Biosynthesis and Degradation of Poly(3-hydroxybutyrate-co-3-hydroxyvalerate) in Alcaligenes sp. SH-69 (Alcaligenes sp. SH-69에서의 Poly(3-hydroxybutyrate-co-3-hydroxyvalerate) 생합성 및 분해)

  • The cyclic metabolism of poly(3-hydroxyhutyrate-co-3-hydroxyvalerate) synthesized from glucose by Alcaligenes sp. SH-69 in the presence or absence of new carbon substrate was investigated. In batch culture, the content and weight average molecular weight of the copolymer already stored in the cell decreased rapidly when there was no other carbon source available. After the depletion of carbon source, the amount of high molecular weight copolymer decreased more rapidly than that of low molecular weight copolymer, and as a result, average molecular weight distribution shifted to the lower value. The addition of a mixture of glucose and levulinic acid when the initial carbon substrate, glucose, was nearly depleted supported the continual increase in cell mass and the accumulation of poly(3HB-co-3HV) with high molar fraction of 3HV. However, solvent fractionation of the polymer with acetone revealed the degradation of pre-existing polyhydroxyalkanoale (PHA) in parallel with the synthesis of PHA from new carbon substrate. Even though PHAs obtained from each substrate alone were the copolymer of 3HB and 3HV, it was found that the polymer accumulated in the cells grown by sequential feeding was mainly physical mixture of two poly(3HB-co-3HV) copolymers containing different molar fractions of 3HV.

Simultaneous Analysis Method for Polar and Non-polar Ginsenosides in Cultivated Wild Ginseng by Reversed-phase HPLC-CAD (HPLC-CAD에 의한 산양삼의 극성 및 비극성 ginsenoside 동시 분석)

  • Cultivated wild ginseng is a widely used dietary supplement and medicinal herb. The aim of this study was to optimize the ginseng using high performance liquid chromatography (HPLC)- charged aerosol detection (CAD) for ginsenoside analysis. CAD measures the physical property of an analyte and responds to almost all non-volatile species, independent of their nature, spectral properties, or particle size. It has become widely employed in pharmaceutical analysis. The cultivated wild ginseng extracts were analyzed for compositions of ginsenosides Rb1, Rd, Rg1, Rf, Re, and Rh1. The optimal analysis condition was set up from an experiment using a gradient. Ten grams of cultivated wild ginseng were extracted with 95% EtOH 100 ml for 24 hr at 80℃. The contents of the 6six major ginsenosides in the cultivated wild ginseng extract were Rb1 (5.48±0.12 mg/g), Rd (5.33±0.14 mg/g), Rg1 (12.80± 0.05 mg/g), Rf (19.08±0.68 mg/g), Re (19.87±0.05 mg/g), and Rh1 (16.47±0.16 mg/g), respectively. HPLC showed that the protopanaxatriol group (Rg1, Rf, Re, Rh1) had more content than the protopanaxadiol group (Rb1, Rd) in cultivated wild ginseng extract. In summary, the ginsenosides were identified with HPLC-CAD analysis, and their presence and quantity imply the importance of quality control, as well as the pharmacological activity of the ginseng root.

Multi-Level Anterior Interbody Fusion with Internal Fixation in Cervical Spine (다분절 경추 유합 및 내고정 수술결과)

