Sorghum extract exerts an anti-diabetic effect by improving insulin sensitivity via PPAR-${\gamma}$ in mice fed a high-fat diet

  • This study investigated the hypothesis that a sorghum extract exerts anti-diabetic effects through a mechanism that improves insulin sensitivity via peroxisome proliferator-activated receptor gamma (PPAR-${\gamma}$) from adipose tissue. Seven C57BL/6 mice were fed an AIN-93M diet with fat consisting of 10% of total energy intake (LF) for 14 weeks, and 21 mice were fed a high-fat AIN diet with 60% of calories derived from fat (HF). From week 8, the HF diet-fed mice were orally administered either saline (HF group), 0.5% (0.5% SE group), or 1% sorghum extract (1% SE group) for 6 weeks (n = 7/group). Perirenal fat content was significantly lower in the 0.5% SE and 1% SE groups than that in the HF mice. Levels of total and low-density lipoprotein cholesterol, triglycerides, glucose, and the area under the curve for glucose were significantly lower in mice administered 0.5% SE and 1% SE than those in HF mice. Serum insulin level was significantly lower in mice administered 1% SE than that in HF mice or those given 0.5% SE. PPAR-${\gamma}$ expression was significantly higher, whereas the expression of tumor necrosis factor-${\alpha}$ was significantly lower in mice given 1% SE compared to those in the HF mice. Adiponectin expression was also significantly higher in mice given 0.5% SE and 1% SE than that in the HF mice. These results suggest that the hypoglycemic effect of SE may be related with the regulation of PPAR-${\gamma}$-mediated metabolism in this mouse model.

  • 본 연구에서는 초과 에탄올 추출물을 이용하여 3T3-L1 지방전구세포에서 지방세포로의 분화 연구를 통해 항비만 활성을 확인하고자 하였다. 초과 에탄올 추출물에 의한 지방세포 분화 억제 활성 및 지방형성에 미치는 영향을 확인하기 위해 3T3-L1 지방전구세포에 분화를 유도하면서 추출물을 농도별로 처리하였고, 그 결과 초과 에탄올 추출물은 3T3-L1 지방전구세포에서 지방세포로의 분화를 농도에 의존적으로 감소시켰다. 이 같은 활성에 대한 작용기전을 알아보기 위해 지방세포 분화에 중요한 역할을 담당하고 있는 peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$)와 CCAAT-enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$), 그리고 지방세포 분화에 관여하는 유전자들의 활성을 확인해 보았다. 실험 결과 초과 에탄올 추출물은 $PPAR{\gamma}$$C/EBP{\alpha}$의 유전자 및 단백질 발현을 유의적으로 감소시켰고 지방세포 분화에 관여하는 adipogenesis marker 유전자들의 발현을 억제 시켰다. 따라서 초과 에탄올 추출물은 지방분화 및 adipogenesis 억제 활성을 통해 항비만 소재로의 가능성이 있을 것으로 기대한다.

Hesperidin Ameliorates TNF-${\alpha}$-Mediated Insulin Resistance in Differentiated 3T3-L1 Cells

  • Adipose inflammation is linked to the development of insulin resistance and type 2 diabetes. Hesperidin (HES) is a flavonoid with antioxidant, anti-inflammatory and anti-diabetic properties. However, whether HES improves inflammation-mediated insulin resistance in adipose tissues remains unclear. The purpose of this study was to investigate whether HES attenuates inflammation-mediated insulin resistance in adipose tissue. Herein, RAW 264.7 cells and differentiated 3T3-L1 adipocytes were pretreated with various concentrations of HES in complete media for 1 h and then cultured in the presence or absence of LPS or TNF-${\alpha}$. Our results demonstrated that HES remarkably inhibited LPS-induced production of IL-6, TNF-${\alpha}$, and NO by RAW 264.7 cells in a dose-dependent manner. Also, HES inhibited TNF-${\alpha}$-induced production of IL-6 and $PGE_2$ in differentiated 3T3-L1 cells, while upregulated TNF-${\alpha}$-suppressed expression of adiponectin and PPAR-${\gamma}$ mRNA. These findings suggest that HES may ameliorate inflammation-mediated insulin resistance in adipose tissue.

