  • Essential oils are mixture of volatile, lipophilic compounds originating from plants. Essential oils have potential biological effects, i.e., antibacterial, antifungal, spasmolytic and antiplasmodial activities and insect-repellent property. In this study, five essential oils, namely R, LG, FR, O, and NM, extracted from various aromatic plants were used to test their antimicrobial activity against the oral microorganisms. The effects of essential oils were investigated against eight important bacteria, Streptococcus mutans (S. mutans), Staphylococcus aureus (S. aureus), Streptococcus sanguis (S. sanguis), Streptococcus anginosus (S. anginosus), Actinobacillus actinomycetemcomitans (A. actinomycetemcomitans), Streptococcus sobrinus (S. sobrinus), Staphylococcus epidermidis (S. epidermidis), and Escherichia coli (E. coli). Essential oils, except NM, effectively inhibited the growth of tested oral pathogenic microorganisms dose-dependently. However, the essential oils didn't show a significant inhibitory effect against E. coli and S. epidermidis. Consequently, these results represented that essential oil-mediated anti-microbial activity was prominent against the oral pathogenic bacteria. For example, minimum bactericidal concentration(MBC) of R, LG, FR oil against A. actinomycetemcomitans was very low as 0.078 mg/mL. In addition, minimum inhibitory concentration (MIC) of R, LG, FR, O oil against S. mutans was low as 0.156 mg/mL in vitro.

  • The purpose of this study was to establish the critical limit of CCP (Critical Control Point) of a HACCP (Hazard Analysis Critical Control Point) system used in the production of seasoned laver products. The hazard analysis examined microbial evaluations and developed a HACCP management plan for the heating process. The results were determined to be capable of reducing the biological element of CCP via the secondary roasting process. This study examined general bacteria and pathogenic microorganisms such as Escherichia coli, Staphylococcus aureus, Salmonella spp., Listeria monocytogenes, Vibrio parahaemolyticus, and Bacillus cereus at temperatures ranging from $170^{\circ}C$ to $230^{\circ}C$ and for 3.0 to 5.5 seconds at a time. Before the secondary roasting process, pathogenic microorganisms were all negative, although the presence of general bacteria was still detected. General bacteria was reduced to $1.0{\times}10CFU/g$ after the temperature was set at $230^{\circ}C$ for a period of 5.5 seconds. In conclusion, it suggested that a HACCP plan was necessary for management standard and systematic approach in the establishment of critical limit, problem resolution, verification method, education, and records management through a secondary roasting process.

  • Studies of immune responses in insects have focused on mechanisms that interact directly with invading microorganisms. However, few studies have examined the immune response to various metabolites produced by microorganisms after they enter the host. Here, we examined immune responses in Protaetia brevitarsis seulensis larvae induced by metabolites produced by symbiotic and pathogenic bacteria. The two types of bacteria were cultured under the same conditions. The bacteria were then removed and the remaining culture supernatant was injected into the larvae. The larvae injected with culture medium (Ch-medium) from symbiotic bacteria remained relatively healthy and did not develop an immune response, whereas more than 60% of the larvae injected with pathogen culture medium (Ec-medium) died after 150 hours and dark brown patches of melanin were observed at the injection site. This immune response was confirmed by the finding of activated lysosomes in insect granulocytes. More than 50% of lysosomes in larvae injected with pathogen culture medium were strongly stained after 12 h, but less than 5% of those injected with symbiotic culture media were stained. Therefore, it is assumed that symbiotic bacteria produce few (if any) substances that induce host immune responses.

