What Event-Related Potential Tells Us about Brain Function: Child-Adolescent Psychiatric Perspectives

  • Electroencephalography (EEG) measures neural activation due to various cognitive processes. EEG and event-related potentials (ERPs) are widely used in studies investigating psychopathology and neural substrates of psychiatric diseases in children and adolescents. The present study aimed to review recent ERP studies in child and adolescent psychiatry. ERPs are non-invasive methods for studying synaptic functions in the brain. ERP might be a candidate biomarker in child-adolescent psychiatry, considering its ability to reflect cognitive and behavioral functions in humans. For the EEG study of psychiatric diseases in children and adolescents, several ERP components have been used, such as mismatch negativity, P300, error-related negativity (ERN), and reward positivity (RewP). Regarding executive functions and inhibition in patients with attention-deficit/hyperactivity disorder (ADHD), P300 latency, and ERN were significantly different in patients with ADHD compared to those in the healthy population. ERN showed meaningful changes in patients with anxiety disorders, such as generalized anxiety disorder, separation anxiety disorder, and obsessive-compulsive disorder. Patients with depression showed significantly attenuated RewP compared to the healthy population, which was related to the symptoms of anhedonia.

The effects on Jowiseungchungtang of patients with early DAT using auditory ERP and K-DRS (알츠하이머형 치매환자에 대한 조위승청탕(調胃升淸湯)의 효능 -청각 ERP 및 K-DRS의 성적 변화를 통하여-)

  • Object : This study was designed to assess the protective effects of Jowiseungchungtang on cognitive decline of the patients with early DAT[Dementia of Alzheimer Type]. Method : 15 patients with early DAT and 10 healthy elderly subjects were measured by using auditory ERP and K-DRS, and after 9 months again. During 9 months the patients with early DAT were administered with Jowiseungchungtang. Result : 1. The score of K-DRS increased in two group but it was not significant. 2. The latency of ERP P300 component increased in 2nd test of the normal group, but did not increase in 2nd test of patients group. 3. The amplitude of ERP P300 component showed the significant difference in electrode location in patients group, but it did not show any typical property. Conclusion : The results suggest that Jowiseungchungtang may have protective effects on cognitive decline in the patients with early DAT.

The Effects on Kunneotang of Patients with Early Dementia of Alzheimer Type -12 Months Clinical Study- (초기 알츠하이머형 치매환자에 대한 건뇌탕(健腦湯)의 효능 -12 개월 임상 연구-)

  • Object : This study was designed to asses the protective effects of Kunneo tang on cognitive decline of the patients with early DAT. Method : Before administered with Kunneotang, 16 patients with early DAT were measured by auditory ERP, MMSE-K and K-DRS. During 12 months the patients with early DAT were administered with Kunneotang. After 12months the patients were measured by the same examinations again. Result : 1. The total score of MMSE-K decreased but it was not significant. 2. The total score of K-DRS increased but it was not significant. Among the five criteria(attention, memory, initiation/perseveration, construction, conceptualization), Initiation/perseveration score signifcantly decreased and there were no significant differences in the other scores. But, memory mean score had a tendency to increase. 3. The latency of ERP P300 component increased but it was not significant and the amplitude of ERP P300 component showed the significant difference. Conclusion : The results suggest that Kunneotang may have protective effects on cognitive decline in the patients with early DAT.

Induction of Ovulation by LHRH-a and Pimozide in the Bullhead, Pseudobagrus fulvidraco (LHRH-a 와 pimozide에 의한 동자개의 배란유도)

  • Effects of intraperitoneal administration of LHRH-a (luteinizing hormone releasing hormone analogue), pimozide and HCG (human chorionic gonadortropin) were studied on ovulation in the bullhead, Pseudobagrus. fulvidraco. Ovulatin was induced in 71.4, 80 and 100% by injection of HCG 5,000, 10,000 and 20,000 IU/kg of body weight, respectively. Also, the majority of fish injected with LHRH-a in the presence or absence of pimozide were ovulated. The ovulating rates in fish injected with LHRH-a alone showed 50, 75 and 100% at a dose 200, 300 and $400^{\mu}$g/kg, respectively. Pimozide enhanced the activity of LHRH-a at $300^{\mu}$g/kg plus pimozide at 1 mg/kg. Pimozide alone was ineffective in inducing ovulation of P. fulvidraco. There were no significant differences in gonadosomatic index (GSI) and pseudo-GSI values among the groups treated with HCG, LHRH-a and LHRH-a plus pimozide (P>0.05). The latency time groups treated with HCG was 17~22 hours, and the group injected with LHRH-a alone and/or in combination with pimozide was ovulated within 23~29 hours.

