Ginsenoside Rg5 promotes muscle regeneration via p38MAPK and Akt/mTOR signaling

  • Background: Skeletal muscles play a key role in physical activity and energy metabolism. The loss of skeletal muscle mass can cause problems related to metabolism and physical activity. Studies are being conducted to prevent such diseases by increasing the mass and regeneration capacity of muscles. Ginsenoside Rg5 has been reported to exhibit a broad range of pharmacological activities. However, studies on the effects of Rg5 on muscle differentiation and growth are scarce. Methods: To investigate the effects of Rg5 on myogenesis, C2C12 myoblasts were induced to differentiate with Rg5, followed by immunoblotting, immunostaining, and qRT-PCR for myogenic markers and promyogenic signaling (p38MAPK). Immunoprecipitation confirmed that Rg5 increased the interaction between MyoD and E2A via p38MAPK. To investigate the effects of Rg5 on prevention of muscle mass loss, C2C12 myotubes were treated with dexamethasone to induce muscle atrophy. Immunoblotting, immunostaining, and qRT-PCR were performed for myogenic markers, Akt/mTOR signaling for protein synthesis, and atrophy-related genes (Atrogin-1 and MuRF1). Results: Rg5 promoted C2C12 myoblast differentiation through phosphorylation of p38MAPK and MyoD/E2A heterodimerization. Furthermore, Rg5 stimulated C2C12 myotube hypertrophy via phosphorylation of Akt/mTOR. Phosphorylation of Akt induces FoxO3a phosphorylation, which reduces the expression of Atrogin-1 and MuRF1. Conclusion: This study provides an understanding of how Rg5 promotes myogenesis and hypertrophy and prevents dexamethasone-induced muscle atrophy. The study is the first, to the best of our knowledge, to show that Rg5 promotes muscle regeneration and to suggest that Rg5 can be used for therapeutic intervention of muscle weakness and atrophy, including cancer cachexia.

SURFACE ROUGHNESS OF COMPOSITE RESIN ACCORDING TO FINISHING METHODS (복합레진 표면의 연마방법에 따른 표면조도)

  • The purpose of this study was to evaluate the difference of surface roughness of composite resin according to composite resin type, polishing methods, and use of resin sealant. Two hundred rectangular specimens, sized $8{\times}3{\times}2mm$, were made of Micro-new (Bisco, Inc., Schaumburg, IL, U.S.A) and Filtek Supreme (3M ESPE Dental Products, St. Paul, MN, U.S.A.), and divided into two groups; Micronew-M group, Filtek Supreme-S group. Specimens for each composite group were subdivided into five groups by finishing and polishing instruments used; M1 & S1(polyester strip), M2 & S2 (Sof-Lex disc), M3 & S3 (Enhance disc and polishing paste), M4 & S4(Astropol) and M5 & S5 (finishing bur), Polished groups were added letter B after the application of resin surface sealant (Biscover), eg, M1B and S1B. After specimens were stored with distilled water for 24hr, average surface roughness (Ra) was taken using a surface roughness tester. Representative specimens of each group were examined by FE-SEM (S-4700: Hitachi High Technologies Co., Tokyo, Japan). The data were analysed using paired t-test, ANOVA and Duncan's tests at the 0.05 probability level. The results of this study were as follows ; 1. The lowest Ra was achieved in all groups using polyester strip and the highest Ra was achieved in M5, S5 and M5B groups using finishing bur. On FE-SEM, M1 and S1 groups provided the smoothest surfaces, M5 and S5 groups were presented the roughest surfaces and voids by debonding of filler on the polished specimens. 2. There was no significant difference in Ra between Micronew and Filtek Supreme before the application of resin sealant, but Micronew was smoother than Filek Supreme after the application of resin sealant. 3. There was significant corelation between Ra of type of composite resin and polishing methods before the application of resin sealant (p=0.000), but no significant interaction between them after the application of resin sealant. On FE-SEM, most of composite resin surfaces were smooth after the application of resin sealant on the polished specimens. 4. Compared with before and after the application of resin sealant in group treated in the same composite and polishing methods, Ra of M4B and M5B was statistically lower than that of M4 and M5, and S5B was lower than that of S5, respectively (p<0.05). In conclusion, surface roughness by polishing instruments was different according to type of composite resin. Overall, polyester strip produced the smoothest surface, but finishing bur produced the roughest surface. Application of resin sealant provided the smooth surfaces in specimens polished with Enhance, Astropol and finishing bur, but not provided them in specimens polished with Sof-Lex disc.

