trolox는 $60{\mu}M$에서 94% 정도의 소거활성을 보였고, 소태나무 잎 추출물은 $10{\mu}g/ml$에서 68%, 편백나무는 $100{\mu}g/ml$에서 89% 정도의 소거활성을 보였다. FRAP 활성 측정결과 소태나무 잎에서 $8.52{\mu}mol/{\mu}g$, 편백나무에서 $1.77{\mu}mol/{\mu}g$ 나타났으며 특히 소태나무 잎에서 높은 $Fe^{2+}$ 함량을 나타내어 total polyphenol contents과 유사한 경향을 보였다. 따라서 본 연구 결과들로부터 소태나무 잎은 높은 폴리페놀 함량에 기인한 강력한 항산화 작용을 가지는 것으로 생각된다.

소태나무 잎 추출물의 항염증 효과 (Anti-Inflammatory Effects of Picrasma Quassioides (D.DON) BENN Leaves Extracts)

  • 본 연구에서는 쌍떡잎식물 쥐손이풀목 소태나무과에 속하는 소태나무 잎 methanol 추출물을 이용하여 항염증 및 항산화 활성을 확인해 보았다. 먼저, 소태나무 잎 methanol 추출의 항산화 활성을 알아보기 위해 폴리페놀, 플라보노이드 함량을 측정하였으며 그 결과, 소태나무 잎 추출물에서 폴리페놀과 플라보노이드의 함량은 각각 $367.52{\mu}g/mg$, $46.41{\mu}g/mg$으로 나타나 플라보노이드 보다는 다량의 폴리페놀을 함유하는 것을 확인하였다. 항염증 활성을 확인하기 위해 염증 매개물질인 NO와 염증성 cytokine인 $PGE_2$를 생성을 측정하였다. 소태나무 잎추출물을 LPS로 염증을 유도한 대식세포에 농도별로 처리한 결과 NO의 생성을 $100{\mu}g/ml$의 농도에서 80%까지 억제하는 것으로 나타났으며, 염증성 cytokine인 $PGE_2$의 생성을 $50{\mu}g/ml$, $100{\mu}g/ml$의 농도에서 각각 85%, 90%까지 생성을 억제하는 것으로 나타났다. 또한, 소태나무 잎 추출물이 염증반응과 관련된 iNOS, COX-2, p-NF-${\kappa}B$, p-$I{\kappa}B$의 단백질 발현에 미치는 영향을 확인한 결과, iNOS 단백질은 농도 유의적으로 그 발현이 감소하는 것을 확인하였으며, COX-2 단백질의 경우 12.5, 25, $50{\mu}g/ml$의 농도에서는 큰 변화 나타나지 않았지만 $100{\mu}g/ml$의 농도에서 그 발현이 크게 억제되는 것을 확인하였다. 또한 iNOS와 COX-2의 발현을 조절하는 NF-${\kappa}B$ signaling을 확인해 본 결과, $I{\kappa}B$와 NF-${\kappa}B$의 인산화를 효과적으로 감소시킴으로써 항염증 활성을 나타내는 것을 확인 할 수 있었다. 이상의 결과와 같이, 소태나무 잎 추출물은 항산화 활성과 우수한 항염증 활성을 나타내는 기능성 소재로의 활용이 가능할 것이라고 사료된다.

설악산 상천 계곡의 황철나무 (On the Populus maximowiczii Forest of Sangcheon Ravine, Mt. Seolag)

  • The Populus maximowiczii dominated forest of the Sangcheon ravine, Mt. Seolag, is distributed in the area with the conglemerates substrate, along the ravine stream from 170m to 550m in altitude. Toward the both slopes of the northern and southern peak from the stream side, the zonal distribution of vegetation was recognized; P. maximowiczii forest of Pinus densiflora forest and deciduous broad leaved forest. The pure community of P. maximowiczii with even more 80-98% in relative basal area (aspen basal area/basal area) was found in the ravine area from 360m to 420m in altitude, the optimal ranges in the species and community by two dimensional ordination with thermal and xeric cline axis. The species compete with Pinus densiflora, in the ravine stream side, and with deciduous broad leaved tree species such as Fraxinus rhynchophylla, Prunus sargentii, Lindera obtusiloba, in the mountain slope sides. On the other hand, the ravine vegetation, including the aspen forest, was classified into Pinus densiflora, Pinus densiflora-Carpinus laxiflora, Pinus densiflora-Populus maximowiczii, jessoensis, Acer mono-Fraxinus rhynchophylla, Carpinus laxiflora-Quercus mongolica, Quercus variabilis-Quercus ariena, Quercus ariena, Quercus variabilis-Stephanandra incisa, Picrasma quassioides-Celtis sinensis, Betula davurica-Zanthoxylum schinifolium and Styrax obassia-Lindera obtusiloba association.

