  • Pig chronic wasting disease, including porcine reproductive and respiratory syndrome (PRRS) and postweaning multisystemic wasting syndrome (PMWS), have made a continuous economic damage in pig farms. Airborne spread of livestock viruses are an important spread factor which is difficult to analyze due to invisible airflow and limitation of measurement. The objective of this study is to analyze airborne disease spread between buildings in the experimental pig farm by means of field experiment and computational fluid dynamics (CFD). The field experiments were conducted to capture airborne virus using air sampler and teflon filter along multi points in the experimental pig farm. The samples were tested in terms of virus detection resulting in positive reaction for PRRS and PCV-2 viruses, which can be a firm evidence of airborne virus spread. The CFD simulation model was developed by considering complex topography, wind conditions, building arrangement, and ventilation systems and was used to analyze airborne virus spread according to different wind conditions. The CFD computed result showed a possibility of airborne virus spread via livestock aerosol from infected pig house to neighboring pig houses according to wind directions. The CFD simulation technique is expected to provide significant data for estimating and making a counterplan against airborne disease spread.

Innate immune recognition of respiratory syncytial virus infection

  • Respiratory syncytial virus (RSV) is the leading cause of respiratory infection in infants and young children. Severe clinical manifestation of RSV infection is a bronchiolitis, which is common in infants under six months of age. Recently, RSV has been recognized as an important cause of respiratory infection in older populations with cardiovascular morbidity or immunocompromised patients. However, neither a vaccine nor an effective antiviral therapy is currently available. Moreover, the interaction between the host immune system and the RSV pathogen during an infection is not well understood. The innate immune system recognizes RSV through multiple mechanisms. The first innate immune RSV detectors are the pattern recognition receptors (PRRs), including toll-like receptors (TLRs), retinoic acid-inducible gene-I (RIG-I)-like receptors (RLRs), and nucleotide-biding oligomerization domain (NOD)-like receptors (NLRs). The following is a review of studies associated with various PRRs that are responsible for RSV virion recognition and subsequent induction of the antiviral immune response during RSV infection.

Serological and genetic characterization of the European strain of the porcine reproductive and respiratory syndrome virus isolated in Korea

  • Porcine reproductive and respiratory syndrome (PRRS) is an economically important disease of swine that occurs all over the swine industry worldwide. It was first observed in the Unite States in 1987 then in Europe in 1990. It has been described in Japan and in Korea in 1993. PRRS virus is divided into two distinct types, North American and European, genetically. Based on our limited knowledge there has been no report on the existence of European PRRSV. But according to the government's Korea Customs Service there has been many importations of breeding pigs from Europe. These seem to make an estimate that European PRRSV could be introduced in Korea by inflow of European breeding pigs. We first detected the European PRRSV could be introduced in Korean pig farms by using polymerase chain reaction (PCR). Further, it is also identified that there are not only North American PRRSV antibody but also a European PRRSV antibody. According to the genetical and serological experiment results, the presence of established North American PRRSV in Korea is due to the use of live vaccines made of North American PRRSV strain as well field virus infection, and the European PRRSV is possibly introduced from imported breeding stock.

전북지역 도축돈 적내장 육안병변 조사 (Survey on the red internal organs gross lesions of slaughtered pigs in Jeonbuk)

  • Respiratory diseases are common in swine industry and have great economic importance. Respiratory disorders cause substantial losses to the swine industry. Losses associated with respiratory disease vary considerably between herds and seasons. In a survey was carry out to investigate the lesion of red internal organs in slaughtered pigs and provided assistant data which are useful for each farm. From november 2012 to december 2013, 1,680 pigs out of 84 farms were sampled in Jeonbuk province. Gross lesions such as swine enzootic pneumonia (SEP), pleuritis, pleuropneumonia, pericarditis, liver milk spot were examined for the pigs. Overall prevalence of SEP was 55.7%. According to season, the incidence occurred higher in fall than winter, spring, and summer. The mean SEP score was 0.91, the highest incidence occurred in fall. The prevalence of pleuropneumonia, pleuritis, pericarditis, and milk spot was 36.4%, 49.7%, 2.3%, and 8.8%, respectively. The positive rate of PRRS and PCV2 was 2.9% and 70.0% by PCR analysis.

