In order to obtain anticancer substances from natural products, extracts of dry herbs, which have long been used to treat cancer or canter-like diseases in oriental countries, were screened. Extracts were made with hot water and/or organic solvents. With the extracts we treated murine leukemic L1210 cells growing in Fischer's medium. After 48 hours of incubation, cells were counted and concentrations of dry extracts to achieve 50 percent inhibition of the control growth, ED$_{50}$ values, were determined. Among the 38 species of medicinal plants tested, water extracts of six species showed ED$_{50}$ values of substantially low. Further extraction with organic solvents could reduce their ED$_{50}$ values within the range of the NCI quality control limit. The promising species as potential sources of anti-cancer substances included Cinnamomum cassia, Citrus trifoliata, Coptis japonica, Panax ginseng, Phellodendron amurense, and Scutellaria baikalensis.
We examine whether Phellodendron amurense (PA) and its major alkaloid compound, berberine (BER), improved memory defects caused by administering scopolamine in rats. Effects of PA and BER on the acetylcholinergic system and pro-inflammatory cytokines in the hippocampus were also investigated. Male rats were administered daily doses for 14 days of PA (100 and 200 mg/kg, i.p.) and BER (20 mg/kg, i.p.) 30 min before scopolamine injection (2 mg/kg, i.p.). Daily administration of PA and BER improved memory impairment as measured by the passive avoidance test and reduced the escape latency for finding the platform in the Morris water maze test. Administration of PA and BER significantly alleviated memory-associated decreases in cholinergic immunoreactivity and restored brain-derived neurotrophic factor and cAMP-response element-binding protein mRNA expression in the hippocampus. PA and BER also decreased significantly the expression of proinflammatory cytokines such as interleukin-$1{\beta}$, tumor necrosis factor-${\alpha}$ and cyclooxygenase-2 mRNA in the hippocampus. These results demonstrated that PA and BER had significant neuroprotective effects against neuronal impairment and memory dysfunction caused by scopolamine in rats. These results suggest that PA and BER may be useful as therapeutic agents for improving cognitive functioning by stimulating cholinergic enzyme activity and alleviating inflammatory responses.

Cortex Phellodendri (CP) is derived from the dried bark of Phellodendron amurense. It has been widely used as a drug in traditional Korea medicine for treating diarrhea, jaundice, swelling pains in the knees and feet, urinary tract infections and infections of the body surface. In this study, preparative centrifugal partition chromatography (CPC) was successfully carried out to separate antibacterial compounds from a CP methanol extract. The optimum two-phase CPC solvent system was composed of n-butanol: acetic acid: water (4:1:5 v/v/v). The flow rate of the mobile phase was 3 $m{\ell}/min$ in ascending mode with rotation at 1,000 rpm. The CPC-separated fraction and purification procedures were carried out by preparatory HPLC. Palmatine weas identified by $^1H$, $^{13}C$-nuclear magnetic resonance and electrospray ionization-mass spectroscopy spectral data analysis.

We evaluated the hepatoprotective activity of crude hot-water extracts of the traditional Korean medicinal herb, Phellodendri Cortex (PC; Bark of Phellodendron amurense RUPR.), in an experimental model of hepatic damage induced by carbon tetrachloride ($CCl_4$). The serum marker of liver damage, sGOT, sGPT and sALP, were significantly decreased in the liver of the PC treated rats, compared with that of $CCl_4$ treated group. The histological observation of liver section of rats, showed the same protective effect of PC treatment. And the protective activity of PC was more significant in the post-treated group than pre-treated group. The significant decrease of malodialdehyde and increase of glutathion, catalase activity were observed in the liver homogenate of PC treated rats. Based on these findings, it is suggested that PC has potent hepatoprotective effects and the mechanism of the protection may be related to antioxidation pathways.
Antifungal activities of the crude extracts of Anemarrhena asphodeloides. Coptis japonica and Phellodendron amurense were tested against Phvtophthora capsici. and the control effect on red-pepper phytophthora hlight and phytotoxicities of red-pepper were investigated. The results were summarized as follows; Mycelial growth and zoosporangial germination of the red-peppcr phytophthora hlight organism P. capsici were inhihited hy thc crude extracts of plant materials. Methanol extracts or plant materials had hctter antifungal activity than water extracts at hoth a room temperature and a hoiling condition. Antifungal activities of three crude extracts were gradually decreased with prolonged storage period. Red-pepper phytophthora hlight was effectively controlled hy the crude extracts of three plant materials. Of these. the crude extract of C. japonica was marvelously effective. Phytotoxic symptom to red-pepper seedling showed hy water cultural method hut not by pot test. Seed germination and radicle growth of red-pepper were inhihited hy the crude extracts of three plant materials. Phytotoxic symptoms in the leaves and fruits of red-pepper were not ohserved with exogenous foliage application of the three crude extracts.
Thirty-four strains of bacteria were isolated from Phellodendron amurense. Using Nectria haematococca as an indicator strain, the best strain, B18, was obtained by the growth rate method. The morphological, physiological and biochemical characteristics of strain B18 and its 16S DNA gene sequence were identified, and the biocontrol effect of strain B18 was assessed in pot and field tests, as well as in a field-control test. Drilling methods were used to determine the antibacterial activity of metabolites from strain B18 and their effects on the growth of pathogen mycelia and spores. The best bacteriostatic rate was 85.4%. B18 can hydrolyse starch and oxidize glucose but does not produce gas; a positive result was obtained in a gelatine liquefaction test. According to 16S DNA gene sequencing, strain B18 is Bacillus methylotrophicus (GenBank accession number: MG457759). The results of pot and field-control trials showed 98% disease control when inoculating $10^8cfu/ml$ of the strain. The disease control effect of the B18 culture liquid (concentrations of $10^8$, $2{\times}10^6$, $10^6$, $5{\times}10^5$ and $2.5{\times}10^5cfu/ml$) in the field-control test was higher than 80%, and the cure rate of the original delivery solution was 96%. Therefore, in the practical forestry production, a $2.5{\times}10^5cfu/ml$ culture liquidshould be applied in advance to achieve good control effects.
General pharmacological properties of ADP, a new pharmaceutical composition, which contains a mixed water extract obtained from the mixture of Phellodendron cortex (Phellodendron amurense) and Anemarrhena rhizoma (Anemarrhena asphodeloides), as the active ingredients, were investigated in experimental animals administering orally and in vitro test system. ADP had no influences on general behavior, pentobarbital sleeping time, spontaneous motor activity, motor coordination of mice, normal body temperature, chemoshock produced by pentylenetetrazole and writhing syndromes induced by 0.8% acetic acid at the dose of 150 and 1500 mg/kg. Gastric secretion of rats and intestinal motility of mice were not also influenced by the administration of ADP at doses of 150 and 1500 mg/kg, with the exception of the significant decrease of free HCI concentration at a dose of 1500 mg/kg in rats. ADP (150 and 1500 mg/kg) did not alter mean arterial blood pressure and heart rate in conscious rats. ADP given to anesthetized rats showed no effect on respiratory rate at the same doses. In in vitro experiments, ADP at the concentration of 150 mg/L did not show direct effect and inhibitory or augmentative action on histamine- or acetylcholine-induced contractions in the isolated ileum of guinea-pig. Taken together, these results indicate that ADP does not induce any adverse effects in experimental animals.

