Hirudin is a potent inhibitor of thrombin, which was originally obtained from the medicinal leech (Hirudo medicinalis) Now it is being produced through the recombinant technology on a large scale. Recombinant hirudin has been assayed for the anticoagulant activity by the measurement of clotting time and the inhibition of thrombin actvity using a chromogenic substrate. The assay range of partial thromboplastin time and thrombin time is within $0.2{\sim}1.0 {\mu}g/mι.$ Thrombin time is more sensitive to the measurement of clot. Ex vivo study showed the level of hirudin in rat plasma was highest in 10 min and then it was eliminated slowly. The half-life of r-hirudin was 80~110 min depending on the assay methods. Intraveneous injection of russel viper venom was used for thrombus induction combined with vents cava ligation. Inhibition of venous thrombosis was observed with i.v. hirudin. It was dependent on the concentration of hirudin.
We investigated the effects of plant vinegar on platelets and blood coagulation system. Plant vinegar inhibited in vitro platelet aggregation in a concentration dependent manner, when platelets were activated by thrombin and collagen. In addition, plant vinegar showed inhibitory effects on the serotonin secretion induced by thrombin in a concentration dependent manner. However, treatment with plant vinegar to platelets did not induce any cytotoxicity, as determined by the release of lactate dehydrogenase. Plant vinegar did not change the coagulation parameters such as activated partial thromboplastin time (aPTT) and prothrombin time (PT) using rat citrated plasma. In vivo study revealed that, treatment with plant vinegar prolonged the bleeding time from mouse tail. All these results suggest that plant vinegar might improve blood hemostasis mediated via anti platelet activities.

An anticoagulant was purified from corn silk which has been used in Oriental Medicine. The anticoagulant from corn silk has a molecular mass of 135 kDa, and purified by 24 folds with a recovery of 11%. It was not sensitive to heat and protease treatment. However, periodate oxidation of the anticoagulant resulted in loss of activity significantly, implying that a carbohydrate was responsible for an anticoagulant activity. Galactose, glucose, mannose, fucose, glucosamine, and galactosamine were detected after acid hydrolysis by thin layer chromatography (TLC) and Bio-LC. It was confirmed that anticoagulant had OH and NH bonds by IR, supporting that the anticoagulant is composed of neutrosugar and aminosugar. Its anticoagulating activity was measured by delay in thrombin time (TT) and prothrombin time (PT) without affecting clotting by snake venom and delay in activated partial thromboplastin time (APTT). TT was more sensitive than PT, and was delayed two and three times at the concentration of 60 and 88 nM, respectively. The anticoagulating activity was reduced in the thrombin-induced clotting assay using purified fibrinogen according to the increase of fibrinogen concentration with the apparent Ki value of 23 nM.
Four-week toxicity of EPO(erythropoietin) was investigated using New Zealand White rabbits according to the established regulations of Korean National Institute of Safety Research. Rabbits were administered intravenously seven days per week for 28 days with dosage of 0, 80, 400 and 2000IU/kg B. W./day. Animals administered with EPO showed no significant changes of body weight, water consumption and feed consumption, and no clinical signs and death. They were not significantly different from the control group in hematological and serum biochemical analysis, urinalysis, prothrombin time, and partial thromboplastin time. In this study, we concluded that EPO had no toxic effect in the New Zealand White rabbits when they were administered intravenously below 2000IU/ kg B.W./ day for 28 days.