The Relationships between Inhibitory Control and Action Monitoring; Event-related Potential Study (억제적 통제 및 행동 감시간의 관계: 사건관련전위 연구)

  • The common features of the behavioral inhibition and the action monitoring that are considered as one of the executive functions were investigated using event-related brain potentials (ERPs) and source localization analysis. The electrophysiological correlates of behavioral inhibition and action monitoring ate analyzed when the subjects performed the Go/NoGo task. Two ERP components of behavioral inhibition termed as N200 and P300 in NoGo condition were differ from those of Go condition, that is the amplitudes of NoGo N200 and P300 are largest on the fronto-central region, which may reflect the inhibitory control of frontal lobe required in NoGo condition. The error-related negativity (ERN) observed on the fronto-central region when the subjects committed error was much larger in amplitude and faster in latency than those of the correct-related negativity (CRN), which may indicate that the signal of action monitoring is much more required for the error response. The correlation analysis for the ERP components of behavioral inhibition and action monitoring revealed the significant negative correlation among the latencies of NoGo N200 and P300 and the amplitude of ERN, which may reflects that the faster subjects inhibit response, the more monitor their own action. The close relationship between behavioral inhibition and action monitoring was also supported by the results of source localization analysis, which showed the common neural sources of NoGo N200 and ERN was anterior cingulate cortex.

  • A number of Brain-Computer Interface (BCI) studies have been performed to assess the cognitive status through EEG signal. However, there are a few studies trying to prevent user from unexpected safety-accident in BCI study. The EEGs were collected from 19 subjects who participated in two experiments (rest & event-related potential measurement). There was significant difference in EEG changes of both spontaneous and event-related potential. Beta power and P300 latency may be useful as a biomarker for prevention of response to safety-accident.

Analgesic and anti-inflammatory activity of a polyherbal formulation (PHFAROGH)

  • The effect of arogh, a polyherbal formulation-PHF [each 3 g powder contained Nelumbo nucifera G. (0.24 g), Hemidesmus indicus R. (0.24 g), Zingiber officinale R. (0.24 g), Terminalia chebula R. (0.24 g), Quercus infectoria O. (0.12 g), Hibiscus rosa-sinensis L. (0.24 g), Rosa damascene M.(0.24 g), Eclipta alba H.(0.24 g), Glycyrrhiza glabra L. (0.24 g)] was investigated in various experimental models of pain and inflammation. Analgesic activity of PHF was studied in mice using acetic acid induced writhing, tail immersion and hot plate methods. Anti-inflammatory activity of PHF was studied in rats using carrageenan induced hind paw edema and formalin induced rat paw edema methods. PHF significantly (P < 0.05) reduced the number of writhings, increased latency to flick tail in tail immersion method and elevated the mean basal reaction time in hot plate method. PHF significantly (P < 0.05) inhibited carrageenan induced hind paw edema and formalin induced rat paw edema. The PHF was tested at dose of 30, 100, 300 and 500 mg/kg.

Neuroprotective effect of Puerariae Radix extract on focal cerebral ischemia in mice (갈근 에탄올추출물의 국소뇌허혈 모델에 대한 뇌신경보호 효과)

  • Objectives : The purpose of this study was to evaluate the neuroprotective effect of Pueraria lobata extract on focal cerebral ischemia in mice. Methods : Focal cerebral ischemia was induced by occlusion of the right middle cerebral artery using the intraluminal filament model. ICR male mice underwent 90 minutes of middle cerebral artery occlusion (MCAo) followed by 24 hours of reperfusion. Mice were administered Pueraria lobata extract orally at the dose of 300mg/kg just prior to reperfusion. Rotarod test and balance beam test were practiced to assess sensory-motor function 23 hours after MCAo. In rotarod test, the latency to fall on the accelerating rotarod was recorded for 5 min. In balance beam test, the score was graded according to number of slips and latency to cross. The infarct volume was measured 24 hours after MCAo using 2% 2,3,5-triphenyltetrazolium chloride (TTC) staining. Results : Pueraria lobata extract treated group showed significant reduction in infarct volume by 27.3% compared to control group (p<0.05). In rotatod test, it also showed significant extension of latency time compared to control group ($67.82{\pm}15.08$ vs. $5.62{\pm}1.06$, p<0.001). In contrast to performance in rotarod test, that in balance beam test did not improve with Pueraria lobata extract treatment. Conclusions : We conclude that Pueraria lobata extract has a significant neuroprotective effect and reduces damage of sensory-motor function in MCAo model. These findings suggest that Pueraria lobata could be a potent neuroprotective agent.

The long-term effect of Interactive Video Game on Cognitive Information Processing the elderly: P300 (장기간의 상호작용적 비디오 게임이 노인의 인지정보처리에 미치는 영향: P300)

  • The objectives of this study was to examine the effect of Interactive Video Game on cognitive information processing the elderly. Sixty elderly were attended in this study. Their ages ranged from 65 to 70, with a mean age of 67.60 years. The subjects were randomly assigned to one of three experimental conditions: (1) interactive video game group (n=20), (2) aerobic exercise group (n=20), (3) control group (n=20). The experimental design of this study was analyzed using two-way ANOVAs with repeated measures of groups and time. Cognitive function was assessed by neuroelectrical response, and ERP analysis. The results of the study showed that the interactive video game group and aerobic exercise group showed no significant statistical differences in the response time, response accuracy, amplitude and potential of the performance of the exercise in cognitive function and ERP analysis, but improved the interaction video game group and aerobic exercise (walking) group over the control group. It was concluded that long-term aerobic exercise like interactive video game is associated with attenuation of cognitive decline in the elderly.
