상엽 유래 퀘세틴과 아스트라갈린의 항염증에 대한 상승효과 (Synergic Effect of Quercetin and Astragalin from Mulberry Leaves on Ani-inflammation)

  • The leaf of mulberry (Morus alba L) has long been used in Oriental medicine for the prevention or treatment of several diseases. However, little is known about the inhibitory effects of a single compound isolated from the mulberry leaves on inflammatory response. We are isolate a single compound of quercetin (3,3',4',5,7-pentahydroxyflavone) and astragalin (kaempferol-3-O-glucopyranoside) from the mulberry leaves, and then investigate the anti-inflammatory effects of quercetin, astragalin or quercetin plus astragalin in lipopolysaccharide (LPS)-stimulated murine peritoneal macrophages. Each compound suppressed the production of inflammatory mediators (NO, $PGE_2$ and IL-6) in LPS-stimulated murine peritoneal macrophages in a dose-dependent manner. Especially, the cotreatment of quercetin (2.5 ${\mu}M$) and astragalin (2.5 ${\mu}M$) markedly suppressed the production and the expression of inflammatory mediators. These suppressive effects were synergistically increased by their combination. These results suggest that the combination of quercetin and astragalin from the mulberry leaves may be useful for therapeutic drugs against inflammatory immune diseases, probably by suppressing the production of inflammatory mediators.

Anti-inflammatory and Anticancer Activities of Ethanol Extract of Pendulous Monkshood Root in vitro

  • Aim: Pendulous monkshood root is traditionally used for the treatment of several inflammatory pathologies such as rheumatisms, wounds, pain and tumors in China. In this study, the anti-inflammatory and anticancer activities and the mechanism of crude ethanol extract of pendulous monkshood root (EPMR) were evaluated and investigated in vitro. Materials and Methods: The cytotoxic effects of EPMR on different tumor cell lines were determined by the MTT method. Cell apoptosis and cell nucleus morphology were assessed by Hoechst 33258 staining. Moreover, nitric oxide (NO) levels and intracellular oxidative stress in peritoneal macrophages were determined to further elucidate mechanisms of action. Results: The data showed that EPMR could produce significant dose-dependent toxicity on three kinds of tumor cells. Furthermore, EPMR displayed obvious anti-inflammatory effects on LPS-induced mouse peritoneal macrophages at the dosage of 4 - 200 ${\mu}g/mL$. The results demonstrated the therapeutic potential of Pendulous Monkshood Root on cancer and inflammatory diseases. Conclusion: Our results indicate that EPMR has anti-inflammatory and anticancer properties, suggesting that pendulous monkshood root may be a useful anti-tumor and anti-inflammatory reagent in the clinic.

복수 환자 2례에 대한 임상적 고찰 (Two Cases Study on the Disease with Ascites)

  • Ascites is commonly caused by portal vein pressure accentuation or peritoneal disease. Also, the use of diuretic and ascitic puncture have been the mainstays of ascites treatment. Objective : Experimental studies were done to research the effects of herbal medication on diseases with ascites (ex. liver cirrhosis, peritoneal carcinoma) on two patients. Methods : For measuring the response about the ascites, we observed this clinical symptoms : weight, abdominal circumference (umbilical circumference, upper abdominal circumference), and lab finding. Result : In both cases, clinical symptoms and ascites improved with herbal medication. Second, abdominal circumference decreased. Third, there was no electrolyte imbalance. Conclusion : This test shows that herbal medication has a strong effect on decreasing the intractable ascites.

Polyclonal gammopathy related to renal bleeding in a peritoneal dialysis patient

  • Polyclonal gammopathy represents the diffuse activation of B cells and is usually related to inflammation or immune-related diseases. However, the mechanisms leading to polyclonal gammopathy are essentially speculative. Generally, infectious, inflammatory, or various other reactive processes may be indicated by the presence of a broad-based peak or band in the gamma region on serum protein electrophoresis results. A 15-year-old girl, who had been receiving peritoneal dialysis, presented with polyclonal gammopathy and massive gross hematuria. Renal artery embolization was performed, after which the continuous bleeding subsided and albumin-globulin dissociation resolved. This is a rare case of polyclonal gammopathy related to renal bleeding.

Treatment of steroid-resistant pediatric nephrotic syndrome

  • Children who suffer from steroid-resistant nephrotic syndrome (SRNS) require aggressive treatment to achieve remission. When intravenous high-dose methylprednisolone fails, calcineurin inhibitors, such as cyclosporine and tacrolimus, are used as the first line of treatment. A significant number of patients with SRNS progress to end-stage renal disease if remission is not achieved. For these children, renal replacement therapy can also be problematic; peritoneal dialysis may be accompanied by significant protein loss through the peritoneal membrane, and kidney allograft transplantation may be complicated by recurrence of SRNS. Plasmapheresis and rituximab were initially used for treatment of recurrent SRNS after transplantation; these are now under consideration as rescue therapies for refractory SRNS. Although the prognosis of SRNS is complicated and unfavorable, intensive treatment in the early stages of the disease may achieve remission in more than half of the patients. Therefore, timely referral of pediatric SRNS patients to pediatric nephrology specialists for histological and genetic diagnosis and treatment is highly recommended.

