Effect of Vacuum Packaging on the Microbial Change of Chicken during Storage

  • To examine the effect of vacuum packaging on the quality of chicken during storage, microbial (total bacterial counts, mold and yeast, E. coli, and Pseudomonas) changes and drip loss were determined. fresh chicken breasts were prepared and packaged using polyethylene film under vacuum and normal atmosphere, respectively. Samples were then stored at 4$^{\circ}C$ for two weeks. At various time intervals during storage, sample was taken homogenized, and diluted with 0.1% sterile peptone water, and followed by microbial counts. Microbial changes of total bacterial counts, mold and yeast, E. coli, and Pseudomonas during storage of chicken were significantly decreased by vacuum packaging. Drip loss was also significantly decreased. These results indicate that vacuum packaging of chicken should be recommended as a suitable storage method in terms of microbial safety as well as quality of chicken.

Production of the Bacteriocin from the Tofu-Residue (두부비지를 이용한 박테리오신 생산)

  • Growth and bacteriocin production by Lactobacillus sp. GM7311 in tofu residue treated with two commercial amylases were investigated. The optimal condition of amylase Ⅰ(liquefied enzyme for sauce) and Ⅱ(multienzyme 2,000) for the enzyme reaction was showed at pH 6.0 and 4.0, respectively. The optimal temperature was 40oC both. At the enzyme dosage 4% and 3% and reaction time 1hr, about 2% of reduced sugar needed bacteriocin production was obtained. The enzymatic treatment of tofu residue enhanced bacteriocin production by lactic acid bacteria, particularly in the tofu residues added 2.0% yeast extract. But, we couldn't see the increment of bacteriocin activity in the tofu residues added other nitrogen sources such as proteose peptone No. 3 and lab lemco powder. Also, in the comparision of amylase I and Ⅱ, bacteriocin activity in the tofu residue treated with amylase Ⅰ was better than that of amylase Ⅱ.

  • In order to use as a starter after drying, optimum growth conditions of Str. thermophilus and L. bulgaricus were investigated. The results were as follows. The growth of Str. thermophilus was reached at stationary phase after 12 hours incubation and L. bulgaricus was after 6 hours. Str. thermophilus was grown well, in the medium containing 10% of skim milk, 2% of sucrose, 0.1% of KCl and 0.015% (v./v.) of soy-sauce. In the case of L. bulgaricus, 10% of skim milk, 2% of glucose, 0.5% of bacto-peptone, 0.1% of $K_2HPO_4$ and 0.02% of pyridoxine was suitable for the maximum growth.

  • 1. Amylase producing bacteria have been isolated from natural sources and was identified out. The above strain is identical to Bacillus subtillis in every details of physiological and morphorogical characteristics by this investigations. 2. Sikworm chrysalis nad chlorella extracts are prove to be further effective for the stimulation of amylase production than any other sources. The former silkworm extract is more stronger in its activity. 3. It is found that effective results had been accepted by a less soybean meal addition per that of wheat bran in the mixture ratio of them, also less addition of corn is needed in the mixture ratio of between corn and whear bran. In the mixing ratio of wheat bran per silkworm or chlorella extracts, the addition of wheat bran is necessary for stimulation of amylase production. 4. Fro the stimulation of amylase production, the effective substances from natural sources and its order of activity in cultral media cultral media is proved as follows, silkworms ext.>chlorella ext.>alcohol>skim-milk and peptone>fermented milk>radish leaf ext.

Suitable Conditions for Mycelial Growth of Phellinus spp.

  • The fungus Phellinus is a mushroom that is widely used medicinally. The optimal conditions for mycelial growth of 13 strains of the fungus were investigated. Mycelial growth was optimal at 25$^{\circ}C$ and was uniformly minimal at 15$^{\circ}C$ and 35$^{\circ}C$. Growth was optimal at pH 6$\sim$7. The mycelial phenotype was best promoted by growth using Potato Dextrose agar, Hamada, Glucose peptone, and Yeast-Malt media, whereas Czapek Dox, Hennerberg, and Lilly media were the most unfavorable for the mycelial growth of Phellinus spp. Glucose, sucrose, fructose, and dextrin were the most suitable carbon sources for mycelial growth, while lactose, maltose, and galactose were unsuitable. Among tested nitrogen sources, ammonium phosphate, potassium nitrate, and arginine best promoted mycelial growth, while alanine, urea, and histidine least promoted mycelial growth.

  • Lactophoricin (LPcin-I) is a cationic amphipathic peptide with 23-mer peptide, and corresponds to the carboxy terminal 113-135 region of Component-3 of proteose-peptone. LPcin-I is a good candidate as a peptide antibiotic, because it has an antibacterial activity, but no hemolytic activity. On the other hand, its shorter analog (LPcin-II), which corresponds to the 119-135 region of PP3, has no antibacterial activity. In order to understand the structure-activity relationship under the membrane environments, we succeed to produce large amounts of LPcin-I and LPcin-II peptides. Peptides were over expressed in the form of fusion protein in Escherichia coli, and purified with several chromatography techniques. In this paper, we introduce the optimizing processes of purification and NMR measurement.