  • According to the microbiological standard, Staphylococcus aureus, Clostridium perfringens and Listeria monocytogenes should not be detected in milk and egg products in Korea. Refrigerated food such as milk must be kept under $10^{\circ}C$ at retail markets. However, temperature abuse of refrigerated foods at such markets is often observed. We compared the growth and survival kinetics of S. aureus and C. perfringens at 10 and $15^{\circ}C$, and the growth kinetics of L. monocytogenes at 4 and $10^{\circ}C$ in whole and skim milk and ready-to-eat (RTE) quail eggs to evaluate their growth possibilities at retail markets. Regardless of storage temperature, the level of S. aureus reached the maximum level ($10^8-10^9CFU/ml$) in whole milk, non-fat milk and RTE quail eggs within the expiration date. Even low contamination levels of S. aureus (10 CFU/mL) grew rapidly in milk and quail eggs to reach the maximum level within the shelf life. Survival of C. perfringens in whole milk was greater than that in non-fat milk, indicating that the fat content in milk influences the survival of C. perfringens. For L. monocytogenes, the population in milk increased by 0.5-1 log CFU/mL at $4^{\circ}C$, while the populations reached the maximum level at $10^{\circ}C$ within the expiration date, regardless of initial contamination levels. In quail eggs, L. monocytogenes grew to the maximum level within the expiration date (60 days) at both temperatures. S. aureus and L. monocytogenes must be controlled to be negative, and proper temperature management should be emphasized at retail markets to protect the consumer. Since C. perfringens did not grow in milk and RTE quail eggs, there is no risk due to the growth of C. perfringens in these products at retail markets.

  • Groundwater is widely used as drinking water supplies around the world. However, microbial contamination of groundwater is a serious environmental problem that degrades drinking water quality and poses a great threat to human health. Among the pathogenic microorganisms such as viruses, bacteria, and protozoa, viruses are not readily removed during transport through soils, having high mobility in groundwater environment due to their smaller size compared to bacteria and protozoa. Studies regarding the fate and transport of viruses in soils and aquifers are necessary to determine the vulnerability of groundwater to microbial contamination and to secure safe drinking water sources. Also, these studies provide important information to establish the regulations and policies related to public health. This review paper presented the field and laboratory studies conducted for the fate and transport of viruses in subsurface environments. Also, the paper provided the factors affecting the fate and transport of viruses, the characteristics of bacteriophages used for virus studies, and virus transport model/colloid filtration theory. Based on this review work, future researches should be performed actively to set up the viral protection zone for the protection of groundwater from viral contamination sources. Especially, the researches should be focused on the development of mathematical models to calculate the setback distance and travel time for the viral protection zone along with the accumulation of information related to the model parameters.

  • Prunus mume has been used for the folk medicine by many old civilizations to treat food-borne diseases or enteric disorders. This study was carried out to examine the antimicrobial effect of juice from Prunus mume against pathogens such as Escherichia coli, Salmonella enteritidis, Staphylococcus aureus, Listeria monocytogenes and Bacillus cereus. The juice of Prunus mume had the strongest antimicrobial activity to Sal. enteritidis. The concentrations of Prunus mume juice for the formation of clear zone were 1% for Sai. enteritidis(15.0mm), 3% for Lis. monocytogenes(14.7mm), and 5% for Bac. cereus(14.75mm), Esc. coli(13.45mm) and Sta. aureux(11.9mm). The growth of all tested microorganisms was inhibited apparently in tryptic soy broth containing 3% and 4% Prunus mume juice. And it was found that the Prunus mume juice showed the highest antimicrobial properties, followed by Sal. enteritidis, Bac. cereus, Sta. aureus, Lis. monocytogenes, Esc. coli.

  • The effects of gamma irradiation (1, 3 and 5 kGy) and packaging methods (air and vacuum) on the growth of microorganisms contaminated in beef was investigated during storage at different temperatures (-20, 4 and $25^{\circ}C$). The initial microbial population of beef was $8.0{\sim}10^2\;CFU/g$ in total aerobic bacteria, $2.0{\times}10^2\;CFU/g$ in total lactic acid bacteria, $8.0{\times}10^1\;CFU/g$ in molds, $6.0{\times}10^2\;CFU/g$ in Pseudomonas sp. and $7.0{\times}10^2\;CFU/g$ in coliforms, respectively. Gamma irradiation at 5 kGy completely eliminated pathogenic bacteria in beef. Gamma irradiation at such dose and subsequent storage at less than $4^{\circ}C$ could ensure hygienic quality prolong the microbiological shelf-life resulting from the reduction of spoilage microorganisms. The different packaging methods of beef caused negligible changes in the growth of microorganisms during storage.

