노랑가지 물추출물에 의한 생쥐 발바닥 부종의 억제효과 (Inhibitory Effect of Mouse Paw Edema by Solanum melongena Aqueous Extract)

  • Solanum melongena L. (Solanaceae) has anti-oxidant, analgesic, and hypolipidemic effects. We previously showed that Solanum melongena (SM) aqueous extract inhibits mast cell-mediated allergic reactions. The activation of protease-activated receptor-2 (PAR-2) induces acute inflammation in rat hindpaw. In the present study, we investigated the effects of the SM aqueous extract on mouse paw edema induced by PAR2 agonists. Trypsin or trans-cinnamoyl-LIGRLO-NH$_2$ (tc-NH$_2$), PAR-2 agonists, was injected into the hind paw of mice to induce paw edema. SM aqueous extract (1, 5, 10, and 100 mg/kg) was orally administered 1 hr before induction of paw edema. SM aqueous extract (5, 10, and 100 mg/kg) significantly inhibited both paw edema and vascular permeability in the dose-dependent manner. Furthermore, SM aqueous extract (10 mg/kg) significantly inhibited PAR-2 agonist-induced myeloperoxidase (MPO) activity and tumor necrosis factor (TNF)-$\alpha$ expression in paw tissue compared to that of saline. These results suggest that SM aqueous extract may be useful for treatment of PAR-2-mediated inflammation.

Carrageenin으로 흰쥐 발 염증으로 Indomethacin에 의한 유도성 nitric oxide synthase의 발현증가 (Potentiation of Inducible Nitric Oxide Expression by Indomethacin in Carageenin-treated Rat Paw Inflammation)

  • Present study was aimed to examine whether indomethacin affected the production of NO in the rat paw exudate by carrageenin. Paw edema and nitrite/nitrate levels in the paw exudate were maximal after 4 h and remained elevated up to 10 h, whereas indomethacin (10 mg/kg, po) significantly inhibited the carrageenin-induced paw edema and levels of nitrate in the paw exudate. However, paw edema and nitrite/nitrite levels were increased thereafter for 10 h. Indomethacin also enhanced the expression of iNOS mRNA and protein 4 h after carrageenin infection. Indomethacin inhibited the level of $PGE_2$ in the paw exudate in a time-dependent manner. These results suggest the possibility that indomethacin may potentiate expression of iNOS and subsequently increase nitrite/nitrate level in the late phase of carrageenin-induced rat paw inflammation possibly by suppressing cycloxygenase activity.

배농산이 프로테이나아제 활성수용체-2에 의한 흰쥐 발바닥 부종에 미치는 항염효과 (Anti-inflammatory Effect of Baenong-san in Proteinase-activated Receptor-2-mediated Paw Edema)

  • The Baenong-san has long been used for treatment of inflammatory in Korea. In this study, the anti-inflammatory effects of Baenong-san water extract (BWX) were investigated in proteinase-activated receptor-2 (PAR2)-mediated rat paw edema. Paw edema was induced by injection of trypsin or trans-cinnamoyl-LIGRLO-NH₂ (to-NH₂) into hindpaw of rats. BWX (10, 50, 100 and 200mg/kg) was orally administered 1 h before induction of inflammation. At doses of 50, 100 and 200 mg/kg, BWX showed significant inhibition of both change in paw volume and vascular permeability. BWX(100mg/kg) significantly also inhibited PAR2 agonist-induced myeloperoxidase (MPO) activity in paw tissue. This study demonstrated that BWX has an anti-inflammatory action for PAR2-mediated paw edema.

율초(葎草) 추출물이 Rat의 paw edema에 미치는 영향 (Anti-inflammaory effects of the MeOH extract of Humulus japonicus in vitro)

  • Objectives : The present study was examined to evaluate the anti-inflammatory effects of the Humulus japonicus MeOH extracts (HJE) in the carrageenan-induced paw edema model in rats. Methods : The effects of HJE on anti-inflammation were measured in the carrageenan-induced paw edema model in rats and infiltrated Inflammatory Cells. Results : 1. HJE (1.0 g/kg) and dexamethasone effectively inhibited paw edema measured 1~4 h after carrageenan injection. HJE (0.3 g/kg) effectively inhibited paw edema measured 1, 3, 4 hr. 2. In histopathological study in rats, 1.0 g/kg HJE and dexamethasone effectively inhibited the increases of hind paw skin thicknesses and inflammatory cell infiltrations induced by carrageenan treatment. But quite similar histopathological changes were detected in 0.3 g/kg HJE treated group as compared with carrageenan control.