  • Objective : The purpose of the present study was to examine neurologic changes, fusion rate and degree of kyphosis from the surgical results of those patients who underwent multi-level anterior interbody fusion and internal fixation. Methods : Among 63 cases of the patients who received multi-level anterior interbody fusion and internal fixation in 5 years between 1995 to 1999 at the neurosurgery department, we performed a retrospective study in 52 cases that could be followed up with dynamic view imaging ; the results were compared and analyzed. The analysis was based on the results of history taking, physical findings and radiologic findings, and Odom criteria were used to classify those cases with neurologic changes. Results : Among those 52 cases in whom the follow-up was possible for at least a year and dynamic view imaging was possible, bone fusion was seen in 93% of the trauma cases and 95% in the non-trauma cases and overall bone fusion was observed in 94% of the cases. Bone fusion was seen in 93% of the autobone cases, 95% of the allobone cases, and 94% of the Mesh cases. Radiologic changes were observed by comparing the lateral view after surgery ; kyphosis was seen in 53% of the autobone cases, in 70% of the allobone cases, and in 35% of Mesh cases ; in 45% and 60% of the non-trauma cases and trauma cases, respectively ; and in 55% of the 2 level fusion cases and 46% of the 3 level fusion cases. Neurologic changes classified according to Odom criteria showed excellent result in 48% of all the cases, good in 23%, fair in 4%, and poor in 25%. Conclusion : Even those cases with multi-level fusion, a high fusion rate could be obtained by performing anterior interbody fusion and internal fixation ; those cases with kyphosis were related more with the presence or absence of posterior compartment injury rather than the fusion level ; and those trauma cases showed not much difference in the fusion rate compared with non-trauma cases but had a high possibility of kyphosis.

Recent Progress in Membrane based Colorimetric Sensor for Metal Ion Detection (색 변화를 활용한 중금속 이온 검출에 특화된 멤브레인 기반 센서의 최근 연구 개발 동향)

  • With a striking increase in the level of contamination and subsequent degradations in the environment, detection and monitoring of contaminants in various sites has become a crucial mission in current society. In this review, we have summarized the current research areas in membrane-based colorimetric sensors for trace detection of various molecules. The researches covered in this summary utilize membranes composed of cellulose fibers as sensing platforms and metal nanoparticles or fluorophores as optical reagents. Displaying decent or excellent sensitivity, most of the developed sensors achieve a significant selectivity in the presence of interfering ions. The physical and chemical properties of cellulose membrane platforms can be customized by changing the synthesis method or type of optical reagent used, allowing a wide range of applications possible. Membrane-based sensors are also portable and have great mechanical properties, which enable on-site detection of contaminants. With such superior qualities, membrane-based sensors examined in the researches were used for versatile purposes including quantification of heavy metals in drinking water, trace detection of toxic antibiotics and heavy metals in environmental water samples. Some of the sensors exhibited additional features like antimicrobial ability and recyclability. Lastly, while most of the sensors aimed for a detection enabled by naked eyes through rapid colour change, many of them investigated further detection methods like fluorescence, UV-vis spectroscopy, and RGB colour intensity.

Weaving the realities with video in multi-media theatre centering on Schaubuhne's Hamlet and Lenea de Sombra's Amarillo (멀티미디어 공연에서 비디오를 활용한 리얼리티 구축하기 - 샤우뷔네의 <햄릿>과 리니아 드 솜브라의 <아마릴로>를 중심으로 -)