Protective Effect of Baicalin on the TNF-${\alpha}$-Mediated Development of Insulin Resistance in Differentiated 3T3-L1 Cells

  • Adipose tissue-derived chronic inflammation contributes to development of insulin resistance in obesity, leading to type 2 diabetes and cardiovascular disease. Baicalin, a flavonoid, has antioxidant, anti-inflammatory, antihyperglycemic, anti-adipogenic, and antiobesity effects. However, whether baicalin attenuates adipose tissue-derived development of insulin resistance remains still unclear. This study was to investigate effect of baicalin on the inflammatory changes involved in the development of insulin resistance in adipose tissue. RAW 264.7 cells and differentiated 3T3-L1 adipocytes were pretreated with various concentrations of baicalin in complete media for 1 h and then cultured in the presence or absence of LPS or TNF-${\alpha}$. Our results demonstrated that baicalin remarkably inhibited LPS-induced production of TNF-${\alpha}$, IL-6, and NO by RAW 264.7 cells in a dose-dependent manner. Baicalin also inhibited TNF-${\alpha}$-induced production of IL-6 and $PGE_2$ in differentiated 3T3-L1 cells in a dose-dependent manner, while upregulated TNF-${\alpha}$-suppressed expression of adiponectin and PPAR-${\gamma}$ mRNA and IRS-1 protein. 3T3-L1 전지방세포에서 발효 삼정환의 지방 분화 억제 효과 (Effect of Probiotics-Fermented Samjunghwan on Differentiation in 3T3-L1 Preadipocytes)

  • 본 연구에서는 3T3-L1 전지방세포를 이용하여 발효 삼정환의 지방 분화 억제 효과를 확인하고자 하였다. 삼정환을 5가지 주요 유산균으로 각각 발효시킨 후, Oil Red O 염색법으로 지방세포 분화 억제 여부를 알아보고, 이의 기전을 알기 위해서 지방생성에 핵심적인 역할을 하는 transcription factor 및 지질 조절 효소의 유전자 발현량을 비교하여 보았다. Oil Red O 염색 분석에서는 Lactobacillus plantarum, Enterococcus faecium으로 발효한 삼정환에서 지방구 수의 감소를 확인할 수 있었다. 또한 transcription factor로 $C/EPB{\alpha}$, $PPAR{\gamma}$, UCP-2 및 콜레스테롤 조절과 관련된 효소인 HMG-CoA reductase의 4가지 유전자 발현 정도를 분석하였는데, 발효시키지 않은 삼정환의 경우 농도가 $100{\mu}L/mL$ 이상일 때 네 가지 항목에서 억제 효과를 나타냈으며, 발효시켰을 때는 유산균의 종류에 따라 효과 억제효과가 있었는데, Lactobacillus plantarum으로 발효한 삼정환이 효과가 가장 좋은 것으로 나타났다. Lactobacillus plantarum으로 발효한 삼정환은 동일 농도의 발효하지 않은 삼정환과 비교했을 때, Oil Red O 실험에서 염색된 지방구 수가 더욱 감소하였으며, $PPAR{\gamma}$, UCP-2, HMG-CoA reductase의 경우 농도가 $200{\mu}L/mL$일 때 각각 35%, 57%, 54%로 5가지 발효 삼정환 중 가장 높은 억제율을 나타내었다. 이에 지방분화 억제 효과를 가진 삼정환을 발효시키면 그 효과가 더욱 증가하는 것으로 나타났는데, 특히 Lactobacillus plantarum으로 발효한 삼정환은 $PPAR{\gamma}$, UCP-2, HMG-CoA reductase의 유전자 발현조절을 통해 지방 분화를 억제시키는 것으로 밝혀져 발효 삼정환은 항비만 혹은 항고지혈증 약물로 개발 가능성이 있는 것으로 사료된다.

