Kiwifruit 과육에 존재하는 단백질분해효소의 특성과 열안정성 (Properties and Thermostability of Gelatin-degrading Proteinases in the Fruit of Actinidia chinensis (Kiwifruit))

  • 본 연구에서는 단백질분해효소의 산업적 이용을 위하여 kiwifruit 과육 속에 들어 있는 gelatin분해활성을 조사하였다. Kiwifruit 과육에는 3개의 단백질분해효소의 활성 밴드(PI, PII, PIII)가 관찰되었다. 단백질분해효소 PI은 220 kD, PII는 51 kD, PIII는 26 kD에 해당하는 것으로 추정할 수 있었다. 이들 단백질분해효소 PI, PII, PIII는 모두 pH 2.0~5.0 범위에서 높은 활성을 보였으며 pH 4.0에서 가장 높게 나타났다. 이들 단백질분해효소 PI, PII, PIII는 모두 cysteine proteinase 저해제인 E-64와 iodoacetate에 의해서 저해되었으며, cysteine proteinase를 촉진하는 DTT, cysteine 및 $\beta$-mercaptoethanol에 의해서 활성이 증가하였다. 그 중 단백질분해효소 PIII는 분자량과 효소의 특성으로 보아 actinidin (EC 3.4.22.14)과 동일한 것으로 판단되었다. 단백질분해효소 PI, PII, PIII는 모두 $Ca^{2+}$, $Mg^{2+}$$Mn^{2+}$에 의해 촉진되었으며 $Zn^{2+}$$Hg^{2+}$에 의해 완전히 저해되는 것으로 나타났다. 하지만, $Co^{2+}$, $Cu^{2+}$, $Al^{3+}$, $Fe^{3+}$ 등 금속이온의 영향이 다소 다르게 나타났다. Kiwifruit 과육의 단백질분해효소 PI, PII, PIII 중에서 PI과 PII는 온도가 증가함에 따라 활성이 점차 낮아졌으나 PIII는 비교적 안정한 것으로 조사되었다. 특히, PIII는 $50^{\circ}C$ 이내의 범위에서 48시간 경과시에도 75% 이상의 활성을 보여 이 범위의 온도에서는 상당 시간 동안 안정한 것으로 나타났다.

초등수학 영재교육 대상자의 원주율 개념에 대한 이해 (Elementary mathematically gifted students' understanding of Pi)

  • 본 연구는 초등수학 영재교육 대상자들이 원주율 개념에 대해서 어떻게 이해하고 있는지를 살펴보고자 하였다. 이를 위해 원주율 계산 방법의 역사 발달 단계를 토대로 세 가지 과제를 개발한 후 6학년 영재교육 대상자 12명을 대상으로 적용하여 그 반응을 분석하였다. 연구결과, 학생들은 '원주율 = 3.14'라는 사고의 고착화로 인하여 원주율의 개념, 근사성, 무한성을 제대로 이해하지 못하였으며, 원주율과 원주율의 근삿값을 혼동하는 오류를 보였다. 또한 학생들은 원주율을 '(원주) ${\div}$ (지름)'의 대수적인 식으로 이해하려는 성향이 강하였으며, 원주율의 상수성과 무한성을 깊이 있게 이해하고 있는 학생은 극히 적었다. 반면에 과제에 대한 토론 활동은 학생들이 원주율의 근사성에 대한 아이디어를 발견할 수 있는 기회를 제공하였다. 이상의 결과를 종합하여, 초등학교에서의 원주율 지도와 관련하여 원주율을 원의 지름을 단위길이로 원의 둘레를 측정하여 얻을 수 있는 값으로 도입할 것과 공학적 도구 등을 이용하여 직관적인 방법을 통해 이해하도록 할 것, 원주율 개념이 가지는 본질적인 의미를 이해할 수 있도록 다양한 상황을 통해 도입할 것을 제안하였다.

구리 산화물 계열 초전도체에서의 스핀 요동에 의한 자체 에너지의 엘리아시버그 계산 (Eliashberg Calculation of the Momentum-Resolved Self-Energy for the Cuprate Superconductors Induced by the Spin Fluctuations)

  • We solve the momentum resolved d-wave Eliashberg equation employing the magnetic excitation spectrum from the inelastic neutron scattering on the LSCO superconductors reported by Vignolle et al. The magnetic excitation spectrum exhibits 2 peaks: a sharp incommensurate peak at 18 meV at momentum (${\pi}$, ${\pi}{\pm}{\delta}$) and (${\pi}{\pm}{\delta}$, ${\pi}$) and another broad peak near 40~70 meV at momentum (${\pi}$, ${\pi}$). Above 70 meV, the magnetic excitation spectrum has a long tail that is shaped into a circle centered at (${\pi}$, ${\pi}$) with ${\delta}$. The sign of the real part of the self-energy is determined by the momentum position of the peaks of the magnetic excitation spectrum and bare dispersion. We will discuss the effects of the each component of the magnetic excitation spectrum on the self-energy, the pairing self-energy.