  • When video composes mise-en-scene during the performance, it reflects the aspect of contemporary image culture, where the individual as creator joins in the image culture through the device of cell phone and computer remediating the former video technology. It also closely related with the contemporary theatre culture in which 1960's and 1970's video art was weaved into the contemporary performance theatre. With these cultural background, theatre practitioners regarded media-friendly mise-en-scene as an alternative facing the cultural landscape the linear representational narrative did not correspond to the present culture. Nonetheless, it can not be ignored that video in the performance theatre is remediating its historical function: to criticize the social reality. to enrich the aesthetic or emotional reality. I focused video in the performance theatre could feature the object with the image by realizing the realtime relay, emphasizing the situation within the frame, and strengthening the reality by alluding the object as a gesutre. So I explored its two historical manuel. First, video recorded the spot, communicated the information, and arose the audience's recognition of the object to its critical function. Second, video in performance theatre could redistribute perceptual way according to the editing method like as close up, slow motion, multiple perspective, montage and collage, and transformation of the image to the aesthetic function. Reminding the historical function of video in contemporary performance theatre, I analyzed two shows, Schaubuhne's Hamlet and Lenea de Sombra's Amarillo which were introduced to Korean audiences during the 2010 Seoul Theatre Olympics. It is known to us that Ostermeir found real social reality as a text and made the play the context. In this, he used video as a vehicle to penetrate the social reality through the hero's perspective. It is also noteworthy that Ostermeir understood Hamlet's dilemma as these days' young generation's propensity. They delayed action while being involved in image culture. Besides his use of video in the piece revitalized the aesthetic function of video by hypermedial perceptual method. Amarillo combined documentary theatre method with installation, physical theatre, and video relay on the spot, and activated aesthetic function with the intermediality, its interacting co-relationship between the media. In this performance theatre, video has recorded and pursued the absent presence of the real people who died or lost in the desert. At the same time it fantasized the emotional aspect of the people at the moment of their death, which would be opaque or non prominent otherwise. As a conclusion, I found the video in contemporary performance theatre visualized the rupture between the media and perform their intermediality. It attempted to disturb the transparent immediacy to invoke the spectator's perception to the theatrical situation, to open its emotional and spiritual aspect, and to remind the realities as with Schaubuhne's Hamlet and Lenea de Sombra's Amarillo.

A Study on How Living Alone or with a Partner Affects the Elderly's Life Characteristics (노인들의 배우자 유무에 따른 삶의 특성 차이에 관한 연구)

  • This study aims to conduct a comprehensive analysis of how the presence or absence of a partner affects the life characteristics of persons aged 65 or older. For this, the subjects were divided into the married group and the single group to investigate sociodemographic characteristics, health conditions, self-rated health level, disease morbidity, limitations in life due to health conditions and health-related quality of life. An assessment was carried out by analyzing local communities' health survey data of 3 years from 2014 to 2016. The results revealed that the single group had more women, a lower household income, poorer health conditions, and a higher chronic disease morbidity rate than the married group, and thus their self-rated heath level was lower as well. These characteristics were found to contribute to a lower quality of life in the single group than the married group. To this day, we have viewed the elderly as one of the groups based on age and only tried to identify their average characteristics, thus overlooking their various inherent problems. The results of this study suggest that physical, emotional and social problems seen among the elderly need a comprehensive measure. In addition, this study found that public health approaches and social welfare systems should be improved to develop personalized support programs for the elderly.

Characteristics of Equilibrium, Kinetic and Thermodynamic for Adsorption of Acid Blue 40 by Activated Carbon (활성탄에 의한 Acid Blue 40 흡착에 있어서 평형, 동력학 및 열역학적 특성)

  • The kinetics and thermodynamics of the adsorption of acid blue 40 from an aqueous solution by activated carbon were examined as a function of the activated carbon dose, pH, temperature, contact time, and initial concentration. The adsorption efficiency in a bathtub was increased at pH 3 and pH 11 due to the presence of sufonate ions ($SO_3{^-}$) and amine ions ($NH_2{^+}$). The equilibrium adsorption data were fitted to the Langmuir, Freundlich and Temkin isotherms. The results indicated that the Langmuir model provides the best correlation of the experimental data. The separation factor of the Langmuir and Freundlich model showed that the adsorption treatment of acid blue 40 by activated carbon could be an effective adsorption process. The adsorption energy determined by the Temkin equation showed that the adsorption step is a physical adsorption process. Kinetics analysis of the adsorption process of acid blue 40 on activated carbon showed that a pseudo second order kinetic model is more consistent than a pseudo second order kinetic model. The estimated activation energy was 42.308 kJ/mol. The enthalpy change (80.088 J/mol) indicated an endothermic process. The free energy change (-0.0553 ~ -5.5855 kJ/mol) showed that the spontaneity of the process increased with increasing adsorption temperature.

