Stilbene Compounds from Cultivated Korean Rhubarb Rhizomes (한국산 재배대황 근경의 스틸벤)

  • In continued studies on cultivated Korean rhubarb rhizomes, three additional stilbene compounds were isolated from water extract by column chromatographic separation using Sepha dex LH-20, ODS-gel. Compound I, desoxyrhapontigenin(3,5-dihydroxy-4`-methoxystilbene), Compound II, rhapontigenin(3,3`,5-trihydroxy-4`-methoxystilbene), and compound III, piceatannol(3,3`,4`,5-tetrahydroxystilbene) were elucidated based on physico-chemical and spectroscopic evidences(UV, IR, $^1H$-NMR, $^13C$-NMR and Mass).

Protective Effects against Brucella abortus 544 Infection in a Murine Macrophage Cell Line and in a Mouse Model via Treatment with Sirtuin 1 Activators Resveratrol, Piceatannol and Ginsenoside Rg3

  • Brucellosis is a contagious zoonotic disease that infects millions of people annually with hundreds of millions more being exposed. It is caused by Brucella, a highly infectious bacterial species capable of infecting humans with an estimated dose of 10-100 organisms. Sirtuin 1 (SIRT1) has been reported to contribute to prevention of viral diseases as well as a chronic infection caused by Mycobacterium bovis. Here, we investigated the role of SIRT1 in the establishment of Brucella abortus infection in both in vitro and in vivo systems using the reported SIRT1 activators resveratrol (RES), piceatannol (PIC), and ginsenoside Rg3 (Rg3). In RAW264.7 cells, SIRT1 activators did not alter the adherence of Brucella or Salmonella Typhimurium. However, reduced uptake of Brucella was observed in cells treated with PIC and Rg3, and survival of Brucella within the cells was only observed to decrease in cells that were treated with Rg3, while PIC treatment reduced the intracellular survival of Salmonella. SIRT1 treatment in mice via oral route resulted in augmented Brucella resistance for PIC and Rg3, but not RES. PIC treatment favors Th2 immune response despite reduced serum pro-inflammatory cytokine production, while Rg3-treated mice displayed high IL-12 and IFN-γ serum production. Overall, our findings encourage further investigation into the complete mechanisms of action of the different SIRT1 activators used as well as their potential benefit as an effective alternative approach against intracellular and extracellular pathogens.

Effect of Stilbenoids on TNF-${\alpha}$-induced Adipokine Secretion

  • Tumor necrosis factor (TNF)-${\alpha}$ is chronically elevated in adipose tissues of obese rodents and humans. Increased levels of TNF-${\alpha}$ have been implicated in both the induction of atherogenic adipokines, such as plasminogen activator inhibitor (PAI)-1, and the inhibition of the anti-atherogenic adipokine, adiponectin. In this study, we investigated the effects of trans-stilbene, piceatannol, rhaponticin, and piceid on the TNF-${\alpha}$-induced atherogenic changes of adipokines in 3T3-L1 cells. Exposure to TNF-${\alpha}$ for 24 hr increased PAI-1 secretion and decreased adiponectin secretion. Among stilbenoids, piceatannol significantly inhibited the increased secretion of PAI-1 induced by TNF-${\alpha}$. Adiponectin secretion decreased by TNF-${\alpha}$ was recovered after trans-stilbene and rhaponticin treatments. Our results showed that stilbenoids exerted different effects on TNF-${\alpha}$-induced changes in adipokines secretion in 3T3-L1 adipocytes according to their structural characteristics.