20(S)-Ginsenoside Rh2 displays efficacy against T-cell acute lymphoblastic leukemia through the PI3K/Akt/mTOR signal pathway

  • Background: T-cell acute lymphoblastic leukemia (T-ALL) is a kind of aggressive hematological cancer, and the PI3K/Akt/mTOR signaling pathway is activated in most patients with T-ALL and responsible for poor prognosis. 20(S)-Ginsenoside Rh2 (20(S)-GRh2) is a major active compound extracted from ginseng, which exhibits anti-cancer effects. However, the underlying anticancer mechanisms of 20(S)-GRh2 targeting the PI3K/Akt/mTOR pathway in T-ALL have not been explored. Methods: Cell growth and cell cycle were determined to investigate the effect of 20(S)-GRh2 on ALL cells. PI3K/Akt/mTOR pathway-related proteins were detected in 20(S)-GRh2-treated Jurkat cells by immunoblotting. Antitumor effect of 20(S)-GRh2 against T-ALL was investigated in xenograft mice. The mechanisms of 20(S)-GRh2 against T-ALL were examined by cell proliferation, apoptosis, and autophagy. Results: In the present study, the results showed that 20(S)-GRh2 decreased cell growth and arrested cell cycle at the G1 phase in ALL cells. 20(S)-GRh2 induced apoptosis through enhancing reactive oxygen species generation and upregulating apoptosis-related proteins. 20(S)-GRh2 significantly elevated the levels of pEGFP-LC3 and autophagy-related proteins in Jurkat cells. Furthermore, the PI3K/Akt/mTOR signaling pathway was effectively blocked by 20(S)-GRh2. 20(S)-GRh2 suppressed cell proliferation and promoted apoptosis and autophagy by suppressing the PI3K/Akt/mTOR pathway in Jurkat cells. Finally, 20(S)-GRh2 alleviated symptoms of leukemia and reduced the number of white blood cells and CD3 staining in the spleen of xenograft mice, indicating antitumor effects against T-ALL in vivo. Conclusion: These findings indicate that 20(S)-GRh2 exhibits beneficial effects against T-ALL through the PI3K/Akt/mTOR pathway and could be a natural product of novel target for T-ALL therapy.

  • 연안해역에서 호기성 광합성 세균 Erythrobacter sp. $S\;\pi-I$I를 분리 및 동정하여 rotifer, Brachionus plicatilis의 먹이로 투여하였을 때 개체수, 체장, 체중 및 지방산 조성과 아미노산 조성을 분석한 결과를 요약하면 아래와 같다. 1. Erythrobacter sp. $S\;\pi-I$ 은 지방산의 함량중 $C_{18:1\omega9}$$36.5\%,\;C_{18:1\omega7}$$13.3\%\;C_{18:3\omega3}\;8.6\%$ 함유하였고 아미노산은 glutamic acid가 $61.6\;mg\%$, aspartic acid가 $45.2\;mg\%$, alanine이 $43.7\;mg\%$, lysine이 $28.7\;mg\%$ 등 높은 함량을 보였다. 2. Erythrobacter sp. $S\;\pi-I$을 rotifer, Brachionus plicatilis에 투여했을 때 (초기 개체수 5개체/$m\ell$) 개체수는 5일째 9개체/$m\ell$, 9일째 20개체/$m\ell$, 13일째는 43개체/$m\ell$로 증가하였다. 또한 체중은 초기 140ng/개체에서 5일째 280ng/개체, 9일째 560ng/개체, 13일째 610ng/개체로 증가하였으며, 체장은 초기 $150{\pm}7{\mu}m\;(length),\;85{\pm}14{\mu}m\;(width)$에서 5일째 $160{\pm}18{\mu}m\;(length),\;92{\pm}15{\mu}m\;(width)$, 9일째 $170{\pm}17{\mu}m\;(length),\;94{\pm}14{\mu}m\;(width)$, 13일째는 $184{\pm}17{\mu}m\;(length),\;100{\pm}12{\mu}m\;(width)$로 성장하였다. 3. Rotifer 개체당 24시간내에 $10^4\~10^5$개체의 세균을 섭취하였다. 4. Erythrobacter sp. $S\;\pi-I$을 rotifer에 먹이로 투여했을때 EPA, DHA가 증가하였다. 비교실험에 사용한 Chlorella sp. PSB, yeast에 비하여 EPA와 DHA의 함량이 높았다. 특히DBHA는 $S\;\pi-I$과 Chlorella sp.에서만 분석되었으며 $S\;\pi-I$이 높았다. 5. Erythrobacter sp. $S\;\pi-I$과 비교로서 PSB, Chlorella sp., yeast를 rotifer에 먹이로 투여했을 때 $S\;\pi-I$에서 lysine이 가장 높았고, PSB, Chlorellaal sp. 순으로 분석되었다. yeast에서는 검출되지 않았다.

