To investigate the effect of intravenous ethanol administration on pancreatic exocrine secretion, we measured volume and protein amount in pancreatic juice and assayed amylase activity and phospholipase $A_2$ activity in pancreatic fragments and serum. Acute pancreatitis induced by obstruction of common bile-pancreatic duct (CBPD) and caerulein infusion (5 $\mu\textrm{g}$/kg/hr) showed typical characteristics, such as hyperamylasemia and pancreatic edema and increase of phospholipase $A_2$ activity in pancreatic fragments and serum. Intravenous ethanol infusion (50 mg/kg/hr) significantly stimulated pancreatic exocrine secretion, but such a stimulatory effect of ethanol disappeared at dose of 100 mg/kg/hr without typical symptoms of acute pancreatitis. In microscopic examination, there were no typical changes of edematous pancreatitis in ethanol administrated rats. These results suggest that acute ethanol administration has dual effect on exocrine pancreatic secretion: low dose of ethanol (50 mg/kg/hr) stimulates pancreatic exocrine secretion, whereas high dose of ethanol (100 mg/kg/hr) does not without typical changes of edematous pancreatitis.

Phospholipase C is a key enzyme of signaling pathways hydrolyzed phosphatidylinositol 4,5-bisphosphate to generate 2 second messengers. Among the PLC, $PLC-{\beta}$ subfamily consisted of 4 isoforms, $PLC-{\beta}$ 1~4. Here, we studied the tissue specific expression of $PLC-{\beta}3$ in olive flounder (Paralichthys olivaceus) following external stimulation like lipopolysaccharide (LPS), concanavalin A (ConA) and environmental stress compared with the inflammatory cytokines IL-1b. $PoPLC-{\beta}3$ gene transcripts has the effect in stimulated tissue compared to control. These results provide what we sure to be a important role for $PLC-{\beta}3$ activity in tissue and verify $PLC-{\beta}3$ as potential immune enzyme for signal transduction.
식물 호르몬의 하나인 앱시스산이 식물의 기공을 닫게 하는 과정 중에 phospholipase C가 활성화되어 inositol 1,4,5-trisphosphate(P3)의 양이 증가함이 보고되었다 (Cot and Crain. 1994). 그러나 아직까지 공변세포에서 phospholipase C의 활성을 조절하는 G-단백질에 대한 보고는 없었다. 그러므로 앱시스산에 의한 기공닫힘과정에 G-단백질이 수반되는지를 조사하고자, G-단백질 활성의 저해제인 pertussis toxin과 촉진제인 cholera toxin을 처리하여 보았다. 닭의장풀(Commelina communis L.)의 잎 뒷면으로부터 얻은 온전한 표피층과 잠두(Vicia faba L)의 잎을 부분 분해하여 공변세포만을 남긴 표피층에 pertussis toxin을 처리하였을 때, 앱시스산에 의한 기공닫힘이 부분적으로 억제됨을 관찰하였다. 그러나 cholera toxin의 경우는 아무런 영향이 없었다. 공변세포만을 지닌 표피층에 pertussis toxin을 전처리한 후 앱시스산을 가했을 때, 앱시스산에 의한 IP3 양의 증가 양상이 억제됨을 확인하였다. 이러한 결과들로부터 앱시스산에 의한 기공닫힘과정에는 pertussis toxin-sensitive, phospholipase C-linked G-protein이 관여하고 있음을 알 수 있었다.

Fresh egg yolk (EY) was enzymatically modified using phospholipase $A_2$ ($PLA_2$) to produce an enzymatically modified-egg yolk powder (EM-EYP). The EM-EYP offered significantly higher emulsifying activity, emulsion stability, protein solubility, and mayonnaise stability than the control EYP. By employing $PLA_2$ in the enzymatic modification process, structural changes occurred in the phospholipids and lipoproteins of the yolk, and cleavage of apo-high density lipoprotein (HDL) components (Mw 105 kDa) was detected by sodium dodecyl sulfate-polyaerylamide gel electrophoresis (SDS-PAGE). Based on its functional properties, EM-EYP has great potential as a replacement for fresh EY in the production of processed food products such as mayonnaise.
Effects of exogenous glycerophospholipids on oleate-dependent phospholipase D (PLD) activity were studied in lymphocytic mouse leukemia L1210 cells and in solubilized microsomal phospholipase D of rat brain. Among the phospholipids tested phosphatidic acid had the most stimulatory effects on both PLD activities up to about 3 folds. Lysophosphatidic acid also showed promoting effect on microsomal PLD activity but much less on L1210 cells compared to that of phosphatidic acid. While phosphatidylethanolamine increased PLD activity slightly, phosphatidylinositides were nearly ineffective in the tested sources. The stimulatory effect of phosphatidic acid observed can be utilized to improve the in vitro assay system for oleate-dependent PLD in mammalian sources.
Phospholipase D2 (PLD2) has been implicated in the tyrosine kinase-mediated signaling pathways, but the regulation events are yet to be identified. Herein, we demonstrate that pleckstrin homology (PH) domain of PLD2 (PLD2-PH) exerts an antitumorigenic effect via the suppression of PLD2 and focal adhesion kinase (FAK). The kinase domain of FAK interacts with PLD2-PH and induces tyrosine phosphorylation and activation of PLD2. Furthermore, PLD2 increased tyrosine phosphorylation of FAK. However, ectopic expression of the PLD2-PH competes for binding to FAK and reduces the interaction between PLD2 and FAK, thereby suppressing FAK-induced PLD activation and tyrosine phosphorylation of FAK. The PLD2-PH suppressed the migration and invasion of glioblastoma cells, as well as tumor formation in a xenograft mouse model. This study uncovers a novel role of PLD2-PH as a negative regulator of PLD2 and FAK.
A gene encoding a putative phospholipase D was isolated from Bacillus licheniformis and cloned into pGEM-T easy vector. The gene was expressed in E. coli BL21 (DE3) using a pET-21(a) vector containing His6 tag. Affinity purification of the recombinant phospholipase D with nickel-nitrilotriacetic acid (Ni-NTA) resin resulted major one-band by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis. The purified enzyme showed a molecular weight of 44 kDa. The optimum activity of enzyme was around pH 7.0 and the enzyme was also the most stable around this condition. The optimum temperature was about $40-45^{\circ}C$ and the enzyme still showed considerable activities at wide range of temperature. Among various detergents, Triton X-100 significantly increased the enzyme activity, resulting in 181% activity of control at 0.6 mM of the detergent. Calcium ion did not significantly affect the enzyme activity, suggesting that the enzyme might be classified into $Ca^{2+}$-independent PLD.
The epidermal growth factor (EGF) is an important signaling ligand for the mitogenesis of many cells. The EGF receptors use signaling molecule multicomplexes and dynamic protein networks for the transmission and amplification of the signals as well as for the regulation of the cellular responses. EGF signaling has been reported to be enhanced in various tumors by the overexpressed EGF receptor and/or the mediators such as phospholipase C-$\gamma$1(PLC$\gamma$1). (omitted)

The role of a phosphoinositide-specific phospholipase C, PLC-deltal, in the bradykinin receptor-mediated signaling pathway was investigated using a clone of stably overexpressed PLC-deltal in rat pheochromocytoma (PC12) cells. Stimulation with bradykinin induced significantly higher [Ca$\^$2+/]i rise in PLC-deltal-overexpressed cells (PC12-D1) than in the wild type (PC12-W) and the vector-transfected (PC12-V) cells.(omitted)
