향나무 심재 메탄올추출물의 염색성과 기능성 (Dyeing Properties and Functionality of Methanol Extract from Juniperus chinensis Heartwood)

  • In this study, we report dyeing properties and functionality of cotton and silk fabrics dyed with methanol extract from Juniperus chinensis heartwood. Non-aqueous dye of Juniperus chinensis heartwood extracted with methanol was expressed reddish color. It showed relatively good affinity to silk than cotton. Dye uptake of cotton and silk continued to increase according to the increase of the dye concentration. Cotton was not affected by the dyeing temperature and time, however, dye uptake of silk was increased as the dyeing time and temperature were increased. Dye uptake of cotton and silk were the highest at pH 8 and pH 6 respectively. Dyed fabrics showed various colors depending on mordant types and mordant concentration. Washing, light and perspiration fastness were not good. However, rubbing and dry cleaning fastness showed relatively good grade. Dyed fabrics of ultraviolet-cut ability and deodorant ability were improved. Also, dyed fabrics showed very good antimicrobial abilities of 99.9%.

  • Saururus chinensis Baill (Saururaceae) is a perennial plant that has been used in the treatment of edema, jaundice and gonorrhea in Korean folk medicine. This study was carried out to investigate the inhibitive effects of Saururus chinensis Baill (SCB) on lipid metabolism in Sprague-DaweIy rat(SD-rat) accutely exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD). After 7 days from TCDD(1$\mu$g/kg) injection, SCB(200mg/kg) was administered into rats intraperitoneally for 4 weeks. (omitted)

오미자 열수추출물의 대장암세포 증식억제 효과 (Effects on Hot Water Extract of Schizandra chinensis on Colon Cancer)

  • 본 실험에서는 대장암 세포 HT-29의 증식을 억제하고 세포 사멸을 유도하는 천연소재 발굴을 목적으로, 오미자(Schizandra chinensis Baillon) 열수 추출물을 이용하여 인체대장암 세포 HT-29의증식에 미치는 영향을 확인하였다. 오미자 열수 추출물이 HT-29 대장암 세포의 apoptosis 유도 효과 및 기전에 미치는 영향을 분자생물학적 방법으로 실험하여 다음과 같은 결론을 얻었다. MTT assay를 통해 인체 대장암세포 HT-29는 오미자 시료농도 0, 1.0, 2.0, 4.0 mg/mL에서 암세포 사멸농도가 각각 0%, 10%, 70%, 88%를 나타내었다. 대장암세포에 오미자 추출물을 처리하고 cell cycle 분석 결과, 시료농도 의존적으로 sub-G1기가 증가하였고, G0/G1기는 감소되는 것을 통해, apoptosis가 일어나 세포 증식을 저해하는 것으로 확인되었다. 대장암세포 핵의 형태학적 변화를 보면, 오미자 추출물 처리 시 농도 의존적으로 세포수가 감소되는 것이 뚜렷이 관찰되었고 cell shrinking, chromatin condensation등 apototic body 등과 같은 형태학적 변화들이 뚜렷하게 관찰되었다. RT- PCR을 통한 유전자 발현은, 오미자 추출물 농도 의존적으로 p53 유전자 발현이 증가되는 것을 통해 대장암세포의 증식억제를 확인할 수 있었다. In vitro 실험에서 오미자 열수추출물이 대장암세포의 성장을 저해하는 효과가 있음을 확인하였으며, 오미자 추출물에 함유된 활성 본체 규명 및 apoptosis를 유도하는 작용기작에 관한 연구가 수행중이다.

ICR 마우스를 이용한 오미자박 추출물의 소핵 시험 (Erythrocyte Micronucleus Test of Pomace Schisandra chinensis Extracts Using ICR Mouse)

  • Objectives : In this study, erythrocyte micronucleus test of pomace Schisandra chinensis extracts was conducted in order to up-cycling to a high value-added industry using by-products discarded in the production process of Schisandra chinensis products and active ingredients such as dibenzocyclooctadiene lignans in Schisandra chinensis. Methods : The micronucleus test was performed according to the 'OECD Guidelines'. Including the negative control group(0 mg/kg) and the positive control group(CPA 70 mg/kg), pomace Schisandra chinensis extracts were orally administered to ICR mouse at doses of 500, 1,000, and 2,000 mg/kg. After sacrificing the experimental animals bone marrow cells were collected and micronucleated polychromatic erythrocyte were counted. And genetic toxicity was confirmed according to the frequency of micronucleus. Results : As a result of the micronucleus test, there were no changes in body weight, clinical signs, or death in any group. But, a significant increase was observed in the frequency of micronucleated polychromatic erythrocyte among polychromatic erythrocytes in the positive control group administered with CPA compared to the negative control group(p<0.05). Whereas, no significant increase was observed in the group administered with pomace Schisandra chinensis extracts compared to the negative control group. Conclusions : Pomace Schisandra chinensis extracts did not induce micronucleus in bone marrow cells of ICR mouse up to a concentration of 2,000 mg/kg, and it was judged that no genetic toxicity was observed.