Secondary Fermented Extract of Chaga-Cheonggukjang Attenuates the Effects of Obesity and Suppresses Inflammatory Response in the Liver and Spleen of High-Fat Diet-Induced Obese Mice

  • Cheonggukjang and chaga mushrooms have numerous health benefits, and have been used in alternative medicine. Therefore, a powder mixture of 98: Cheonggukjang and 2: Chaga extracts was fermented with Lactobacillus acidophilus KCTC3925 (FCC) and its anti-obesity effects in high-fat diet (HFD)-induced obese mice were determined. Five-week-old male ICR mice were fed a normal diet or HFD in the presence or absence of 3% and 5% FCC by weight (n = 10 per group). After 12 weeks, the mice were sacrificed, and the serum and tissue samples were collected for analysis. Body weight and epididymal fat pad weight were significantly lowered in the 3% and 5% FCC groups compared with those in the HFD control group (p < 0.01). FCC supplementation suppressed serum triglyceride and increased serum HDL-C levels (p < 0.01). Serum GOT, GPT, and leptin levels, hepatic COX-2 mRNA expression, and splenic COX-2 and IL-4 mRNA expression were significantly higher in the HFD groups than in the control group (p > 0.05); however, except for splenic IL-4 levels, the increases were significantly attenuated by FCC supplementation. Expression of ICAM-1, an aortic inflammatory marker, was significantly increased in the HFD group; this effect was suppressed in the 3% FCC group (p < 0.01) but not in the 5% FCC group. FCC suppressed the body weight and epididymal fat pad weight gain, as well as inflammatory responses in the liver and spleen of HFD-fed mice. Thus, FCC supplementation will be beneficial for the treatment of obesity-related effects.

A Study on the Characteristics of Clinker and Cement as Chlorine Content (염소 함량에 따른 클링커 및 시멘트의 물성에 관한 연구)

  • In this study, a clinker was prepared using raw materials with CaCl2. The characteristics of the chlorine-added clinker and cement were analyzed. The clinker modulus were set to Lime Saturation Factor (LSF) 92, Silica modulus (SM) 2.5, and Iron Modulus (IM) 1.5. The physical properties of cement using the chlorine-containing clinker were characterized. As the chlorine content increased, the free-CaO content in the clinker decreased, and that in the 2000 ppm clinker was reduced by approximately 40% compared to that in the 0 ppm clinker. There was an increase in the amount of chlormayenite, with a content of up to 3.4% present in the 2000 ppm clinker. The amounts of alite and belite also slightly increased. The compressive strength of mortar at 3 days and 7 days increased as the chlorine content increased. This trend was presumed to arise from the effect of hydration, which was promoted by the presence of chlorine. The compressive strength of 1000 ppm mortar increased by approximately 20% compared to that of 0 ppm mortar.

A Characteristic Analysis of Glass Beads in Geumgwan Gaya, Korea (I) (금관가야 유리구슬의 특성 분석 (I))

  • This study examined the physical attributes and heat treatment characteristics of glass beads excavated from the Gimhae area, which is the location of Geumgwan Gaya. This enabled classification of surface characteristics of the beads based on the investigation of the color, size, and shape. The glass beads were classified into eight color systems, with purplish-blue beads as the representative color. Bead size was categorized into three types depending on the outer diameter and how it increased over time. Bead shapes were categorized as round, tubular, or doughnut-shaped based on the inner diameter and length, with round being the typical shape. According to the degree of heat treatment, there are three types of cross-section for glass beads that are manufactured by the drawing technique, most of which are the HT-III type. In addition, it is estimated that the heat treatment technology has more considerable effects than other methods. Through non-destructive analysis, the chemical composition was obtained and categorized as flux, stabilizer, and colorant. Analysis confirmed the presence of 63 and 9 pieces in the potash and soda glass groups, respectively. Overall findings from the study highlighted a correlation between the chemical composition and the external factors such as color, size, shape, and manufacturing technology of glass beads recovered from Geumgwan Gaya, revealing characteristics related to that time and region.