지실 추출물의 전사인자 SREBP-1 활성에 의한 지질 생성 촉진 (Ponciri Fructus Extract Induces Lipogenesis through Transcription Factor SREBP-1 Activation)

  • This study was to explore the lipogenic effect by ethanol extract of ponciri fructus (EPF) and possible molecular mechanisms in sebocyte. When SZ95 sebocyte cell line were treated with the EPF, lipid droplets were accumulated in the majority of cells. EPF increased expression of sterol regulatory element-binding protein-1 (SREBP-1) and fatty acid synthase (FAS) in the SZ95 cells. EPF augmented expression of PPAR-${\beta}$ and PPAR-${\gamma}$ but not that of PPAR-${\alpha}$. These results suggest that EPF induces lipogenesis in SZ95 cells through SREBP-1, PPAR-${\beta}$ and PPAR-${\gamma}$ activations.

천궁(川芎)의 정유 추출물이 3T3-L1 세포의 분화 및 지방 생성에 미치는 영향 (Effects of Essential Oils Extracted from Cnidii Rhizoma on Differentiation and Adipogenesis in 3T3-L1 Adiopocytes)

  • Objectives We investigated anti-obesity effects of essential oils extracted from Cnidii Rhizoma (CR) in immature adipocytes to magnify it's clinical therapeutic usage. Methods Essential oil of CR was extracted with ethyl acetate or petroleum ether and through steam distillation, respectively. Oil red-O staining for monitoring its inhibition effect on adipogenesis and differentiation in murine 3T3-L1 adipocytes and 3-(4,5-methylthiazol-2-yl)-2,5-diphenyletetra zolium bromide (MTT) assay for cell safety were done. Also phospho-adenosine monophosphate (AMP)-activted protein kinase (P-AMPK), AMP-activated protein kinase, phospho-acetyl-CoA carboxylase (P-ACC), acetyl-CoA carboxylase, peroxisome proliferator-activated receptor-${\alpha}$ (PPAR-${\alpha}$), peroxisome proliferator-activated receptor-${\gamma}$ (PPAR-${\gamma}$) and CCAAT/enhancer binding protein ${\alpha}$ (C/EBP-${\alpha}$) expressions as obesity-related factors were measured by western blot analysis. Results Protein expressions of P-AMPK, P-ACC and PPAR-${\alpha}$ were increased in essential oils-treated adipocytes compared to those of control group, respectively. Furthermore, protein expressions of PPAR-${\gamma}$ and C/EBP-${\alpha}$ were decreased in essential oils-treated adipocytes compared to those of control group, respectively. Conclusions These results demonstrate that essential oils of CR inhibit adipogenesis and differentiation. Also they promote the oxidation of fatty acids in adipocytes. Thus, results suggest that essential oils of CR could be used as a valuable material for anti-obesity therapeutics via control of lipid metabolism.

Zinc-chelated Vitamin C Stimulates Adipogenesis of 3T3-L1 Cells

  • Adipose tissue development and function play a critical role in the regulation of energy balance, lipid metabolism, and the pathophysiology of metabolic syndromes. Although the effect of zinc ascorbate supplementation in diabetes or glycemic control is known in humans, the underlying mechanism is not well described. Here, we investigated the effect of a zinc-chelated vitamin C (ZnC) compound on the adipogenic differentiation of 3T3-L1 preadipocytes. Treatment with ZnC for 8 d significantly promoted adipogenesis, which was characterized by increased glycerol-3-phosphate dehydrogenase activity and intracellular lipid accumulation in 3T3-L1 cells. Meanwhile, ZnC induced a pronounced up-regulation of the expression of glucose transporter type 4 (GLUT4) and the adipocyte-specific gene adipocyte protein 2 (aP2). Analysis of mRNA and protein levels further showed that ZnC increased the sequential expression of peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) and CCAAT/enhancer-binding protein alpha (C/$EBP{\alpha}$), the key transcription factors of adipogenesis. These results indicate that ZnC could promote adipogenesis through $PPAR{\gamma}$ and C/$EBP{\alpha}$, which act synergistically for the expression of aP2 and GLUT4, leading to the generation of insulin-responsive adipocytes and can thereby be useful as a novel therapeutic agent for the management of diabetes and related metabolic disorders.

