Biochemical Application of IgG Fc-Binding Peptide: From Biochip to Targeted Nano Carrier

  • FcBP consisting of 13 amino acids specifically binds to Immunoglobulin G Fc domain. Initially, we utilized this peptide for preparation of antibody chip as a PEG composite for enhanced solubility. After then, the peptide conjugate was immobilized on agarose resin, resulting in highly efficient affinity column for antibody purification. The efficiency was comparable to commercial Protein A column. Recently, this peptide was conjugated with cell penetrating peptide (CPP) on a backbone of GFP, affording antibody transducer, which carries antibody into live cells by simple mixing of antibody and the transducer in cell culture media. Antibody transduction into cells was monitored by live cell imaging. More recently, the FcBP was fused to ferritin cage, which consists of 24 ferritin protein molecules. The FcBP-ferritin cage showed greatly increased binding affinity to human IgG. Its binding was analyzed by QCM and SPR analysis. Finally, it was selectively delivered by Herceptin to SKBR3, a breast cancer cell, over MCF10A, non-tumorigenic cells (Fig. 1). Fig. 1. Performance of self-curing concrete as affected by different curing regimes

  • In this study, polyethylene glycol (PEG) and polyacrylamide (PAM) have been used as self-curing agents to produce self-curing concrete (SC). Compressive strength, ultrasonic pulse velocity (UPV), bulk electrical resistivity, chloride ion penetrability, water permeability, and main microstructural characteristics were examined under different curing regimes, and compared to those of the control concrete mixture with no self-curing agents. One batch of a control mixture and one batch of a SC mixture were air-cured in the lab to act as non-water-cured samples. The water curing regimes for the control mixture included continuous water curing for 3, 7, and 28 days and periodical moist curing using wetted burlap for 3 and 7 days. Curing regimes for the SC mixtures included 3 days of water curing and periodical moist curing for 3 and 7 days. SC mixtures showed better microstructure development and durability performance than those of the air-cured control mixture. A short water curing period of 3 days significantly improved the performance of the SC mixtures similar to that of the control mixture that was water cured for 28 days. SC concrete represents a step towards sustainable construction due to its lower water demand needed for curing and hence can preserve the limited water resources in many parts of the world.

Production and Characterizations of Somatic Hybrids between Brassica campestris L. ssp pekinensis and Brassica of oleracea L. var capitata

  • Protoplasts isolated from inbred lines of Brassica oleracea L. var capitata (cabbage) and Brassica campestris L. ssp. pekinensis (Chinese cabbage) were fused by PEG-mediated method, and somatic hybrid cells were differentiated into plants. for the identification of somatic hybrid plants, ploidy level, plant morphology, and cytological analysis were performed. All of the regenerated plants derived from fused protoplasts were shown to be 2X-4X, or higher ploidy level, presumably due to somatic hybridization or chromosome doubling. The morphology of leaves, petioles, and flowers showed an intermediate phenotype between Chinese cabbage and cabbage. Chromosome numbers in these somatic hybrids ranged mostly from 33 to 38. According to Genomic in situ hybridization (GISH) pattern, signals from both fusion parents of B.campestris or B.oleracea were detected in different colors when chromosomes of putative somatic hybrids were observed.

  • A. oryzae에 있어서 protoplast를 이용한 형질전환이 아닌 세포벽이 부분적으로 분해된 cell을 이용하여 electroporation으로 형질전환시켰고, novozyme234, hemicellulase와 celluclast를 사용하여 형질전환 효율이 어떻게 다른지를 비교 분석 하였다. Hemicellulase를 $^{\sim}10^8$ cell에 처리하여 A. oryzae에서 83 transformants/10ug of DNA를 얻었고, novozyme234와 celluclast를 사용하였을 때는 4.3 transformants/10ug of DNA를 얻었다.

