Effect of Maengjong-Juk (Phyllostachys Pubescens) Extract Coated Rice Diet on Antioxidative System of C57BL/6 Mice Fed Atherogenic Diet (맹종죽(Phyllostachys Pubescens)추출물 코팅쌀이 Atherogenic 식이를 섭취한 C57BL/6 마우스의 항산화 시스템에 미치는 영향)

  • To evaluate the antioxidative effect of maengjong-juk (Phyllostachys pubescens) extract coated rice in vivo system, maengjong-juk extract coated rice diets were fed to C57BL/6 mice for 16 weeks. Plasma total antioxidative capacity, hepatic lipid peroxidation, protein oxidation, activities of antioxidative enzymes and total glutathione content were measured. Plasma total antioxidative capacity was elevated significantly in maengjong-juk extract diets supplemented group in a dose dependant manner. Hepatic TBARS contents were significantly decreased in maengjong-juk extract diets supplemented group compared to high cholesterol group. Maengjong-juk extract coated rice diets suppressed the protein oxidation significantly in liver. Activities of hepatic antioxidative enzymes such as total SOD, CuㆍZn-SOD, Mn-SOD, GSH-Px and catalase activities of maengjong-juk extract coated rice diets were significantly higher than those of high cholesterol diet. Total hepatic glutathione content was significantly increased by maengjong-juk extract coated rice diets administration. According to this study, numerous antioxidative materials and phytochemicals containing in maengjong-juk extracts appear to protect antioxidative systems in C57BL/6 mice fed bamboo extract coated rice diet.   • To determination of antioxidant substance homoorientin, from Phyllostachys bambusoides leaves, the ultrasonic extraction and HPLC on-line $ABTS^+$ screening method were empolyed. Also, the various experimental variables such as the frequency and time of ultrasonic system were investigated and homoorientin was extracted efficiently at the low frequency 35 kHz and the extraction time 60 min. The values were positive peak 1574.71 (relative area, 23.67%) and negative peak 6924.34 (relative area, 1.23%), respectively. This HPLC on-line $ABTS^+$ screening method was rapid and efficient to search for antioxidants from natural products. These results will provide a database for investigating the constituents of natural products and the resources of pharmaceutical and cosmetic products.

Anti-inflammatory Effects of the Water Extract of Phyllostachys Folium via NF-κB Inhibition (죽엽 열수추출물의 염증억제 효과)

  • Objectives : Phyllostachys Folium is leaves of Phyllostachys nigra var. henesis $S_{TAPF}$. In the East Asian traditional medicine, the herb has been used to treat nasal bleeding, dysuria, epilepsy and etc. The present study was conducted to evaluate the anti-inflammatory effects of the Phyllostachys Folium water extracts (PFE) in vitro and in vivo model. Methods : Cell viability was measured by MTT assay after the treatment of PFE and NO production was monitored by measuring the nitrite content in culture medium. iNOS, COX-2, $I{\kappa}B$, $p-I{\kappa}B{\alpha}$ amd $NF{\kappa}B$ were detected by immunoblot analysis, and levels of cytokine were analyzed by sandwich ELISA kit. Anti-edema effect of PFE was determined in the carrageenan-induced paw edema model in rats. Results : LPS increased NO and cytokines levels compared with control, these increases were attenuated by PFE. In addition, LPS-induced pro-inflammatory proteins such as iNOS, COX-2 were down regulated by PFE. These anti-inflammatory effect of PFE results from inhibition of phosphorylation of $I{\kappa}B$ and translocation of $NF-{\kappa}B$. Conclusion : These results show that PFE has some anti-inflammatory effects which might play a role in gram-negative bacterial infection inflammation and $NF{\kappa}B$ activated diseases.

