Objective : To investigate the effect between Fructus Immaturus Ponciri (FIP, the immature fruit of Poncirus trifoliata) and Fructus Ponciri (FP, the ripe fruit of Poncirus trifoliata) on mast cell-mediated immediate-type allergic reactions. Methods : We performed anaphylactic reaction, histamine release, cAMP, $TNF-{\alpha}$, IgE. Results : The aqueous extract of FIP dose-dependently inhibited systemic and local allergic reaction was induced by compound 48/80 or anti-dinitrophenyl (DNP) IgE in a murine model. FIP also significantly inhibited mast cell-dependent ear swelling response induced by topical application of compound 48/80. When mice were orally pretreated with FIP, the plasma histamine levels were reduced in a dose-dependent manner. FIP dose-dependently inhibited histamine release from the rat peritoneal mast cells (RPMCs) was activated by compound 48/80 or anti-DNP IgE. The level of cAMP in RPMCs, when FIP was added, increased compared with that of a normal or control. In addition, FIP had a significant inhibitory effect on anti-DNP IgE-induced tumor necrosis factor-a ($TNF-{\alpha}$) production from the RPMCs and IgE produced by lipopolysaccharide-stimulated murine whole spleen cells or U266B1 as human IgE-bearing B cells. However, FP showed the lower inhibition rate than those of FIP in above all allergic reactions. Conclusion : These data have important implications for our understanding of the clinical effects of FIP and FP on allergic diseases, and FIP is more effective than FP on the allergic reaction.
Pruritus is a unpleasant symptom in the skin that provokes the act of or desire to scratch. Mast cells are important effector cells in allergic reactions such as pruritus and inflammation. The purpose of this study was undertaken to investigate the synergic anti-pruritic and anti-inflammatory effects of Scutellariae Radix (SB) plus Flos Loncerae (FL) extracts in rat peritoneal mast cells (RPMCs), pruritogen-induced scratching mice and 2,4-dinitrofluorobenzene (DNFB)-induced allergic mice. We investigated the effect of SB, FL and SB plus FL extracts on the production of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-$1{\beta}$, and histamine in RPMCs, on the scratching behavior in ICR mice, and skin clinical serverity and inflammatory mediators in DNFB-induced allergic hairless mice. RPMCs stimulated with PMA plus A23187 or compound 48/80 significantly increased TNF-${\alpha}$, IL-$1{\beta}$ or histamine production compared with media control. However, TNF-${\alpha}$ IL-$1{\beta}$ or histamine levels increased by PMA plus A23187 or compound 48/80 treatment were significantly inhibited by SB, FL in a dose-dependent manner. Especially, SB plus FL pretreatment had a synergic inhibitory effects on stimulator-induced cytokines (TNF-${\alpha}$ and IL-$1{\beta}$) and histamine production. Moreover, SB plus FL administration had a synergic inhibitory effects on the scratching behavior induced by pruritogen (compound 48/80, histamine, serotonin, substance P) in ICR mice. Furthermore, SB plus FL administration had a synergic inhibitory effects on skin damage, inflammatory mediators, leukocyte and mast cell infiltration induced by DNFB in hairless mice. These results suggest that SB plus FL administration has a potential use as a medicinal plant for treatment against itching and inflammation-related skin disease.
Purpose: This study was performed to investigate the effects of Dangguijakyaksan on the development of experimentally-induced endometriosis in rats. Methods: Endometriosis was induced in rats by autotransplanting uterine tissue to the peritoneum and divided them into three groups: (1) sham-operated group(n=8). (2) surgically induced endometriosis and untreated control group(n=8). (3) surgically induced endometriosis and Dangguijakyaksan treated group. Dangguijakyaksan was orally administrated for 15 days after operation. Then we measured the body weight. the volume of endometriotic implants. the weight of uterus and ovary. and analysed the concentration of cytokines (MCP-1, TNF-$\alpha$, IL-1$\beta$, IL-6) in peritoneal fluids. Histopathology, immunohistochemistry for COX-2 and VEGF. and histochemistry for mast cell in transplanted uterine tissue were also performed. Results: - The volume($mm^3$) of endometriotic implants in Dangguijakyaksan treated group was significantly decreased compared with control group. - The concentration(pg/ml) of MCP-1, TNF-$\alpha$ and IL-l$\alpha$ in peritoneal fluids in Dangguijakyaksan treated group was significantly decreased compared with control group. - Histopathologically, proliferation of endometriotic epithelia. infiltration of inflammatory cells and angiogenesis in transplanted uterine tissue of Dangguijakyaksan treated group were weakly observed than those of control group. - The percentage of positive epithelial layers for COX-2 in Dangguijakyaksan treated group was significantly decreased compared with control group. - The VEGF expression of endometriotic epithelia, neovascular endothelia and stromal cells in transplanted uterine tissue of Dangguijakyaksan treated group were weakly observed than those of control group. - The number of mast cells in transplanted uterine tissue in Dangguijakyaksan treated group was significantly decreased compared with control group. Conclusion: On the basis of these results. we concluded that Dangguijakyaksan has inhibiting effects on the development of transplanted uterine tissue. And these effects may be related with decreased production of MCP-1, TNF-$\alpha$ and IL-1$\beta$, and decreased expression of COX-2 and VEGF, and inhibition of mast cell infiltration by administration of Dangguijakyaksan.
