Characteristics of Cytosolic Calcium-Independent Phospholipase $A_2$ Isolated from Rat Liver

  • A calcium-independent phospholipase $A_2$ ($iPLA_2$) was identified from the cytosolic fraction of rat liver cells. On gel filtration chromatography, the $iPLA_2$ activity was eluted as broad peaks of 150 to 500 kDa. The enzyme was maximally active at pH 7.5, retained 75% of its original activity after heating at $50^{\circ}C$ for 5 h, and was inhibited by $Ca^{2+}$, $Mg^{2+}$, and $Zn^{2+}$ ions, but was not affected by $Na^+$ and $K^+$ ions. The enzymatic activity was increased up to 150% by 1 to 4 mM DTT and was inhibited up to 25% by 0.1 to 1 mM PMSF. The $iPLA_2$ activity had preference for the head group of phospholipids, where phosphatidylethanolamine was preferred to phosphatidylcholine. The results suggest that the $iPLA_2$ may be a novel enzyme distinct from the previously reported $iPLA_2s$.

  • 자연계에 존재하는 수은중 유기수은은 생태계 먹이사슬을 통하여 체내의 여러장기에 축적되어 조직손상을 일으키는 것으로 잘 알려져 있다. 그러나 이러한 세포독성에 대한 정확한 생화학적 기전에 대해서는 자세히 알려진 바가 없다. 포스포리파아제 $A_2$(PLA$_2$)는 세포막의 인지질로부터 Arachidonic acid (AA)와 Lysophospholipid를 유리시키는 효소로 최근 세포손상과 관련하여 그 역할이 주목되고 있으며, 극히 최근, 일차배양 소뇌신경세포를 이용한 연구에서 메칠수은처리에 의해 세포독성의 지표인 Lactate dehydrogenase (LDH)의 유리와 함께 AA 유리가 증가되는 것이 관찰되었으나 여러형태의 PLA$_2$중 어느형태의 효소가 관련되어 있는지, 또한, 그 자세한 기전에 대해서는 불분명한 점이 많다. 본 연구에서는 신장세포의 일종인 MDCK세포를 이용하여 메칠수은의 처리에 의한 PLA$_2$의 활성화 및 그 생화학적인 기전을 구명하고자 하였다. [$^3$H]AA를 MDCK세포의 배양액에 첨가하여 라벨링한 후 메칠수은을 처리하였을때 [$^3$H]AA가 대조군에 비해 농도의존적 및 경시적으로 현저하게 증가하였으며 동시에 LDH의 유리도 함께 관찰되었다. 이러한 [$^3$H]AA의 유리 증가는 세포질 PLA$_2$에 특이적인 저해제로 알려진 AACOCF$_3$의 전처리에 의해 거의 완전히 억제되었으나 LDH의 유리는 오히려 증가하였다. 또한, 글루타치온(GSH)의 전구체인 NAC (N-Acetyl Cysteine)에 의해 [$^3$H]AA의 유리는 부분적으로 감소하였으나, LDH의 유리는 변함이 없었다. 돼지비장이나 MDCK 세포에서 얻어진 세포질 PLA$_2$에 메칠수은을 직접 처리하였을때는 오히려 PLA$_2$의 활성은 감소되었다. 위의 결과들로부터 메칠수은에 의한 [$^3$H]AA의 유리 증가는 세포질 PLA$_2$효소에 대한 직접적인 작용이 아니라 세포내 -SH기의 차단이나 Oxidative Stress에 의해 간접적으로 활성화되는 것으로 예상되며, 세포질 PLA$_2$에 의해 유리된 AA의 세포독설과 관련된 세포내의 역할에 대해 의문이 제기되었다.

