Long-Term Effects of Growth Regulators and Nitrogen Sources on Proliferation and Turnover of Cell Wall Polysaccharides in Suspension Culture of Kidney Bean (Phaseolus vulgaris L.) (강낭콩의 현탁배양시 증식과 세포벽 다당류 전환에 미치는 생장조절제 및 질소원의 장기간 효과)

  • To underatand in vitro regulation of differentiation, the effects of growth regulators and nitrogen source on metabolism of cell wall polysaccharides in suspension culture of kidney bean (Phaseolus vulgaris L.) were investigated. The suspension cells (cell clusters) were directly induced from the epicotyl segments of the seedlings, which were cultivated in MS medium supplemented with 1.0mg/L of 2,4-D and 0.5 mg/L of kinetin. When compared with cell wall sugar contents of the epicotyl segments, the cellulose content of the suspension-cultured cells decreased; while the pectin and hemicellulose content increased; suggesting increases of rhamnogalacturonan I and arabinogalactan IIduring the dedifferentiation, respectively, The effects of growth regulators(2,4-D, 1.0mg/L and kinetin, 0.5mg/L) and nitrogen source (potasium nitrate, 19.0mg/L and ammonium nitrate, 16.5 g/L) in the medium on the proliferation and the turnover of the cell wall polysaccharides were investigated for 30 days. In the medium with growth regulators and without nitrogen source, the proliferation rate was extremely high (16 folds). Growth regulators and nitrogen source increased the pectin content. Analysis of neutral sugar composition of pectin fraction showed that nitrogen source enhanced rhamnose level remarkably, suggesting that rhamnogalacturonan I was the one most likely synthesized. In hemicellulose fraction, growth regulators reduced arabinose level, suggesting that arabinogalactan II was degraded. And nitrogen source reduced galactose level, suggesting that xyloglucan was also degraded.

  • Changes in activity of gibberellin $3{\beta}-hydroxylase$ which converts $[^3H]GA_{20}\;to\;GA_1$ were studied during seed maturation using partially purified enzyme preparations of two cultivars, Kentucky Wonder (normal) and Masterpiece (dwarf) of Phaseolus vulgaris. The specific activity of $3{\beta}-hydroxylase$ per seed reached maximum at 21 days after flowering and subsequently decreased during seed maturation in both cultivars. The ratios of conversion of $[17-^{13}C,\;^3H_2]GA_{20}\;to\;GA_1.\;GA_5,\;and\;GA_6$ by the same amount of $3{\beta}-hydroxylase$ were almost identical. Epoxidation of $GA_5\;to\;GA_6$ is also catalyzed by the partially purified $3{\beta}-hydroxylase$ preparation(Kobayashi et aI., 1991) and the conversion was inhibited by the substrates of $3{\beta}-hydroxylase$. These results suggest that the same enzyme might catalyze $3{\beta}-hydroxylase{\;}of{\;}GA_{20}\;to\;GA_1$ and epoxidation of $GA_5\;to\;GA_6$..

Role of Plant Hormones in the Senescing Detached Leaves of Phaseolus vulgaris (강낭콩 잎의 노화에 있어서 식물 호르몬의 역할)

  • Role of plant hormones on the leaf senescence of Phaseolus vulgaris were investigated by measuring the disassembly of chlorophyll-protein complexes in detached leaves treated with NAA, GA$_{3}$ , or BA. The loss of chlorophyll that was characteristic of leaf senescence induced disassembly of chlorophyll-protein complexed. During dark-induced senescence, PSI complex was rapidly degraded after the early stage, whereas RC-Core3 was slightly increased until the middle stage and slowly decreased thereafter. And gradual degradation of trimeric LCHII progressed after the late stage of senescence. Exogenous application of NAA and GA$_{3}$ had little or no effect in protecting disassembly of chlorophyll-protein complexes during leaf senescence compared to control. However exogenous BA application strongly leaves. In the simultaneous treatment of plant hormones and light, BA application under illumination of light was most effective in the stability of chlorophyll-protein complexes, particularly PSI, LHCII, RC-Core2, RC-Core3 and SC-1. these results suggest, therefore, that simultaneous application of BA and light induced synergistic effect on the stability off chlorophyll-protein complexes during leaf senescence.