포유 및 이유자돈의 질병발생 동향 (Investigation of diseases incident to pre- and pos-weaning piglets)

  • This experiment was carried out to investigate the tendency of incident diseases in pre- and post- weaning piglets which ages were 1 to 7 weeks old by laboratory diagnosis and in order to minimize death in preweaning piglets and of stunted growth in postweaning piglets. The result of this experiment used as the basic data for the preventive programs in pre- and post- weaning piglets and were as follows: 23 different diseases diagnosed in 331 cases were studied in relation to age, season, and etiology. The most prevelent diseases of pre- and post- weaning piglet were Colibacillosis(79 case, 23.9%) and the major diseases were Salmonellosis(44 cases, 13.3%), Anemia(37 cases, 11.2%). Unknown viral disease(20 case, 6.1%), Rota viral infection(19 case, 5.8%), Porcine reproductive & respiratory syndrome(PRRS; 15 case 4.5%), Transmissible gastroenteritis(TGE; 12 case, 3.6%). The gastrointestinal disease, such as Colibacillosis, Salmonellosis, Swine dysentery, Clostridial infection, Rotaviral infection, TGE, Porcine epidemic diarrhea(PED) and Ballantidiosis occured pro- dominently in the period of pre- and post- weaning, which were 178 cases(53.8%) and not related to occurrence according to age and season. The respiratory diseases were Atrophic rhinitis(AR), Swine enzootic pneumonia, Pneumonic pasteurellosis, Pleuropneumonia, Branchopneumonia, PRRS and which were 48 cases(14.5%) and higher prevalent in spring and summer. The viral diseases was 73 cases(22.1%) that occurred in the period of 5 weeks piglet and prevalent mainly in spring. The bacterial diseases were 188 cases(56.8%) that were not related to occurrence according to age and season. Salmonellosis was prevalent in 3 to 5 weeks piglet and mainly occurred in summer. Viral septicemia and rotaviral infection occurred after 5 weeks piglets intensively and 3 to 5 weeks, respectively. And the both occurred without relation with season. PRRS occurred after 4 weeks piglet and prevalent in summer. TGE occurred 1 to 7 days old piglets and prevalent in spring and winter. Hematologic values of anemia was decrease in number of Red Blood Cell, concentration of Hemoglobin and Hematocrit. Amikacin, cephalothin, colistin, norfloxacin were effective to E coli, and amikacin, cephalothin, nortloxacin, neomycin were effective to Salmonellra spp. but clindamycin, erythromycin, penicillin, sulfonamides were resistant to E coli and Salmonella spp.

3D8 scFv 형질전환 돼지 개발 및 PRRS 저항성 평가 (Production of a transgenic pig expressing 3D8 single chain variable fragment (scFv) and its evaluation of PRRS resistant)

  • In this study, we have developed 3D8 scFv transgenic pig (TG) by microinjection of fertilized one-cell pig zygotes (2.17%). The effect of 3D8 scFv TG on porcine reproductive and respiratory syndrome virus (PRRSV) resistance were evaluated through PRRSV VR2332 (1×103 TCID50/mL) challenge and transmission experiments. As a result, the average daily weight gain (ADWG) of TG increased compared to the wild type pigs (WT) in PRRSV challenge groups and the serum viremia levels of the TG was significantly lower than of WT on the 7 day and 21 day after infection, meaning that the viral shedding was suppressed by 3D8 scFv expression. These results suggest that the expression of 3D8 scFv in pig could suppress spreading of infected virus to pigs sharing a room.

  • 축산물위해요소중점관리기준(HACCP)은 식품의 안전성에 영향을 미치는 위해발생을 사전에 방지하거나 제거하기 위하여 생산단계부터 제조, 가공 등 모든 단계를 과학적이고 체계적으로 사전 위생 관리하는 제도이다. 지난해 12월 축산물HACCP기준원으로부터 HACCP 농장으로 지정받은(주)금보육종(대표 장성훈) 원주 GP. 지난해 양돈협회에서 실시한 HACCP 교육을 통해 농림부 HACCP 인증 농장으로 다시 태어났다. 농장주와 임직원 모두가 HACCP 제도를 정확히 이해하고 이를 수행하는 한편, 국내 소모성 질환 극복을 위해서는 PRRS 청정화가 반드시 필요하다는 목표로 우수한 위생방역 시스템을 갖추고 있는 청정 강원도의 양돈전문기업 금보육종을 찾아보았다.