Dimensional Stability of Bentwoods by Treatment Conditions

  • This study was carried out to investigate the dimensional stability of bentwoods by three treatments: steaming, urethane varnish coating, and polyethylene glycol (PEG) treatment. Bentwood processing employed a bending-jig with only 4 cm radius of curvature (ROC). The used species were bitter wood (Picrasma quassioides), painted maple (Acer mono), and birch (Betula schmidtiii). The bending properties of these are well-known in bentwood production (Jung et al., 2002). The bentwoods were treated repeated at room temperature [20℃, RH 80% (12 hours) and 40℃ under RH 10% (12 hours)]. To estimate the dimensional stability of bentwoods, we measured the radius of curvature and end-distance. The best results could be attained with PEG treatment. Steaming was the worst treatment. Comparing the properties of the different species, the dimensional stability of bitter wood was excellent. It was concluded that the steaming treatment was unsuitable for dimensional stability of bentwoods.

망개나무림(林)의 군집(群集) 구조(構造)와 개체군(個體群) 구조(構造) (Community and Population Structure of Berchemia berchemiaefolia Forest)

  • 경상북도 군위군 장곡리에서 발견된 희귀식물인 망개나무군락에 대하여 군집 구조와 개체군 구조를 구명하였던 바 다음과 같이 요약되었다. 첫째, 군집 구조를 볼 때, 조사지의 망개나무림은 소태나무군, 털대사초군 및 전형군의 3개 유형으로 분류되었고, 층위별 종구성의 경우 망개나무는 교목층 42.82%, 아교목층 34.43%, 관목층에는 5.28%를 각각 점유하였으며, 기타 굴참나무, 굴피나무 및 졸참나무의 중요치가 높게 나타났다. 군집유형별 종다양도는 소태나무군이 1.6669로사 가장 높았다. 둘째, 개체군 구조를 볼 때, 망개나무 개체군은 직경이 6 cm~10cm 사이에 가장 많은 개체가 분포하였고, 전반적으로 역J자형의 구조를 나타내었으며, 수고급은 최고 14m에 이르는 개체들로 구성되어 있었으며, 교목층과 아교목층의 수령구조는 11년에서부터 60년까지 범위로 전반적으로 정규분포형을 나타내었으며, 망개나무 분지수는 최고 13개이며, 거의 대부분 4개 이하로 나타났다. 한편, 망개나무의 치수는 상층임관이 열려있는 임도에 집중적으로 발생하고 있었으므로 앞으로 망개나무림의 관리대책을 수립할 때 이러한 점은 반드시 고려되어야 할 것으로 사료되었다.

Dimensional Stability of Plastic Processing Wood Material - Compression Wood and Bentwood -

  • This study was carried out to assess the dimensional stability of wood material treated by plastic processing for bentwood and compression wood. The evaluation method was different between two wood materials, but the treatments for them were very similar to each other. One of the main methods is heat treatment with sufficient water vapor. In bentwood, the used species were painted maple (Acer mono), bitter wood (Picrasma quassioides) and birch (Betula schmidtii). Steaming was the worst treatment method for dimensional stabilization of bentwood. The best results could be attained with PEG treatment for dimensional stabilization of bentwood. Dimensional stability of bitter wood was found to be conspicuous. However the steaming treatment at lower temperatures, i.e., about $130^{\circ}C$ was not suitable for dimensional stability of bentwood. In compression wood, the used specimen was Italian poplar wood (Populus euramericana). Two heat compressive pressing conditions, an open-press system and an air-tighten closed-press system, were used. The recovery rate was measured after boiling and/or absorbing in water to estimate the dimensional stability of heat compressed wood. The best dimensional stability of compressed wood in the air-tighten closed-press system was found to be better at $200^{\circ}C$ than $180^{\circ}C$. The best compression rate for dimensional stability was 73 percent.

한국약용식물의 최종당화산물 생성저해활성 검색 (IX) (Screening of Korean Herbal Medicines with Inhibitory Activity on Advanced Glycation End Products Formation (IX))

  • In this study, 64 Korean herbal medicines have been investigated with an in vitro evaluation systems using glycation end products (AGEs) formation inhibitory activity. Of these, 30 herbal medicines ($IC_{50}<50{\mu}g/ml$) were found to have significant AGEs formation inhibitory activity. Of these, four herbal medicines ($IC_{50}<5{\mu}g/ml$) were found to have significant AGEs formation inhibitory activity. Particularly, Cornus controversa (branches and leaves), Acer ginnala (stems and leaves), Platycarya strobilacea (flowers) and Picrasma quassioides (stems), showed more potent inhibitory activity (approximately 17-27 fold) than the positive control aminoguanidine ($IC_{50}=77.04{\mu}g/ml$).