한방에서 사용되는 약재 중 가자와 지모, 단삼은 면역조절 및 항암효과, 항염증효과가 있다고 알려졌다. 그러나 비만세포와 관련된 아나필락스를 이해할 수 있는 세포학적 기전에 대한 가자와 지모, 단삼의 효과에 대해서는 알려진 바 없다. 본 연구에서는 가자와 지모, 단삼의 메탄올 추출물이 아나필락시스 반응에 대해 어떠한 영향을 끼치는지를 조사하였다. 시험관내 실험과 생체 실험을 통하여, 가자와 지모, 단삼의 메탄올 추출물이 compound 48/80에 의한 아나필락시스와 비만세포 활성화 및 IgE에 의한 피부반응을 관찰하였다. 실험결과는 다음과 같다. 1) 가자와 지모, 단삼의 메탄올 추출물은 compound 48/80에 의한 전신성 아나필락시스와 귀부종 반응, IgE에 의한 피부반응을 억제하였다. 2) Compound 48/80에 의한 비만세포 탈과립과 흰쥐 복강 비만세포로부터 히스타민 유리가 가자와 지모, 단삼의 메탄올 추출물 전처리에 의해 억제되었다. 3) Compound 48/80에 의한 흰쥐 복강 비만세포내로의 칼슘 유입이 가자와 지모, 단삼의 메탄올 추출물 전처리에 의해 현저하게 억제되었다. 이상의 결과로 가자와 지모, 단삼의 메탄올 추출물은 비만세포 탈과립과 비만세포로부터 히스타민 유리, 비만세포내로 칼슘유입을 억제함으로써 compound 48/80에 의한 아나필락시스와 IgE에 의한 피부반응을 억제한다는 것을 알 수 있었다. 이로써 가자와 지모, 단삼은 알레르기 질환에 대한 효과적인 치료제로 이용될 수 있을 것으로 생각된다.
$80^{\circ}C$에서 30% 주정으로 추출된 소엽추출물 분말의 로즈마린산 함량은 12.3 mg/g이었으며, 소엽 30% 주정추출물분말(PF-E30)은 생쥐모델 실험에서 ant-DNP IgE으로 활성화된 local allergy 반응과 compound 48/80으로 유도된 mast cell-mediated immediatetype allergy 반응에 대한 억제 효과를 나타내었다. 더욱이, PFE30(0.1-0.5 mg/kg BW)의 투여는 혈장 히스타민 수준을 유의성있게 감소시켰으며, compound 48/80 또는 anti-DNP IgE으로 활성화된 복막 비만세포로부터 히스타민 방출을 억제하였다. 특히 PFE30는 antigen-induced IgE 의 생산을 농도 의존적으로 억제하였다. 이런 결과는 소엽 주정추출물이 in vivo 및 in vitro 실험에서 mast cell-mediated immediate-type allergy 반응을 저해한다는 것을 제시한다. 소엽 주정추출물이 알레르기 반응을 억제하는 기능성 식품 소재로서의 개발을 위해서는 mast cell mediated-type allergy 반응에 대한 보다 많은 연구가 필요할 것이다.

