Thermotropic Liquid Crystalline Behavior of Aliphatic Acid Esters of N,O-Hydroxypropyl Chitosans (N,O-히드록시프로필 키토산 지방산 에스터들의 열방성 액정 거동)

  • Two kinds of N,O-hydroxypropyl chitosans (HPCTOs) with degree of substitution (DS) and molar substitution (MS) ranging from 2.15 to 2.39 and 2.9 to 4.1, respectively, and five kinds of aliphatic acid esters of HPCTOs (HPCTOAms, m=0,2,4,7,9, the number of methylene units in aliphatic substituent) based on the HPCTOs were synthesized, and the thermotropic liquid crystalline properties of the derivatives were investigated. All the derivatives formed enantiotropic cholesteric phases whose optical pitches (${\lambda}_m$'s) increased with increasing temperature. However, the glass and clearing temperatures, the magnitude of ${\lambda}_m$ of the mesophase at the same temperature, and the temperature dependence of ${\lambda}_m$ of the investigated derivatives highly depended on MS and m. The thermotropic mesophase properties of HPCTOAms were significantly different from those reported for the aliphatic acid esters of hydroxypropyl celluloses. The results indicate that the secondary amino group in the C-2 position plays an important role in the thermal stabilization and temperature dependence of ${\lambda}_m$ of the cholesteric mesophase.

Physical Properties of Covered Stent in Gastric Acid Environment: In Vitro Study (위산 환경에서 피막형 스텐트의 물성 변화)

  • In membrane covered stent, occlusion and fracture from membrane degradation by gastric acid sometimes occurred. Therefore, we investigated the physical properties of membrane covered stent according to its ingredient and concentration in gastric acid environment. Membrane covered stents consisted of silicone and polyurethane with 15%, 18%, 20% concentrations were used. After incubating stents in a condition of pH 1.2, we checked any changes at every 3 weeks for 18 weeks. The changes of membrane surface, radial expansion and recovery force of stent were investigated. Coating thickness increased proportionally to an increase in ingredient concentration. Surface was evenly coated with silicone compared to the case with polyurethane and its homogeneity was excellent in a high concentration. Degradation was much severe in the case of polyurethane. The radial force of silicone was higher than polyurethane, and the decrease of radial and recovery force was higher in the case of polyurethane. In conclusion, high concentration of silicone membrane was more stable than polyurethane in acid environment of in vitro study.

Preparation of Elastic Branched Copolyester for Toner Binder: Effects of Branching Agents (토너 바인더용 분지화된 탄성 폴리에스테르 공중합체의 합성: 분지제의 영향)

  • The branched copolyester was synthesized and its molecular weight, $T_g$, 1/2 method temperature ($T_{1/2}$) and rheological properties were characterized for the application of toner binder. The linear copolyester had low molecular weight and melt elasticity obtained by dimethylterephthalate (DMT), ethylene glycol (EG) and 2,2-bis(4-(2-hydroxypropoxy) phenyl)propane (HPP). The branched copolyesters prepared with various branching agents such as 2-(hydroxymethyl)-2-ethylpropane-1,3-diol (trimethylol propane, TMP), 2,2-bi(hydroxymethyl)-1,3-propanediol (pentaerythritol, PER), 1,2,4-benzenetricarboxylic anhydride (trimellitic anhydride, TMA) and glycerol to improve the physical properties of the linear copolyester. The effect of branching agents on the molecular weight and melt elasticity of the branched copolyester was examined. The branched copolyesters prepared by adding over 15 mol% of branching agent showed relatively high molecular weight and melt elasticity, and $T_{1/2}$ value of $140^{\circ}C$. Therefore, the highly branched copolyesters were deemed suitable as a hot-melt toner of laser print process.

Physical Characterizations and In Vitro Skin Permeation of Elastic Liposomes for Transdermal Delivery of Polygonum aviculare L. Extract (마디풀 추출물의 경피 전달을 위한 탄성 리포좀의 물리적 특성 및 In Vitro 피부 투과 연구)

  • In this study, Polygomun aviculare L. (P. aviculare L.) extract loaded elastic liposomes (ELPs) were investigated to enhance the transdermal delivery of P. aviculare L. extract composed of various flavonoids. ELPs were composed of egg phospholipids (PC) and edge activator ($Tego^{(R)}$ care 450) and the physical properties and in vitro permeation studies of ELPs were performed. Particle size ranged from 148.1 to 262.2 nm and deformability index was recorded as 11.5~25.4. Loading efficiency was from 53.1 to 66.3%. In vitro skin permeation studies using Franz diffusion cell demonstrated that ELP-4 having ratio of 85:15 for PC to $Tego^{(R)}$ care 450 exhibited the higher skin permeability than ELP-1, the general liposome without $Tego^{(R)}$ care 450. It was visually seen by fluorescence image restoration microscopy. The findings suggest that ELP-4 selected as the optimal formulation could be used as useful formulation for transdermal delivery of the extract.

