Nutritional quality of Peucedanum japonicum Thunb. leaves in relation to ripening time, growing condition and blanching (재배조건, 수확시기 및 열처리에 따른 갯기름나물의 영양성분 변화)

  • This work aimed to investigate the changes in the nutrient contents and antioxidant activity of Peucedanum japonicum Thunb. by ripening stage, growing condition, and blanching. The crude protein content of the young leaves (3.6~4.3%) was higher than that of the mature leaves (3.1~3.9%). Higher calcium contents were observed in the greenhouse-cultivated samples (225.9~259.2 mg/100 g) compared to the field-cultivated samples (178.5~199.5 mg/100 g). The vitamin C and folate contents (18.1~83.8 and 175.8~220.2 mg/100 g, respectively) of the field-cultivated samples were significantly (p<0.05) higher than those of the greenhouse-cultivated samples (13.1~57.7 and 133.0~148.8 mg/100 g, respectively). The growing condition and blanching were significant factors affecting the changes in the vitamin and polyphenol contents. The ${\beta}$ carotene contents of the blanched samples increased 2.6-fold compared to those of the raw samples. The total polyphenol contents (10.2~17.1 mg/g extract) and DPPH radical scavenging activity ($IC_{50}=2.0{\sim}3.0mg/ml$) of the blanched samples were significantly (p<0.05) higher than those of the raw samples (1.8~4.3 and $IC_{50}=16.2{\sim}21.1mg/ml$, respectively).

Peucedanum japonicum Thunberg Leaf Alleviates the Symptoms of Dextran Sulfate Sodium Induced Ulcerative Colitis in Mice (식방풍 잎의 DSS로 유도한 궤양성 대장염 완화 효과)

  • In general, Reucedani Radix (Peucedanum japonicum Thunbergis: PJ) is the Korean traditional herbal medicine used to remove dampness, to relieve pain, and spasm. So, PJ folium is believed to have the same effects. The aim of this study is to investigate the alleviation of dextran sulfate sodium (DSS) induced ulcerative colitis in mice by PJ folium. 25 mice were divided into 5 groups: normal, DSS, DSS + 100 mg/kg PJ folium (PJ100), DSS + 500 mg/kg PJ folium (PJ500), and DSS + 150 mg/kg 5-amino salicylic acid (5-ASA) groups. Body weights, colon lengths, histological changes in colon tissue, and spleen weights were observed. Inflammatory cytokines such as IL-$1{\beta}$, IL-6, and TNF-${\alpha}$ level in plasma were measured by ELISA. IL-$1{\beta}$, IL-6, and TNF-a mRNA expression in colon tissue were detected by RT-PCR. In the results, body weight lose was inhibited in PJ100, PJ500, and 5-ASA groups, but it was not different compared with DSS group, significantly. PJ500 group showed the preventive effects of colon length shorten and histological changes in colon tissues as good as 5-ASA group. The weight of spleen was increased in DSS group but it reduced in PJ100, PJ500, and 5-ASA groups. Moreover, IL-$1{\beta}$ and TNF-${\alpha}$ cytokine levels in plasma were reduced in PJ500 and 5-ASA groups. IL-$1{\beta}$, IL-6, and TNF-a mRNA expression in colon tissue were inhibited in PJ100, PJ500, and 5-ASA groups and it was significantly different compared with DSS group. In conclusion, PJ folium showed the alleviative effect on DSS induced ulcerative colitis in mice.

Comparison of Volatile Compounds Identified in Different Parts of Peucedanum japonicum Thunberg by Harvest Time (방풍나물의 수확시기에 따른 부위별 향기성분 비교 분석)

