Drug Release from Hollow Suppository(I) - Release Rate of Indomethacin from Witepsol H-15 Suppository - (중공 좌제의 약물방출 (I) - Witepsol H-15 기제로부터의 인도메타신의 방출속도 -)

  • In order to study drug release from the suppository, three types of hollow suppositories and one conventional suppository were prepared using indomethacin(IDM) as a model drug and Witepsol H-15 as a base. The 4 types of suppository prepared are as follows: type I, conventional suppository containing 50 mg of IDM powder, type II, hollow supository containing 50 mg of IDM powder in the cavity, type III, hollow suppository containing 25 mg of IDM powder in the base and IDM microcapsules (25 mg as IDM powder) in the cavity, and type IV, hollow suppository containing IDM microcapsules (25 mg as IDM powder) in the base and 0.5 ml of 5%(w/v) IDM-PEG 300 solution in the cavity. The drug amount released(%) from type II and I within 24 hrs was 46.7% and 66.9%, respectively. Comparing with the drug amount released from four types of suppository within initial 2 hrs and 24 hrs, that of type IV was high as 32.7% and 76.6%, respectively. IDM-ethycellulose microcapsules passed through 270 mesh sieve and the IDM content was 20.95%.

  • High temperature superconducting films deposited on metal Ag wire were prepared with YBCO powders by electrophoretic deposition method. $I_2$was used as additives for surface charge of YBCO particles. When 2~3 wt.% $BaF_2$ was added in the YBCO suspension, the pores and cracks of film surface were decreased and film density could be increased. In case of YBCO films, the critical current density ($J_{c}$) was calculated at the value of $1458{\;}A/\textrm{cm}^2$ (77K, 0K) by 4 point prove method.

Psychological and Physical Effects of Schroth and Pilates Exercise on Female High School Students with Idiopathic Scoliosis

  • Purpose: The purpose of this study is to investigate effects of psychological factors on Schroth exercise and Pilates exercise to female high school students with idiopathic scoliosis. Methods: Sixteen subjects with over 20 of Cobb's angle were randomly divided into Schroth exercise group (SEG, n=8) and Pilates exercise group (PEG, n=8). Both training programs were carried out 3 day a week for 12 weeks. Psychological factors were measured using depression scale, self esteem scale and body esteem scale. Results: Both groups showed significant differences in depression scale, self esteem scale and body esteem scale. Schroth exercise group showed greater improvement than Pilates group. Conclusion: Having correct body image affects psychological factors to growing students. Schroth exercise is more effective method to physical and psychological approach of idiopathic scoliosis.

Efficacy of Egg Yolk Immunoglobulin (IgY) against Edwardsiella tarda Infection (계란난황항체의 Edwardsiella tarda에 대한 효능)

  • The present study evaluated effect of egg yolk immunoglobulin (IgY) from the hen immunized with Edwardsiella tarda. The purification of anti-E. tarda IgY was performed by polyethylene glycol (PEG). Purified IgY had heavy chain of 64 kDa and light chain of 27 kDa size. The IgY was instable against olive flounders digestive factors and artificially modulated pH 2 and 3. Nevertheless, some activity of IgY appeared in intestine. IgY was orally administered with viable E. tarda to the olive flounders and the efficacy of protection against E. tarda infection was evaluated. Orally administered IgY at a dose of 20 mg/fish delayed infection period of E. tarda cannulated at $10^{6{\sim}8}CFU/fish$ to small size (30~40 g) and middle size (110~120 g) flounder. Moreover, orally administered IgY at dose of 20 mg/fish inhibited the penetration of E. tarda cannulated at $10^8CFU/fish$ into the liver, kidney, spleen and gill via intestine. The fish orally administered with IgY showed increased survival rate. These results suggest that egg yolk containing anti-E. tarda IgY is effective in preventing edwardsiellosis.

Extraction and Purification of Agar from Gelidium amansii (우뭇가사리로부터 한천의 추출 및 정제)

  • The effect of different treatments on the quality and yield of purified agar produced from Gelidium amansii has been studied, and the extraction condition ol agar produced from G. amansii has been examined. The contents of ash, sulfate in agar produced from g. amansii collected from different places were $2.63\~2.92\%\;and\;1.38\~1.78\%$, respectively. Yields and gel strength of agar produced from G. amansii collected from different places were $31.6\~46.8\%\;and\;496\~887g/cm^2$, respectively. It was effective to extract agar at $120^{\circ}C\;for\;2\~3hrs$. Agar was purified by D.W. washing, EDTA washing, chitosan treatment, CPC treatment, PEG treatment, ethanol precipitation, acetone precipitation and propanol precipitation. The mineral contents of agar produced from Gelidium amansii were Na (2934ppm), Ca (2472ppm), Mg (2259ppm), K (2527ppm), P1(81.1ppm), Fe (66.4ppm), Al (71.7ppm), Zn (29.7ppm) and Pb (ND: not detected), respectively.

