양제근 추출물 및 분획의 항산화 활성과 Tyrosinase 저해 활성 (Antioxidant and Tyrosinase Inhibitory Activity of Extract Rumex japonicus HOUTT Root and Its Fractions)

  • Background: We investigated the antioxidative and tyrosinase inhibitory activities of 70% ethanol extract, and its fractions, of the root of Rumex japonicus HOUTT. Methods and Results: The total phenolic compound contents of the 70% ethanol extract and ethyl acetate fraction were 168.99 mg/g and 651.78 mg/g, respectively. The antioxidant activity was compared through the DPPH radical and nitric oxide (NO) scavenging assays. The ethyl acetate fraction showed the highest DPPH radical and NO scavenging abilities, which confirmed the antioxidant activity. Specifically, the ethyl acetate fraction showed a higher DPPH radical scavenging ability than ascorbic acid. These results were related to the total phenolic compound content of the ethyl acetate fraction. Moreover, in the tyrosinase inhibition assay, the ethyl acetate fraction exhibited stronger inhibitory activity than arbutin, which was used as the positive control. The cell viability of L929 cells was analyzed by MTT assay after treatment with 70% ethanol extract and all fractions; no changes in viability were observed, which demonstrated the nontoxic nature of the extract and fractions. Conclusions: These results suggested that the extract from the root of R. japonicus and its ethyl acetate fraction could be a novel resource for the development of a cosmetic with antioxidant and tyrosinase inhibitory activity.

민들레 추출물의 항산화 활성 및 세포독성 효과 (Antioxidant Activity and Cytotoxicity Effect of Extracts from Taraxacum mongolicum H.)

  • This study was investigated antioxidant and anticancer activity of water, methanol extract from upper and root part of Taraxacum mongolicum H. Total phenolic compound contents of methanol and water extracts from upper part were $51.95{\pm}0.18$ mg/g and $48.16{\pm}0.89$ mg/g respectively, and total flavonoid compound contents were estimated as $20.57{\pm}1.12$ mg/g in methanol extract and $6.55{\pm}1.20$ mg/g in water extract. $EC_{50}$ values for DPPH radical scavenging activity of methanol and water extract from upper part were $138.47{\pm}3.78{\mu}g/mL$ and $204.38{\pm}5.32{\mu}g/mL$, and methanol and water extracts from root part were as $512{\pm}8.11{\mu}g/mL$ in methanol extract and $1315.05{\pm}11.98{\mu}g/mL$ in water extract. Reducing power and hydroxyl radical $({\bullet}OH)$ scavenging activity estimated that methanol extract of each part were higher than water extracts. The cell viability showed that the methanol extract from upper part had a cytotoxicity in the growth of colon carcinoma cell (44.58%). Both water extract $(51.97{\pm}11.43%)$ from upper part and methanol $(53.46{\pm}19.77%)$, water $(52.79{\pm}13.53%)$ extracts from root part had quite higher cytotoxicity than that of methanol extract $(88.25{\pm}2.02%)$ from upper part. Based on the results, It was suggested that the methanol extract of Taraxacum mongolicum H. were potential materials for use as functional food and medicine.

복분자 추출액 급이 사양꿀과 일부 시판꿀의 항산화활성 비교 (Comparison of Some Antioxidative Activities of Feeding Honey from the Mixture of Extract of Rubus Coreanus Miquel and Sugar with Three Types of Honeys on the Market)

  • To make sure of the usefulness of extract of Rubus coreanus Miquel for producing functional feeding honey, we compared some antioxidative indicators of feeding honey using extract of Rubus coreanus Miquel with acacia honey, SueBee Clover honey(USA), feeding honey on the market. The water content of four honeys were 16.6~26.5%, pH were 3.18~3.70, and titratable acidity ranged 0.018~0.022%. The phenolic compound contents of SueBee Clover honey and feeding honey using extract of Rubus coreanus Miquel were 8.3 mg/100 g and 7.3 mg/100 g, respectively, and were significaltly higher thgheacacia honey and feeding honey on the market. The flavonoid contents per 100 g of honey ranged from 2.3 mg(acacia honey) to 15.0 mg(SueBee Clover honey). DPPH anion scavenging activity of four honeys were not high. 0.5~2.0 g/ml of feeding honey using extract of Rubus coreanus Miquel was 16~36% and showed a concentration-reliant figure. At the concentration of 0.25~0.75 mg/ml, the reducing power of four honeys increased concentration-dependently, and the power of 0.25 mg/ml of feeding honey using extract of Rubus coreanus Miquel was corresponding to thgt of $150{\mu}g$/ml of a vitamin C solution. ABTS radical scavenging activity of feeding honey using extract of Rubus coreanus Miquel was 43.3~68.4%; the highest activity amongst all samples. When plotting the dose-response curve, ABTS radical scavenging activity also increased as their concentration increased from 62.5 mg/ml to 500 mg/ml. The heat treatment reduced the phenolic compound contents of acacia honey and feeding honey using extract of Rubus coreanus DPPH anion scavenging activity of feeding honey on the market and feeding honey using extract of Rubus coreanus Miquel did not changed significaltly after heating for 20 min, and the three honey except SueBee Clover honey maintained the reducing power with the same treatment.