The most absorbance of Phellodendron. sol-ution is 330nm berberine is 430nm. 2. The best and proper temperature to extrat Phe,. was 80 and dyeing solution for 24 hours. 3. The pH effect was stable in the absorbances and color differences change. 4. Mordanted group increased the absorbances compared to non-mordanted sample. In case depth the most absorbance of Al. Sn. K. Cu and C. A solution were in 3% Fe and Cr were in 5%. In the color difference change they became dark reduced greenish and increased yellowish when their absorbances were in 3% 5. K./S was recoginzed by tainnin and fe, K/S was more increased pre-mordants than post-mordants. 6. Fe is the unchangable mordant in the color difference change and pre-mordants is more stable than post-mordants in the difference color. 7. Generally light C. F indicated more than grade 2 and light C. F, was much improved in post-mordanted by alumium potassium potass-ium dichromate. Washing C.F was improved in mordanted by tainnin and ferrous suulfate mordanted group was better than non-mordanted sample. Perspir-ation C,F was more stable in acid than alkali. Dry cleaning C.F indicated more than grade. 4.

In this study, among 10 medicinal plants extracted with 50%-ethanol, antimicrobial activities measured by paper disc method were showed in Scutellaria baicalensis (Sb) on Leu mesenteroides and in Phellodendron amurene (Pa) on Lac. plantarum and Leu. mesenteroides. While 0.05∼0.2% of Pa extracts showed also relatively strong growth inhibition of both strains of lactic acid bacteria cultured in MRS broth for 24 hours at 3$0^{\circ}C$, Sb extract at concentration of 0.1% showed similar inhibitory effect on Leu mesenteroides to that of 0.05% of Pa extract. Addition of 0.02∼0.04% of Pa extracts to kimchi lowered effectively extents of pH decrease and acidity increase, and numbers of lactic acid bacteria and total bacteria compared to those of control during fermentation at 4$^{\circ}C$. This extending effect on fermentation period was obtained by adding 0.04% in case of Sb. Mixed extracts of Pa and Sb (Pa-Sb) by ratio 1 : 1 delayed also fermentation of kimchi at 0.03∼0.04%. In sensory evaluation kimchi containing 0.04% of Pa extract were less sour than kimchi containing 0.02% of Sb extract and/or control at late stage of fermentation.
To investigate bioactive materials for development of natural conservative agent on organic cultural heritage, methanol extracts from 11 wood species which populate in Korea were screened for antifungal and insecticidal activity against 9 wood contaminant fungi and adult of Lasioderma serricorne. Antifungal activity of extracts was tested by using paper disc soaking method and feeder strip test against wood-rot fungi and wood surface contaminant fungi. Among these extracts, the most significant antimicrobial activity was observed from the extract of Phellodendron amurense at 5 mg/disc against Coniophora puteana, Lentinus lepideus, Tyromyces palustris and Aspergillus niger. The insecticidal activity of extracts was examined by topical application against L. serricorne adults. The extracts from Taxus cuspidata and Abies holophylla gave 56.7 % mortality at 25 ${\mu}g$/adult for 24 hr. From these result, the methanol extract from T. cuspidata, A. holophylla and P. amurense described could be useful for conservation of organic cultural heritage against biological deterioration by insect and wood contaminant fungi.