대하 (Fenneropenaeus chinensis) 양식장 사육수에 미치는 Probiotics의 효과 (Effect of Probiotics on Water Quality in the Shrimp (Fenneropenaeus chinensis) Ponds)

  • Applications of probiotics to shrimp ponds were carried out to determine their effects on water quality. Fermented solutions consisting of Bacillus spp. and Nitrosomonas spp. were applied to a 4 ha shrimp (Fenneropenaeus chinensis) pond from July to September, 2000. In the pond treated with probiotics, daily variations of DO and pH, and concentrations of DIN and DIP were lower than those in the ponds without probiotic treatment. Concentration of phytoplankton was less variable and the number of species was more variable in the probiotic-treated pond than those in the control pond. Variation of bacterial numbers and the number of Vibrio spp. were lower in the treated pond than those in the control pond. It is confirmed that the probiotics can be used to improve water quality of the shrimp ponds.

Stock Characterization of the Fleshy Prawn (Penaeus chinensis) in the Yellow Sea by Intraspecific Sequence Variation of the Cytochrome c Oxidase Subunit I Gene

  • To determine the amount of genetic variation among populations of Penaeus chinensis (Osbeck) in the Yellow Sea, 342 bp region of the mitochondrial cytochrome c oxidase subunit I gene was amplified and sequenced. Six haplotypes, which differ by from one to four nucleotide sustitutions, were detected from 34 individuals of 4 populations examined. Mean sequence divergence between pairs of haplotypes was $0.68\%$. Most individuals from 4 populations were shared by the most common genotype. This genotype was distributed evenly in the Korean and Chinese populations. This result is in accordance with findings observed using RFLPs analysis of mtDNA (Hwang et al., 1997). Therefore, it is suggested that P. chinensis should be treated as one unit stock in the Yellow Sea.

Viscozyme L aided flavonoid extraction and identification of quercetin from Saururus chinensis (Lour.) Baill

  • In order to enhance the extraction efficiency of flavonoid from Saururus chinensis, carbohydrate-hydrolyzing enzyme Viscozyme L aided extraction techniques have been studied. Then flavonoid composition, as well as quercetin, were also identified using UV/Vis, HPLC/MS, and 1H-NMR. The results showed that favorable extraction conditions were Viscozyme L concentration of 0.25 mg/g, pH 4.2, reaction at 45 ℃ for 12 h. Under the favorable extraction condition, total flavonoid yield (37.9 mg/g) and quercetin yield (0.86 mg/g) increased by about 2.0 and 9.6 times, respectively, compared to control group. Interestingly, as a significant flavonoid of S. chinensis, flavonoid glycones rutin was hydrolyzed to aglycones quercetin by Viscozyme L. These findings provide scientific and theoretical support for the development quercetin-rich products, which was quickly absorbed by the human body than rutin.

흑오미자(Schizandra nigra Max) 즙액의 추출조건과 추출물의 특성 (The properties and extracting conditions of juice preperation from Schizandra nigra Max.)

  • 흑오미자를 이용하여 음료를 제조하는데 필요한 여러 가지 가공적성 가운데 가공제품의 생산과 상품성에 관련이 깊은 몇 가지 특성을 한국산, 중국산 오미자와 비교 조사하였다. 흑오미자로부터 온도(80, 4$0^{\circ}C$), 추출시간 (3, 4, 5, 7시간), 추출용매(증류수, 20% 에탄올)를 사용하여 즙액의 추출 수율과 색도, 당도(가용성 고형분) pH을 측정하여 한국산 오미자와 중국산 오미자를 비교한 결과, 수율을 기준으로 한 흑오미자의 추출조건은 20% 에탄올 수용액으로 8$0^{\circ}C$ 물 중탕에서 3시간 추출이 가장 적합하였다. 추출액의 색도 측정에서 L값(투명 도)은 8$0^{\circ}C$로 4시간, 20%에탄올 추출액이 낮고 4$0^{\circ}C$로 물 추출액이 높았다. a값(적색도)은 8$0^{\circ}C$, 4시간 20% 에탄올 추출물이 가장 높았으며, b값(황색도)은 온도가 높고 (8$0^{\circ}C$, 5시간)장시간 증류수 추출액이 높아 적색색소의 추출방법과 차이를 나타내었다. 흑오미자 추출물의 당도는 2.0에서 2.6%Brix 범위로 오미자 추출물보다는 낮았으며, pH는 3.39­3.62 범위로 산성을 나타내었고 오미자 즙액 의 안정한 적색을 나타내는데 충분한 pH 범위이었으며, 흑오미자 줄기의 즙액도 증량제로 이용할 수 있는 가치가 있는 것으로 사료되었다.