와송의 이화학적 특성 및 NDMA 생성에 관한 연구 (Studies on the Physicochemical Characteristics and NDMA Formation of Orostachys japonicus A. Berger)

  • The purpose of this study was to analyze the changes in physicochemical characteristics and NDMA formation of Orostachys japonicus A. Berger according to harvest times. The results showed moisture, carbohydrate, crude fiber and crude protein contents of <$48.5{\pm}1.4%$, $38.2{\pm}1.2%$, $l5.8{\pm}0.4%$ and $13.2{\pm}0.6%$ respectively. The total mineral content was 4,817.1 mg/100 g, where Ca was highest($2,577.0{\pm}3.2$ mg/100 g), followed by K, Mg, Na, P and Fe. The composition amino acid of O. japonicus A. Berger showed high levels of glutamic acid and aspartic acid. And among the five free sugars detected, galactose and glucose were most abundant at $32.2{\pm}0.02$ mg/100 g and $25.3{\pm}0.1O$ mg/100 g, respectively. Upon comparing O. japonicus A. Berger extracts that came from plants harvested during $August{\sim}October$, total phenolic compound($2,703.l{\pm}5.2l{\sim}2,428.0{\pm}3.52$ mg/100 g) and flavonoid($634.2{\pm}2.33{\sim}1,324.6{\pm}1.87$ mg/100 g) contents were higher in the methanol extract than in the water extract. Also, within a reaction system, nitrite scavenging ability and NDMA inhibition were most effective at pH 2.5, and increased in proportion to the extract concentration. Finally, the methanol extract of O. japonicus A Berger harvested during $August{\sim}October$ had the highest nitrite scavenging and NDMA inhibition effects.

아피오스 분말 첨가 소시지의 품질 특성 및 항산화 활성 (Quality Characteristics and Antioxidant Activities of Sausage Added with Apios (Apios americana M.) Powder)

  • This study investigated the effect of apios powder on the quality characteristics of sausages. The results were as follows: pH values of the sausages were not significantly different among the samples. The amount of moisture content in the sausages decreased with increasing amount of apios powder. Increase in apios powder increased the lightness (L) value but decreased redness (a) and yellowness (b) values. Increase in apios powder increased the hardness and chewiness of sausage but decreased the adhesiveness. Apios powder had no significant effect on gumminess and springiness of the sausages among all samples. Total phenolic compound content and DPPH radical scavenging activity of apios powder added sausage significantly increased with increasing apios powder. Total bacterial counts of apios powder added sausage increased in all the samples as storage period passes and after 4 weeks of storage, the more adding ratio of apios powder was increased, the total bacterial count decreased. In view of above results, apios powder was considered to be food material suitable not only for functionality but also for developing sausage product of which preservation property is improved. From the results, 100% apios powder can be used to make optimum level for production of sausage.

Waterlogging induced oxidative stress and the mortality of the Antarctic plant, Deschampsia antarctica

  • We investigated the mortality and the oxidative damages of Deschampsia antarctica in response to waterlogging stress. In field, we compared the changes in the density of D. antarctica tuft at the two different sites over 3 years. The soil water content at site 2 was 6-fold higher than that of site 1, and the density of D. antarctica tuft decreased significantly by 55.4% at site 2 for 3 years, but there was no significant change at site 1. Experimental results in growth chamber showed that the $H_2O_2$ and malondialdehyde content increased under root-flooding treatment (hypoxic conditions-deficiency of $O_2$), but any significant change was not perceptible under the shoot-flooding treatment (anoxic condition-absence of $O_2$). However, total chlorophyll, soluble sugar, protein content, and phenolic compound decreased under the shoot-flooding treatment. In addition, the catalase activity increased significantly on the 1st day of flooding. These results indicate that hypoxic conditions may lead to the overproduction of reactive oxygen species, and anoxic conditions can deplete primary metabolites such as sugars and protein in the leaf tissues of D. antarctica. Under present warming trend in Antarctic Peninsula, D. antarctica tuft growing near the shoreline might more frequently experience flooding due to glacier melting and inundation of seawater, which can enhance the risk of this plant mortality.

식품부패세균에 대한 가자(Terminalia chebula Retz.) 추출물의 항균활성 (Antibacterial Activity of Terminalia chebula Retz. Extract Against Food Spoilage Microorganisms)

  • 가자 물 추출물과 70% 에탄올 추출물을 acetone, hexane, chloroform, butanol 용매로 순차 분획하였다. 용매 순차 분획물중 butanol분획물이 E. coli, S. typhimurium, P. aeruginosa, B. subtilis, S. aureus, L. monocytogenes균종에 대해 가장 좋은 항균활성과 추출수율을 나타내었다. 또한 polyphenol 표준물질 중 pyrogallol이 항균활성 실험결과 가장 좋은 활성을 나타내었다. 가자의 물 추출물과 에탄올 추출물을 13가지 표준품을 사용하여 HPLC 분석한 결과, pyrogallol의 수치가 가장 높게 나타났다.