대시호탕(大柴胡湯)이 3T3-L1 지방전구세포와 고지방식이 유도 비만쥐에 미치는 영향 (The Effects of Dai-saiko-to (Da-Chai-Hu-Tang) on 3T3-L1 Preadipocytes and High-Fat Diet-Induced Obese Mice)

  • Objectives This experimental study was designed to investigate the effects of Dai-saiko-to (DSH) on differentiation of 3T3-L1 preadipocytes and body weight, serum lipid levels in high-fat diet-induced obese mice. Materials and Methods Cells were incubated with DSH at an indicated concentration (0.01-1 mg/ml) for 24h, then the growth rate was assessed by MTS assay. 3T3-L1 preadipocytes were incubated in DMEM for 2 days with the indicated concentrations of DSH. On Day 6, the cells were fixed and the cellular lipid contents were assessed by Oil-Red-O staining. The expression of peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$) and cytidine-cytidine-adenosine-adenosine-thymine (CCAAT)/enhancer-binding proteins ${\alpha}$ ($C/EBP{\alpha}$) as adipocyte-specific proteins were determined by real time RT-PCR and western blotting. Four-weeks old mice (wild-type C57BL/6) were used for all experiments. Body weight gain and serum lipid levels were measured in the obesity-induced mice. Results DSH did not show toxicity even at the concentration of 1 mg/ml and DSH significantly inhibited the differentiation of 3T3-L1 preadipocytes in a dose-dependent manner. Also, DSH significantly reduced the expressions of $PPAR{\gamma}$ and $C/EBP{\alpha}$ in a dose-dependent manner. Furthermore, DSH significantly reduced body weight gain, serum glucose, total cholesterol and LDL-cholesterol contents in obesity-induced mice. Conclusions These results demonstrated that DSH inhibited 3T3-L1 preadipocyte differentiations and high-fat diet-induced obesity in mice.

상엽(桑葉) 함유 한약복합제 추출물의 항비만(抗肥滿)효과 연구 (The Study on Anti-obesity Effects of Mulberry Leaves Contained Herbal Mixture)

  • Objectives This experimental study was designed to investigate the effects of Mulberry leaves contained herbal mixture (MLHM) on body weight, serum lipid level and adipocyte differentiation in high fat diet-fed obese mice. Methods Four-week old mice (wild-type C57/BL6) were used for all experiments. Cells were incubated with MLHM at the indicated concentration (0.04-4mg/ml) for 24h, and growth rate was assessed by MTT ((3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. 3T3-L1 preadipocytes were incubated in DMEM for 2 days with the indicated concentrations of MLHM, and on Day 6, the cells were fixed and the cellular lipid contents were assessed by Oil-Red-O staining. The expression of peroxisome proliferator-activated receptor ${\gamma}$ (PPAR ${\gamma}$) and cytidine-cytidine-adenosine-adenosine-thymine (CCAAT)/enhancer-binding proteins ${\alpha}$ (C/EBP ${\alpha}$) as adipocyte-specific proteins were determined by real time RT-PCR and western blotting. In addition, body weight gain and serum lipid levels were measured in the mice with obesity induced by the high fat-diet for four weeks. Results Though MLHM did not show toxicity even at the concentration of 4mg/ml, MLHM significantly inhibited the differentiation of 3T3-L1 preadipocites in a dose-dependent manner. Also, MLHM significantly reduced the expressions of PPAR ${\gamma}$ and C/EBP ${\alpha}$ in a dose-dependent manner. Furthermore, MLHM significantly reduced body weight gain and LDL-cholesterol contents in high fat diet-fed obese mice. Conclusions These results demonstrate that MLHM exerts anti-obesity effect in 3T3-L1 cells and mice with obesity by high-fat diet.
