Antibaterial Activity and Skin Moisturizing Effect of Cedrela sinensis A. Juss Shoots Extracts (참죽나무 새순 추출물의 항균 활성과 피부 보습효과에 관한 연구)

  • In this study, we investigated the antibacterial activity and the skin moisturizing effect of cream contaning Cedrela sinensis A. Juss shoots extracts. MIC values of ethyl acetate fraction from Cedrela sinensis A. Juss shoots extracts on P. acnes and S. aureus were 0.13 %, 0.03% respectively and P. ovale and E. coli were 0.25 % equally. The results showed that the antibacterial activity of the ethyl acetate fraction on P. acnes and S. aureus were higher than the methyl paraben. The cream containing the ethyl acetate fraction from Cedrela sinensis A. Juss shoots extracts was formulated and its skin moisturizing effect was evaluated in a clinical trial. The skin hydration and transepidermal water loss (TEWL) were investigated after topical application of the cream. The skin hydrating effect of the cream containing Cedrela sinensis A. Juss shoots extract was 10 ~ 15 % higher than the placebo cream, and transepidermal water loss was decreased to 6.7 g/$m^2h$ in the subjects applied with cream containing the ethyl acetate fraction of Cedrela sinensis A. Juss shoots extracts. These results indicate that ethyl acetate fraction from Cedrela sinensis A. Juss shoots extracts could be applicable to new functional antibacterial cosmetics.

  • Increasing the oil volume in the oil : water mixtures using isopropyl myristate, oleic acid as oils, the solubilities of methyl, propyl and butyl parabens and the MICs (minimum hibitory concentrations) to E. coli were increased. But in liquid paraffin, the solubility was reversed. The relation between MIC and solubility of parabens had constant factor of 0.7 as saturation fraction percent. The solubility of methyl paraben was crossed over in those of propyl and butyl parabens, when the oil volume was increased. When the oil was used over the cross-over point, methyl paraven was useful to preserve the systems, and below the cross-over point the more lipophilic propyl and/or butyl parabens were effective.

Chemical Sensors Using Polymer/Graphene Composite and The Effect of Graphene Content on Sensor Behavior (고분자/그래핀 복합재료의 센서 응용 및 그래핀 함량이 센서 거동에 미치는 영향)

  • In this study, a polymer/graphene hybrid composite was prepared by a simple roll-method and a simple sensor was produced by a convenient surface engineering procedure. The sensor performance was examined and the effect of graphene content on the sensing behavior was monitored. A polymer (polydimethylsiloxane, PDMS) paste containing graphene powder was prepared by a three-roll apparatus and polymer/graphene hybrid composite was produced by a two-roll technique. The sensing medium, cyclodextrin (CD) was introduced by a convenient bio-conjugation method. The efficacy of surface modification was confirmed by FT-IR spectroscopy and the ohmic relation was observed on composite surfaces. An analyte (e.g., methyl paraben, MePRB) at a 10 nM concnetration could be detected. When the graphene loading was low, the sensor performance was relatively poor. This was attributed to the absence of graphene alignments, which were observed for the composites having a high graphene loading. This indicates that the sensor performance was influenced by physical alignments of the filler. This article can provide important information for future research on developing sensing devices.

Development of a Quantitative Analytical Method for Determining the Concentration of Human Urinary Paraben by LC-MS/MS

  • Parabens, the esters of p-hydroxybenzoic acid, have been widely used as antimicrobial preservatives in cosmetic products, drugs, and processed foods and beverages. However, some parabens have been shown to have weak estrogenic effects through in vivo and in vitro studies. Because such widespread use has raised concerns about the potential human health risks associated with exposure to parabens, we developed a simultaneous analytical method to quantify 4 parabens (methyl, ethyl, propyl, and butyl) in human urine, by using solid-phase extraction and high-performance liquid chromatography coupled with triple quadrupole mass spectrometry. This method showed good specificity, linearity ($R^2$ > 0.999), accuracy (92.2-112.4%), precision (0.9-9.6%, CV), and recovery (95.7-102.0%). The LOQs for the 4 parabens were 1.0, 0.5, 0.2, and 0.5 ng/mL, respectively. This method could be used for quick and accurate analysis of a large number of human samples in epidemiological studies to assess the prevalence of human exposure to parabens.

Identification of Toxic Chemicals Using Polypyrrole-Cyclodextrin Hybrids (폴리피롤-사이클로덱스트린 혼성체를 이용한 유해화합물질의 검출)

  • Polypyrrole is a typical electrical conducting polymer, which has an excellent charge transport property. Cyclodextrins are a group of toxic-free and cyclic oligosaccharide molecules, capable of capturing low molecular weight chemicals. Considering these advantages, hybrid materials of polypyrrole and cyclodextrin can be used to detect hazardous compounds. Cyclodextrin molecules can accommodate toxic chemicals by the formation of host-guest complexes and generate electric signals, which are effectively delivered by polypyrrole backbone. In this study, the polypyrrole/cyclodextrin hybrid material was prepared using a facile wet method and included into a hydrogel. Subsequently, it was applied to a simple sensor system with a gold-patterned electrode for the detection of potentially hazardous material, methyl paraben. Compared with pristine polypyrrole, it was found that the polypyrrole/cyclodextrin hybrid showed an improved performance. This study can be an example of using environmentally benign conducting polymer/cyclodextrin hybrids as sensing media.

