Dietary Supplementation with Acanthopanax senticosus Extract Modulates Gut Microflora in Weaned Piglets

  • This study was conducted to investigate the effects of Acanthopanax senticosus extract (ASE) as a dietary additive on gut microflora in weaned piglets. A total of sixty pigs were weaned at 21 d of age (BW = $5.64{\pm}0.23kg$) and allocated on the basis of BW and litter to three dietary treatments in a randomized complete block design. The dietary treatments were: control group (basal diet), antibiotics group (basal diet+0.02% colistin), and ASE group (basal diet+0.1% ASE). On d 7, 14 and 28 after consuming the experimental diets, five piglets per group were sacrificed and then the contents from the jejunum, ileum and cecum were collected to determine changes in the microbial community by using a polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) technique and estimating the contents of Lactobacillus and E. coli by in vitro culturing methods. The results showed that the ASE promoted the microflora diversity in the cecum. Enumeration of bacteria in the gut contents showed that the number of Lactobacillus increased (p<0.05), while that of E. coli decreased (p<0.05) when compared with the other 2 groups as the days of age progressed post-weaning. These findings suggested that the ASE, as a substitute for dietary antimicrobial products, could improve the development of the normal gut microflora and suppress bacterial pathogens, and effectively promote a healthy intestinal environment.

Effect of increased dietary crude protein levels on production performance, nitrogen utilisation, blood metabolites and ruminal fermentation of Holstein bulls

  • Objective: This study investigated the effect of dietary crude protein (CP) supplementation on nutrient intake, nitrogen (N) utilisation, blood metabolites, ruminal fermentation and growth performance of young Holstein bulls. Methods: Twenty-one young bulls weighing $277{\pm}11.2kg$ were equally divided into three groups and were offered diets formulated with low CP (LCP; 10.21% CP and 4.22% rumen degradable protein [RDP]), medium CP (MCP; 12.35% CP and 5.17% RDP) and high CP (HCP; 14.24% CP and 6.03% RDP). Yellow corn silage was used as a unique forage source and was mixed with concentrate. This mixed feed was given ad libitum to the young bulls included in the study. Results: Results showed that CP intake, blood urea nitrogen, N intake, total N excretion and N balance increased linearly with an increase in dietary CP level (p<0.05). However, no significant difference was observed in nutrient digestibility among the bulls receiving the different diets. Ruminal pH (p<0.05) and ammonia nitrogen ($NH_3-N$) concentration (p<0.01) were significantly higher in the bulls receiving the MCP and HCP diets than in those receiving the LCP diet. The bulls receiving the HCP diet showed significantly higher ruminal bacterial protein level, propionate, acetate and total volatile fatty acid (TVFA) concentrations than bulls receiving the LCP diet (p<0.05). Moreover, dietary CP level exerted a significant positive effect on the final body weight, average daily gain and gain-to-feed ratio of the bulls (p<0.05). Conclusion: High dietary CP level is optimal for achieving maximum growth and high profitability without exerting a negative effect on the physiology of growing Holstein bulls.

119 구급서비스 지역별 출동특성 및 출동거리와 현장도착시간과의 관계 (The Regional Characteristics of 119 Ambulance Dispatch, the Distance and Response Time to the Scene)

  • 본 연구는 응급구조학과 학생들의 구급차 동승실습으로 구축된 구급활동일지를 바탕으로 응급환자의 지역별 특성을 파악하고, 지역 간의 출동거리와 현장도착시간 간에 의미있는 영향이 있는 지를 알아보고자 하였다. A대학 4학년 학생 총 31명이 서울, 대전, 충남 및 세종지역의 소방서에서 4주간의 실습으로 얻어진 총 1133개의 구급활동일지를 후향적으로 분석하였다. 연구결과 환자평가 중 맥박, 호흡수, 산호포화도는 모든 지역에서 80%이상을 실시하고 있었다. 중증도 분류에 있어서는 대전이 응급으로 표시한 경우가 가장 많았으나 환자에 대한 응급처치는 모든 지역에서 단순한 기본적인 처치가 대부분이었다. 신고부터 현장까지 도착하는 시간은 서울이 평균 5분 40초(${\pm}$2분 25초)로 가장 빨랐고, 충남세종은 10분 이상 걸리는 경우도 35.7%에서 나타났다. 주된 신고 이유는 서울과 대전에서는 질병이었고, 충남은 손상의 비율이 높았다. 이러한 결과는 응급구조학과 학생들의 실습지역에 따라 실습내용이 다소 차이가 있을 수 있어 실습기준에 대한 표준지침이 필요할 것으로 사료되며, 빠른 시간내에 적절한 응급처치를 받을 수 있는 119 안전센터의 배치나 구급차 배치를 고민하고, 지역별 구급의 질 적 차이를 최소화하기 위해 노력해야 할 것이다.