  • Detection of pathogenic microorganisms takes several days by conventional methods. It is necessary to assess microorganisms in a timely manner to reduce the risk of spreading infection. For this purpose, bacteriophages are chosen for use as a biosensing tool due to their host specificity, wide abundance, and safety. However, their lytic cycle limits their efficacy as biosensors. Phage proteins involved in binding to bacteria could be a robust alternative in resolving this drawback. Here, a fragment of tail protein J (residues 784 to 1,132) of phage lambda fused with 6X His-tag (6HN-J) at its N-terminus was cloned, overexpressed, purified, and characterized for its binding with microorganisms. The purified protein demonstrated a size of about 38 kDa in sodium dodecyl sulfate - polyacrylamide gel electrophoresis (SDS-PAGE) and bound with anti-His monoclonal antibodies. It bound specifically to Escherichia coli K-12, and not Salmonella typhimurium, Bacillus subtilis, or Pseudomonas aeruginosa in dot blotting. Binding of the protein to E. coli K-12 inhibited about 50% of the in vivo adsorption of the phage lambda to host cells at a concentration of $1{\mu}g/ml$ 6HN-J protein and almost 100% at $25{\mu}g/ml$ 6HN-J. The results suggest that a fusion viral protein could be utilized as a biosensing element (e.g., protein chips) for detecting microorganisms in real time.

  • Disposable contact lenses, which are one type of soft contact lenses, provide convenience in use, but also cause various ocular infectious diseases. Microorganisms that cause eye diseases include Acanthamoeba, bacteria, Fungi, and so on. It is impossible to prevent microorganism contamination completely due to the use of hands as wearing contact lenses. The contamination by various microorganisms leads to infectious keratitis, but it is not well known for the exact microorganisms that affect the disease. For this reason, to identify the microorganisms, two groups that are commonly used for disinfection of lenses were divided: normal saline solution and multiple purpose solution. Using these solutions the degree of microorganism contamination was observed according to the days of 1, 3, 5, 10, and 15. Twenty students by two groups from Ophthalmic Optics department at D college in Daegu Metropolitan city participated in the experiment after their ocular health conditions were checked. During they wore one-day disposable lenses for 1, 3, 5, 10, and 15 days, bacteria were cultured in media. The results, which were Gram stained by selecting the cultured colonies, show as followings: Gram positive cocci 33%, Gram-negative cocci 2%, Gram positive bacilli 34%, and the Gram negative bacilli 31%, respectively. As for the identification of potential pathogens, VITEK system and API kit methods were used. Keratitis caused by bacteria known as Staphylococcus aureus, Pseudomonas aeruginosa were detected as a result of wearing contact lenses. This study examined the distribution of bacteria as wearing one-day disposable contact lenses and pathogenic bacteria according to the duration of wearing them. In conclusion, the importance of hygiene when using contact lenses is suggested.

  • Nanoparticles have wide-scale applications in various areas, including medicine, chemistry, electronics, and energy generation. Several physical, biological, and chemical methods have been used for synthesis of silver nanoparticles. Green synthesis of silver nanoparticles using plants provide advantages over other methods as it is easy, efficient, and eco-friendly. Nanoparticles have been extensively studied as potential antimicrobials to target pathogenic and multidrug-resistant microorganisms. Their applications recently extended to development of antivirals to inhibit viral infections. In this study, we synthesized silver nanoparticles using Cinnamomum cassia (Cinnamon) and evaluated their activity against highly pathogenic avian influenza virus subtype H7N3. The synthesized nanoparticles were characterized using UVVis absorption spectroscopy, scanning electron microscopy, and Fourier transform infrared spectroscopy. Cinnamon bark extract and its nanoparticles were tested against H7N3 influenza A virus in Vero cells and the viability of cells was determined by tetrazolium dye (MTT) assay. The silver nanoparticles derived from Cinnamon extract enhanced the antiviral activity and were found to be effective in both treatments, when incubated with the virus prior to infection and introduced to cells after infection. In order to establish the safety profile, Cinnamon and its corresponding nanoparticles were tested for their cytotoxic effects in Vero cells. The tested concentrations of extract and nanoparticles (up to 500 μg/ml) were found non-toxic to Vero cells. The biosynthesized nanoparticles may, hence, be a promising approach to provide treatment against influenza virus infections.