Wood Characteristics of Phyllastachys bambusoides, Phyllostachys nigra var. henonis, and Phyllastachys pubescens Grown in Damyang District (담양지역(潭陽地域) 왕대, 분죽 및 맹종죽의 재질특성(材質特性))

  • This study was performed to investigate the basic wood characteristics of Phyllostachys bambusoides, Phyllostaehys nigra var. henonis, and Phyllostaehys pubeseens grown in Damyang district in Korea. There were no significant differences in the anatomical structure of three species and they showed rediculate vessels and tylosis. The bulk density of P. bambusoides, P. nigra var. henonis, and P. pubeseens was 0.61g/$cm^3$, 0.66g/$cm^3$, and 0.58g/$cm^3$ P. respectively. P. pubescens showed the highest cleavage resistance and P. nigra var. henonis showed the lowest cleavage resistance, and therefore P. nigra var. henonis was fittest for the cleaving prosessing of bamboo.

Genetic Diversity and Phylogenetic Relationship of Genus Phyllostachys by RAPD Markers (RAPD분자 마커를 이용한 왕대속 대나무의 유전적 다양성 및 계통 관계)

  • Genus Phyllostachys is a long-lived woody species primarily distributed throughout South East Asia. Many species of this genus has been regarded as medically and ecologically important in the world. We evaluated representative samples of the four taxa with RAPD to estimate genetic relationships within the genus Phyllostachys. The percentages of polymorphic loci were 8.9-33.3% at the species level. P. bambusoides was found to show lower genetic diversity (H=0.018) than other species. Total genetic diversity ($H_T$) was 0.315, genetic diversity within populations ($H_S$) was 0.043, the proportion of total genetic diversity partitioned among populations ($G_{ST}$) was 0.659 and the gene flow (Nm) was 0.0263. As some Korean populations were isolated and patchily distributed, they exhibited low levels of genetic diversity. The four taxa of the genus Phyllostachys analyzed were distinctly related to a monophyletic. P. nigra var. henonis. Stapf was found to be more closely related to P. pubescens than to P. nigra. P. bambusoides was quite distinct from the remaining species.

Anti-hypertensive Effects of ethanol extract of Phyllostachys Pubescens via Antioxidant Activity (맹종죽의 항산화활성을 통한 항고혈압 효능)

  • Phyllostachys pubescens (Maengiong-Juk), a kind of the bamboo, was reported to have many beneficial pharmacological actions. in this study, of using 70% ethanol extract of Phyllostachys pubescens we investigated its efficacy on angiotensin converting enzyme (ACE) and antioxidant enzyme activities. In addition, vasorelaxant effect was examined in rat aortic rings. The inhibitory effect of ACE activity by Phyllostachys pubescens extract (PPE) was dose-dependently increased by 61.42% at 10mg/ml. PPE relaxed the pre-contracted rat aortic rings with 10$^{-6}$M phenylephrine, showing about 88% at 4.0mg/ml. Sprague Dawley (SD) rats were given different concentrations of PPE mixed in the drinking water for 10 weeks. PPE did not show any difference with control group in blood pressure, body weight (BW) and food intake. However, it revealed the highest total antioxidative effect at dose of 1.0 g/100 g BW in plasma by TEAC assay. Thiobarbituric acid reactive substance (TBARS) and protein carbonyl levels which are markers of tissue peroxidation, were significantly lowed at the same dosage. Furthermore, hepatic antioxidant enzymes such as total superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and catalase activities were also significantly increased by PPE (1.0 g/100 g BW). In conclusion, we suggest that PPE might have antihypertensive effect through increasing antioxidant activities.

A Morphological Study of Bamboos in Mt. Jiri by Vascular Bundle Sheath (지리산(智異山) 죽류(竹類)의 유관속초(維管束鞘)에 의(依)한 형태학적(形態學的) 연구(硏究))