Comparison of Indigenous Browses and Sunflower Seed Cake Supplementation on Intake and Growth Performance of Dual-purpose Goats Fed Buffel Grass (Cenchrus ciliaris) Hay

  • A study to compare the effects of supplementing Delonix elata, Grewia similis, Tamarindus indica and sunflower seed cake on intake and growth rate of dual-purpose goats fed low quality Buffel grass (Cenchrus ciliaris) hay was carried out. Twenty-eight male goats aged five to seven months (mean weight 12.93${\pm}$3.94 kg) were randomly allocated to four dietary groups in a completely randomised design. The diets were hay plus Grewia similis, hay plus Delonix elata, hay plus Tamarindus indica and hay plus sunflower seed cake. All diets were supplemented with maize bran. The experimental period was 90 days. Voluntary dry matter intake of the supplements was higher for Tamarindus indica (275.5 g/day) and Grewia similis (201.8 g/day) and lowest for sunflower seed cake (81g/day). Goats supplemented with Grewia similis had the highest hay intake (183.8 g/day) while those supplemented with sunflower seed cake had the lowest hay intake (98.9 g/day). Animals fed browse supplements gained significantly more weight (p<0.001) than those with sunflower seed cake. There were no significant differences in live weight change between goats fed the different browses. However, those fed Tamarindus indica gained an average of 20.79 g/d which was slightly higher than the gains for those on Grewia similis and Delonix elata while those fed sunflower seed cake lost weight. Correspondingly, goats supplemented with browse leaf meals had higher feed conversion ratios than those supplemented with sunflower seed cake and required 23.91 to 35.06 g DM of feed to produce one g of weight gain per day. In a separate study, the DM disappearance pattern indicated that Grewia similis and Delonix elata were highly degradable compared to Tamarindus indica. At 24 h of incubation, DM degradability was 627, 588 and 345 g/kg DM for Grewia similis, Delonix elata and Tamarindus indica, respectively. In another study in vivo DM digestibility ranged from 46.1% (for hay alone) to 56.2% (for hay plus Grewia similis). It was concluded that the addition of Tamarindus indica, Grewia similis and Delonix elata leaf meals to Cenchrus ciliaris hay resulted in increased total DM intake, in vivo digestibility and growth rate. Therefore, leaf meals of indigenous browses particularly Tamarindus indica and Grewia similis could be used as supplementary feeds for small ruminants grazing on poor quality roughages during the dry season rather than use of expensive, less effective and intermittently available sunflower seed cake.

Influence of Dietary Supplementation of Condensed Tannins through Leaf Meal Mixture on Intake, Nutrient Utilization and Performance of Haemonchus contortus Infected Sheep

  • The study assessed the effect of dietary supplementation of leaf meal mixture (LMM) containing condensed tannins (CT) on feed intake, nutrient utilization and performance of sheep infected with Haemonchus contortus. Eighteen adult sheep of similar age and body weight ($25.03{\pm}1.52$) were included in this study and out of these, 12 sheep were infected with single dose of infective third stage larvae of H. contortus at 2,000 larvae per sheep. The experimental sheep were allocated in three different groups' i.e. negative control (NC; no infection), control (C; H. contortus infected) and treatment (T; H. contortus infected+CT at 1.5% of the DM through LMM) and the experiment was conducted for a period of 90 d. The intake of dry matter (DM), organic matter (OM) and digestibility of DM, OM, neutral detergent fibre (NDF) and acid detergent fibre (ADF) were comparable among three animal groups. However, digestibility of crude protein (CP) and ether extract (EE) were significantly (p<0.05) higher in NC group as compared to both C and T groups. Nitrogen (N) retention (g/d or % of N intake) was significantly (p = 0.038) lower in C group as compared to T and NC groups. Daily intake (g/kg $W^{0.75}$) of digestible crude protein (DCP), digestible organic matter (DOM) and total digestible nutrient (TDN) did not differ significantly (p<0.05) in the three groups. Haemoglobin (Hb) and packed cell volume (PCV) were significantly (p<0.001) higher in treatment group as compared to control. The level of Hb and PCV reduced (p<0.001) after 30 days of experimental feeding. CT significantly (p<0.001) reduced serum urea in T group as compared to NC and C groups. Serum proteins differed significantly (p<0.01) among the three groups. The activity of serum enzymes AST, ALT, ALP and LDH were also statistically non significant (p<0.05) among treatments. The weight of abomasal lymph nodes (ALN) in T group was higher (p<0.05) than in C group. Treatment group had lower (p<0.05) total worms and fecal egg count compared to control group. It may be concluded that dietary supplementation of CT through LMM significantly improved the N retention, and inhibited the different developmental stages of Haemonchus contortus in experimental sheep.