Annexin I의 구조와 결합에 관한 분광학적 연구

  • Annexin I is a member of the in family of calcium dependent phospholipid banding proteins and is an in vitro phospholipase $A_2$ (PLA$_2$) inhibitor. The mechanism of PLA$_2$ inhibition by annexin I is still ambiguous. The structure of annexin I was studied at the atomic level by using nuclear magnetic resonance (NMR), circular dichrotsm (CD) and fluorescence spectroscopy. Recombinant human annexin I and N-terminally truncated annexin I (1-31 deleted: d-annexin I) were purified and their NMR spectra were compared. The NMR spectra of the two were similar. When $Ca^{2+}$ ion added to annexin I ad d-annexin I, peak broadening occurred, but no significant spectroscopic change was observed. When porcine pancreatic PLA$_2$ was added to deuterium labeled annexin I, an interaction of annexin I with PLA$_2$ was observed as indicated by the disappearance and shift of several peaks in the NMR spectrum. This result supports a protein-protein interaction mechanism for PLA$_2$ inhibition by annexin I.I.

  • We previously reported that phospholipase$A_2$ ($PLA_2$)-related pathway and capacitative calcium entry (CCE) via store-operated calcium channel (SOC) were involved in the regulation of muscarinic receptor- mediated sAPP release. We also observed that stimulation of muscarinic receptor associated with the inositol phosphate cascade resulted not only in increase of CCE but also in activation of PLA$_2$ in SH-SY5Y cells. In this study, we further investigated whether the $PLA_2$ isoforms differently regulate the muscarinic receptor-mediated sAPP release, and examined the relationships between activation of $PLA_2$ isoforms and CCE mediated by muscarinic receptors in SH-SY5Y cells.   • Poly(lactic acid)(PLA) nanocomposites containing various nanofillers were synthesized using the solution intercalation method. Organically modified bentonite clay (NSE), octadecylamine-graphene oxide (ODA-GO), and an NSE/ODA-GO complex were utilized as nanofillers in the fabrication of PLA hybrid films. PLA hybrid films with varying nanofiller contents in the range of 0-10 wt% were examined and compared in terms of their thermomechanical properties, morphologies, and oxygen permeabilities. Transmission electron microscopy (TEM) confirmed that most of the NSE and ODA-GO nanofillers were dispersed homogeneously throughout the PLA matrix on the nanoscale, although some agglomerate NSE/ODA-GO complex particles were also formed. Among the three nanofillers for PLA hybrid films, the NSE/ODA-GO complex showed the best improvement in film thermal stability. In contrast, NSE and ODA-GO exhibited the best improvement in tensile mechanical properties and oxygen barrier properties of the PLA hybrid films, respectively.

D609, an Inhibitor of Phosphatidylcholine-specific Phospholipase C, Inhibits Group IV Cytosolic Phospholipase A2

  • As an inhibitor of phosphatidylcholine-specific phospholipase C (PC-PLC), D609 has been widely used to explain the role of PC-PLC in various signal transduction pathways. This study shows that D609 inhibits group IV cytosolic phospholipase $A_2$ ($cPLA_2$), but neither secretory $PLA_2$ nor a $Ca^{2+}$-dependent $PLA_2$. Dixon plot analysis shows a mixed pattern of noncompetitive and uncompetitive inhibition with $K_i=86.25{\mu}M$ for the $cPLA_2$ purified from bovine spleen. D609 also time- and dose-dependently reduces the release of arachidonic acid from a $Ca^{2+}$- ionophore A23187-stimulated MDCK cells. In the AA release experiment, $IC_{50}$ of D609 was ${\sim375}{\mu}M$, suggesting that this reagent may not enter the cells easily. The present study indicates that the inhibitory effects of D609 on various cellular responses may be partially attributable to the inhibition of $cPLA_2$.

Cytosolic Phospholipase A2 Activity in Neutrophilic Oxidative Stress of Platelet-activating Factor-induced Acute Lung Injury (Platelet-activating Factor에 의한 급성폐손상에서 호중구성 산화성 스트레스에 관여하는 Cytosolic Phospholipase A2 활성도의 변화)