  • Headspace volatile compounds of leaf, stem, and root of Peucedanum japonicum Thunberg (PJT) harvested in March and July were isolated, separated, and identified by using a combined system of solid phase microextraction and gas chromatography/mass spectrometry. Identified numbers of volatile compounds were 72, 75, and 63 in March PJT as well as 78, 73, and 69 in July, respectively. Total compounds identified from PJT consisted of alcohols (1~7), aldehydes (7~15), esters (1~4), hydrocarbons (5~9), ketones (1~2), monoterpenes (13~16), monoterpene alcohols (1~5), monoterpene aldehydes (2~4), monoterpene esters (1~4), monoterpene ketones (1~2), sesquiterpenes (16~24), and miscellaneous compounds (2~3). Major volatile compounds of PJT were monoterpenes at all harvest times and in all parts of PJT. The amounts of ${\beta}$-pinene in leaf and stem harvested in March were highest in monoterpenes, followed by ${\alpha}$-pinene and ${\beta}$-myrcene. However, amounts of volatile compounds of monoterpenes in leaf and stem from July PJT were in the decreasing order of ${\beta}$-pinene, ${\beta}$-phellandrene, and ${\beta}$-myrcene. Of the 39 sesquiterpenes identified in PJT, (E)-caryophyllene in March leaf was the most abundant volatile compound, followed by acoradiene in March leaf, ${\beta}$-elemene, and ${\alpha}$-copaene in July stem.

Changes in Growth and Yield of Peucedanum japonicum Thunberg by Planting Time, Mulching, and Planting Density (식방풍(植防風)의 파종기(播種期), 피복(被覆), 재식밀도(栽植密度)에 따른 생육(生育)과 수양(收量) 변화(變化))

  • This experiment was carried out in the field to investigate the effects of sowing time, mulching materials and planting density on the growth and root yield of Peucedanum japonicum. The optfimum planting time for yield increase at Kyungbuk area whether the plant was grown under non-mulching or black P. E. mulching condition. Black polyethylene (P. E) film mulching was effective to promote emergence and growth, and dry root yield in the P. E. mulching was 10% higher than that in the non-mulching condition. The yield in the $20{\times}15cm$ planting density was 50% higher than that in the conventional $30{\times}20cm$.

  • Peucedanum japonicum $T_{HUNB}$ used as a edible and medicinal plants was investigated for in uitro regeneration. Callus formation occurred on leaf and stem explant cultures and showed spontaneous embryogenic and organogenic capability on MS basal medium supplemented with 0.1~5 mg/L NAA and 0~10 mg/L BA in dark. The regeneration was highest on the condition supplemented with 2.5 mg/L NAA and 10 mg/L BA. Development of the somatic embryo progressed through the globular, heart-shaped, torpedo-shaped and cotyledonary stage, typical of zygotic embryos. When the first somatic embryos was cultured on the medium supplemented with 0.2 mg/L NAA, secondary somatic embryo were induced with higher frequency on the hypocotyl then on the cotyledon and root.t.

  • A new Sikbangpoong(Peucedanum japonicum Thunberg) variety, 'Sikbangpoog 1', was developed through a pure line selection at the Kyougpuk Provincial RDA during the period of 1990 to 1995. The variety was characterized to have higher stem height, leaf number and seed production, and larger root diameter compared with the check variety of Bonghwa Sikbangpoong but plant height of 'Siknagpoong 1' was similar to that of the check variety and it was also more tolerant to water and heat stress. Root color of 'Sikbangpoong 1' was yellowish brown and root shape of the variety was straight and conical. The root yields of Sikbangpoong 1 in yield trial, regional adaptation trial and farmer's field trial were always 10-28% higher than those of the check variety.

Antinociceptive effect of intrathecal sec-O-glucosylhamaudol on the formalin-induced pain in rats

  • Background: The root of Peucedanum japonicum Thunb., a perennial herb found in Japan, the Philippines, China, and Korea, is used as an analgesic. In a previous study, sec-O-glucosylhamaudol (SOG) showed an analgesic effect. This study was performed to examine the antinociceptive effect of intrathecal SOG in the formalin test. Methods: Male Sprague-Dawley rats were implanted with an intrathecal catheter. Rats were randomly treated with a vehicle and SOG ($10{\mu}g$, $30{\mu}g$, $60{\mu}g$, and $100{\mu}g$) before formalin injection. Five percent formalin was injected into the hind-paw, and a biphasic reaction followed, consisting of flinching and licking behaviors (phase 1, 0-10 min; phase 2, 10-60 min). Naloxone was injected 10 min before administration of SOG $100{\mu}g$ to evaluate the involvement of SOG with an opioid receptor. Dose-responsiveness and ED50 values were calculated. Results: Intrathecal SOG showed a significant reduction of the flinching responses at both phases in a dose-dependent manner. Significant effects were showed from the dose of $30{\mu}g$ and maximum effects were achieved at a dose of $100{\mu}g$ in both phases. The ED50 value (95% confidence intervals) of intrathecal SOG was 30.3 $(25.8-35.5){\mu}g$ during phase 1, and 48.0 (41.4-55.7) during phase 2. The antinociceptive effects of SOG ($100{\mu}g$) were significantly reverted at both phases of the formalin test by naloxone. Conclusions: These results demonstrate that intrathecal SOG has a very strong antinociceptive effect in the formalin test and it seems the effect is related to an opioid receptor.