Preparation of Coil-Embolic Material Using Syndiotactic Poly(vinyl alcohol) Gel Spun Fibers (교대배열 PVA 젤 섬유를 이용한 고분자 색전 코일 제조)

  • The structure, morphology, and physical properties of syndiotatic poly(vinyl alcohol) (s-PVA) gel spun fibers were investigated to prepare polymeric embolization coils. S-PVA was prepared by saponification of the poly(vinyl acetate)/poly(vinyl pivalate)(PVAc/PVPi) copolymer. The viscosity of s-PVA solutions showed shear thinning behavior and the solution formed a homogeneous phase. Based on shear viscosity change with concentration, the optimum dope concentration was selected as 13 wt%, after which s-PVA fibers were spun and the solvent was removed. The fibers were then drawn with a maximum draw ratio of 15. A polymeric embolization coil was made of the s-PVA gel-spun fibers. The fibers were wound densely onto rigid rod and then annealed at different annealing temperatures. The polymeric embolization coil annealed at $200^{\circ}C$ was similar to metallic coils and its shape was maintained well after extension. Overall, gel-spun PVA fibers performed well for the preparation of primary and secondary coils to replace metallic coils.

  • Pigs have been considered as an ideal source of donor organs because of their plentiful supply and their numerous anatomical and physiological similarities to the human in xenotransplantation. However, for the public health risks associated with the potential for porcine endogenous retrovirus(PERV) infection through xenograft from pig to human, the investigation of methods for elimination and/or control of PERV has been required. In this study we developed the detection and classification methods for PERV based on PCR using specific primers. PERV-A and PERV-B were found in all pigs including Berkshire, Duroc, Landrace, Yorkshire, miniature pig, and Korean native black pig from Jeju by PCR with type-specific primers for PERV. However, PERV-C was detected only from Duroc, miniature pig, and Korean native black pig from Jeju. PERV-A and PERV-B could be distinguished by PCR-RFLP with BamHI. These methods for PERV will be useful in rapid screening of safe organ for xenograft, furthermore, helpful in monitoring of PERV during and after xenotransplantation.

Effect of Sodium Lignosulfonate Treatment on the Dispersion of CaCO3 in CaCo3/Polypropylene Composite (Sodium Lignosulfonate 표면처리가 탄산칼슘/폴리프로필렌 복합체에서 탄산칼슘의 분산에 미치는 영향)

  • The dispersion of calcium carbonate ($CaCO_3$) in polypropylene (PP) and the effect of $CaCO_3$ size on the crystallinity of PP were studied. Polymer composite usually suffers from the brittleness when reinforced with inorganic fillers. The problem is generally related to the size and dispersion of fillers. First, the dispersion was studied for the nanosize $CaCO_3$ with 15~40 nm average diameter. To enhance the dispersibility in PP, the surface of the $CaCO_3$ was treated with sodium lignosulfonate (SLS). $CaCO_3$/PP composites were prepared via melt compounding. The $CaCO_3$ coated with more than 3 wt% SLS was uniformly distributed within the PP matrix, while the uncoated $CaCO_3$ formed aggregated structures in the PP. Even with 30 wt%, the SLS-$CaCO_3$ was well dispersed in the PP matrix. Also, the transition enthalpy of $CaCO_3$/PP increased and the full-width of half maximum of the crystallization peak decreased regardless of SLS coating and size of $CaCO_3$. However, the crystallinity of PP was more influenced by nano $CaCO_3$. These results imply that the nano $CaCO_3$ coated with SLS may reduce the brittleness of polymer composites.

Involvement of Peripheral Benzodiazepine Receptor on the Contractility of Canine Trachealis Muscle (기관근의 수축성에 대한 말초성 Benzodiazepine 수용체의 역할)