  • Present study was performed to observe the surface ultrastructures of Stictadora furcatum (Trematoda: Heterophyidae). Adult worms were recovered from the cat experimentally infected with metacercariae, and were prepared for scanning electron microscopy. The body was leaf-like and ventrally concave. The oral sucker armed with prominent tegumental spines on the inner surface, and it's lips were obscure. Sensory papillae (type I) in single or grouped forms symmetrically arranged around the oral sucker. The ventrogenital opening retained protruding gonotyl spines and sperms discharged from the genital pore . The body surface was covered with scale-like multipointed tegumental spines. The density and digitated point of spines were gradually decreased toward the posterior end of the body. The digitated points of spines on the anterior portion were 9-12, on the middle were 7-8 and 5-6, and on the posterior were 2-3 and peg-like. Although the tegumental ultrastructure of S. fuscatum was generally similar to those of other heterophyid flukes , the oral sucker with tegumental spines on the inner surface and without obvious lips, and the ventrogenital opening with protruding gonotyl spines were suggested to be the characteristic features.

  • High viscous hyaluronic acid complex from Klebsiella sp. L-10 NTG 50 mutant was purified by two-phase extraction system using PEG-K2HP04 and its physicochemical properties were Investigated. Viscosity of the purified hyaluronic acid complex was decreased as temperature and salts concentration were Increased and also showed low viscosity at below pH 5.0 and above pH 11.0. Hardness, cohesiveness and adhesiveness of the purified hyaluronic acid complex were 1, 20kg, 1.91 and 0.62, respectively. Water holding capacity was 6.9ml per gram of the purified hyaluronic acid complex powder.

  • EVA microsphere was prepared by a thermally induced phase separation. EVAL microsphere was made by a saponification on sheath of EVA microsphere. And microcapsule with EVA core-PU shell structure was synthesized by interfacial polymerization using diisocyanates with PEG in gelatin aqueous solution as the stabilizing agent. The effects of chemical structure of diisocyanate on the average particle size and distribution, morphology, color strength and friction fastness of core-shell particles were investigated to design microcapsule. The friction fastness of the fabrics printed with EVA core-PU shell microcapsules had the 4-5 grade.

  • OMZ과 HP-$\beta$-CD 포접화합물의 유형은 $A_{L}$ type의 가용성 복합체이며, CD, UV, IR, DSC, XRD pattern 및 $^1$H-NMR 방법으로 측정한 포접화합물의 결함형태는 OMZ의 benzimidazole부분이 HP-$\beta$-CD 공동에 포접된 형태이었고, OMZ과 HP-$\beta$-CD의 결합조성몰비는 1:1, 안정도 상수는 약 34 M$^{-1}$이었다. 또한 cholestyramine 수지와 OMZ 복합체는 방출양상이 신속하였다. OMZ-cholestyramine 수지염은 안정성이 양호하고 용출율이 우수한복합체로 경구용 재제를 개발할 수 있었으며, 용출시험 및 산 저항성이 양호하고 pellet의 제조에 있어서 core의 제조는 정제수 : 에탄올(7: 3) 결합액을 이용하여 lactose를 주로 사용하고 기타 PEG 6000, sorbitol, Avicel PH101, sodium lauryl sulfate 및 무수 PVP K-30을 적절히 배합하여 제조하고, 장용코팅의 내산성은 90.5% 이었으며, 소장에서의 용출경향이 85%이상이 10분 이내에 용출되어 규정에 적합하였다.

  • ${\alpha}-amylase$ was immobilized on the surface of polyethylene glycol(M.W. 2000) grafted polyurethane film using diisocyanate in an attemp to develop enzyme immobilized polymeric materials. The surface morphology of the modified polyurethane film was examined by SEM. Effects of pH and temperature on the activity of the immobilized ${\alpha}-amylase$ were investigated. The optimal pH range of the activity was $7.0{\sim}7.5.$ The immobilized ${\alpha}-amylase$ demonstrated high thermal stability and maintained consistent activity during long-term storage.