Effect of the water extract of Peucedani Japonici Radix on ovalbumin-induced allergic asthma in mice (난알부민으로 유도된 천식 마우스에서 식방풍(植防風)의 알레르기 면역반응 조절효과 연구)

  • Objectives : The root of Peucedanum japonicum Thunberg (Peucedani Japonici Radix; PJR) has been traditionally used as an herbal medicine for the treatment of anti-headache, anti-paralysis, anti-cancer, vascular protection, and blood pressure regulation. In this study, we investigated the anti-allergic effect of PJR water extract on ovalbumin (OVA)-induced allergic asthma in mice. Methods : Mice were sensitized at days 1, 8 and 15 with OVA and airway challenged at days 22, 24, 26, 28, and 30 to induced allergic asthma. PJR-W extract at doses of 100 and 300 mg/kg/body weight (bw) was orally administered during OVA challenge once per a day. The levels of allergic mediators such as immunoglobulin (Ig) E, and Th1/Th2 cytokines (IFN-${\gamma}$ and IL-4) were measured in the sera of mice by ELISA. The histological change of lung tissue was observed with hematoxylin and eosin (H&E) staining. Results : The administration of PJR-W extract significantly decreased the serum levels of IgE, IL-4, and IFN-${\gamma}$ compared with those of OVA control group. In H&E staining, PJR-E extract inhibited OVA-induced airway inflammation and the inflammatory cells infiltration in the peribronchial regions of the lung. Conclusions : These results indicate that PJR-W extract has an anti-inflammatory and anti-allergic effect on allergic response through the down-regulation of allergic mediators, suggesting that this herb may be used as a useful source for the treatment of allergic inflammatory diseases such as asthma.

Comparative Study of Bang-poong (root of Saposhnikovia divaricata Schischkin) and Related Species on Neuroprotective and Acetylcholinesterase Inhibitory Effects (방풍류(防風類) 약재(藥材)의 신경세포보호효과 및 아세틸콜린에스터라제 저해 효과 비교)

  • Objectives : Bang-poong (Saposhnikovia divaricata; SD) was traditionally used to treat inflammatory disorders. In this study, we aimed to investigate whether Bang-poong and related species including SD, Glehnia littoralis (GL), and Peucedanum japonicum (PJ) possess neuroprotective effects and acetylcholinesterase (AChE) inhibitory activities. Methods : Roots of SD, GL and PJ were extracted with distilled water (DW) or 70% ethanol (EtOH). We assessed 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activities of the extracts. To examine neuroprotective effects, we measured cell viability in PC12 or HT22 cells after treatment of the extracts with $H_2O_2$ or amyloid-beta ($A{\beta}$). To assess anti-neuroinflammatory effects, we measured the nitric oxide (NO) levels after treatment with the extracts and lipopolysaccharide (LPS) in BV2 microglial cells. In addition, we performed AChE inhibition assay to explore effects of the extracts on the cholinergic system. Results : DW and EtOH extracts of SD, GL and PJ showed mild DPPH free radical scavenging activities. Also, DW extracts of GL and PJ showed protective effects against $H_2O_2$-induced toxicity in PC12 cells. In LPS-activated BV2 cells, EtOH extracts of SD, GL and PJ exerted inhibitory effects on NO production. Meanwhile, DW extracts of SD, GL and PJ inhibited the $A{\beta}$-induced cell death in HT22 cells. In addition, DW and EtOH extracts of GL exhibited remarkable inhibitory activities on AChE. Conclusions : We demonstrated that SD, GL and PJ exert anti-oxidative, anti-neuroinflammatory and AChE inhibitory activities. These results indicate that SD, GL and PJ could be potential candidates for neurological disorders.