Antithrombotic and fibrinolytic activity of natto was evaluated on platelet aggregation in vitro and in vivo. Natto showed inhibitory effects on platelet aggregation induced by adenosine 5’diphosphate (ADP) and collagen. Orally administered natto also showed fibrinolytic activity in hypercholesterolemia rats. Normal levels of natto, when administered for four weeks, shortened euglobulin clot lysis time (ECLT) and prolonged partial thromboplastin time (PATT) significantly compared to non-treated group. In addition, the natto treatment decreased total cholesterol in serum. These results showed that intake of normal levels of natto can elicit antithrombotic and fibrinolytic effects, suggesting its consumption may improve blood circulation.
The in vitro anticoagulant and fibrinolytic activities of crude extract from insects which have been used as traditional medicines. The extracts of Formica, Huechys and Eupolyph-aga/Steleophaga prolonged activated partial thromboplastin time and thrombin time compared to the value of the control. The fibrinolytic activity of insect extracts was also tested by fibrin plate method. We found that the extracts of Cicadae Periostracum, Eupolyphaga/Steleophdga, Mantidis $O{\ddot{o}}otheca$ and Huechys directly could hydrolyze the fibrin clot without the activation of plasminogen by plasminogen activators.

Synthetic coumarin and furocoumarin derivatives were evaluated for anticoagulant activity and the effect on liver and kidney function. It was found that all of the compounds under investigation proved to be neither toxic nor lethal up to 500 mg/100 g body weight as a single dose for 24 hrs. All tested compounds showed a significant increase in prothrombin time (PT) in the acute model but failed to show a significant action in the chronic model. Furthermore, all tested compounds revealed a significant increase in activated partial thromboplastin time (APTT) as compared to control value in both acute and chronic model. Also, all tested compounds did not cause any significant changes on liver and kidney functions in rats.
Curcumin, a polyphenol responsible for the yellow color of the curry spice turmeric, possesses antiinflammatory, antiproliferative and antiangiogenic activities. However, anticoagulant activities of curcumin have not been studied. Here, the anticoagulant properties of curcumin and its derivative (bisdemethoxycurcumin, BDMC) were determined by monitoring activated partial thromboplastin time (aPTT), prothrombin time (PT) as well as cell-based thrombin and activated factor X (FXa) generation activities. Data showed that curcumin and BDMC prolonged aPTT and PT significantly and inhibited thrombin and FXa activities. They inhibited the generation of thrombin or FXa. In accordance with these anticoagulant activities, curcumin and BDMC showed anticoagulant effect in vivo. Surprisingly, these anticoagulant effects of curcumin were better than those of BDMC indicating that methoxy group in curcumin positively regulated anticoagulant function of curcumin. Therefore, these results suggest that curcumin and BDMC possess antithrombotic activities and daily consumption of the curry spice turmeric might help maintain anticoagulant status.
Thrombogenesis and atherosclerosis are mainly caused by platelet aggregation, blood coagulation, and hyperlipidemia. Platelet aggrelation, activated platelet thromboplastin time (APTT) were measured as indexes of blood coagulation and lipid contents in the subjects who have taken red ginseng products (e.g. water extract, tea, drink etc.) for 4 to 5 years. The platelet aggregation in the red ginseng-taking group was significantly decreased, as compared with the non-red ginseng-intaking group, when platelets were stimulated by 100 $\mu\textrm{g}$/ml of collagen (P<0.01). The atherogenic index and the ratio of triglyceride to HDL-cholesterol in blood, the risk factors of atherosclerosis, were decreased in the subjects of ginseng group, compared with that in control group. APTT was also prolonged to greater extent in ginseng group than in control group. These results suggest that long-term intake of ginseng products may help to prevent the risks of thrombogenesis and atherosclerosis.
The 26 patients were randomly selected among patients who underwent valve replacement from February 1992 through September 1993 at National Medical Center. They were divided into two groups, one control group[n=13], the other aprotinin group[n=13]. The aprotinin group recieved 15000 KIU/kg aprotinin in the CPB priming volume and 35000 KIU/kg aprotinin intravenously during operation. We checked preoperative and postoperative hemoglobin, platelet, prothrombin time, activated partial thromboplastin time, creatinine phosphokinase and lactic dehydrogenase. There was no difference in the patient`s above clinical parameters between both groups. The platelet count in both groups decreaced after operation. These findings demonstrated that the effectiveness of aprotinin was not associated with platelet number but probably associated with a protection of platelet function and a prevention of hyperfibrinolysis. The intraoperative and postoperative blood loss and requirement of blood were significantly reduced in aprotinin group.