Mycobacterial Heparin-binding Hemagglutinin Antigen Activates Inflammatory Responses through PI3-K/Akt, NF-${\kappa}B$, and MAPK Pathways

  • Background: Mycobacterium tuberculosis (Mtb) heparin binding hemagglutinin (HBHA) is an Ag known to evoke effective host immune responses during tuberculosis infection. However, the molecular basis of the host immune response to HBHA has not been fully characterized. In this study, we examined the molecular mechanisms by which HBHA can induce the expression of proinflammatory cytokines in macrophages. Methods: HBHA-induced mRNA and protein levels of proinflammatory cytokines were determined in bone marrow-derived macrophages (BMDMs) using RT-PCR and ELISA analysis. The roles of intracellular signaling pathways for NF-${\kappa}B$, PI3-K/Akt, and MAPKs were investigated in macrophage proinflammatory responses after stimulation with HBHA. Results: HBHA robustly activated the expression of mRNA and protein of both TNF-${\alpha}$ and IL-6, and induced phosphorylation of NF-${\kappa}B$, Akt, and MAPKs in BMDMs. Both TNF-${\alpha}$ and IL-6 production by HBHA was regulated by the NF-${\kappa}B$, PI3-K, and MAPK pathways. Furthermore, PI3-K activity was required for the HBHA-induced activation of ERK1/2 and p38 MAPK, but not JNK, pathways. Conclusion: These data suggest that mycobacterial HBHA significantly induces proinflammatory responses through crosstalk between the PI3-K and MAPK pathways in macrophages.

  • To improve the performance of PID controller of high order systems by model reduction, we proposed a new model reduction method in frequency domain. A new model reduction method we proposed, considered four points (${\angle}G(jw)=0$, $-{\pi}/2$, $-{\pi}$, $-3{\pi}/2$) in stead of two points (${\angle}G(jw)=-{\pi}/2$, and $-{\pi}$) in Nyquist curve. And for high order systems that it have not two point (${\angle}G(jw)=-{\pi}/2$, and $-{\pi}$) in Nyquist curve, we proposed a method to annex very small dead time. This method has a annexed very small dead time on the base model for reduction, and we cancel it after to get the reduced model. It is shown that the performance of proposed method is better than any other methods.

The existence of solutions of a nonlinear suspension bridge equation

  • In this paper we investigate a relation between the multiplicity of solutions and source terms in a nonlinear suspension bridge equation in the interval $(-\frac{2}{\pi}, \frac{2}{\pi})$, under Dirichlet boundary condition $$ (0.1) u_{tt} + u_{xxxx} + bu^+ = f(x) in (-\frac{2}{\pi}, \frac{2}{\pi}) \times R, $$ $$ (0.2) u(\pm\frac{2}{\pi}, t) = u_{xx}(\pm\frac{2}{\pi}, t) = 0, $$ $$ (0.3) u is \pi - periodic in t and even in x and t, $$ where the nonlinearity - $(bu^+)$ crosses an eigenvalue $\lambda_{10}$. This equation represents a bending beam supported by cables under a load f. The constant b represents the restoring force if the cables stretch. The nonlinearity $u^+$ models the fact that cables expansion but do not resist compression.

Effects of Amino Substitution on the Excited State Hydrogen Transfer in Phenol: A TDDFT Study

  • When isolated phenol or a small phenol-solvent cluster is excited to the $S_1\;state\;of\;{\pi}{\pi}^*$ character, the hydrogen atom of the hydroxyl group dissociates via a ${\pi}{\sigma}^*$ state that is repulsive along the O-H bond. We computationally investigated the substitution effects of an amino group on the excited state hydrogen transfer reaction of phenol. The time-dependent density functional theory (TDDFT) with B3LYP functional was employed to calculate the potential energy profiles of the ${\pi}{\pi}^*$ and the ${\pi}{\sigma}^*$ excited states along the O-H coordinate, together with the orbital shape at each point, as the position of the substituent was varied. It was found that the amino substitution has an effect of lowering the ${\pi}{\sigma}^*$ state and enhancing the excited state hydrogen transfer reaction.

Phosphatidylinositol 3-kinase (PI3KCA) Oncogene Mutation Analysis and Gene Expression Profiling in Primary Breast Cancer Patients

  • Background: The phosphatidylinositol 3-kinase (PI3K) pathway plays a significant role in apoptosis, cellular proliferation and motility. The aim of the present study was to analyze mutations and gene expression profiles of the PI3KCA gene to determine any role in breast carcinomas. Materials and Methods: We analyzed 38 breast cancers for mutations in the two PIK3CA hotspots in exons 9 and 20 by direct sequencing of DNA obtained from biopsy samples. We have also analyzed expression of the PI3KCA gene in 38 breast carcinoma tumor and corresponding control tissue samples at the mRNA level by RT-PCR. The Fisher's exact test ($2{\times}2$ only) was performed using MedCalc software for to examine associations with mRNA levels. Results: In the present study a total of 13 cases demonstrated somatic mutations. In 9/13 cases 1633 G>A (E545K) were found in exon 9, whereas in exon 20, 4/13 cases had 3140A>G mutation. Our combined analysis showed PI3KCA mutations present in 34% of human breast cancer patients. In our study, we have also clearly found significantly higher expression in breast cancer tissues in comparison with control tissues (p=0.001). Conclusions: PIK3CA mutation is an emerging tumor marker that, in the future, might be used in the process of choosing a treatment. The detection of PI3KCA mutation might have important clinical implications for diagnosis, progression and therapy.