  • Potentilla chinensis Ser. (Rosaceae) has long been used for a remedy of diarrhea and inflammation in Korea. In this study, the anti-inflammatory effects of the Potentillae chinensis Herba water extract (PCX) was investigated in proteinase-activated receptor-2 (PAR2)-mediated rat paw edema. Paw edema was induced by injection of trypsin or trans-cinnamoyl-LIGRLO-$NH_2$ (tc-$NH_2$) into the hind paw of rats. PCX (10, 50, 100 and 200 mg/kg) was orally administered 1 h before the induction of inflammation. At doses of 50, 100 and 200 mg/kg, PCX showed significant inhibition on both change in paw volume and vascular permeability. PCX (100 mg/kg) significantly inhibited PAR2 agonists-induced myeloperoxidase (MPO) activity in paw tissue. 흰쥐에서 carrageenan에 의한 족부종(足浮腫)에 대한 nifedipine의 영향 (Effect of nifedipine against carrageenan-induced paw oedema in rats)

  • The effects of nifedipne and verapamil were compared on carrageenan-induced paw edema in rats and the results are summarized as follows: Carrageenan induced severe acute paw edema within 30 minutes and the maximum effect was achieved around 4 hours after administration. The carrageenan-induced paw edema was prominantly reduced by pretreatments of indomethacin (2mg/kg, p.o.) and nifedipine (10 and 20mg/kg, i.p.), whereas verapamil had no effect on the carrageenan-induced paw edema. These results suggest that calcium antagonists, nifedipine and verapamil, have a different effect on the inflammatory response induced by carrageenan.

Analgesic and anti-inflammatory effect of the aqueous extract of Angelica dahurica

  • Background: Angelica dahurica has been used in various clinical cases. Its taste is hot and its property is warm, dry and nonpoisonous. Its efficacy is to remove wind-damp, cure swelling and edema, exhaust pus, stop itching, rhinitis and leukorrhea. Object: To test through experiment Angelica dahurica's analgesic and anti-inflammatory efficacy. Method: Inject acetic acid as a pain-inducing substance to the mice and measure visceral pain bywrithing reflex. Inject carrageenan that is an edema-inducing substance to the rat's paw and measure volume of edema. Take thermal pain to mice with plantar test and measure paw withdrawal latency. Normal group is non Angelica dahurica-treated group and treated group is Angelica dahurica-treated group. Results: In acetic acid-induced visceral model, treatment with Angelica dahurica suppressed writhing reflex significantlyand dose-dependently. In carrageenan-induced paw edema model, treatment with Angelica dahurica suppressed carrageenan-induced paw edema. In plantar test model, no significant effect on the withdrawal latency of thermal stimulation-induced nociception was observed. Conclusion: Angelica dahurica has analgesic and anti-inflammatory efficacy.

The protective mechanism of melatonin on carrageenan-induced paw edema generation in rats

  • The influence of melatonin on carrageenan-induced paw edema in Sprague-Dawley rats has been studied. The injection of 1% carrageenan (given at 0.1ml/paw) into the intraplantar induced an inflammatory response, and the maximal increase of paw volume, edema, was observed at 4hour. The levels of nitric oxide (NO), malondialdehyde (MDA) or prostaglandin E$_2$ (PGE$_2$) were increased after edema generation. Also, the expression of the inducible NO synthase (iNOS) were increased in the western blot. (omitted)

  • 패장 (Patrinia villosa Jussieu, Valerianaceae)의 뿌리는 한국에서 오래전부터 감염성 질환의 치료에 사용되어왔다. 본 논문에서는 패장근의 물 추출물 엑스 (PVWX)가 proteinase 활성수용체-2 (PAR2)에 의하여 유발된 흰쥐 발바닥 부종에 대한 항염증효과를 연구하였다. 발바닥 부종은 trypsin이나 $trans-cinnamoyl-LIGRLO-NH_2\;(tc-NH_2)$를 쥐의 발바닥에 주사하여 유발시켰다. PVWX (10, 50, 100 and 200 mg/kg)는 부종유발 1시간 전에 경구로 투여하였다 50, 100 및 200 mg/kg의 PVWX 투여시 부종의 부피와 혈관투과성의 변화에 유의성 있는 억제를 나타냈다. PVWX (100 mg/kg)은 발바닥 조직에서 PAR2 작동약으로 유발된 myeloperoxidase (MPO)활성을 유의성 있게 억제하였다. 이러한 결과들은 PVWX가 PAR2로 유발된 쥐의 발바닥 부종에서 항염증효과가 있음을 보여준다.

평위산 전탕액의 보관 온도 및 기간에 따른 항염증 효능 비교 (Changes in Anti-inflammatory Effect of Pyungwi-san Decoction According to the Preservation Temperature and Period)

  • Objectives : To provide the information of preservation method for herbal decoction, we evaluated the anti-inflammatory effects according to preservation temperature and period of Pyungwi-san (PWS, Ping wei-san) decoction. Methods : The anti-inflammatory activity of PWS was investigated by carrageenin-induced paw edema in SD rats. At 0 month, PWS extract was administrated 100, 300 and 1000 mg/kg/day orally for seven days prior to induction of edema. Edema was induced by subcutaneous injection of 1% carrageenin into the right hind paw. The paw volume was measured at 4 hr following carrageenin-induced paw edema in rats. At 6 and 12 months, PWS extracts according to preservation temperatures (room temperature, $4^{\circ}C$ and $-20^{\circ}C$) were administrated in rats and then carrageenin-induced paw edema volume was measured. Results : PWS showed inhibitory effect on carrageenin-induced paw edema in rats and the optimal dose was 1000 mg/kg/day at 0 month of storage. At 6 month of storage, PWS storaged at $4^{\circ}C$ and $-20^{\circ}C$ were inhibited paw edema but that storaged at room temperature was not reduced paw edema in rats. Conclusions : These results suggest that PWS decoction pouch has anti-inflammatory activities at both $4^{\circ}C$ and $-20^{\circ}C$ during 6 month storage.
