Anti-inflammatory Effects of MeOH Extract of Corylopsis gotoana Uyeki (히어리의 항염증 효능)

  • Corylopsis gotoana (Hamamelidaceae) has been used as a traditional medicine for the treatment various diseases including cold, edema and vomiting. However, previous studies regarding component analysis and pharmacological actions of C. gotoana are extremely limited until now. In this study, we investigated anti-inflammatory activities of the methanolic extract of the twigs of Corylopsis gotoana (MCG) both in vitro and in vivo. MCG effectively inhibited excessive NO production in IFN-${\gamma}$ and LPS-stimulated mouse peritoneal macrophages without notable cytotoxicity. In addition, we also found that MCG could attenuate the expression of inflammatory mediators such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). We further tested in vivo anti-inflammatory activity of MCG using paw edema mouse model. Herein, MCG demonstrated significant suppression on the paw edema induced by both of trypsin and carrageenan. These results indicated that MCG has potent anti-inflammatory potential and may be useful for prevention and treatments of inflammatory diseases.

Anti-inflammatory and anti-nociceptive effects of extract of Astragalus membranaceous

  • The root of Astragalus membranaceous (AM) has been used to treat edema and arthritis in the traditional Korean medicine. To elucidate the anti-inflammatory and anti-nociceptive effects of ethanol extract of AM, the carrageenan-induced paw edema using a plethysmometer and thermal hypersensitivity using the plantar test were measured. Ibuprofen was used as a control drug. Pretreatment with AM (400 mg/kg p.o.) significantly reduced paw edema, compared to the carrageenan-treated rats. In the plantar test, the thermal withdrawal latency in AM-treated group was significantly increased than the carrageenan-treated group. The results indicate that AM could have be the anti-inflammatory and anti-nociceptive properties.

Effects of Achyrantes japonica on Carrageenan-Induced Arthritis Rat Model (Carrageenan으로 유발한 관절염 쥐에서의 우슬추출물 효과)

  • Achyrantes japonica (AJ) has been used to treat edema and arthritis in the traditional Korean medicine. Analgesic and anti-inflammatory activity of a polyherbal formulation (PHFAROGH)

  • The effect of arogh, a polyherbal formulation-PHF [each 3 g powder contained Nelumbo nucifera G. (0.24 g), Hemidesmus indicus R. (0.24 g), Zingiber officinale R. (0.24 g), Terminalia chebula R. (0.24 g), Quercus infectoria O. (0.12 g), Hibiscus rosa-sinensis L. (0.24 g), Rosa damascene M.(0.24 g), Eclipta alba H.(0.24 g), Glycyrrhiza glabra L. (0.24 g)] was investigated in various experimental models of pain and inflammation. Analgesic activity of PHF was studied in mice using acetic acid induced writhing, tail immersion and hot plate methods. Anti-inflammatory activity of PHF was studied in rats using carrageenan induced hind paw edema and formalin induced rat paw edema methods. PHF significantly (P < 0.05) reduced the number of writhings, increased latency to flick tail in tail immersion method and elevated the mean basal reaction time in hot plate method. PHF significantly (P < 0.05) inhibited carrageenan induced hind paw edema and formalin induced rat paw edema. The PHF was tested at dose of 30, 100, 300 and 500 mg/kg.

The Experimental study on the Anti-inflammatory and Analgesic Effects of Gold injection Aqua-acupuncture (금주사액약침자극(金注射液藥鍼刺戟)의 항염증(抗炎症) 및 진통(鎭痛)에 관한 실험적(實驗的) 연구(硏究))

  • Objectives : This study was purposed to investigate the Anti-inflammatory and Analgesic Effects of Gold injection Aqua-acupuncture on the experimental model of rheumatoid arthritis. Methods : The experimental groups were divided into 4 groups : Control group (group injected with normal saline), J-NS (group injected with normal saline into bilateral Choksamni(ST36)), J-GS (group injected with Gold Injection into bilateral Choksamni(ST36)), and N-GS (injected with Gold Injection into the blank locus of the root of mouse tail). In addition, Diclofenac-Na as a comparative medicine is injected into bilateral Choksamni(ST36) and the blank locus of the root of mouse tail. So we measured the mice paw edema induced by Carrageenin and Dextran, the chronic rat paw edema induced by adjuvant, vascular permeability induced by Acetic acid in mice, the writhing syndrome induced by Acetic acid in mice, the heat-induced pain threshold in mice. Results : The following result have been obtained. 1. The mice paw edema induced by Carrageenin was significantly decreased in J-GS as compared with the control group. 2. The mice paw edema induced by Dextran was significantly decreased in J-GS and N-GS as compared with the control group. 3. The chronic rat paw edema induced by Adjuvant was significantly decreased in J-GS and N-GS as compared with the control group. Serum Iron content was significantly decreased in J-GS and N-GS as compared with the control group. But the effect on the Serum Copper contents has no significance statistically. 4. Vascular permeability induced by Acetic acid in mice was significantly decreased in J-GS and N-GS as compared with the control group. 5. The level of Acetic acid-induced Writhing syndrome and Heat-induced Pain Threshold in mice were all significantly decreased in J-GS and N-GS as compared with the control group. Conclusion : According to the result, gold injection aqua-acupuncture has significant anti-inflammatory and analgesic effects on the experimental model of rheumatiod arthiritis.