전북지역 도축 출하돈 병변 조사 (Survey on the gross lesions of slaughtered pigs in Jeonbuk area, Korea)

  • Respiratory disease in pigs is common in modern pork production worldwide and is often referred to as porcine respiratory disease complex (PRDC). PRDC is polymicrobial in nature, and results from infection with various combinations of primary and secondary respiratory pathogens. The control of swine respiratory disease requires an understanding of the interactions between the organisms that can cause this illness, the pig and management of the environment. This study was carried out to investigate the lesion of red internal organs in slaughtered pigs and provided assistant data for pig farms. A total of 900 lung samples, 45 farms were collected randomly from slaughtered pigs in Jeonbuk province from April to December in 2014. Gross lesions such as swine enzootic pneumonia (SEP), pleuritis, pleuropneumonia, pericarditis, liver white spots were examined for the pigs. Overall prevalence of SEP was 70.8%. According to season, the incidence occurred higher in summer than winter, fall and spring. The mean SEP score was 1.4, the highest incidence occurred in fall. The prevalence of pleuropneumonia, pleuritis, pericarditis, and milk spot was 26.1%, 71.4%, 2.8%, 21.6%, respectively. In the detection of pathogens, PRRS was not detected, PCV2 was positive in 87.6%.

경기도 북부지역 야생멧돼지(Sus scrofa) 전염성 질병 감염 실태 조사 (A survey for prevalence of infectious diseases in wild boar (Sus scrofa) in northern Gyeonggi province, South Korea)

  • This study was conducted to investigate the infection of livestock diseases using 500 blood samples from wild boars captured in six cities and one county in northern Gyeonggi province, South Korea. We examined 239 cases of classical swine fever virus (CSFV), and each of 500 cases of foot and mouth disease virus (FMDV), porcine reproductive and respiratory syndrome virus (PRRSV), porcine circovirus type 2 (PCV2), Mycoplasma hyopneumoniae (MH), Actinobacillus pleuropneumoniae (APP), Pasteurella multocida type A (PMA), Hemophilus parasuis (HP), Salmonella (Sal.) spp. infections. Antibodies were detected against CSFV (23.4%), PRRSV (4.0%), PCV2 (60.4%), MH (3.0%), APP (69.2%), PMA (52.8%), HP (11.8%), and Sal. spp. infections (37.2%). No antibodies were detected against FMDV. As a result of antigenic analysis of 68 positive cases (13.6%) out of 500 PRRS antigen tests, 61 North American cases, 6 European cases, 1 North American-European complex case. PCV2 has 158 positive cases (31.6%) out of 500 antigen tests, and the results indicate that a considerable number of individuals are infected. To our knowledge, this is the first seroprevalence report of MH, APP, PMA, HP, and Sal. spp. infections in wild boars in South Korea.

Nursery depopulation 기법에 의한 돼지 호흡기질병 상재돈군의 호흡기 병인체 전파방지에 관한 연구 (Elimination of respiratory pathogens in endemically infected swine herds by nursery depopulation)

  • Recently new technologies for the establishment of high health herds are becoming efficient tools in the control of PRRS virus and secondary infections. Medicated early weaning(MEW) and nursery depopulation(ND) have shown to be one of the most successful procedures in the eradication and control of pathogens. Indirect evidence of the role of PRRSV in precipitating secondary infection comes from successful improvement in growth and in decreasing mortality on farms that have eliminated PRRSV through ND. Hence the present experiments were conducted in an effort to compare ND with MEW procedures as a means of eliminating PRRSV controlling secondary pathogens and improving performance of pigs in endemically infected swine herds. Following MEW and ND procedures practiced in the farms, some benefits obtained were as follows: 1. A decrease in PRRSV circulation in the nursery, but no entire elimination. 2. Decrease in the frequency of secondary bacteria and in the use of antibiotics. 3. Mycoplasma hyopneumoniae infection was prevented during the nursery stage. 4. ND protocol had a lower cost and management changes than MEW techniques. 5. Nursery performance was improved after the depopulation, cleaning and disinfection procedures, even though PRRSV still being cycled in the old nursery rooms. These studies revealed that the MEW and ND protocols are not always successful for PRRS virus elimination but it's great effect on control of secondary pathogens and improvement of performance make MEW and ND an efficient tools for the establishment of healthier and more efficient herds.