  • Non-neuronal high affinity binding sites for benzodiazepines have been found in many peripheral tissues including cardiac muscle and vascular smooth muscle, and have been designated as 'peripheral benzodiazepine receptor'. Benzodiazepines have been shown to induce relaxation of the ileal, vesical, and uterine smooth muscles. However, it is still unclear about possible involvement of peripheral benzodiazepine receptor on the contractility of trachealis muscle. This study was performed to investigate the role of the peripheral benzodiazepine receptor on the contractility of canine trachealis muscle. Canine trachealis muscle strips of 15 mm long were suspended in an isolated organ bath containing 1 ml of physiological salt solution maintained at $37^{\circ}C$, and aerated with $95%\;O_2/5%\;CO_2$. Isometric myography was performed, and the results of the experiments were as follows: Ro5-4684, FGIN-1-27 and clonazepam reduced a basal tone of isolated canine trachealis muscle strip concentration dependently, relaxant actions of RoS-4684 and FGIN-1-27 were antagonized by PK11195, a peripheral benzodiazepine receptor antagonist. Flumazenil, a central type antagonist, did not antagonize the relaxant action of Peripheral type agonists. Saturation binding assay of [3H]Ro5-4864 showed a high affinity$(Kd=5.33{\pm}1.27nM,\;Bmax=\;867.3{\pm}147.2\;fmol/mg\;protein)$ binding site on the canine trachealis muscle. Ro 5-4684 suppressed the bethanechol-, 5-hydroxyoyptamine- and histamine- induced contractions. Platelet activating factor (PAF) exerted strong and prolonged contraction in trachealis muscle strip. Strong tonic contraction by PAE was attenuated by Ro 5-4684, but not by WEB 2086, a PAF antagonist. Based on these results, it is concluded that the peripheral benzodiazepine receptor mediates the inhibitory regulation of contractilty of canine trachealis muscle.

Expression characterization and transcription regulation analysis of porcine Yip1 domain family member 3 gene

  • Objective: The Yip1 domain family (YIPF) proteins were proposed to function in endoplasmic reticulum (ER) to Golgi transport and maintenance of the morphology of the Golgi, which were homologues of yeast Yip1p and Yif1p. YIPF3, the member 3 of YIPF family was a homolog of Yif1p. The aim of present study was to investigate the expression and regulation mechanism of porcine YIPF3. Methods: Quantitative realtime polymerase chain reaction (qPCR) was used to analyze porcine YIPF3 mRNA expression pattern in different tissues and pig kidney epithelial (PK15) cells stimulated by polyinosine-polycytidylic acid (poly [I:C]). Site-directed mutations combined with dual luciferase reporter assays and electrophoretic mobility shift assay (EMSA) were employed to reveal transcription regulation mechanism of porcine YIPF3. Results: Results showed that the mRNA of porcine YIPF3 (pYIPF3) was widely expressed with the highest levels in lymph and lung followed by spleen and liver, while weak in heart and skeletal muscle. Subcellular localization results indicated that it expressed in Golgi apparatus and plasma membranes. Upon stimulation with poly (I:C), the level of this gene was dramatically up-regulated in a time- and concentration-dependent manner. pYIPF3 core promoter region harbored three cis-acting elements which were bound by ETS proto-oncogene 2 (ETS2), zinc finger and BTB domain containing 4 (ZBTB4), and zinc finger and BTB domain containing 14 (ZBTB14), respectively. In which, ETS2 and ZBTB4 both promoted pYIPF3 transcription activity while ZBTB14 inhibited it, and these three transcription factors all played important regulation roles in tumorigenesis and apoptosis. Conclusion: The pYIPF3 mRNA expression was regulated by ETS2, ZBTB4, and ZBTB14, and its higher expression in immune organs might contribute to enhancing ER to Golgi transport of proteins, thus adapting to the immune response.

Adhesion Behavior of Chondrocyte and Osteoblast on Surface-Modified Biodegradable PLLA Films and Scaffolds (표면개질된 생분해성 PLLA 필름 및 지지체의 연골세포와 조골세포 점착거동)

  • Surface-modified poly(L-lactic acid) (PLLA) films and scaffolds were treated with plasma discharge in oxygen gas and subsequently subjected to $in$ $situ$ grafting of acrylic acid (AA) in order to increase the cell compatibility. The surface of AA-grafted PLLA was converted to hydroxyapatite (HA)-deposited PLLA in stimulated body fluid (SBF). After the samples were immersed in phosphate-buffered saline (PBS), fetal bovine serum (FBS), normal saline, or cell medium, the water contact angles were significantly reduced on the surface of HA-deposited PLLA. Chondrocyte and osteoblast showed a higher attachment and cell proliferation on HA-deposited surfaces and in particular, it was confirmed that chondrocyte was considerably influenced by HA. However, osteoblast showed better cell proliferation on the surfaces immersed in FBS, cell medium or HA-deposited surface. In addition, the cell proliferation in 3D scaffolds was much higher than that on film type, irrespective of chondrocyte and osteoblast. Therefore, such surface-modified PLLAs are expected to be useful as organic-inorganic hybrid scaffolds in the regeneration of cartilage and bone.