국내산 유채 종자의 품종별 지방 조성 및 페놀 추출물의 항산화 활성 (Lipid Composition of Korean Rapeseed (Brassica napus L.) Cultivar and Antioxidant Capacity of Phenolic Extract)

  • 본 연구는 국내산 유채 종자 내한, 탐라, 목포111호, 영산, 탐미, 한라와 목포68호의 품종별 지방산 조성, 인지질, 당지질, 토코페롤 함량을 조사하고, 유채 종자를 탈지한 후 crude phenolic compound를 추출하여 총 페놀 함량과 항산화 활성을 분석하였다. 유채 종자의 조지방 함량은 29.3~33.2%였고, 품종들 간의 유의적인 차이는 없는 것으로 나타났다. 주요 지방산은 oleic(n-9), linoleic(n-6)과 ${\alpha}$-linolenic acid(n-3)였으며, n-6/n-3 지방산의 비율은 2.20~3.68이고 이 중 내한종이 가장 낮고 한라종이 가장 높았다. Neutral lipids 함량은 내한종과 탐라종, 인지질 함량은 탐미종에서 가장 높고, 소량 함유된 당지질은 내한, 탐라, 한라와 목포68호 간 함량 차이가 없었으며 목포111호에 가장 낮게 함유되었다. 토코페놀 함량 수준은 ${\gamma}$ > ${\alpha}$ > ${\beta}$ > ${\delta}$-tocopherol이며, 내한종의 총 tocopherol 함량이 가장 높고 다음으로 탐라, 목포111의 순이었으며 영산, 탐미, 한라 목포68호는 유의적인 함량 차이를 보이지 않았다. 품종별 총 페놀 함량은 탐라> 영산> 탐미, 한라, 내한> 목포68호, 목포111호의 순으로 나타났으며, 탐라종의 DPPH 라디칼 소거능과 FRAP value가 가장 높아 7개의 품종 중 항산화 활성이 가장 높은 것으로 확인되었다. 탈지 후의 유채박에 함유된 phenol compounds의 용매별 분획할 결과 30% ethanol fraction에 가장 많이 추출되며 이 fraction의 항산화 활성이 가장 높게 나타났다.

감귤박 에탄올추출물과 열수추출물의 영양성분 및 항산화 활성 (Antioxidant Activity and Nutrient Content of Ethanol and Hot-Water Extracts of Citrus unshiu Pomace)

  • 본 연구는 감귤박의 생리기능성을 탐색하고자 통과 감귤박과, 과피를 제거한 과육 감귤박을 열수와 에탄올로 각각 추출하여 일반성분 및 항산화 활성을 측정하였다. 감귤박의 일반성분을 측정한 결과, 열량과 탄수화물은 통과 감귤박이 더 높았으며 수분은 과육 감귤박이 더 높은 것으로 나타났다. 총 폴리페놀 함량은 추출 용매에 따른 차이는 없었으나, 과피를 포함하고 있는 통과 감귤박이 더 높은 함량을 나타냈다. ESR을 이용한 DPPH 라디칼 소거능과 alkyl 라디칼 소거능 측정 결과, 과육보다는 통과 감귤박의 활성이 더 높았으며, 열수추출물보다는 에탄올추출물이 활성이 높게 측정되었다. Liquid chromatography를 이용한 플라보노이드 함량 분석 결과, naringin은 통과 에탄올추출물에서만 검출되었으며, 또한 nobiletin과 tangeretin의 함량이 통과 에탄올추출물에서 높게 나타났다. GC 분석 결과를 토대로 감귤박 추출물의 항산화 활성은 플라보노이드뿐만 아니라 HMF와 maltol에 기인할 수 있음을 나타냈다. 따라서 과피를 포함하고 있는 통과 감귤박은 기능성 식품을 개발하는 데에 있어서 높은 활용 가치가 있는 것으로 사료된다.

용규 추출물의 항산화 활성 탐색 (Antioxidant Activities of Extracts of Solanum nigrum L.)

  • In this study, the antioxidant activities of fractions (methanol, hexane, methylene chloride, butanol, ethyl acetate and water) of Solanum nigrum L. extract was investigated. The contents of total phenolic compounds of each fractions of methanol, butanol, methylene chloride, ethyl acetate, hexane and water are $4.41{\pm}0.23%$, $5.57{\pm}0.35%$, $9.89{\pm}0.19%$, $9.86{\pm}0.19%$, $1.89{\pm}0.04%$, and $3.18{\pm}0.06%$, respectively. For assay of antioxidant activity, 1,1-diphenyl-2-picryhydrazyl (DPPH) radical scavenging activity, reducing power and nitrite scavenging activity are evaluated. In DPPH radical scavenging activity, the highest effect was obtained from the fraction of ethyl acetate. Reducing power is ordered as ethyl acetate > methylene chloride > methanol. In nitrite scavenging activity, the highest activity was 5.5% (butanol fraction), whereas hexane fraction did not detected. Overall, antioxidant activities are closely related the content of phenolic compound in extracts of S. nigrum L.