Production of Bioactive 3'-Hydroxystilbene Compounds Using the Flavin-Dependent Monooxygenase Sam5

  • The flavin-dependent monooxygenase Sam5 was previously reported to be a bifunctional hydroxylase with coumarate 3-hydroxylase and resveratrol 3'-hydroxylase activities. In this article, we showed the Sam5 enzyme has 3'-hydroxylation activities for methylated resveratrols (pinostilbene and pterostilbene), hydroxylated resveratrol (oxyresveratrol), and glycosylated resveratrol (piceid) as substrates. However, piceid, a glycone-type stilbene used as a substrate for bioconversion experiments with the Sam5 enzyme expressed in Escherichia coli, did not convert to the hydroxylated compound astringin, but it was converted by in vitro enzyme reactions. Finally, we report a novel catalytic activity of Sam5 monooxygenase for the synthesis of piceatannol derivatives, 3'-hydroxylated stilbene compounds. Development of this bioproduction method for the hydroxylation of stilbenes is challenging because of the difficulty in expressing P450-type hydroxylase in E. coli and regiospecific chemical synthesis.

Effect of Stilbene Derivatives from Rheum undulatum on Carrageenan-Induced Acute Edema in Rats (Carrageenan 유발 족부종에 미치는 종대황 스틸벤 유도체의 효과)

  • In order to examine the anti-ohyul (anti-inflammatory) activity of rhubarb, we investigated the effects of stilbene derivatives obtained from rhizomes of Rheum undulatum on the carrageenan-induced hind paw edema in rats. Administration of stilbene aglycon (rhapontigenin) at doses of 50 and 100 mg/kg exhibited the anti- edema effect in dose dependent manner and stilbene glycosides $(rhaponticin,\;piceatannol-3'-O-{\beta}-D-glucopyranoside)$ at a dose of 100 mg/kg also showed inhibitory effect on the carrageenan-induced edema. These inhibitory effects may ascribed, at least in part, to the anti-ohyul activity of rhubarb.

Composition and Quantitative Analysis of Stilbenoids in Mulberry (Morus alba L.) Leaves and Fruits with DAD/UV HPLC (DAD/UV HPLC를 이용한 뽕잎과 오디(Morus alba L.)에 함유된 Stilbenoids 조성 및 함량 분석)

  • This study was aimed to analyze the composition and to quantify the contents of stilbenoids in the leaves and fruits of Morus alba L. using high performance liquid chromatography with phodtodiode array detector and UV detector. Optimal wavelength for the detection of various stilbenoids such as resveratrol, piceatannol, rhapontigenin, astringin, pterostilbene, piceid, rhaponticin and vitisin A was screened by DAD detector and set to 308 nm. Seven kinds of stilbenoids except vitisin A were identified in fruits, while 5 kinds of stilbenoids in leaves. Total stilbenoids contents were $609.15{\pm}7.24$ mg/100 g d.w. in fruits and $188.57{\pm}1.70$ mg/100 g d.w in leaves. Stilbenoids contents in fruits were 3 times higher than those in leaves. Rhaponticin was the most profound stilbene, analyzed to $389.26{\pm}5.22$ mg/100 g d.w. (63.8% of total stilbenoids) in fruits and $99.17{\pm}2.79$ mg/100 g d.w. (52.5% of total stilbenoids) in leaves. Astringin and piceatannol were only detected in fruits and vitisin A was not detected. Contents of piceid and rhaponticin were higher than those of aglycone forms, rhapontigenin and resveratrol.

In Vitro and In Vivo Effects of Piceatannol and Resveratrol on Glucose Control and TLR4-NF-κB Pathway (피세아테놀과 레스베라트롤의 혈당조절 및 TLR4-NF-κB 경로 조절 작용)

