Physiological Characteristics and GABA Production of Lactobacillus plantarum K255 Isolated from Kimchi

  • As a major inhibitory neurotransmitter of the central nervous system in animals, ${\gamma}$-aminobutyric acid (GABA) has several physiological functions, such as anti-hypertensive, diuretic, tranquilizer and anti-stress effects in human. In order to determine strains with high GABA producing ability and glutamate decarboxylase (GAD) activity, 273 bacteria were isolated from various types of Kimchi. Strain K255 contained $386.37{\mu}g/mL$ of GABA in MRS broth containing 1% MSG, $600.63{\mu}g/mL$ of GABA in MRS broth containing 2% MSG and $821.24{\mu}g/mL$ of GABA in MRS broth containing 3% MSG. It showed that K255 had the highest GABA production ability compared to other commercial lactic acid bacteria. K255 was identified as Lactobacillus plantarum based on its API carbohydrate fermentation pattern and 16S rDNA sequence. K255 was investigated for its physiological characteristics. The optimum growth temperature of K255 was $37^{\circ}C$and cultures took 13 h to reach the pH 4.4. K255 showed more sensitive to bacitracin in a comparison of fifteen different antibiotics, and showed most resistance to kanamycin and vancomycin. Moreover, it was comparatively tolerant to bile juice and acid and displayed resistance to Escherichia coli, Salmonella Typhimurium, Staphylococcus aureus, with rates of 30.8%, 29.7%, and 23.4% respectively. These results demonstrate that K255 could be an excellent strain for the production of functional products.

  • This paper describes the design of implantable 8-channel telemetering system to get physiological signals. The internal circuits of this system are designed not only to achieve as small size and low power dissipation as possible, but also to enable continuous measurement of physiological signals. Its main functions are to enable continuous measurement of physiological signals and to accomplish on-off power switching of an implantable battery by receiving appropriate command signals from an external circuit. To integrate implantable biotelemetry system, we performed layout of internal system using Lambda based $2{\mu}m$ n-well design rules. This system, used together with appropriate sensors, is expected to be capable of measuring and transmitting such significant parameters as pressure, pH, and temperature.

  • In Pulmonary functions which is important Place which national physical power, especially, in under growth course whom aged from 13 to 16 young boys that whole members of 207 person, and had taken pulmonary functions eyamination record, and took as the following results. 1) Frequency of breathing: Pertaingof the frequency of breathing are 18 times per minute at boys and girls whom age is 13.41 (primary student 6 grade), and at 14.18 and with age 16.41 (middle student, & high school student) are per minute was about 16 times. 2) Vital capacity: Measures of vital capacity are in case of boy student, whom was a primary student was 3 liter, and middle student was 3.2 liter, and at high school students was 3.9 liter, and which different boys and girls student was more low quantify at girls student with comparison student. 3) Breathing holding time. Breathing holding time was nor less in boys student and with girls student, but, that prinary student which middle student and high school student was shown, that is to say, more higher age shown high quantity.

The Effects of Exercise in the Frail Elderly (재가 허약노인을 위한 운동 프로그램 연구)

  • Purpose: This study was conducted to examine the effects of exercise on physiological, physical and psychological functions of the frail elderly. Methods: The research design was a nonequivalent control group pretest-posttest design. Data were collected from September 9 to October 10, 2009. The subjects were 56 frail elders who agreed to participate in this study and each subject was randomly assigned to either the experimental group (n=28) or the control group (n=28). The exercise program was provided to each subject in the experimental group twice in a week for 6 weeks. Data were analyzed with frequency, ${\chi}^2$-test, Kolmogorov-Smirnor test and independent t-test using SPSS/WIN 12.0. Results: There was a significant improvement in psychological functions in the experimental group compared to the control group (t=1.726, p=.045). Conclusion: The exercise program showed the effects to improve the psychological functions of the frail elderly with chronic disease. In recommendation, this exercise program could be utilized as a health promoting program for the frail elderly.

  • Taurine is present in a variety of tissue and exhibits many important physiological functions in many tissues. Although it is known that many tissues mediate taurine transport, its functions of taurine transport in bone have not been identified yet. In the present study, we investigated the expression of taurine transporter (TauT) and taurine uptake using mouse stromal ST2 cells and osteoblast-like MC3T3-El cells, which is bone related cells. Detection of TauT mRNA expression in these cells were performed by reverse transcription polymerase chain reaction (RT-PCR). The activity of TauT was assessed by measuring the uptake of [$^3$H]taurine in the presence or absence of inhibitors. TauT mRNA was detected in these cells. [$^3$H]Taurine uptake was dependent upon the presence of extracellular sodium, chloride and calcium ions, and inhibited by cold-taurine and ${\beta}$-alanine. These results suggest that taurine has biological functions in bone and some effect on the bone cells.

Do the Types of Seat Surface influence the pulmonary Functions during Prolonged Sitting?

  • Purpose: The purpose of this study was to identify the effects of the types of seat surface (static or dynamic seat surface) on the pulmonary functions during prolonged sitting. Methods: Thirty-four participants (20 males and 14 females) were recruited, and distributed randomly into dynamic prolonged sitting (DPS, n=17) and static prolonged sitting (SPS, n=17) groups. The DPS group was seated on a chair with a dynamic air cushion, and the SPS group was seated on a chair without a dynamic air cushion. The pulmonary function was assessed before sitting, and after participants had been seated for one hour. The pulmonary function [forced vital capacity (FVC), forced expiratory volume in 1 second (FEV1), and Peak expiratory flow (PEF)] was measured using a spirometer. Results: Statistical analyses revealed significant differences in the time x group interactions of FVC, FEV1, PEF, and FEV1/FVC. The DPS group were significantly different in FVC, FEV1, PEF, and FEV1/FVC after prolonged sitting for one hour, compared to the SPS group (p<0.05). Conclusion: These findings suggest that dynamic sitting can prevent a decrease in the physiological function, such as pulmonary functions, rather than static sitting during prolonged sitting.

