Vegetable oils are a rich source of bioactive substances. Phytosterols in those have been known for many years for their properties for reducing blood cholesterol levels, as well as their other beneficial health effects. Phytosterols are triterpenes that are important structural components of plant cell membranes just as cholesterol does in animal cell membranes. The aim of this study was to provide consumers with information about phytosterol contents in vegetable oils in Korea market. The contents of major phytosterols (campesterol, stigmasterol, β-sitosterol) in 50 vegetable oils of 10 kinds (perilla oil, peanut oil, avocado oil, olive oil, pine nut oil, sesame oil, canola oil, coconut oil, grape seed oil, and sunflower oil) were analyzed by gas chromatography with flame ionization detector. The average contents of vegetable oils containing 5 or more samples were in the order of sesame oil (334.43 mg/100 g), perilla oil (262.16 mg/100 g), grape seed oil (183.71 mg/100 g), and olive oil (68.68 mg/100 g). Phytosterol content of sesame oil and perilla oil was high among vegetable oils.
Two lupane-type triterpenes, lupenone (1) and lupeol (2), a phytosterol, ${\beta}-sitosterol$ (3), two ursane-type triterpenes, $3{\beta}-acetoxy$ ursolic acid (4) and ursolic acid (5), a lignan, (-)-lyoniresinol $3a-O-{\beta}-D-xylopyranoside$ (6), and two flavanol glycosides, $catechin-7-O-{\beta}-D-xylopyranoside$ (7) and $catechin-7-O-{\beta}-D-apiofuranoside$ (8) were isolated from the stem bark of Sorbus commixta (Rosaceae). Their chemical structures were identified by physicochemical and spectroscopic methods.
To elucidate the composition of phytosterols in the fruit body of Pholiota spp. 6 species(P. adiposa, P. aurivella, P. highlandensis, P. nameko, P. squarrosa and Pholiota sp.) were analyzed with Gas chromatography(GC). Pholiota spp. were contained campesterol, stigmasterol, and $\beta$-sitosterol as the major phytosterols.
Contents of ginsenosides 7 subordinations of two-year ginseng (fresh ginseng) is 1.27% and three-year ginseng is 2.09%, so the three-year ginseng root increased 64.9% compared to the two-year root. Compared with the comparison group, ginsenosides component content of KEM+SAF-applied group increased 24% in case of the two-year root and 20% in case of the three-year root. In vitamin C content, two-year root showed 59.4% higher and three-year root showed 37.7% higher in KEM+SAF applied group compared with the comparison group. In case of vitamin E, the two-year root indicated 5.6% higher and three-year root indicated 1.5% higher in KEM+SAF applied group compared with the comparison group, but there is no significant difference. In phytosterol three components (campesterol, stigmasterol, sitosterol), two-year root showed 25.3, 3.6, 14.1% higher for each, and three-year root showed 23.6, 6.8, 12.9% higher in KEM+SAF applied group and 14.4% was higher on average. In DPPH, two-year root indicated 34.4% higher and three-year root indicated 42.4% higher in KEM+SAF applied group compared to the comparison group. To sum up the results, KEM+SAF applied group showed (1)22% ginsenosides components content, (2)48.6% vitamin C content, (3)3.6% vitamin E content, (4)14.4% phytosterol content, (5)38.4% DPPH higher averagely compared to the comparison group.
Background: In this study, the fermentation of ginseng seeds was hypothesized to produce useful physiologically-active substances, similar to that observed for fermented ginseng root. Ginseng seed was fermented using Bacillus, Pediococcus, and Lactobacillus strains to extract ginseng seed oil, and the extraction yield, color, and quantity of phenolic compounds, fatty acids, and phytosterol were then analyzed. Methods: The ginseng seed was fermented inoculating 1% of each strain on sterilized ginseng seeds and incubating the seeds at $30^{\circ}C$ for 24 h. Oil was extracted from the fermented ginseng seeds using compression extraction, solvent extraction, and supercritical fluid extraction. Results and Conclusion: The color of the fermented ginseng seed oil did not differ greatly according to the fermentation or extraction method. The highest phenolic compound content recovered with the use of supercritical fluid extraction combined with fermentation using the Bacillus subtilis Korea Food Research Institute (KFRI) 1127 strain. The fatty acid composition did not differ greatly according to fermentation strain and extraction method. The phytosterol content of ginseng seed oil fermented with Bacillus subtilis KFRI 1127 and extracted using the supercritical fluid method was highest at 983.58 mg/100 g. Therefore, our results suggested that the ginseng seed oil fermented with Bacillus subtilis KFRI 1127 and extracted using the supercritical fluid method can yield a higher content of bioactive ingredients, such as phenolics, and phytosterols, without impacting the color or fatty acid composition of the product.
