Seasonal Variations of Primary Productivity Analyzed by Phyto-PAM Chlorophyll Fluorometry in the Beopsu Marsh, Haman-gun, Gyeongsangnam-do (경상남도 함안군, 법수늪에서 엽록소 형광광도계(Phyto-PAM)에 의한 일차생산의 계절변동)

  • The seasonal variations of primary production were investigated by phyto-PAM chlorophyll fluorometry as well as classical methods (standing crops of phytoplanktons and chlorophyll ${\alpha}$) in the Beopsu Marsh, Haman-gun, Gyeongsangnam-do. The amounts of turbidity, SS, T-N, T-P, BOD, COD, Ca$^{2+}$ and Cl$^-$ were the highest at the station 3, where located in flowout site of wastewater treated by the filtration plants. The water quality was the third level by the standard of BOD and COD. The amount of chlorophyll a (268.8 mg L$^{-1}$) was the highest at the station 2 in April because the cell density (2,677 cells mL$^{-1}$) of Micractinium pusillum increased suddenly from February (180 cells mL$^{-1}$). The patterns of primary production of phytoplankton by phyto-PAM chlorophyll fluorometry were fallen in with those of standing crops and chlorophyll a of phyto-planktons. The primary production was varied according to stations and seasons. The water environments of the Beopsu Marsh as a natural mounument should be under the control of a regular examination in order to preserve the ecosystem.

Synthesis of Pyto-patch as Silver Nanoparticle Product and Antimicrobial Activity (은나노 제품인 Pyto-patch의 제조공정 및 Pyto-patch의 고추 탄저병 방제효과)

  • The commercial product "pyto-patch" prepared as nano sized silver particle less than 5 nm, has effective antifungal activity against Collectotrichum gloeosporioides, Botrytis cinerea, Sclerotinia sclerotiorum in vitro. As a fungal growth inhibiton mechanism, it can reduce spore germination rate and mycelial growth. As s promising fungicide, Phyto-patch can control anthracnose effectively. The spore of C. gloeosporioides dipped in 5 ppm phyto-patch dilute suppressed germination rate to 13.2%, and mycelial growth stopped for 15 days at 10 ppm. The spore postinoculated on 10 ppm phyto-path smeared PDA surface could not germinate for 3 days and prohibit pathogen infection effectively. In field test, the anthracnose development of 4 ppm phyto-patch treated plot was less than 7% after 21 days compaired to 40% of it in untreated plot. In heavy rainfall season, pepper anthracnose effectivly contrrolled by regular 10 ppm phytopatch spraying every 7 days. The diseased pepper fruit decreased to 5.8% compaired to 94.6% in untreated plot. During drying period, the diseased pepper fruit havested in phyto-patch treated plot was 24.2%, but pepper fruit havested in untreated plot destroyed to 100% within 3 days. The nano silver particle coated on multching textile prevented late blight of pepper effectively and disease occurance delayed about month.

Effects of Phyto-Extract Mixture on the Nicotine Decomposition (식물추출혼합물의 니코틴 분해능에 미치는 효과)

  • Cotinine, one of nicotine metabolites, has been blown to reduce 4-(methylnitro samino)-1-(3-pyridyl)-1-butanone(NNK)- induced $O^{6}$MeG DNA adducts significantly in A/J mice when administered together with NNK. In order to examine the effects of phyto-extract mixture on the conversion of cotinine from nicotine, cellular and clinical experiments were carried out. When the phyto-extract mixture was added to culture media, human liver cells (FLCFR5) produced cotinine from nicotine 2~3 times compared to the control. The phyto-extract mixture which was microinjected into Xenopus oocyte along with nicotine showed the almost similar production of cotinine compared with the results of hepatic cell culture. In clinical test employing 17 to 20 healthy men, concentrations of urinary cotinine derived from smoking after taking photo-extract mixture increased up to 2 times compared to the control group. These results indicatethat the phyto-extract mixture can increase the metabolic efficiency of nicotine to cotinine, leading to the reduced formation of $O^{6}$MeG DNA adducts.

Antioxidant Activities of Methanol Extracts from Root Parts of Korean Salad Plants

  • Phenolics level, total flavonoid content, and antioxidant were determined from the methanol extracts of the eight medicinal plants using roots. Total phenolics were found as the highest levels in the methanol extracts of Arctium lappa, and followed by Youngia sonchifolia and Cirsium japonicum. Total amount of the each phenol compounds were detected in C. japonicum extracts ($319.2\;mg\;kg^{-1}$) as the greatest component, and followed by A. lappa ($96.3\;mg\;kg^{-1}$) and Y. sonchifolia ($22.9\;mg\;kg^{-1}$). Total flavonoid content showed the highest amount in methanol extracts from A. lappa (68.1 mg $100\;g^{-1}$) and followed by Y. sonchifolia (11.2 mg $100\;g^{-1}$) Lycoris radiate extracts (87.2%) had the highest nitrite scavenging activity and followed by A. lappa (81.5%) and Y. sonchifolia (77.5%). Methanol extracts of A. lappa at 25 mg $100\;g^{-1}$ exhibited the highest DPPH radical scavenging activity by 90.8%, even though less activity than synthetic antioxidants Vitamin C or butylated hydroxytoluene (BHT). Level of polyphenols was highly correlated with anti oxidative activity ($r^2$ = 0.85). The results suggest that several medicinal plants selected had the potent biological activities, and that their activities were differential depending on plant species.