Auxin-responsive SMALL AUXIN UP RNA genes : recent research progress and its application for crop improvement (옥신 반응 SMALL AUXIN UP RNA 유전자의 최근 연구 동향 및 작물 개량을 위한 적용)

  • Auxin is a key plant hormone which regulates overall plant growth development. A number of researches to investigate auxin signaling identified three major classes of early auxin response genes: AUX/IAA, GH3 and SMALL AUXIN UP RNA (SAUR). Among these genes, in planta functions of SAUR gene family are largely ambiguous, while both AUX/IAA and GH3 genes are analyzed to mediate negative feedback on auxin response. SAUR genes encode small plant-specific proteins. SAUR gene products are highly unstable and transiently expressed in the tissue- and developmental-specific manners in response to auxin and various environmental stimuli. In the decades, molecular and genetic approaches to elucidate in planta functions of SAURs have been hampered by several factors such as the unstable molecular features and functional redundancy among them. However, a series of recent studies focusing on several subgroups of SAUR gene family made significant progress in our understanding of its biochemical and physiological functions. These works suggest that many SAUR proteins mainly regulate auxin-related cell expansion and auxin transport. In this review, the recent progress in SAUR research and prospects for crop improvement through its genetic manipulation are discussed.

Identification of Protein Phosphatase 4 Inhibitory Protein That Plays an Indispensable Role in DNA Damage Response

  • Protein phosphatase 4 (PP4) is a crucial protein complex that plays an important role in DNA damage response (DDR), including DNA repair, cell cycle arrest and apoptosis. Despite the significance of PP4, the mechanism by which PP4 is regulated remains to be elucidated. Here, we identified a novel PP4 inhibitor, protein phosphatase 4 inhibitory protein (PP4IP) and elucidated its cellular functions. PP4IP-knockout cells were generated using the CRISPR/Cas9 system, and the phosphorylation status of PP4 substrates (H2AX, KAP1, and RPA2) was analyzed. Then we investigated that how PP4IP affects the cellular functions of PP4 by immunoprecipitation, immunofluorescence, and DNA double-strand break (DSB) repair assays. PP4IP interacts with PP4 complex, which is affected by DNA damage and cell cycle progression and decreases the dephosphorylational activity of PP4. Both overexpression and depletion of PP4IP impairs DSB repairs and sensitizes cells to genotoxic stress, suggesting timely inhibition of PP4 to be indispensable for cells in responding to DNA damage. Our results identify a novel inhibitor of PP4 that inhibits PP4-mediated cellular functions and establish the physiological importance of this regulation. In addition, PP4IP might be developed as potential therapeutic reagents for targeting tumors particularly with high level of PP4C expression.

  • Probiotics are live microorganisms that can be consumed by humans in amounts sufficient to offer health-promoting effects. Owing to their various biological functions, probiotics are widely used in biological engineering, industry and agriculture, food safety, and the life and health fields. Lactobacillus acidophilus (L. acidophilus), an important human intestinal probiotic, was originally isolated from the human gastrointestinal tract and its functions have been widely studied ever since it was named in 1900. L. acidophilus has been found to play important roles in many aspects of human health. Due to its good resistance against acid and bile salts, it has broad application prospects in functional, edible probiotic preparations. In this review, we explore the basic characteristics and biological functions of L. acidophilus based on the research progress made thus far worldwide. Various problems to be solved regarding the applications of probiotic products and their future development are also discussed.

Effect of Adenosine Receptor Antagonists on the Renal Function and Renin Release in Rabbits (가토의 신장기능과 Renin 분비에 미치는 Adenosine수용체 차단제의 영향)

  • Recently it was suggested that the endogenous adenosine might be the mediator for the intercellular communication in the regulation of tubuloglomerular feedback control and renin release. Even though the previous data showed more important regulatory roles in the renal hemodynamics and renin release for the A1 adenosine receptor, it has not yet been settled down about the functional subclassification of renal adenosine receptors. The purpose of the present experiment was to clarify the importance of the renal adenosine receptors for the regulations of the hemodynamic, excretory and secretory functions. Experiments have been done in unanesthetized rabbits. Intrarenal arterial infusion of A1 adenosine antagonist, 8-phenyltheophylline, $3{\sim}30\;nmole/min$, increased urine flow, renal hemodynamics and urinary excretion of sodium. Intrarenal arterial infusion of Al antagonist, 1-3-diethyl-8-phenylxanthine (DPX), $10{\sim}100\;nmole/min$, increased renal hemodynamics and excretory functions. Non-specific adenosine antagonist, theophylline, $30{\sim}300\;nmole/min$, resulted in dose dependent increases in renal hemodynamics and excretory function. All of the three adenosine antagonists for the increases in renal hemodynamics, excretory and secretory functions was 8-phenyltheophylline > DPX > theophylline. These results suggest that the endogenous adenosine is important for the intrinsic regulatory roles for the renal functions through the adenosine receptors, and that the A1 adenosine receptor is more important than the A2 receptor in the regulation of renal hemodynamics, excretory and renin secretory functions.