후숙 온도에 따른 오미자의 이화학적 특성 변화 (Changes of Physicochemical Characteristics of Schizandra chinensis during Postharvest Ripening at Various Temperatures)

  • 본 실험에 사용하였던 미숙성 오미자의 일반성분은 수분 $79.6{\pm}0.8%$, 조단백질 $1.7{\pm}0.5%$, 조지방 $1.2{\pm}0.1%$, 회분 $0.6{\pm}0.0%$이었다. 후숙기간 동안 착즙수율은 $4^{\circ}C$에서는 $55.3{\pm}0.6-56.3{\pm}0.6%$로서 큰 변화가 없었고, $25^{\circ}C$와 실온에서는 6-7% 정도 감소하였다. 가용성 고형분 함량은 $25^{\circ}C$와 실온 저장시 6일째 $12.2{\pm}0.15%$로서 가장 높게 나타났으며, 그 이후에는 약간 감소하였다. pH는 $4^{\circ}C$와 실온 저장시 후숙기간 동안 pH 2.76-2.81로서 큰 변화가 없었으며, $25^{\circ}C$ 저장시 초기 pH $2.81{\pm}0.02$에서 점차 낮아져 저장 8일째 pH $2.68{\pm}0.03$을 나타내었다. 적정산도는 $25^{\circ}C$와 실온 저장시 저장 6일째 각각 $7.49{\pm}0.03%$$7.20{\pm}0.03%$로서 가장 높게 나타났으며 그 이후 감소하였다. 색도의 변화는 $25^{\circ}C$와 실온 저장시 a값(적색도)은 초기 5.04에서 8일째 각각 6.77 및 7.65로 증가하였으며, L값(명도)은 후숙기간 동안 모든 처리구에서 증가하였다 유리당은 fructose(0.55%), glucose(0.56%), sucrose(0.50%)가 주로 검출되었으며, 후숙이 진행되면서 fructose와 glucose 함량은 증가하였고, sucrose 함량은 감소하였다. 총 유리당의 함량은 $25^{\circ}C$와 실은 저장시 초기 1.61%에서 저장 6일째까지 각각 2.53%와 2.13%로 증가하였고, 후숙 8일째에는 각각 1.94%와 2.08%로 감소하였다. 유기산은 후숙 초기에 citric acid, malic acid 및 succinic acid의 함량이 각각 0.17, 0.07 및 1.21%로서 succinic acid의 함량이 가장 많았고, 후숙기간 중 변화는 유리당의 경우와 비슷하였다. 오미자의 후숙을 위해서는 가용성 고형분 함량과 총 유리당 함량을 기준으로 할 때 $4^{\circ}C$에서 저장하는 경우 8일동안, 그리고 $25^{\circ}C$ 및 실온에서 저장하는 경우 6일 동안 후숙시키는 것이 바람직한 것으로 판단되었다.

Seasonal Variations in Voluntary Intake and Apparent Digestibility of Forages in Goats Grazing on Introduced Leymus chinensis Pasture

  • The nutrient composition of pasture, voluntary intake and digestibility of diet ingested by goats grazing on an introduced Leymus chinensis pasture were measured across spring (May), summer (July), autumn (October) and winter (March). In each season, 12 Inner Mongolian Cashmere goats (6 wethers and 6 does with an average live weight of $22.2{\pm}1.3$ kg and $19.5{\pm}0.8$ kg, respectively) were used to graze on a 2 hectares size paddock. Diet selection was observed and the plant parts selected by grazing goats and whole plant L. chinensis were sampled simultaneously. The alkane pair $C_{32}:C_{33}$ and $C_{36}$ were used to estimate intake and digestibility, respectively. The results showed that the plant parts selected by goats had higher crude protein (CP) and lower acid detergent fiber (ADF) and neutral detergent fiber (NDF) than the whole plant, especially in the autumn and winter. The voluntary intake of dry matter (DM), CP, ADF, NDF, and metabolizable energy (ME) by goats was highest in summer (p<0.05). The goats ingested more CP, ME, and less ADF in spring than in autumn (p<0.05). The intakes of DM, CP, and ME were lowest in winter (p<0.05). There were significant differences in nutrient intake between wethers and does in each season, except for the ADF and ME intake per metabolic weight ($LW^{0.75}$). The nutrient digestibilities were higher in spring and summer, and decreased significantly during the autumn and winter (p<0.05). Goats, especially wethers, had a relative constant NDF digestibility across seasons, however, the apparent digestibility of CP in both wethers and does, decreased to negative values in winter. The grazing goats experienced relatively sufficient nutrients supply in spring and summer, and a severe deficiency of CP and ME in winter.