The immature fruits of Poncirus trifoliata L. or Ponciri fructus (PF), well known as 'Jisil' in Korea, have been used against allergic diseases for generations, and still occupy an important place in traditional Oriental medicine. Anti-allergic effects of this fruit have been investigated in a few experimental models. Immunoglobulin E (IgE) is the principal immunoglobulin involved in immediate hypersensitivities and chronic allergic diseases. The effect of an aqueous extract of PF on in vivo and in vitro IgE production was investigated. PF dose-dependently inhibited the active systemic anaphylaxis and serum IgE production induced by immunization with ovalbumin, Bordetelia pertussis toxin and aluminum hydroxide gel. PF strongly inhibited interleukin 4 (IL-4)-dependent IgE production by lipopolysaccharide-stimulated murine whole spleen cells. In the case of U266 human IgE-bearing B cells, Ponciri fructus also showed an inhibitory effect on the IgE production. On the other hand, mast cell hyperplasia can be causally related with chronic inflammation. Stem cell factor (SCF), the ligand of the c-kit protooncogene product, is a major regulator and ohernoattractant of mast cells. Ponciri fiuctus (1 mg/mL) significantly inhibited the SCF-induced migration of rat peritoneal mast cells (RPMCs). RPMCs exposed to SCF (50 ng/mL) resulted in a drastic shape change with a polarized morphology while the cells exposed to Ponciri fructus (1 mg/mL) remained resting, with little or no shape alteration. The drastic morphological alteration and distribution of polymerized actin were blocked by pretreatment with Ponciri fructus. In addition, Ponciri fructus inhibited both TNF-alpha and IL-6 secretion from RPMCs stimulated with SCF. These results suggest that Ponciri fructus has an anti-allergic activity by inhibition of IgE production from B cells. These findings also provide evidence that Ponciri fructu inhibits chemotactic response and inflammatory cytokines secretion to SCF in mast cells.
02PS15 extracts (BuOH, $H_2O$, and crude extracts) significantly inhibited IL-4 and IL-6 secretion from the phytohemagglutinin (PHA)-plus phorbol 12-myristate 13-acetate (PMA)-induced peripheral blood mononuclear cleas (P<0.05). 02PS15 extracts (BuOH and crude extracts) also significantly inhibited the histamine release from rat peritoneal mast cells (P<0.05). Significant reduced levels (P<0.05) of PMA- and A23187-induced IL-8 were observed in the human mast cell line, HMC-1, with O2PS15 extracts (BuOH and crude extracts). 02PS15 extracts (BuOH and crude extract) downregulated the expression of IL-6 and IL-8 in the activated HMC-1. These results suggest that O2PS15 has the inhibitory effect of atopic allergic reaction anil this might be useful for clinical application to treat several allergic diseases such as atopic dermatitis.
The current study evaluates the capacity of eugenol to regulate immediate allergic reaction by control of histamine release. Administrations of eugenol (1M/kg, i.p.) at 60 min b efore and 5, 10 min after the compound 48/80 treatment (8mg/kg, i.p.) were shown the mortality rates as 0, 44.4, and 77.8%, respectively. A 60 min before administered group revealed a significant inhibition of serum histamine release compared with those of 5 and 10 min after the compound 48/80 injection. Eugenol (6-48mM) was also showed a dose-dependent activity on the compound 48/80-induced histamine release from the highly purified population of Alcian Blue-positive peritonea] mast cells. These results indicate that in vitro treatment with exogenous eugenol inhibited the active response of mast cell populations and modulated its characteristics.
The purpose of this research was to investigate the effects of Kamihyungbangjihwang-tang (KHT) on immediate-type allergic reaction. KHT was composed of hyungbangjihwangtang (HJT) and Rubus coreanus Miquel (RC) and Aspalathus linear is (AL). KHT and HJT (500 mg/kg) inhibited the systemic anaphylaxis induced by compound 48/80 and inhibited the passive cutaneous anaphylaxis (PCA) induced by anti-dinitrophenyl (DNP)-IgE and DNP-human serum albumin (HSA) in vivo. In addition, KHT, HJT, RC and AL inhibited the release of histamine and increased the release of cAMP from rat peritoneal mast cells. The anti-allergic action of KHT was more potent than those of HJT. These results indicate that KHT may be useful for the prevention and treatment of type I allergy related disease via inhibition of histamine release from mast cells.
We investigated the effect of Saururus chinensis (Lour.) Baill (SCB) on allergy in mice. We conformed compound 48/80-induced mesenteric mast cell degranulation, active systemic anaphylatic shock and histamine release. Also observed acetic acid-induced vascular permeability and anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis. SCB inhibited mesenteric mast cell degranulation and active systemic anaphylatic shock induced by compound 48/80 dose-dependently. When SCB was pretreated by intra-peritoneal injection, the plasma histamine levels were reduced. SCB also significantly inhibited acetic acid-induced vascular permeability and anti-DNP IgE-mediated passive cutaneous anaphylaxis. In addition, SCB reduced IL-10 mRNA expression of the lung on ovalbumin-induced allergy. These results indicate that SCB inhibits allergy.
