Studies on the Improvement of Butt Welding Characteristic of Polyethylene Pipes (PE 이중벽관의 맞대기 융착 용접 강도 향상에 관한 연구)

  • Waste water is disposed to sewage disposal plant by underground PE double wall pipes. Various processes have been introduced to join PE pipes, but most of these methods have many disadvantages such as costs, lack of reliability and difficulties in joining, etc. Recently butt welding has been paid much attention to joint PE pipes as this process has many advantages such as cost, safety and reliability. In this study, newly developed heat plate, a patent-pending heat plate with a groove, was used to butt-weld PE double wall pipes with different misalignment gaps to simulate real underground conditions, and the butt welding temperature of PE pipe was determined by thermal analysis (Thermal Gravimetric Analysis, Differential Scanning Calorimetry and Dynamic Mechanical Analysis). The resulting joining characteristics of double wall pipes were compared with those from a conventional heat plate, in terms of stiffness, flattening and leakage tests. The results from the stiffness and flattening test showed that there were no big differences between the butt-welded joints made from these two plates. From the leakage test, although PE pipes welded with a conventional heat plate did leak below the required test conditions (10 min. at 0.75kgf/cm2), the pipes welded with a patent-pending grooved heat plate did not show any leakage even at a pressure 1.5 times higher than the required conditions. It was noted that by utilizing a grooved heat plate more complete fusion at the pipe joints could be obtained, which in turn induced a high quality joints.

Effects of Flavonoid-rich Plant Extracts on In vitro Ruminal Methanogenesis, Microbial Populations and Fermentation Characteristics

  • The objective of this study was to evaluate the in vitro effects of flavonoid-rich plant extracts (PE) on ruminal fermentation characteristics and methane emission by studying their effectiveness for methanogenesis in the rumen. A fistulated Holstein cow was used as a donor of rumen fluid. The PE (Punica granatum, Betula schmidtii, Ginkgo biloba, Camellia japonica, and Cudrania tricuspidata) known to have high concentrations of flavonoid were added to an in vitro fermentation incubated with rumen fluid. Total gas production and microbial growth with all PE was higher than that of the control at 24 h incubation, while the methane emission was significantly lower (p<0.05) than that of the control. The decrease in methane accumulation relative to the control was 47.6%, 39.6%, 46.7%, 47.9%, and 48.8% for Punica, Betula, Ginkgo, Camellia, and Cudrania treatments, respectively. Ciliate populations were reduced by more than 60% in flavonoid-rich PE treatments. The Fibrobacter succinogenes diversity in all added flavonoid-rich PE was shown to increase, while the Ruminoccocus albus and R. flavefaciens populations in all PE decreased as compared with the control. In particular, the F. succinogenes community with the addition of Birch extract increased to a greater extent than that of others. In conclusion, the results of this study showed that flavonoid-rich PE decreased ruminal methane emission without adversely affecting ruminal fermentation characteristics in vitro in 24 h incubation time, suggesting that the flavonoid-rich PE have potential possibility as bio-active regulator for ruminants.

  • This study was performed to purify fig pectinesterase(F-PE) and characterize its in situ activity. Three kinds of F-PE were partially separated by using ammonium sulfate fractionation, Q-Sepharose column, CM-cation exchanger column chromatography, and HPLC. One of those was anionic protein and the others were cationic proteins. All of them had approximate molecular weight of 27,000 and lost rapidly their activity during storage. Therefore alternative crude enzyme was prepared by suspending the freeze dried and milled fig powder in 0.1 M NaCl at pH 7.5. F-PE had the optimum pH of 8.5, the optimum temperature of $50^{\circ}C$ with activation energy of 7,671 cal $mol^{-1}K^{-1}$ and stability up to $55^{\circ}C$ with 10 minutes heating. Optimum activity was obtained in $0.2{\sim}0.4$ M NaCl with optimum solubility at above 0.8 M NaCl.

  • An experimental investigation was conducted to clarify the combustion characteristics of Polyethylene-GOX(PE-GOX) hybrid motor using a single-port fuel grain configuration. Data from the experiments were analyzed to evaluate the length-averaged regression rate of PE-GOX propellants. Based on the existing theories, the empirical regression rate formulas provided from Marxman[3,4] and Altman[14] showed good concordance with the PE-GOX experiments. The accuracy of the regression rate was then evaluated and compared with the measured one. As a result, Marxman's model was somewhat more precise than Altman's model in these experiments. Moreover, the consideration of the empirical regression rate showed that O/F ratio has minor variation due to the quasi constant inflow of the fuel during motor firing.

  • Ethyl-branched polyethylene[PE(2)] containing 2mole% ethyl branch and three ethylene-propylene rubbers(EPR's) haying the same ethylene(E)-propylene(P) molar ratio(E/P=50/50) with different stereoregularity, that is, random EPR(r-EPR), alternating-EPR(alt-FPR) and isotactic-alternating-EPR(iso-alt-EPR) were mixed for the investigation of their properties depending on the stereoregurarity. The three blends were immiscible at room temperature, and showed the simple additivity effect in density behavior. The melting point depression with blend composition increased in order of PE(2)/r-EPR>PE(2)/alt-EPR>PE(2)/iso-alt-EPR. In the tensile test, this blend systems have the lowest value or the breaking strength at FE(2) fraction of 0.5. This phenomenon results from the greastest separated phase morphology at this blend composition.