Effects of Chitosan on Body Weight Gain, Growth Hormone and Intestinal Morphology in Weaned Pigs

  • The study was conducted to determine the effects of chitosan on the concentrations of GH and IGF-I in serum and small intestinal morphological structure of piglets, in order to evaluate the regulating action of chitosan on weaned pig growth through endocrine and intestinal morphological approaches. A total of 180 weaned pigs (35 d of age; $11.56{\pm}1.61kg$ of body weight) were selected and assigned randomly to 5 dietary treatments, including 1 basal diet (control) and 4 diets with chitosan supplementation (100, 500, 1,000 and 2,000 mg/kg, respectively). Each treatment contained six replicate pens with six pigs per pen. The experiment lasted for 28 d. The results showed that the average body weight gain (BWG) of pigs was improved quadratically by dietary chitosan during the former 14 d and the later 14 d after weaned (p<0.05). Furthermore, dietary supplementation of chitosan tended to quadratically increase the concentration of serum GH on d 14 (p = 0.082) and 28 (p = 0.087). Diets supplemented with increasing levels of chitosan increased quadratically the villus height of jejunum and ileum on d 14 (p = 0.089, p<0.01) and 28 (p = 0.074, p<0.01), meanwhile, chitosan increased quadratically the ratio of villus height to crypt depth in duodenum, jejunum and ileum on d 14 (p<0.05, p = 0.055, p<0.01) and 28 (p<0.01, p<0.01, p<0.01), however, it decreased quadratically crypt depth in ileum on d 14 (p<0.05) and that in duodenum, jejunum and ileum on d 28 (p<0.01, p<0.05, p<0.05). In conclusion, these results indicated that chitosan could quadratically improve growth in weaned pigs, and the underlying mechanism may due to the increase of the serum GH concentration and improvement of the small intestines morphological structure.

Effects of inclusion level and adaptation period on nutrient digestibility and digestible energy of wheat bran in growing-finishing pigs

  • Objective: This experiment was to evaluate the effect of different inclusion levels and adaptation periods on digestible energy (DE) and the apparent total tract digestibility (ATTD) of chemical constituents in diets supplemented with wheat bran. Methods: Thirty-six crossbred barrows with an initial body weight of $85.0{\pm}2.1kg$ were allotted to 6 diets in a completely randomized block design with 6 pigs per diet. Diets included a cornsoybean basal diet and 5 additional diets which were formulated by replacing corn and soybean meal in control diet with 15%, 25%, 35%, 45%, or 55% wheat bran. The experiment lasted for 34 d, and feces were collected from d 8 to 13, 15 to 20, 22 to 27, and 29 to 34 respectively. Results: The results showed no interaction effects between inclusion level and adaptation period on the concentration of DE and the ATTD of gross energy (GE) and crude protein (CP) in wheat bran. The DE value and ATTD of GE in wheat bran decreased (p<0.05) significantly as the inclusion level of wheat bran increased, but no difference in the ATTD of CP was observed. The ATTD of CP in wheat bran increased (p<0.10) significantly as the adaptation period for pigs was prolonged. In addition, the concentration of DE and the ATTD of GE in wheat bran decreased linearly (p<0.05) when pigs were fed either an increased level of wheat bran or given an increased adaptation period to the diets. Conclusion: Wheat bran showed a negative effect on the concentration of DE and ATTD of GE and CP as the inclusion level increased. A longer adaptation period can gradually increase the DE value and ATTD of GE and CP in wheat bran, and at least 14 to 21 d of adaptation might be recommended for growing-finishing pigs fed the high-fiber diets with wheat bran.