  • Background: The present investigation was performed in rats and isolated human neutrophils in order to confirm the presumptive role of the positive feedback loop of cytosolic phospholipase $A_2$ ($cPLA_2$) activation by plateletactivating factor (PAF). Methods: The possible formation of the positive feedback loop of the $cPLA_2$ activation and neutrophilic respiratory burst was investigated in vivo and in vitro by measurement of the parameters denoting acute lung injury. In addition, morphological examinations and electron microscopic cytochemistry were performed for the detection of free radicals in the lung. Results: Five hours after intratracheal instillation of PAF ($5{\mu}g/rat$), the lung leak index, lung myeloperoxidase (MPO) activity, the number of neutrophils and the concentration of cytokine-induced neutrophil chemoattractant (CINC) in bronchoalveolar lavage fluid were increased by PAF as compared with those of control rats. The NBT assay and cytochrome-c reduction assay revealed an increased neutrophilic respiratory burst in isolated human neutrophils following exposure to PAF. Lung and neutrophilic $cPLA_2$ activity were increased following PAF exposure and exposure to hydrogen peroxide increased $cPLA_2$ activity in the lung. Histologically, inflammatory findings of the lung were observed after PAF treatment. Remarkably, as determined by $CeCl_3$ cytochemical electron microscopy, increased production of hydrogen peroxide was identified in the lung after PAF treatment. Conclusion: PAF mediates acute oxidative lung injury by the activation of $cPLA_2$, which may provoke the generation of free radicals in neutrophils.

Fabrication of PLA/TiO2 nanofibers using melt-electro-spinning (용융전기방사를 이용한 PLA/TiO2 나노섬유의 개발)

  • Electrospun webs have been widely investigated for applying to drug delivery system (DDS) because of their high specific surface area and high porosity. In this study, the composite webs of PLA (poly(lactic acid)) and $TiO_2$ were fabricated by melt-electro-spinning method for applying to drug delivery system. The morphologies of PLA/$TiO_2$, webs were observed using scanning electron microscope (SEM) and field emission transmission electron microscope (FE-TEM). The crystal structures of PLA/$TiO_2$ composite webs were confirmed by X-ray diffractometer (XRD).

Prevalence and Genotype Distribution of Human Papillomavirus among Women from Henan, China

  • Human papillomavirus (HPV) infection has been implicated as a causative of cervical cancer. In the present study, a total of 578 samples from females attending the gynecological outpatient clinic in Henan province, China, were collected and the HPV genotypes were detected by gene chip and flow-through hybridization. Overall, 44.5% (257/578) females were found to be HPV DNA positive, and the high risk HPV (HR-HPV) rate was 35.1% (203/578). The first peak of HR-HPV infection appeared in the >60 year-old group (55.0%), and the second was within the 51-55 year-old group (50.0%) (${\chi}^2$=19.497, p<0.05). HPV 16 was the most prevalent genotype (9.2%), followed by HPV 52 (7.8%), HPV 6 (6.9%), HPV 11 (5.9%) and HPV 42 (5.0%). The single type HPV infection was 30.4%, with the five majority prevalent genotype HPV 16 (16.5%), HPV 52 (14.3%), HPV 6 (12.6%), HPV 42 (8.6%), HPV 31 (5.1%). The multiple-type HPV infections were 14.0%, and HPV 16 was the most prevalent type (29.6%), followed by HPV 52 (24.7%), HPV 6 (22.2%), HPV 11 (22.2%), HPV 42 (17.3%) and HPV 39 (17.3%).

Toughening of PLA stereocomplex by Impact modifiers (충격보강제에 의한 PLA stereocomplex의 강인화 연구)

  • We tried to blend PLLA and PDLA at overall compositions to form PLA stereocomplexes (SC). The presence of the SC crystalline phase in the PLLA matrix was verified by differential scanning calorimetry (DSC). As a result, a various PDLA composition of the PLA SC blends can influence PLA SC formation. And the largest amount of PLA SC crystallites was formed when PLLA/PDLA ratio is 50/50. In addition, we have tried to do PLA SC toughening with two impact modifiers in 92/8, 85/15 ratio of PLLA/PDLA to enhance the mechanical properties such as impact strength. Thermal and mechanical properties of PLA SC were investigated by DSC, HDT, Izod impact tester and UTM. PLA SC formation decreased when 10-20 wt% of Strong120 (impact modifier) was added. On the other hand, there is no effect on PLA SC formation when 10-20% of Elvaloy (impact modifier) was added. HDT values dramatically increased over $100^{\circ}C$ with the addition of PDLA. However, HDT decreased as Strong120 and Elvaloy content increased. Finally, we could find well balanced composition of toughened PLA SC with 10wt% of impact modifier in flexural modulus and impact strength.