Evaluation of Phytochemical econtents and antioxidant activity of Korean common bean (Phaseolus vulgaris L) landraces (한국 재래종 강낭콩 유전자원의 phytochemical 및 항산화 활성 평가)

  • The Korean common bean (Phaseolus vulgaris L.) has been receiving increased attention as a functional food. The objective of this study was to reveal the phytochemicals genetic variation and antioxidant activity of 209 Korean common bean landraces. Antioxidant activity was evaluated with the DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate), ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid), ferric reducing antioxidant power (FRAP), and superoxide dismutase (SOD) assay. Antioxidant activities among common bean accessions showed wide variation. Four flavonoids (kaempferol, myricetin, quercetin, and naringenin) of the 209 Korean common bean landraces were measured using HPLC. Among them, kaempferol had the highest phytochemicals compared to the other three flavonoids. Using the relative antioxidant capacity index (RACI), it was found out that the IT104587 had the highest antioxidant activity. Meanwhile, in clustering analysis, the Korean common bean landraces were classified into three clusters. Among them, cluster II contained 64 landraces with higher antioxidant activities and phytochemicals than the other clusters, except DPPH. The results could provide information on the valuable Korean common bean landraces for the development of new common bean varieties.

Effects of Decursin and Decursinol on the Germination and Growth of Plants (Decursin과 Decursinol이 식물의 생장과 발아에 미치는 영향)

  • Biological activities of decursin and decursinol, natural coumarin derivatives, on the germination, growth and adventitious root formation of several plants were observed. In 10 ppm of decursin or decursinol, the growth of Avena coleoptile sections was inhibited, and the activity of IAA-oxidase was gradually enhanced by the increase of its concentrations. Inhibition effect on seed germination was observed from 100 ppm of each chemcials, and the activity of amylase in the germinating seeds was also gradually decreased. However, in the higher concentrations of decursin, the inhibited germination rate of wheat was slightly reduced. Decursin and decursinol also promoted the adventitious root formation in seeds of Phaseolus vulgaris.

  • L-PHA, a lectin having lymphoagglutinating activity but devoid of erythroagglutinability which is contained in Korean white kidney bean (Phaseolus vulgaris L.), was isolated and purified through several techniques such as ion exchange chromatography, hydroxyapatite column and affinity chromatography. Purified L-PHA was identified as a single band by polyacrylamide disc gel electrophoresis. The molecular weight of the L-PHA was estimated about 125, 000 daltons by polyacrylamide gel electrophoresis and it was turned out having one subunit (probably dimer of the Yachnin's model) of Mr~60, 000. Immunochemical studies also tried and these results reconfirmed the purity of this L-PHA.

  • Rhizofiltration for cesium uptake by sunflowers (Helianthus annuus L.) and beans (Phaseolus vulgaris var.) was investigated for groundwater contamination. The cesium removal by sunflowers was greater than 98% of the total cesium in solution, and the uptake by beans was also greater than 99% within 24 hours of the rhizofiltration, showing that the rhizofiltration has a great capability to remove cesium from the contaminated water system. Experiments at various pH of solution indicated that a solution of pH $5{\sim}9$ yielded very high cesium accumulation in two plants. From the results of the analysis for cesium accumulation in plant parts, about 80% of cesium transferred into the plant from solution was accumulated in the root part and less than 20% of cesium existed in the shoot part (including leaves). Results suggest that only the roots of the fully grown plant used for rhizofiltration should be disposed or post-treated and thus the cost and time to treat massive amounts of grown plants could be dramatically reduced when sunflower and bean are used in the real field. The results of SEM and EDS analyses indicated that the most of cesium were accumulated in the root surface as a ionic phase rather than a soil precipitation phase.