The Anthropometric Status and Nutrient Intakes of Physical Education Majors and Non-major Male College Students (체육전공 남학생과 비전공 남학생의 신체조성 및 영양소 섭취에 관한 연구)

  • The study was performed to investigate the relationship of regular exercise to body composition and nutrient intakes of male college students by comparing anthropometric measurements and one-day dietary intakes by 24-hour recall of physical education (PE) majors and non-major male college students. The subjects were 253 students residing in the Gyeonggi area. Only 64.1% and 32.1% of PE majors and non-major students exercised over 4 times a week and PE major students did stronger exercise than the non-major students did. Mean BMI and WHR (Waist Hip Ratio) of the two groups were not different. But percentages of body fat and lean body mass in PE major students were lower and higher, respectively, than those of non-major students. PE major students consumed significantly more energy and other nutrients except iron, niacin, vitamin C, and vitamin E than non-major students. Nutrients consumed less than KRDA was calcium in PE major students and energy, calcium, zinc, vitamin $B_{12}$, folate, and vitamin C in non-major students. Mean adequacy ratio (MAR) of PE major students (1.00) was significantly higher than that of non-major students (0.94). Index of Nutritional Quality (INQ) of all nutrients except vitamin A of PE major students was also significantly higher than that of non-major students. Nutrients below 1.00 in INQ of both groups were calcium, zinc, vitamin B2, and folate. Our data supports that regular exercise may positively influence on body composition and increase diet quality in male college students. Therefore, an educational program focused on regular exercise along with proper dietary behavior would be needed.

Biocontrol Potential of a Lytic Bacteriophage PE204 against Bacterial Wilt of Tomato

  • Bacterial wilt caused by Ralstonia solanacearum is a devastating disease of many economically important crops. Since there is no promising control strategy for bacterial wilt, phage therapy could be adopted using virulent phages. We used phage PE204 as a model lytic bacteriophage to investigate its biocontrol potential for bacterial wilt on tomato plants. The phage PE204 has a short-tailed icosahedral structure and double-stranded DNA genome similar to that of the members of Podoviridae. PE204 is stable under a wide range of temperature and pH, and is also stable in the presence of the surfactant Silwet L-77. An artificial soil microcosm (ASM) to study phage stability in soil was adopted to investigate phage viability under a controlled system. Whereas phage showed less stability under elevated temperature in the ASM, the presence of host bacteria helped to maintain a stable phage population. Simultaneous treatment of phage PE204 at $10^8$ PFU/ml with R. solanacearum on tomato rhizosphere completely inhibited bacterial wilt occurrence, and amendment of Silwet L-77 at 0.1% to the phage suspension did not impair the disease control activity of PE204. The biocontrol activities of phage PE204 application onto tomato rhizosphere before or after R. solanacearum inoculation were also investigated. Whereas pretreatment with the phage was not effective in the control of bacterial wilt, post-treatment of PE204 delayed bacterial wilt development. Our results suggested that appropriate application of lytic phages to the plant root system with a surfactant such as Silwet L-77 could be used to control the bacterial wilt of crops.

Characteristics of Pectinesterase (PE) in Cucumbers (오이의 Pectinesterase 에 관한 연구)

  • This study was attempted to investigate the occurrence and the partial characteristics of pectinesterase (PE) in cucumbers. And the involvement of endogenous cucumber PE with the formation of insoluble pectic acid, in the presence of endogenous or added calcium ions, was also studied. The results of this study are as follows: 1) PE activity was detected in whole cucumber homogenate. 2) The optimum pH and temperature of this enzyme were found to be 8.5 and $50^{\circ}C$, respectively 3) When PE extract was added to 0.25% pectin solution, pectin gel was formed. The time required for the formation of pectin gel was reduced when the pectin solution was adjusted to the optimum conditions for PE activity(pH 8.5, $50^{\circ}C$) and $CaCl_2$ was added. 4) Cucumber juice which was heated to $100^{\circ}C$ for 10 minutes fatted to form any insoluble pectate precipitate. The formation of precipitate in cucumber juice was accelerated by preheating to $50^{\circ}C$, adjusting to pH 8.5 and addition of $CaCl_2$. 5) These results can be interpreted that endogenous PE in cucumber juice demethylates pectin, allowing interlinking of pectin molecules via divalent cation $(Ca^{++})$ and thus forming insoluble Ca-pectate. Therefore additional firming effects of cucumber can be expected to be obtained through activation of PE in conjunction with calcium ions.

  • Surface modification of hydrophobic polymeric materials to be hydrophilic or to have specific functional groups is of great importance for a diversity of applications of the materials. In this study, polyethylene (PE) film surfaces were modified by vapor phase photografting of hydrophilic vinyl monomers with different functional groups. The functional monomers were introduced on PE films by introducing the monomers in vapor phase using a vapor phase photografting apparatus designed by our laboratory. Functional monomers used were acrylic acid (negatively chargeable), acrylamide and allylalcohol (neutral), and allylamine and N,N-dimethyl aminopropyl acryamide (positively charged). The functional monomer-grafted PE film surfaces were characterized by the measurement of water contact angles and the attenuated total reflectance Fourier-transform infrared spectroscopy. The vapor phase photografting seems to be effective means for introduction of various functional groups onto polymeric substrates.