  • I have investigated and compared the morphology of vascular bundle shown in the section of culm wall of bamboo trees growing on Mt. Jiri which were classified by Grosser and Liese with their methods of morphological classification. The results obtained were as follows: 1. It was shown that there are no b.g.i. types of bamboo classified by Grosser and Liese among the bamboo trees on Mt. Jiri (Phyllostachys and Sasa). 2. As for the thickness of the culm wall in the culm, it was shown that the culm wall of the Phyllostachys becomes thinner in proportion to its nearness to the upper part of the tree, but no distinctive difference appeared in the Sasa. 3. The c, d, and e types of Sasa were the same as those of the Phyllostachys, but there was a vascular bundle type of the a' type, which was quite different from that of the Phyllostachys. 4. It was shown that the a', d, and e types of Sasa were distributed in a zone less than 500m above sea level, but no a' type was distributed in the high mountain area except for the c, d and e types which ranged from 600m to 1000m above sea level. Such facts mean that the vascular bundle sheath has changed in quantity because of the height of mountain. 5. In general, as compared with the Phyllostachys, the Sasa (types a, c, d and e which included a new type a) have fewer vascular bundles. 6. Considering the above results, it is thought that not by the current Sasa classification method based on observation of the the study of Sasa form the outside, but by a new method of classification based on the aspect of the physiological construction as seen from the inside wall is advanced. I believe this new method of classification to be a first step towards an epoch-making methodological advance and encourage the further study of it.

Impact of hexeneuronic acid on kappa number determination in 3 different bamboo soda-anthraquinone(AQ) pulps (대나무류 3종 소다-안트라퀴논 펄프 내 존재하는 hexeneuronic acid가 카파 값에 미치는 영향)

  • Hexeneuronic acid in soda-anthraquinone(AQ) pulps from Moso bamboo(Phyllostachys pubescebs), Timber bamboo(Phyllostachys bambusoides) and Henon bamboo(Phyllostachys nigra var. henonis) was investigated with mercuric chloride hydrolysis and UV spectroscopic quantification. Concentration of hexeneuronic acid in bamboo pulps was $36.6-45.4{\mu}mol/g$, which contributed to 3.1-3.9 value increase of kappa number. Lower concentration of 4-O-methylglucuronic acid in bamboo xylan contributed to lower hexeneruonic acid content in bamboo pulps than those of hardwood(yellow poplar or eucalyptus) but higher than softwood(red pine).

Isozymic Variation of Local Populations in the 3 Species of Phyllostachys (왕대속 3종의 지역개체군간 동위효소변이)

  • Intraspecific and interspecific isozyme variations and their relationship of 16 local populations in 3 species of Phyllostachys, that is, P. bambusoides, P. nigra var. henonis and P. pubescens were investigated by multi-variate analysis. Leaf isozymes of Phyllostachys such as 6-PGD (6-phosphogluconate dehydrogenase), MDH (malate dehydrogenase), PGI (phosphoglucoisomerase), PRX (peroxidase), PGM (phosphoglutamase), IDH (isocitrate dehydrogenase) showed electrophoretic variations in the number of zymotypes (7, 6, 6, 9, 3 and 5, respectively). In the cluster analysis on the isozymic characteristics, 16 populations were classified into 3 species at the euclid genetic distance of 2.041. P. nigra var. henonis and P. bambusoides were clustered first at 2.813 and then P. pubescens at 3.001. So far, 3 local types of intraspecific ariation were found in P. nigra var. henonis and P. bambusoides.

Dynamic Viscoelasticity of Heat-Treated Bamboo (열처리(熱處理) 죽재(竹材)의 동적점탄성(動的粘彈性))

  • This study was undertaken to investigate the effect of heat treatment on the dynamic viscoelasticity of three species of Phyllostachys bambusoides, Phyllostachys nigra var. henonis and Phyllostachys pubescens, grown in southern Korea. The bamboo was treated for 3~24 hours at $60{\sim}180^{\circ}C$, and then was treated in a climatic chamber for 3~48 hours at $40^{\circ}C$ and 95% relative humidity. The results obtained are summarized as follows : 1. Dynamic Young's modulus decreased with increasing temperature and duration of the heat treatment. 2. Internal friction decreased with increasing treatment duration. 3. Moisture absorption decreased with increasing temperature and duration of the heat treatment. 4. Dynamic viscoelasticity decreased, whereas internal friction slowly increased, with increasing moisture content.
