Phytophthora root and stem rot reduce soybean yields worldwide. The use of R-gene type resistance is currently crucial for protecting soybean production. The present study aimed to identify the genomic location of a gene conferring resistance to Phytophthora sojae isolate 2457 in the recombinant inbred line population developed by a cross of Daepung × Daewon. Singlemarker analysis identified 20 single nucleotide polymorphisms associated with resistance to the P. sojae isolate 2457, which explained ~67% of phenotypic variance. Daewon contributed a resistance allele for the locus. This region is a well-known location for Rps1 and Rps7. The present study is the first, however, to identify an Rps gene locus from a major soybean variety cultivated in South Korea. Linkage analysis also identified a 573 kb region on chromosome 3 with high significance (logarithm of odds = 13.7). This genomic region was not further narrowed down due to lack of recombinants within the interval. Based on the latest soybean genome, ten leucine-rich repeat coding genes and four serine/ threonine protein kinase-coding genes are annotated in this region, which all are well-known types of genes for conferring disease resistance in crops. These genes would be candidates for molecular characterization of the resistance in further studies. The identified R-gene locus would be useful in developing P. sojae resistant varieties in the future. The results of the present study provide foundational knowledge for researchers who are interested in soybean-P. sojae interaction.
Soybean cultivars susceptible to Phytophthora root and stem rot are vulnerable to seed rot and damping-off of seedlings and young plants following an infection by Phytophthora sojae. In this study, the disease responses of Japanese soybean cultivars including currently grown main cultivars during the early growth stages were investigated following infections by multiple P. sojae isolates from Japanese fields. The extent of the resistance to 17 P. sojae isolates after inoculations at 14, 21, and 28 days after seeding varied significantly among 18 Japanese and two US soybean cultivars. Moreover, the disease responses of each cultivar differed significantly depending on the P. sojae isolate and the plant age at inoculation. Additionally, the treatment of 'Nattosyo-ryu' seeds with three fungicidal agrochemicals provided significant protection from P. sojae when plants were inoculated at 14-28 days after seeding. These results indicate that none of the Japanese soybean cultivars are completely resistant to all tested P. sojae isolates during the first month after sowing. However, the severity of the disease was limited when plants were inoculated during the later growth stages. Furthermore, the protective effects of the tested agrochemicals were maintained for at least 28 days after the seed treatment. Japanese soybean cultivars susceptible to Phytophthora root and stem rot that are grown under environmental conditions favorable for P. sojae infections require the implementation of certain practices, such as seed treatments with appropriate agrochemicals, to ensure they are protected from P. sojae during the early part of the soybean growing season.
Since the cultivation of resistance cultivar is essential for organic agriculture, Phytophthora late blight resistance of 16 advanced potato clones obtained from the potato breeding laboratory at the National Institute of Highland Agriculture was evaluated. Resistance of the clone was examined through artificial inoculation in the laboratory and in the field. Dominant isolates of the pathogen in Gangwon province produced abundant sporangia on leaves of most clones showing susceptibility. The number of sporangia ranged from $10^{7{\sim}8}sporangia/ml$ in the susceptible clones at 7 days after inoculation. However, one clone is resistant in organic farming fields. Disease incidence was 2.3% using the resistant clone. The area under disease progress curve(AUDPC) was 75.5. Contrarily, disease incidence of the susceptible variety was 100% and the AUDPC was 1773.5 during the same cultivation period. The resistance clone named as 'Haryeong' is considered suitable for organic potato cultivation in Gangwon province in Korea.
Chitinase 유전자가 삽입된 감자 Belchip 품종의 형질전환체와 대조구에 곰팡이 병원균을 접종하였다. 7개 계통의 형질전환식물체를 12cm정도 키운 후 병원균인 Phytophthora infestans의 zoospore를 접종하여 인공적으로 역병을 유발시켰다. 그 결과 발병율에 따라서 세 그룹으로 분리되었는데 대조구에 비하여 감염 정도가 심한 것 2개 계통. 비슷한 3개 계통, 발병 정도가 약한 2개 계통으로 구분되었다. 대조구에 비하여 저항성이 높았던 2개 계통과 발병이 심했던 1개 계통만을 대상으로 하여 2차실험을 실시한 결과, 1차실험과 비슷한 경향을 나타냈다. 포장에서 생육된 2개의 저항성 계통은 자연적으로 발생한 역병에 대해서도 역시 대조구에 비하여 역병저항성이 더 높았다.
고추 역병은 그 동안 우리나라 고추 생산에 있어서 심각한 수량감소의 피해를 주어 왔으며, 2004년 처음으로 고추 역병 저항성 품종이 보급되기 시작하면서, 국내 건고추 육종에 있어서 역병 저항성의 도입은 필수불가결한 것으로 되었다. 그러므로 저항성 식물체 선발의 정확성 제고 및 육종의 효율 향상을 위하여 역병 저항성과 연관된 분자표지의 개발이 요구된다. 지금까지 고추 역병 저항성 주동 유전자에 연관된 분자표지가 일부 개발되어 있지만, 부분적으로 밖에 사용할 수 없다는 문제점이 있다. 따라서 본 연구에서는 역병 저항성 소재에 상관없이 저항성을 구별해 낼 수 있는 분자표지를 개발하고자 하였다. 이를 위해 '수비초' ${\times}$ 'CM334' 조합의 $F_2$ 분리집단($SF_2$)과 역병 저항성 시판 $F_1$품종('독야청청')을 자가수정한 $F_2$ 분리집단($DCF_2$)을 만들었다. BSA-AFLP 방법으로 총 1,024 프라이머 조합을 사용하여 역병 저항성과 연관된 세 개의 AFLP 분자표지(AFLP1, AFLP2 및 AFLP3)를 선발하였으며, 이를 CAPS 분자표지(M1-CAPS, M2-CAPS 및 M3-CAPS)로 전환하였다. 이 중 M3-CAPS 분자표지를 10개의 역병 저항성 계통, 14개의 이병성 계통, 'CM334'를 화분친으로 사용한 5개의 $F_1$ 조합, 그리고 53개의 시판 $F_1$ 품종에 적용해 본 결과, 역병 저항성 표현형과 M3-CAPS 분자표지의 마커형이 잘 일치하였고, P5-SNAP과 Phyto5.2-SCAR 분자표지보다 높은 실용성을 보이는 결과를 얻었다. 따라서, M3-CAPS 분자표지는 역병 저항성 고추 계통 육성에 많은 도움을 줄 수 있을 것으로 생각한다.