Production of Anti-dementia Acetylcholinesterase Inhibitors from the Wild Yeasts Saccharomyces cerevisiae WJSL0113 and Wickerhamomyces anomalus JSF0128

  • In this paper, the screening of potent acetylcholinesterase (AChE) inhibitor - producing yeasts from wild yeasts and the condition for the production of anti-dementia AChE inhibitors are described. Among one hundred and seven non-pathogenic wild yeast strains from the waters and soils of three main rivers in Daejeon metropolitan city and midstream of Yeongsangang river in Sangju, sporogenous Saccharomyces cerevisiae WJSL0113 and asporogenous Wickerhamomyces anomalus JSF0128 were selected as useful strains for the production of potent AChE inhibitors. The AChE inhibitors of S. cerevisiae WJSL0113 and W. anomalus JSF0128 had a maximum yield when they were incubated in yeast extract-peptone-dextrose media (pH 6.0 in S. cerevisiae WJSL0113 and pH 5.0 in W. anomalus JSF0128) for 18 hr at $30^{\circ}C$, respectively.

Establishment of Sample Preparation Method to Enhance Recovery of Food-borne Pathogens from Produce (농산물 중 식중독세균 검출을 위한 전처리법 확립)

  • To establish sample preparation method for detection of food-borne pathogens from lettuce, perilla leaves, cucumber, pepper, and cherry tomato, the influences of diluent composition, processing time, and proportion of diluent to sample were examined. Each produce was inoculated with 6.0 log $CFU/cm^2$ of Escherichia coli O157:H7, Salmonella Typhimurium, Listeria monocytogenes, Staphylococcus aureus, and Bacillus cereus. Each produce was treated with 0.1% peptone water, and D/E neutralizing broth. Processing time of produce was 30, 60, 90, and 120s, and the proportion of diluent to sample was 2 : 1, 4 : 1, 9 : 1, and 19 : 1. The number of bacteria after treatment of D/E neutralizing broth was higher than that of 0.1% peptone water (P<0.05). In cherry tomato, the population of S. typhimurium recovered from treated with D/E broth was higher than that recovered from treated with 0.1% peptone water by 1.05 log $CFU/cm^2$ (P<0.05). No difference in numbers of pathogens was observed in processing time. Optimum proportion of diluent to perilla leaf, iceberg lettuce, cucumber, green pepper, and tomato was 9 : 1, 4 : 1, 2 : 1, 2 : 1, and 2 : 1, respectively. These data suggest that D/E neutralizing broth should be recommend as diluent, and the diluent volume applied to produce should be determined in proportion to produce surface area per weight (g).

  • The characteristics of the six different agricultural soil from Michigan were as follows. Colwood and Capac soil were loam, Gilford and Ceresco were sandy clay loam, Sission was clay loam, and Spinks was sandy loam. pH of Gilford sandy clay loam was 6.6 whereas that of the soil ranged $5.4{\sim}5.9$. Gilford sandy loam found to contain a relatively higher amount of organic matters as compared to other soils. Furthermore, the numbers of bacteria in Gilford sandy clay loam were significantly higher than those in other soils. The populations of fungi in Gilford sandy clay loam and Colwood loam soils were significantly greater than those in other soils. On the other hand, the densities of actinomycetes in Gilford sandy clay loam and Ceresco sandy clay loam soils were significantly different from those in other soils. The population of anaerobic bacteria varied depending on the soils; Ceresco sandy clay loam, Capac loam, Colwood loam soils have higher numbers of bacteria, whereas Gilford sandy clay loam was very lesser than the other soils. In the $^{14}C-glucose$ respiration by soil microorganisms after 10 hrs, the respiration rate was decreasing in the order of Ceresco sandy clay loam, Spinks sandy loam, Colwood loam, Sission clay loam, Capac loam and Gilford sandy clay loam. Germination of test propagules on natural soil soil was $0{\sim}5%$, and it was germinated $90{\sim}98%$ on autoclaved soil and PDA. The propagules differed in thier germination response to nutrients added to the soils. In general, more nutrients were required to promote germination on Capac loam and Gilford sandy loam soil than Spinks sandy loam soil. Especially Mortierella n. sp. required more nutrients for germination to obtain the same ratio as Helminthosporium victoriae.

  • Penicillium sp I which produces a powerful hydrolysing enzyme was isolated from putrefid and dry Jerusalem artichoke medium. The strain was used to study on the optimum culture conditions for enzyme production. The results obtained are as follows: 1. Penicillium sp I was a vigorous strain to produce inulase. 2. The optimum culture conditions of the strain was examined in the Jerusalem artichoke extract medium and the synthetic medium. 3. Inulase productivity in the Jerusalem artichoke extract medium was higher than that of the synthetic medium. 4. The optimum culture period of the Jerusalem artichoke extract medium was four days, whereas that of the synthetic medium was five days. 5. The optimum temperature, pH and concentration in the Jerusalem artichoke extract medium were $30^{\circ}C$, 5.0 and 4.0% (W/V), respectively. Meanwhile, the optimum temperature, pH and concentration in the synthetic medium were $30{\sim}33^{\circ}C$, $5.0{\sim}6.0$, and $1.0{\sim}1.5%$ (W/V), respectively. 6. Corn steep liquor, peptone, $(NH_4)_2HPO_4,\;NH_4H_2PO_4,\;(NH_4)_2SO_4$, etc. were favorable as nitrogen sources. Of these, especially, Corn steep liquor and peptone as organic nitrogen sources caused an increase in inulase production in the synthetic medium. 7. All sugars except for inulin have no effect upon the inulase production. 8. KCl, $MgSO_4\;and\;FeSO_4$ were favourable mineral sources for inulase production.