Bio-Derived Poly(${\gamma}$-Glutamic Acid) Nanogels as Controlled Anticancer Drug Delivery Carriers

  • We have developed a novel type of polymer nanogel loaded with anticancer drug based on bio-derived poly(${\gamma}$-glutamic acid) (${\gamma}$-PGA). ${\gamma}$-PGA is a highly anionic polymer that is synthesized naturally by microbial species, most prominently in various bacilli, and has been shown to have excellent biocompatibility. Thiolated ${\gamma}$-PGA was synthesized by covalent coupling between the carboxyl groups of ${\gamma}$-PGA and the primary amine group of cysteamine. Doxorubicin (Dox)-loaded ${\gamma}$-PGA nanogels were fabricated using the following steps: (1) an ionic nanocomplex was formed between thiolated ${\gamma}$-PGA as the negative charge component, and Dox as the positive charge component; (2) addition of poly(ethylene glycol) (PEG) induced hydrogen-bond interactions between thiol groups of thiolated ${\gamma}$-PGA and hydroxyl groups of PEG, resulting in the nanocomplex; and (3) disulfide crosslinked ${\gamma}$-PGA nanogels were fabricated by ultrasonication. The average size and surface charge of Dox-loaded disulfide cross-linked ${\gamma}$-PGA nanogels in aqueous solution were $136.3{\pm}37.6$ nm and $-32.5{\pm}5.3$ mV, respectively. The loading amount of Dox was approximately 38.7 ${\mu}g$ per mg of ${\gamma}$-PGA nanogel. The Dox-loaded disulfide cross-linked ${\gamma}$-PGA nanogels showed controlled drug release behavior in the presence of reducing agents, glutathione (GSH) (1-10 mM). Through fluorescence microscopy and FACS, the cellular uptake of ${\gamma}$-PGA nanogels into breast cancer cells (MCF-7) was analyzed. The cytotoxic effect was evaluated using the MTT assay and was determined to be dependent on both the concentration and treatment time of ${\gamma}$-PGA nanogels. The bio-derived ${\gamma}$-PGA nanogels are expected to be a well-designed delivery carrier for controlled drug delivery applications.

Strengthening Treatment of Aged Hanji with Water Soluble Polymers (수용성 고분자를 이용한 열화한지의 보강처리)

  • In this study, 13 water soluble polymers(6 natural polymer, 7 synthetic polymer) were treated on Hanji. Mechanical properties, morphology and oxidation index with thermal aging were measured on the aged Hanji, dewaxed Hanji and polymer treated Hanji. 3 natural polymer(such as CMC, EC, MC) and 3 synthetic polymers(such as PVA 1500, 2000, PEG 1500) treated Hanji had higher strength than other polymer treated Hanji. The oxidation index of 3% methyl cellulose solution treated Hanji did a little increase with thermal aging. Finally, methyl cellulose was found to be the most efficient method for strengthening the dewaxed Hanji. The best aging safety and thermal stability were obtained at the methyl cellulose 3% water solution.

Studies on Practical Methods to Control Seed Vigour in Several Food Crops (주요식량작물 종자세 개선에 관한 연구)

  • To improve seed vigour in rice, barley and soybean, several methods of presowing treatment, using chemicals and priming in polyethylene glycol solution, were investigated. Gibberellic acid($GA_3$) slightly improved germination of rice, but other chemical treatments showed no beneficial effect on seed vigour. Aged seeds were primed in polyethylene glycol solution then rinsed and germinated with drying back. In general mean germination time increased and percentage germination decreased with in- creasing water potential of the priming solution, but there were no significant effects on spread of germination times. Priming also showed no marked improvement in germination under cold, wet, or osmotic conditions. None of the treatments used was successful in practically improving the seed germination and vigour of the tested crops. However, seed treated with GA$_3$ gave the best overall germination response.

Effect of Robot-Assisted Hand Rehabilitation on Hand Function in Chronic Stroke Patients (손 재활 로봇의 적용이 만성 뇌졸중 환자의 손 기능 향상에 미치는 영향)

  • The purpose of this study was to investigate effect of robot-assisted hand rehabilitation(Amadeo(R)) on hand motor function in chronic stroke patients. This study used a single-subject experimental design with multiple baselines across individuals. Three chronic stroke survivors with mild to sever motor impairment took part in study. Each participants had 2 weeks interval of starting intervention. Participants received robot-assisted therapy(45min/session. 3session/wk for 6wks). Finger active range of motion(AROM) was assessed by Range of Assessment program in Amadeo(R), and test-retest reliability was verified using Pearson correlation analysis. To investigate effect of Amadeo(R), finger AROM was measured immediately after each sessions and Fugl-Meyer Assessment of Upper extremity, Motor Activity Log, Nine hole peg board test and Jebsen-Taylor hand motor function test were assessed at pre-post intervention. Results were analyzed by visual analysis and comparison of pre-post tests. The test-retest reliability of Range of Assessment was good(r=.99). After robot-assisted therapy, finger AROM of participant 1, 2, and 3 was respectively improved by 18%, 3.6%, and 6% each. Hand motor function of participant 1, 3 was improved on all four tests, but not effect in participant 2. Robot-assisted hand rehabilitation could improve finger AROM and effect on hand motor function in chronic stroke patients.

Effect of Salinity on Orobanche cernua Seed Germination

  • Seeds of broomrape (Orobanche cernua) were exposed to 0, 25, 50, 75, and 100 mM NaCl solutions during their preconditioning period (14 days of moisture) under laboratory conditions and induced to germinate by synthetic germination stimulant (GR24). There was significant reduction in seed germination with increased salt concentration as shown in 35.2, 32.5, 23.6, 14.3, and 9.2% germination, respectively. Exposure of Orobanche cernua seeds to 0.0, 1.0, 1.25, and 1.5 M levels of NaCl for 9 hours resulted in 29.4, 21.3, 20.5, and 17.4% germination, respectively. Water preconditioned seeds showed Heavier protein profile bands of 6.5-14.2 KDa than those of dry seeds. Seeds treated with 0.75 M NaCl showed profile similar with that of water preconditioned ones, plus an extra band at 29-36 KDa. The protein profiles of 1.0 and 1.5 M NaCl treated seeds showed weaker bands with the absence of 29-36 KDa band.