  • Piceatannol (PIC) is a natural hydroxylated analog of resveratrol (RSV), which is a polyphenol known to extend lifespan by stimulating sirtuins. The aim of this study was to investigate the effects of PIC and RSV on the toll-like receptor 4 (TLR4)-nuclear factor kappa B ($NF-{\kappa}B$) pathway in mouse hepatocytes and an obese/diabetic KK/HlJ mouse model. AML12 mouse hepatocytes in the absence or presence of palmitic acids (PA) were treated with PIC ($50{\mu}M$) or RSV ($50{\mu}M$). Male KK/HlJ mice at 20 weeks of age were divided into three subgroups as follows: 1) obese and diabetic control (KK), 2) KK_PIC, and 3) KK_RSV. PIC and RSV were administered orally at a dose of 10 mg/kg/d for 4 weeks. Four weeks of PIC and RSV treatment did not affect body weight or food intake in KK mice. Serum fasting blood glucose was significantly reduced in KK_PIC, and 2 h oral glucose tolerance test area under the curve was significantly reduced by PIC and RSV treatment in KK mice. PIC tended to improve homeostasis model assessment of the insulin resistance index (HOMA-IR) and HOMA beta-cells in diabetic KK mice. TLR4 and $NF-{\kappa}B$ were down-regulated by PIC and RSV treatments in hepatocytes in the absence or presence of PA. Insulin receptor, AMP-activated protein kinase, peroxisome proliferator-activated receptor gamma, nucleotide oligomerization domain-like receptor family pyrin domain-containing 3, interleukin-1, and $NF-{\kappa}B$ were altered in PIC-treated livers. Collectively, PIC and RSV inhibited the $TLR4-NF-{\kappa}B$ pathway, and PIC seems to be more effective than RSV in the regulation of analyzed targets, which are involved in insulin signaling and inflammation in vivo.

  • In order to find the antioxidative components, fractionation of Cyperus amuricus (Cyperaceae) methanol extract was performed by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging effect. Three compounds, 3,4-dimethoxy benzoic acid (1), 4-hydroxybenzoic acid (2), and piceatannol (3) were isolated from the active ethylacetate soluble fraction of C. amuricus through repeated silica gel and Sephadex LH-20 column chromatography. Among them, compound 3 showed the significant antioxidative effect on DPPH free radical scavenging test. These compounds are reported for the first time from this plant.

Identification of Soluble Epoxide Hydrolase Inhibitors from the Seeds of Passiflora edulis Cultivated in Vietnam

  • Soluble epoxide hydrolases (sEH) are enzymes present in all living organisms, metabolize epoxy fatty acids to 1,2-diols. sEH in the metabolism of polyunsaturated fatty acids plays a key role in inflammation. In addition, the endogenous lipid mediators in cardiovascular disease are also broken down to diols by the action of sEH that enhanced cardiovascular protection. In this study, sEH inhibitory guided fractionation led to the isolation of five phenolic compounds trans-resveratrol (1), trans-piceatannol (2), sulfuretin (3), (+)-balanophonin (4), and cassigarol E (5) from the ethanol extract of the seeds of Passiflora edulis Sims cultivated in Vietnam. The chemical structures of isolated compounds were determined by the interpretation of NMR spectral data, mass spectra, and comparison with data from the literature. The soluble epoxide hydrolase (sEH) inhibitory activity of isolated compounds was evaluated. Among them, trans-piceatannol (2) showed the most potent inhibitory activity on sEH with an IC50 value of 3.4 μM. This study marks the first time that sulfuretin (3) was isolated from Passiflora edulis as well as (+)-balanophonin (4), and cassigarol E (5) were isolated from Passiflora genus.

  • The sutdies were carried out to evaluate the consituents in the rihzomes of the cultivated Korean Rhubarb (Polygonaceae). From the acetone fraction ofl methanol extract Compound I (1, 8-dihydroxy-3-methyl anthraquinone, chrysophanic acid), Compound II($chrysophanol-8-O-{\beta}-D-glucopyranoside$), Compound III ($emodin-8-O-{\beta}-D-glucopyranoside$) and Compound IV ($aloe-emodin-8-O-{\beta}-D-glucopyranoside$), and from the ether fraction Compound V(1, 8-dihydorxy-3-emthyl-6-methoxy anthraquinone, physcion) and Compound VI (1, 6, 8-thitydroxy-3-emthyl anthrauinone, emodin), and also from the n-buthanol fraction Compound VII ($rhapontigenin-3-O-{\beta}-D-glucopyranoside, rhaponticin$) and Compound VIII ($piceatannol-3'-O-{\beta}-D-glucopyranoside$), were isolated and identified on the basis of their physico-chemical and spectroscopic evidences (UV, IR, H-NMR, C-NMR, EI-MS), respectively.