A New Disinfestation Approach Against Some Greenhouse Pests Using Ethyl Formate Fumigation (훈증제 에틸포메이트를 이용한 몇 가지 시설하우스 해충에 대한 새로운 방제 전략)

  • Ethyl formate (EF) is a rapid kill, environmentally safe, and low mammalian toxicity fumigant, registered to disinfest quarantine insect pests from imported agricultural products. A new concept for controlling insect pests of agricultural crops was tested in a fumigation chamber with EF. Control efficacy of and phyto-toxicity due to EF fumigation were evaluated against four pests (Thrips palmi, Bemisia tabaci, Myzus persicae, and Tetranychus urticae) and on seedlings of four fruit vegetables (FVs; yellow melon, cucumber, tomato, and pepper). Ethyl formate fumigation at a dose of 1.5 g m-3 for 12 h produced >93.3% mortality in T. palmi, B. tabaci, and M. persicae. However, T. urticae was tolerant to fumigation, showing only 20% mortality at 2.0 g m-3. In terms of concentration × time (CT) products, at least 8.9 g·h m-3 CT at 20 ± 1.5℃ was needed to achieve > 90% mortality against the three susceptible insect pests. Fumigation at 1.5 g m-3 for 12 h caused no phyto-toxicity to any of the four FV seedlings. Ethyl formate application, as a new disinfestation method in greenhouses, could be an alternative to reduce the use of conventional insecticides. However, further studies are needed to determine the efficacy of this method at different pest developmental stages and in different greenhouse environments. Additionally, research is needed to elucidate the phyto-toxicity of EF application at different growing stages of a wide variety of crops.

Beneficial Effects of Phyto-Extract Complex (CME) on Degenerative Arthritis (식물추출복합물(CME)의 퇴행성관절염 개선효과)

  • Objectives : Degenerative arthritis arises from several physiological factors. The purpose of this study is to investigate the beneficial effects of Phyto-extract Complex (CME) on degenerative arthritis. Methods : CME is composed of extracts of mulberry (Morus alba L.) fruit, mulberry leaves and black beans (Glycine max (L.) Merr.). To measure the toxicity of CME, we performed the single-dose toxicity study. For the evaluation of its effects on degenerative arthritis, we examined the inhibition of cyclooxygenase-2 (COX-2) activity, using in vitro enzyme activity assay, the reduction of protein expression of COX-2, 5-lipoxygenase (5-LO), and inducible nitric oxide synthase (iNOS) in RAW264.7 cells which were stimulated by lipopolysaccharide (LPS). We also examined the serum level of prostaglandins (PGs) and injury of the knee joint cartilage, using animal model of degenerative arthritis induced by mono-sodium iodoacetate (MIA). Results : CME did not have any toxicity in single-dose toxicity study. The CME inhibited the activity of COX-2 and could reduce the protein expression of COX-2, 5-LO and iNOS in RAW264.7 cells. The CME also reduced the serum level of PGs and prevented from the cartilage injury of knee joint in animal model of degenerative arthritis induced by MIA. Conclusions : Taken altogether, the CME could be useful for the improvement of degenerative arthritis through its various anti-inflammatory activities and prevention from the cartilage injury of knee joint.

Validity Test for Molecular Markers Associated with Resistance to Phytophthora Root Rot in Chili Pepper (Capsicum annuum L.) (고추의 역병 저항성과 연관된 분자표지의 효용성 검정)

  • Phytophthora root rot has been causing a serious yield loss in pepper production. Since 2004, the year in which commercial cultivars resistant to the disease were firstly commercialized, it has been necessary to introduce the resistance into domestic pepper cultivars for dried red pepper. Therefore, developing molecular markers linked to the resistance is required for an accurate selection of resistant plants and increasing breeding efficiency. Until now, several markers associated with the major dominant gene resistant to Phytophthora root rot have been reported but they have some serious limitations for their usage. In this study, we aimed to develop molecular markers linked to the major dominant gene that can be used for almost of all genetic resources resistant to Phytophthora root rot. Two segregating $F_2$ populations derived from a 'Subicho' ${\times}$ 'CM334' combination and a commercial cultivar 'Dokyacheongcheong' were used to develop molecular markers associated with the resistance. After screening 1,024 AFLP primer combinations with bulked segregant analysis, three AFLP (AFLP1, AFLP2, and AFLP3) markers were identified and converted into three CAPS markers (M1-CAPS, M2-CAPS, and M3-CAPS), respectively. Among them, M3-CAPS marker was further studied in ten resistants, fourteen susceptibles, five hybrids and 53 commercial cultivars. As a result, M3-CAPS marker was more fitted to identify Phytophthora resistance than previously reported P5-SNAP and Phyto5.2-SCAR markers. The result indicated that the M3-CAPS marker will be useful for resistance breeding to Phytophthora root rot in chili pepper.