Paw Edema was Reduced in Carrageenan Induced Acute Inflammation in Stat4 Deficient Mice

  • Signal transducer and activator of transcription 4 (STAT4) is one of the important mediators in generating inflammation and immune responses. To address the role of Stat4 in carrageenan induced acute inflammation, we performed paw edema measurement and 7.4 k mouse cDNA microarray analysis in carrageenan induced acute inflammation in Stat4 knockout (-/-) mice. Male BALB/c (n=8) and Stat4 -/- (n=5) were used and paw edema was induced with injection of $30\;{\mu}L$ of 1% carrageenan into plantar surface of right hind paw. Next, we isolated the mRNA in mouse whole brain and analyzed cDNA microarray profiles for the changes of the brain expression in Stat4 -/- mice. Interestingly, the increase in paw volume of Stat4 -/- mice was reduced by about 30% as compared to that of wild type. The cDNA microarray analysis revealed the altered expressions of several cytokines (Tnf, Il6, and Il4) and pain-associated proteins (Ptgs2, Gabra6, and Gabbr1) in Stat4 -/- mice. Our results suggest that Stat4 may be related to the inhibitory responses on carrageenan induced acute inflammation.

Anti-Inflammatory Effects of 15Hz Electroacupuncture on the Carrageenan-Injected Rat (Carrageenan 유발염증에 대한 15Hz 전침의 효과에 대한 연구)

  • Objective : The aim of this study is to investigate the anti-inflammatory effects of 15Hz electroacupuncture(EA) on carrageenan-injected rats. Inflammation was induced by an intraplantar injection of 1% carrageenan into the right hind paw. Methods : Bilateral EA stimulation with 15 Hz were delivered at those acupoints corresponding to Zusanli and Sanyinjiao in man via the needles for a total of 30 min duration in carrageenan-injected rats. Results: The developing edema was measured 30 minutes interval afer carrageenan injection and 15 Hz EA stimulation presented significant edema inhibition. Three hours after carrageenan injection, prostaglandin $E_2(PGE_2)$ and nitric oxide(NO) levels were measured. The 15Hz EA stimulation significantly inhibited $PGE_2$ and NO production in the right paw. The pro-inflammatory mRNA expression such as cyclooxygenases(COX)-2 and interleukin(IL)-$1{\beta}$ were slightly down-regulated by EA stimulation. The number of COX-2, IL-$1{\beta}$, tumor necrosis factor-${\alpha}$ immunoreactive cells were abundantly observed in paw edema. But these cells were decreased in nmber according to anti-edema effect of 15Hz EA. Conclusions: These results indicate that 15Hz EA stimulation have an alleviation action against carrageenan-induced edema and local inflammation.

Anti-inflammatory Activity of the Ethanol Extract from Magnoliae Flos on PAR2-mediated Edema (신이 에탄올 추출물의 PAR2-유발 부종에 대한 항염증 활성)

  • The flower of Magnolia denudata Desrousseaux(Magnoliaceae) has long been used for treatment of nasal disorder in Korea. The physiological activity of the Magnoliae Flos ethanol extract (MFX) was investigated. MFX showed antimicrobial activity. At doses of 100 and 200 mg/kg, MFX showed significant inhibition on both change in paw volume and vascular permeability. MFX (100 mg/kg) significantly inhibited PAR2 agonist-induced myeloperoxidase (MPO) activity in paw tissue. These results indicate that MFX has anti-inflammatory activity in PAR2-mediated paw edema.

Multiple 5-Hydroxytryptamine(5-HT) Receptors Are Involved in the Melittin-induced Nociceptive Responses in Rat I. Role of Peripheral 5-HT Receptor

  • Melittin-induced tonic pain model is characterized by local inflammation, edema, spontaneous flinchings, and sustained mechanical hypersensitivity. These nociceptive responses are mediated through selective activation of capsaicin-sensitive primary afferent fibers by melittin. The present study was undertaken to elucidate the role of peripheral 5-hydroxytryptamine(5-HT) receptors in the melittin-induced nociceptive responses. Changes in mechanical threshold, flinching behaviors and paw thickness were measured in rat intraplantarly injected with melittin($40{\mu}g/paw$) alone or treated together with melittin and 5-HT receptor antagonists. WAY-100635($100{\mu}g\;&\;200{\mu}g/paw$), isamoltane hemifumarate($100{\mu}g\;&\;200{\mu}g/paw$), methysergide maleate($60{\mu}g,\;120{\mu}g\;&\;200{\mu}g/paw$) and ICS-205,930($100{\mu}g\;&\;200{\mu}g/paw$) were intraplantarly injected 20 min before melittin injection. All 5-HT receptor antagonists tested in this experiment significantly attenuated the ability of melittin to reduce mechanical threshold and to induce flinching behaviors. 5-HT receptor antagonists, except ICS-205,930, had mild inhibitory effect on melittin-induced edema. These experimental findings suggest that multiple peripheral 5-HT receptors are involved in the melittin-induced nociceptive responses.

Experimental Studies on the Analgesic and Antiphlogistic Effects of Violae Herba (자화지정(紫花地丁)의 진통(鎭痛) 및 소염작용(消炎作用)에 관(關)한 실험적(實驗的) 연구(硏究))

  • Experimental studies were done to investigate analgesic and antiphlogistic actions of Violae Herba. Its water extract showed a significant dose dependent analgesic effect against writhing syndrome induced by acetic acid as well as in hot plate test in mice. The extract also showed a significant inhibitory effect against paw-edema induced by egg-white in rats.