생약 열수추출물의 근력향상 효능 평가 (Evaluation on the Muscular Strength Activity of Medicinal Herb Hot-Water Extracts)

  • 생약 열수추출물의 근력 향상 효능을 평가하기 위하여 근력 측정 및 혈액의 피로요소를 분석한 결과 간 글리코겐 함량은 모든 식이군들이 대조군에 비하여 높은 수치를 나타내었다. 특히 맥문동, 오미자, 오가피, 함초 식이군의 경우 높은 글리코겐 함량을 나타내었다. 4주간 측정한 근력 향상효능은 오가피, 함초, 홍삼 열수추출물 섭취군의 경우 다른 열수추출물 섭취군에 비하여 높은 근력 향상 효과를 보여주었다. 무기인산은 맥문동, 오미자, 오가피, 두충, 구기자, 홍삼 열수추출물 섭취군의 경우 대조군에 비하여 낮은 수준을 나타내었다. 젖산 함량은 모든 생약재 열수추출물 식이군에서 대조군과 비교하여 낮은 함량을 나타내어 생약 열수 추출물을 이용한 근력 향상용 기능성 식품의 개발 가능성을 보여주었다.

허베이스피릿호 원유유출 사고 이후 태안주민의 식생활 변화 - 포커스 그룹 면담 결과 - (Changes of Eating Habits of the Residents of Taean, Korea after the Herbei Oil Spill Accident Based on Focus Group Interviews)

  • Hazardous chemicals, such as polycyclic aromatic hydrocarbons (PAHs), volatile organic compounds (VOCs) and heavy metals, are known as being harmful to human health were included in oils released by the Herbei Spirit Oil Spill accident in December 2007. To investigate changes of eating habits by the exposure to harmful substances, we conducted 5 focus group interviews for residents at Taean coast areas, who had experienced the oil spill accident. Participants included 46 women (mean age: $57.2{\pm}10.9$) who were mainly responsible for preparing family meals. Focus group discussions were audio-taped, transcribed and categorized by themes. Participants expressed more frequent illness symptoms such as dizziness, vomiting, visual loss, and skin diseases after the accident. They mentioned that their worries about economic hardship and worsened health status since the accident induced mental problems, such as depression about their children. Regarding eating habits, participants reported less intakes of fishes and meats and relatively more intakes of vegetables and kimchi due to the lack of household incomes after the accident. Although the participants had been used to collecting or catching fish or shellfish for their consumption previously, they mainly purchased these foods from local markets after the accident. Changes of eating habits induced by the accident included drinking boiled water and having steamed or fried seafood rather than raw seafood. Changes of food intakes occurred less frequently in older adults due to their longterm fixed eating habits, although they felt uncomfortable for having raw fish. The findings of this study clearly present that the exposure of hazardous substances by the oil spill accident had a significant impact on changes in eating habits besides economic, physical, and mental problems among the residents in Taean. Continuous health and nutrition monitoring and support are needed.

A Collaborative Study on Korean Standard JE Vaccine for Potency Assay

  • The objective of this collaborative study was to establish a Korean standard of Japanese encephalitis (JE) vaccine (mouse brain-derived, formalin-inactivated) for potency assay. A candidate preparation proposed as a Korean standard was provided by GreenCross Vaccine, and six laboratories, including one national control laboratory and five manufacturers of JE vaccine, participated in the study. The potency of the candidate preparation and a reference standard obtained from Japan was estimated by mouse immunogenicity assay using the in vitro plaque reduction neutralization test (PRNT). The results of 72 assays conducted by the 6 laboratories showed that the overall mean potency estimate of the candidate preparation was 2.455 log median plaque reduction neutralization antibody titer per 0.5-ml dosage administered twice in mice at 7-day intervals, and that the mean potency ratio of the candidate preparation relative to the reference standard was 1.074. The potency estimates were quite variable among laboratories irrespective of the preparation. The variability of assays assessed by Z scores and coefficient of variation (CV) were in general within the level of acceptance (Z scores within $\pm3$ and $CV\;\leq\;15%$). Therefore, we concluded that the candidate preparation would be suitable as a national standard for testing the potency of JE vaccine (inactivated).