Chrysanthemum morifolium inhibits inflammatory responses in IFN-γ and LPS-induced mouse peritoneal macrophages

  • Chrysanthemum morifolium (CM) is a herb widely used in medicine for the treatment of a variety of diseases. In this study, using mouse peritoneal macrophages, we have examined whether CM affects nitric oxide (NO), tumor necrosis factor $(TNF)-\alpha$ and interleukin (IL)-6 induced interferon $(IFN)-\gamma$ and lipopolysaccharide (LPS). CM inhibits $IFN-\gamma$ and LPS-induced NO in dose dependent manner. We also found that CM inhibits pro-inflammatory cytokine, $TNF-\alpha$ and IL-6. The expression of cyclooxygenase-2 was reduced by CM. These finding means that CM can be used in controlling macrophages-mediated inflammatory disease.

The Anti-inflammatory Mechanism of Xanthoangelol E is Through the Suppression of NF-${\kappa}B$/Caspase-1 Activation in LPS-stimulated Mouse Peritoneal Macrophage

  • Angelica keiskei has exhibited numerous pharmacological effects including antitumor, antimetastatic, and antidiabetic effects. However, the anti-inflammatory effects and mechanisms employed by xanthoangelol E isolated from Angelica keiskei are incompletely understood. In this study, we attempted to determine the effects of Xanthoangelol E on the lipopolysaccharide (LPS)-stimulated mouse peritoneal macrophage. The findings of this study demonstrated that xanthoangelol E inhibited the production of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-6, and prostaglandin $E_2$ ($PGE_2$). Xanthoangelol E inhibited the enhanced levels of cyclooxygenase (COX)-2 and inducible nitric oxide synthase (iNOS) caused by LPS. Additionally, we showed that the anti-inflammatory effect of xanthoangelol E is through the regulation of the activation of nuclear factor (NF)-${\kappa}B$ and caspase-1. These results provide novel insights into the pharmacological actions of xanthoangelol E as a potential candidate for the development of new drugs to treat inflammatory diseases.

Paragonimiasis in the Abdominal Cavity and Subcutaneous Tissue: Report of 3 Cases

  • Paragonimiasis is a parasitic disease caused by the lung fluke, Paragonimus spp. Lung flukes may be found in various organs, such as the brain, peritoneum, subcutaneous tissues, and retroperitoneum, other than the lungs. Abdominal paragonimiasis raises a considerable diagnostic challenge to clinicians, because it is uncommon and may be confused with other abdominopelvic inflammatory diseases, particularly peritoneal tuberculosis, and peritoneal carcinomatosis. Also, subcutaneous paragonimiasis does not easily bring up clinical suspicion, due to its rarity. We herein report 2 cases of abdominal paragonimiasis and 1 case of subcutaneous paragonimiasis in Korea.

  • Proinflamamtory mediators such as prostaglandins (PGs), cyclooxygenase-2 (COX-2), and inducible nitric oxide synthase (iNOS) are known to be key mediators in pathogenesis of inflammatory diseases. Capsaicin, the major ingredient of hot pepper, is considered to elicit anti-inflammatory property. 복분자 자죽염의 마우스 대식세포주 및 복강 대식세포에 대한 면역증진 효과 (Immunostimulatory Effects of Purple Bamboo Salts Composed with Rubus coreanus in Raw264.7 Cells and Mouse Peritoneal Macrophages)

  • 본 연구에서는 전통적인 기능성 식품이자 의약품으로 사용되었던 자죽염(PubS)과 복분자를 조합한 복분자 자죽염(PuBS-R)을 사용해 비특이적, 선천성 면역에 중요한 역할을 하는 대식세포에서의 영향을 확인하였다. PuBS-R은 마우스 대식세포인 Raw264.7 세포에서 $1,000{\mu}g/mL$ 농도처리 시 세포의 증식을 유발하였으며, 그에 반해 PuBS를 $1,000{\mu}g/mL$ 처리 시 유의하게 세포 증식률이 억제되었다. 이어서 면역 관련 사이토카인 $TNF-{\alpha}$, $IFN-{\gamma}$, IL-12, IL-10 등을 유전자 단계와 단백질 단계에서 평가하였다. 그 결과 PuBS-R은 50, 100, $500{\mu}g/mL$에서 유의한 증가율을 보였고, 무엇보다 이것은 PuBS 군에 비해서도 월등한 증가율을 확인할 수 있었다. 마지막으로 마우스의 복강에서 분리한 peritoneal macrophage에 PuBS-R을 처리 후 $TNF-{\alpha}$, $IFN-{\gamma}$, IL-12, IL-10, iNOs를 단백질 단계에서 평가한 결과, Raw264.7 세포에서 얻은 결과와 동일한 결과를 확인할 수 있었다. 이러한 결과로 미루어 보아 PuBS-R은 인체의 대식세포 활성의 증가를 통해 비특이적, 선천성 면역력을 증가시킬 것으로 판단되며, 이러한 결과를 바탕으로 추후 실험동물을 이용한 in vivo 후속 연구를 통해 PuBS-R의 면역증강 기능성 식품 개발이 가능할 것으로 생각된다.