Recent consumption of germinated brown rice is increasing due to its health-beneficial effects. To clarify germination-dependent changes in functional compound contents, 5 different types of (ordinary, glutinous, green-kerneled, red-colored and black-colored) rices were selected and their pre- and post-germination content changes in vitamin E (${\alpha}-$, ${\beta}-$, ${\gamma}-tocopherols$ and ${\alpha}-$, ${\gamma}-$, ${\delta}-tocotrienols$), squalene, and phytosterols (campesterol, stigmasterol, sitosterol) were evaluated. In the case of vitamin E isomer contents, germination generally resulted in decreasing tendency around 10% in most isomers as well as in total tocopherol and tocotrienol contents in all types of rices. In the case of squalene, significant increase could be observed in glutinous rice (by 31%), while other types of rices exhibited no changes or slight reduction by germination. Regarding stigmasterol contents, all tested types of germinated brown rice exhibited significant increment by 19 to 27% compared to nongerminated cases, except for green colored-rice which showed statistically not significant 5% increment. No changes or slight reductions, however, could be observed in campesterol and sitosterol, as well as in total phytosterol contents. As a conclusion, most of tested pharmaceutical compounds exhibited statistically not significant changes except for stigmasterol which were increased by the germination process.
Response surface methodology was used for monitoring and optimizing the extraction conditions of campesterol, stigmasterol, ${\beta}$ -sitosterol, and total sterols from the safflower seed. The conditions of phytosterol extraction were optimized by using central composite design with the temperature(35∼75$^{\circ}C$, X$_1$), the time (1∼11hr, X$_2$), and the preheating temperature(60∼100$^{\circ}C$, X$_3$) as three variables. The extraction conditions for maximum campesterol content were 59.01$^{\circ}C$(X$_1$), 2.88hr(X$_2$), and 75.04$^{\circ}C$(X$_3$). But stigmasterol, ${\beta}$ -sitosterol and total sterols were not significantly different under designed extraction condition in this study. Besides, oil was extracted from safflower seed at various conditions and yields were 23.44% at 35$^{\circ}C$ and 20.05% at 80$^{\circ}C$, respectively. Total tocopherol content increased from 0.172% to 0.207% as the extraction temperature increased from 35$^{\circ}C$ to 80$^{\circ}C$. A structured lipids(SL) was synthesized enzymatically by extracted safflower oil and conjugated linoleic acid(CLA). After 24hr reaction, 31.79 mol% CLA was incorporated into the extracted safflower oil.
To obtain the oil sesame, walnut, whole wheat, and rice bran were extracted for 1, 3, or 6 hr by a shanking water bath(35$^{\circ}C$ and 100 rpm), and by soxhlet extractor(80$^{\circ}C$) for 1, 3 or 6 hr, respectively. The highest yield of extracted oil was obtained from the walnut(63.07% weight) and the whole wheat showed the lowest extraction yield of oil(1.13% weight). Major fatty acids from the extracted oils were linoleic, oleic, and palmitic acid. The maximum contents of total phytosterol in sesame, walnut, whole wheat, and rice bran were 0.44, 1.57, 2.25, and 2.03(% weight), respectively. Besides, total tocopherol contents in sesame, walnut, whole wheat, and rice bran were maxima 3.42, 0.16, 2.92, and 0.07(% weight), respectively. From the extracted oils, structured lipids(SL) were synthesized by the interesterification reaction with conjugated linoleic acid(CLA) in a shanking water bath at 55$^{\circ}C$. When the reactions(1:3 substrate molar ratio, extracted oil:CLA) were conducted for 24 hr, maxima 23.75 mol% of CLA incorporation was obtained from walnut oil and, in other cases, 16.28 - 19.15 mol% of CLA was found in the produced SL triacylglycerol molecules.
This study was done to investigated the phytosterol compositions of safflower (Carthamus tinctorius L.) seed. The phytoestrogen activity was also determined using CAT-ELISA Kit in ethanol extract of safflower seed. The phytosterol of safflower seeds was identified using gas chromatography-mass spectrometry after saponification of the oils. The phytosterol content and composition of safflower seed oils were 4% and identified stigmast-5-en-3-ol (3$\beta$, 24S)-form, ${\gamma}$-sitosterol (clionasterol) with Wiley MS spectrum library. The synergistic effect of human estrogen receptor (hER) has been investigated using a minimal chimeric promoters composed of the TATA region of the adenovirus-2 major late promoter (A22MLP) and two consensus perfectly polindromic Xenopus vitellogenin A2 gene estrogen responsive elements (XVEREl19). Transient transfection experiments in tile human breast adenocarcinoma cell line MCF-7, which is known to express the estrogen receptor endogenously, revealed that phytoestrogen from Carthamus tinctorius L. acts as estrogen. We have observed the transcriptional activities stimulated methanol and ethanol extract of safflower seed in MCF-7, were 0.43 and 0.37 respectively, compared to that by $\beta$-estradiol as 1.0. Our data showed that safflower seeds have estrogenic activity methanol and ethanol extracts and ethanol lower than that of $\beta$-estradiol. This result provides the first evidence that the beneficial effect of safflower seeds may be mediated, at least in part, by the stimulating effect of phytoestrogen ell bone-protecting.