Effects of Practical Variations in Fasting, Stress and Chilling Regime on Post-slaughter Metabolic Rate and Meat Quality of Pork Loin (절식, 스트레스 및 냉각속도가 돼지도체의 사후 대사속도와 등심근의 육질에 미치는 영향)

  • The study was conducted to investigate the effects of practical variations in feed restriction, pre-slaughter handing and chilling regime on pork quality during ageing. A total of twenty male landraces were allocated into three treatments(i.e., pre-slaughter feeding, stress and chilling regime) in a factorial arrangement. pH, temperature, free calcium ions, WB-shear force, sarcomere length, cooking loss, drip loss and objective color were determined during rigor development and/or 1, 3, 7 d postmortem. Pre-slaughter chasing stress for approximately 15 min had no effects on pH/temperature profile and objective meat quality. There was an interaction(P < 0.05) between the fasting treatment and chi1ling regime for muscle temperature at pH 6.2. Sarcomere length indicated that the current experiment conditions did not induce muscle shortening, with 1.7 to 1.8 ${\mu}m$, in spite of a significant effect of the fasting treatment (P<0.01). Pigs fed until the morning of slaughter showed a low WB-shear force(P < 0.05) until 3 d at I "C. The treatment also resulted in a higher Hunter L* and a*(P < 0.05) at 24 h and 7 d. Fasted pigs showed a significantly(P < 0.05) reduced cooking loss. The current results indicated that feeding upon the morning of slaughter became detrimental on meat color and the negative effect on cooking loss were linearly elevated with increased ageing time. On the other hand, WB-shear force did not distinguishable after 3 d. Collectively, it appeared that feed restriction from a day before slaughter could produce more a desirable meat quality at the time of consuming. However, the limited effect of animal handling and chilling rate on meat quality is not necessarily to extend to that these do not affect pork quality, as that largely depends on experimental design.

Hydro-geochemical Nature and Nitrates Contamination Charecters of Groundwater in the Youngdong, Chungbuk Province (충북 영동지역 지하수의 수리지화학적 특성 및 질산염 오염 특성)

  • Major ions and nitrogen isotopic analyses were performed to determine the geochemical characters and to identify the source of nitrate of the shallow groundwater around agricultural field in the Youngdong area. The pH value of groundwater ranges from 60. to 8.2 (pH 7.2, mean). The average of EC, Eh and DO is 369 ${\mu}S/cm$ (70~729 ${\mu}S/cm$), 165.6 mV (29~383.2 mV), 4.3 mg/L (1.8~8.0 mg/L) respectively. The ion concentraion of groundwater was in the order of $Ca^{2+}$>$Na^{2+}$>$Mg^{2+}$>$K^{2+}$ and ${HCO_3}^-$>${NO_3}^-$>${SO_4}^{2-}$>$Cl^-$>$F^-$. Most of groundwater is Ca-$HCO_3$ type. The groundwater was affected by water-rock interaction in the shallow depth. Some groundwater is Ca-Cl or Na-$HCO_3$ (2.5%) type that was due to agricultural activities. The $NO_3$_N concetration of grondwater range from 10.2 mg/l to 26.9 mg/l, which show that this area is under nitrate pollution. ${\delta}^{15}N-NO_3$ value of the groundwater is the origins of are a combination of animal wastes and man-made fertilizers.