Inhibitory Effects of Phyto Extract Mixture (PEM381) on Type I Allergic Reaction (식물추출 복합물(PEM381)의 제I형 알레르기 반응 억제 효과)

  • The purpose of this study is to investigate the anti type I allergic effects and mechanisms of the phyto extract mixture (PEM381) which contains Camellia sinensis (leaf), Psidium guajava (leaf), and Rosa hybrida (flower). PEM381 was tested for its inhibitory effects on arachidonic acid cascade related enzymes (5-lipoxygenase and cyclooxygenase), the mast cell mediated allergic reaction and passive cutaneous anaphylaxis. $IC_{50}$ value of PEM381 against 5-lipoxygenase was $14.11{\pm}0.51ppm$ while that of positive control (nordihy-droguaiaretic acid) was $0.54{\pm}0.08ppm$. PEM381 also exhibited considerable selective inhibition of cyclooxygenase-2. PEM381 could inhibit both degranulation and histamine release in a dose dependent manner from rat peritoneal mast cells activated by compound 48/80. In addition, oral administration of PEM381 showed an inhibitory effect on passive cutaneous anaphylaxis reaction activated by anti-dinitrophenyl IgE antibody in mice. These results suggest that PEM381 may be useful for the prevention and treatment of type Ⅰ allergy related diseases.

Proximate Compositions and Selected Phytoestrogens of Iranian Black Pomegranate Extract and Its Products (이란산 흑석류 농축액과 그 제품의 성분 및 함유된 Phyto 에스트로겐류에 관한 연구)

  • Phytoestrogens are non-steroidal compounds fecund In a variety of plants, which exert estrogenic effects in animals. In this study, the physico-chemical properties of Iranian black pomegranate extract and its products as preliminarily research for the developing of natural estrogen supplement were evaluated. The chemical components of Iranian black pomegranate extracts and its product (Forever 120) were analyzed. Proximate compositions of pomegranate extracts were as follows; crude lipid 0.4%, crude protein 0.9%, crude ash 1.4% and carbohydrate 42.0%. Major amino acids of pomegranate extracts are glutamic acid (1310.0ppm), aspartic acid (896.2ppm), arginine (877.7ppm) and phenylalanine (57.5ppm). Fatty acid compositions of pomegranate extract 1ibid extracted by chloroform-methanol (2:1) were myristic (13.1%), stearic (69.4), oleic acid (6.8%) and palmitic acid (8.3%). Mineral elements were ferrous (6640.0ppm) and potassium (2550.8ppm). Vitamins were composed of ascorbic acid(20.0mg/100g), Vit. B$_1$(0.12mg/100g) and niacin (0.80mg/100g). 20 phytoestrogens and 20 estrogens of pomegranate extracts were detected Daidzein (0.29ppm), quercetin (9.75ppm) genistein (0.29ppm) and 17 $\beta$ -estradiol(0.15ppm). Above the chemical components of pomegranate extracts were compared with that of pomegranate its product or other isoflavon concentrates.

Effect of Phyto-Extract Fermented Mixture (MP119) on the Sexual Functions and on the Toxicities of Cadmium (식물추출복합발효물(MP119)이 성기능에 미치는 영향 및 카드뮴 독성에 대한 효과)

  • This study was aimed to investigate the effect of phyto-extract fermented mixture (MP119) on the male sexual functions. The MP119 was evaluated for anti-impotency and anti-hypertensive effects via ACE (angiotensin converting enzyme) or PDE (phosphodiesterase) inhibition assay. $IC_{50}$ values of MP119 against ACE and PDE were 241.3${\pm}$35.5 ppm and 372.2${\pm}$33.8 ppm, respectively. To investigate the effect of testosterone expression by MP119, we performed cell media test using mouse Leydig-derived TM3 cells. Production of testosterone in TM3 cell was increased by MP119. Also, NO (nitric oxide) production of HUVEC (human umbilical vein endothelial cell) was increased when MP119 was added to the cultures. Forty male ICR mice were divided into 4 groups. MP119 was orally intubated for 7 days to group 1 and 3, and same volume of vehicle to group 2 and 4 as controls. After that, group 3 and 4 were intraperitoneally injected cadmium chloride at a single dose of 2 mg/kg. On the 8th experimental day, weights of testis, epididymis and seminal vesicle, number of sperm, concentrations of serum testosterone and cGMP were determined. The number of sperm, the concentrations of testosterone and cGMP were significantly increased in two experimental groups (group 1, 3). These results suggest that MP119 enhanced the sexual function of male mice, and could protect the sexual organs from the cadmium chloride as one of the endocrine disrupters.