감미종(甘未種)고추에 역병저항성(疫病抵抗性)을 도입하기 위하여, 품질이 우수하면서 국내 적응성이 뛰어난 피만계 품종 Keystone Resistant Giant #3과 역병에 저항성이며 과실이 피만계와 가까운 PI201232를 교배하여 그 후대(後代)의 유전적인 분리 양상을 조사하였다. $F_1$에서는 저항성이 우성(優性)으로 나타났으나 $F_2$ 집단의 경우 고사(枯死)한 개체가 살아남은 개체보다 월등히 많아 저항성이 한 개 혹은 두 개의 우성유전자(優性遺傳子)에 의해 지배된다는 기존의 보고와는 다른 양상으로 나타났다. 뿌리에 대한 조사 결과를 볼 대 지상부가 건전한 개체간에도 뿌리 썩음 정도에 변이(變異)가 나타나기 때문에 저항성 품종을 육성하는 과정에서 지상부와 뿌리의 상태를 함께 관찰한 후 선발(選拔)하는 것이 유리할 것으로 사료되었다. 전반적으로 발병(發病)이 심한 것은 품질친(品質親)이 역병에 매우 약하여 분리(分離) 집단(集團)에서 높은 이병성(罹病性)의 유전적(遺傳的)인 배경을 형성한 것과 동시에 접종(接種) 후의 기상 및 환경조건이 역병 발생에 최적조건을 만든 영향으로 사료된다. $BC_1$ 개체가 모두 고사(枯死)하였으므로 $F_2$에서 저항성 개체를 선발한 후 여교잡(戾交雜)과 자식(自殖)을 반복하는 순환적 여교잡(戾交雜)을 실시하는 것이 저항성의 수준을 유지하면서 품질친의 실용 형질을 회복할 수 있을 것으로 사료된다.
식물은 병원균이나 여러 가지 환경스트레스에 내하여 사기 방어기작을 가지며, 특히 PR 단백질은 병원균의 침입시에 동물의 면역반응과 유사한 생체방어반응을 나타내는 중요한 역할을 하는 것으로 알려져 있다. 본 연구에서는 고추에서 항균 특성을 나타내는 PR5 유전자를 클로닝하고 이들의 특성을 구명하였다. 고추에서 서로 다른 3종의 PR5 유전자, CAPR5-1, CAPR5-2, CAPR5-3를 클로닝하였다. 이들 유전자의 특성을 조사하고 아미노산 수준에서 유사성을 비교하여 본 결과, 서로간에는 90% 이삳의 상동성을 나타내었고 이들의 2차구조를 비교한 결과 중요한 domain은 높은 상동성을 나타내어 PR5 유전자들이 항균 특성을 나타내는데 매우 중요한 motif로 작용할 것으로 사료된다. CAPR5-1, CAPR-2, CAPR5-3 유전자들의 항균성 정도를 조사하기 위하여 이들 유전자를 대장균에서 발현시켜 단백질을 분리하여 고추 역병균인 phytophthora capsici에 처리한 결과, 균사의 성장이 억제되어 CAPR5-1, CAPR5-2, CAPR5-3 단백질들이 항균성을 지니고 있는 것으로 나타났다.
Recently days, soybean production in paddy field is increasing, from 4,422 ha in 2016 to 10,658 ha in 2021 in Korea. It is easy for Phytophthora stem and root rot (PSR) occurring in paddy field condition, when it is poorly drained soils with a high clay content, and temporary flooding and ponding. Therefore PSR resistant soybean cultivar is required. The objective of this study is to identify QTL region and candidate genes relating to PSR resistance of the race in main soybean cultivation area in Korea. 210 soybean materials including cultivars and germplasm were used for inoculation and genome-wide association study (GWAS). Inoculation was conducted using stem-scar method with 2 replications in 2-year for the race 3053 from Kimje and 3617 from Andong. 210 materials were genotyped with Soya SNP 180K chip, and structure analysis and association mapping were conducted with QTLMAX V2. The results of inoculation showed that survival ratio ranged from 0% to 96.7% and mean 9.7% for 3053 and ranged from 0% to 100% and mean 7.6% for 3617. Structure analysis showed linkage disequillibrium (LD) was decayed below r2=0.5 at 335kb of SNP distance. Significant SNPs (LOD>7.0) were identified in Chr 1, 2, 3, 4, 5, 11, 14, 15 for 3053 and Chr 1, 2, 3, 7, 10, 14 for 3617. Especially, LD blocks (AX-90455181;15,056,628bp~AX-90475572;15,298,872bp) in Chr 2 for 3053 and 3067 were duplicated. 29 genes were identified on these genetic regions including Glyma.02gl47000 relating to ribosome recycling factor and defense response to fungus in Soybase.