Antimicrobial Activities of Ethosome-Encapsulated Palmitoyl Tripeptide (Ethosome에 캡슐화된 Palmitoyl Tripeptide의 항균효과)

  • Palmitoyl tripeptide (M330) showed higher antimicrobial activities than methyl paraben or phenoxy ethanol through minimum inhibitory concentration (MIC) test. However, when the M330 was added into cosmetic formulation, white precipitates formed due to the electrostatic interaction between M330 and carbopol (carboxy vinyl polymer) as a thickener in cosmetics, and the viscosity of cosmetics decreased sharply. Also, the antimicrobial activities of M330 in cosmetics became lower than those of methyl paraben or phenoxy ethanol. Thus, the encapsulation of M330 in ethosome vesicle was attempted in order to recover the declined antimicrobial activities of M330 in cosmetics and prevent the precipitates from forming. When ethosome-encapsulated M330 was added into cosmetics, the precipitates did not form, and the decrease in the viscosity of cosmetics was not large compared to the addition of unencapsulated M330. Challenge tests showed that antimicrobial activities against gram negative bacteria were improved by the encapsulation of M330, but the encapsulation was not effective against gram positive bacteria and fungus. A combination of M330 with EDTA showed synergistic inhibitory potential against C. albicans. After coencapsulation of M330 and EDTA in ethosome, antimicrobial activities proved to be higher than those of unencapsulated M330 and EDTA.

Mineralogical Study of the Granite Weathering in the Seoul Area: Water-Rock Interaction in the Namsan Granite (남산 화강암의 풍화 및 광물-물 반응에 관한 연구)

  • The weathering of the Namsan granite was studied in terms of sorption process. The Namsan granite consists mainly of quartz, alkali feldspar, plagioclase (${Ab_{85}An_{15}}-Ab_{100}$ and biotite with small amounts of sericite, magnetite and ilmenite. The kinetic factors for altering the granite body are the proton and hydroxyl ions derived from the reaction of water and mineral. There are two different types of pH variation curves for rocks of different mineral assemblages. when powdered granite was dispersed in distilled water under ambient condition. The sorption-process proceeds by three steps for fresh granite; (1) the initial rapid pH-rise to 10 by the uptake of proton by negatively charged mineral surfaces, (2) the gradual pH-down, and (3) the stable pH tail between 7.1-7.5. For somewhat weathered granites, the sorption proceeds; (1) the initial rapid pH-down to 4.8, (2) the slight pH-rise and slow ph-down, and (3) the stable pH tail between 5.0-5.3. The reaction rate is controlled by the density of adsorbable sites, the solubility of the mineral, pH of the system and formation of amorphous gel and gibbsite. Amorphous gel floates on the surface of the solution while stirring the powdered granite and then is transformed into gibbsite in an hour or so. The pH saturation values for -325 mesh fresh granite from 5 m depth is about pH 10 when rock/water ratio is over 10g/200 ml.

Role of Lectins in Host Plant-Rhizobium Interactions (근류균과 숙주식물의 상호작용에 관한 렉틴의 역할)

  • Experiments were carried out to elucidate the specific interactions between host plant, Phaseolus vulgaris, and symbiotic bacteria, Rhizobium Phaseoli. Purified P. vulgaris lectins and six species of cultured Rhizobium were subjected to agglutination test. Lectins from bean and R. phaseoli showed relatively high agglutination activity indicating that host plant lectins recognize carbohydrate moieties on the compatible Rhizobium cell surface. The specific carbohydrate receptors for binding of the lectins on the cell surface of R. phaseoli were found as mannose and galactose. The minimum concentration of sugars for the inhibition was 6.25mM. The lectin content of cultured plant roots was measured after germination and was maximum in 5-day seedlings. The nodulation was competitively inhibited by lectins for the plants cultured with Rhizobium cells. By immunochemical studies, there was some relationship in antigenic determinants between R. phaseoli and R. japonicum but no relationships were observed with other Rhizobium species. The results suggest that the infection by rhizobia to the roots of leguminous plants may be caused by the specific interaction of lectins with rhizobia.