A Study on the Evaluation of Antimicrobial Effect of Orostachys Japonicus A. Berger Ethyl Acetate Fraction (와송 에틸아세테이트 분획물의 항균효능 평가에 관한 연구)

  • In this study, a study was conducted to utilize Orostachys japonica A. Berger EtOAc fraction extract as an antibacterial activity and cosmetic ingredient. As a result of measuring the antimicrobial activity of Orostachys japonica A. Berger EtOAc, the growth of S. aureus, S. epidermidis, and P. aeruginosa was inhibited. Among them, S. aureus was an extract of 18.35 ± 1.5 mm Orostachys japonica A. Berger EtOAc fraction at a concentration of 0.5 g / mL, showing superior antibacterial activity than methyl paraben (16.83 ± 1.0 mm), and was shown as a positive control. As a result of evaluating the MIC of the Orostachys japonica A. Berger EtOAc fraction extract through MIC measurement, the remaining strains excluding Candida. A showed a MIC of 17.5 mg/mL. As a result of evaluating the cosmetic preservation effect through the challenge test applied to the cosmetic emulsion formulation, the growth inhibitory effect of S. aureus in the emulsion containing 0.3% Orostachys japonica A. Berger EtOAc fraction extract 7 days after microbial inoculation was 100%.

Simultaneous Analysis Method for 27 Endocrine Disrupting Chemicals in Human Urine using UPLC-MS/MS (UPLC-MS/MS를 이용한 소변 시료 중 내분비계 교란물질 27종 동시분석법 확립)

  • Endocrine disrupting chemicals (EDCs) are compounds that come from outside the body and disrupt hormone action within the body's endocrine system. Examples include parabens, benzophenones, bisphenols, and phthalates, which are currently used in a wide range of applications. However, continuous exposure to them can have negative effects on glycemic control, reproduction, metabolism, nervous system development, pregnancy, childbirth, and growth. In this study, human samples (urine) were pretreated using liquid-liquid-extraction to determine the exposure level of EDCs and then analyzed effectively and rapidly by UPLC-MS/MS. In this way, the analytical conditions were established and the reliability of the simultaneous analysis method was evaluated through method validation. The results showed that the accuracy ranged from 75.28 to 122.36% and the precision ranged from 2.16 to 22.74%. The analytical method established in this study can be used as a methodology for future studies to evaluate and monitor the exposure of EDCs in human samples.

Uterotrophic and Hershberger Assay for Butyl ρ-Hydroxybenzoic Acid

  • Butyl $\rho$-hydroxybenzoic acid (butyl paraben, BP) is a homologous series of parabens and is widely used as a preservative in cosmetic and pharmaceutical products. The purpose of this study was to investigate the estrogenic/antiandrogenic activities of BP in animals. For that, we performed an uterotrophic assay and a Hershberger assay in rats. In uterotrophic assay, BP was administered subcutaneously to immature female SD rats (18 days old) for 3 consecutive days. The wet and dry uterus weights were significantly increased in the groups treated with BP in dose­dependent manner. In case of Hershberger assay, BP significantly reduced the weight of seminal vesicle of castrated rats. And other accessory organ/glands - prostate, Cowper's glands, bulbocavernosus muscle and glans penis were also slightly decreased. The results of this study suggested that BP showed estrogenic and anti-androgenic activities in vivo.

Simultaneous HPLC Determination of Preservatives in Cosmetics (화장품중 살균ㆍ보존제의 동시분리 및 정량)

  • A high-performance liquid chromatographic method for the simultaneous quantitative analysis of methylparaben (MP), ethylparaben (EP), propylparaben (PP), butylparaben (BP) and imidazolidinyl urea(IU) or diazolidinyl urea(DU) in cosmetics was studied by using a cyano-propyl column and 0.05M hexanesulfonic acid at 228 nm. Calibration curves were found to be linear in the 60-1000 $\mu\textrm{g}$/mL range (parabens), 100-1,250 $\mu\textrm{g}$/mL range (IU) and the 120-2000 $\mu\textrm{g}$/mL range (DU). Linear regression analysis of the data demonstrates the efficacy of the method in terms of precision and accuracy. An extraction method is developed and validated in order to apply this chromatographic method to a commercial cosmetic cream. The precision of this method, calculated as the relative standard deviation (RSD) of the recoveries (0.46-2.71%) was excellent for all compounds.

Biodegration of Formaldehyde-Releasing Preservatives

  • A strain of Pseudomonas aeruginosa was isolated from the spoiled product and its characteristics on various formaldehyde-releasing preservatives were investigated. This strain, P. aeruginosa FR, could utilize 1.0% of imidazolidinyl urea and 0.2% of DMDM hydantoin as a sole carbon and nitrogen source in the minimal salts medium. With the growth of the strain in minimal salts medium containing imidazolidinyl urea, formic acid was initially accumulated according to the decrease of formaldehyde concentration. It was suggested that formaldehyde dehydrogenase was involved in this oxidation process and could catalyze formaldehyde, imidazolidinyl urea, DMDM hydantoin and quaternium-15, but not bronopol. MICs of this strain to each preservation were 0.03% in formaldehyde, 1.0% in imidazolidinyl urea, 0.2% in DMDM hydantoin, 0.2% in quaternium-15 and 0.1% of EDTA-2Na. But the MICs were diminished about ten times when 0.01% of EDTA-2Na was added to the preservative systems. In actual challenge test, the eyeliner and the pack which contained paraben and imidazolidinyl urea were not able to be protected from this strain, but when 0.05% EDTA-2Na was added the products were sufficiently preserved.