Purification and Characterization of an Intracellular NADH: Quinone Reductase from Trametes versicolor

  • Intracellular NADH:quinone reductase involved in degradation of aromatic compounds including lignin was purified and characterized from white rot fungus Trametes versicolor. The activity of quinone reductase was maximal after 3 days of incubation in fungal culture, and the enzyme was purified to homogeneity using ion-exchange, hydrophobic interaction, and gel filtration chromatographies. The purified enzyme has a molecular mass of 41kDa as determined by SDS-PAGE, and exhibits a broad temperature optimum between $20-40^{\circ}C$, with a pH optimum of 6.0. The enzyme preferred FAD as a cofactor and NADH rather than NADPH as an electron donor. Among quinone compounds tested as substrate, menadione showed the highest enzyme activity followed by 1,4-benzoquinone. The enzyme activity was inhibited by $CuSO_4,\;HgCl_2,\;MgSO_4,\;MnSO_4,\;AgNO_3$, dicumarol, KCN, $NaN_3$, and EDTA. Its $K_m\;and\;V_{max}$ with NADH as an electron donor were $23{\mu}M\;and\;101mM/mg$ per min, respectively, and showed a high substrate affinity. Purified quinone reductase could reduce 1,4-benzoquinone to hydroquinone, and induction of this enzyme was higher by 1,4-benzoquinone than those of other quinone compounds.

Crack growth analysis and remaining life prediction of dissimilar metal pipe weld joint with circumferential crack under cyclic loading

  • Fatigue crack growth model has been developed for dissimilar metal weld joints of a piping component under cyclic loading, where in the crack is located at the center of the weld in the circumferential direction. The fracture parameter, Stress Intensity Factor (SIF) has been computed by using principle of superposition as KH + KM. KH is evaluated by assuming that, the complete specimen is made of the material containing the notch location. In second stage, the stress field ahead of the crack tip, accounting for the strength mismatch, the applied load and geometry has been characterized to evaluate SIF (KM). For each incremental crack depth, stress field ahead of the crack tip has been quantified by using J-integral (elastic), mismatch ratio, plastic interaction factor and stress parallel to the crack surface. The associated constants for evaluation of KM have been computed by using the quantified stress field with respect to the distance from the crack tip. Net SIF (KH + KM) computed, has been used for the crack growth analysis and remaining life prediction by Paris crack growth model. To validate the model, SIF and remaining life has been predicted for a pipe made up of (i) SA312 Type 304LN austenitic stainless steel and SA508 Gr. 3 Cl. 1. Low alloy carbon steel (ii) welded SA312 Type 304LN austenitic stainless-steel pipe. From the studies, it is observed that the model could predict the remaining life of DMWJ piping components with a maximum difference of 15% compared to experimental observations.

Inhibitory Potential of Thelephoric Acid on CYP2J2 Activities in Human Liver Microsomes (Thelephoric acid의 CYP2J2 효소 활성 저해제 평가)

  • Cytochrome P450 2J2 (CYP2J2) is an enzyme mainly found in human extrahepatic tissues, with predominant expression in the cardiovascular system. CYP2J2 plays important roles in the metabolism of endogenous metabolites and therapeutic drugs, such as arachidonic acid, astemizole, ebastine, and terfenadine. CYP2J2 is also overexpressed in human cancer tissues and cancer cell lines and may represent a potential target for therapy of human cancers. In this study, 10 natural products obtained from plants and microorganisms were screened as potential CYP2J2 inhibitors. Among them, thelephoric acid showed strong inhibition of astemizole O-demethylation activity ($IC_{50}=3.23{\mu}M$) in a dose-dependent manner. Evaluation of the substrate dependency of the inhibitory activity of thelephoric acid showed that it strongly inhibited CYP2J2-mediated ebastine hydroxylation ($IC_{50}=5.32{\mu}M$) and terfenadine hydroxylation ($IC_{50}=3.27{\mu}M$) in a substrate nondependent manner. The present data suggest that this compound might be a potential candidate for further evaluation for anticancer activity.
