Flow injection analysis technique for monitoring of xylitol concentrations in biological processes has been developed using xylitol oxidase (XYO) immobilized on VA-Epoxy Biosynth carrier. The immobilized XYO cartridge has been integrated into a FIA system with an oxygen electrode and systematically investigated with regards to the factors which can affect the activity of the immobilized XYO, such as pH, temperature, salt concentration etc. The activity of the immobilized XYO increased with the temperature ($19.0 - 29.0^{circ}C$) and sample injection volume ($75-250\muL$) and molarity of potassium phosphate buffer (0.1-1 M), but it reached the highest value at pH 8.5. The XYO-FIA system has been also applied for on-line monitoring of xylitol concentrations in a reactor and showed good operational stability and agreement with off-line data measured with HPLC.
A computational fluid analysis was performed on an LOx line system of a liquid rocket engine. The model was created with 3D CAD and imbedded to the 3D CFD program. Before the full scale analysis on the system was carried out, each components with simplified models was analyzed to save time and cost. As a result, the inlet pressure of the gas generator should be compensated with a certain device unless the inlet pressure of the line system is sufficiently high. The flow pattern of the exit of the system was dependant upon the location of the orifice as well as the size. As a whole the line system analyzed met the requirements, and will be tested and confirmed after being manufactured.
In this study, optical emission spectroscopy was used to monitor the plasma produced during the RF magnetron sputtering of a $BaTiO_3$ target. The intensities of chemical species were measured by real time monitoring with various discharge parameters such as RF power, pressure, and discharge gas. The emission lines of elemental and ionized species from $BaTiO_3$ and Ti targets were analyzed to evaluate the film composition and the optimized growth conditions for $BaTiO_3$ films. The emissions from Ar(I, II), Ba(I, II) and Ti(I) were found during sputtering of the $BaTiO_3$ target in Ar atmosphere. With increasing RF power, all the line intensities increased because the electron density increased with increasing RF power. When the Ar pressure increased, the Ba(II) and Ti(I) line intensity increased, but the $Ar^+$ line intensity decreased with increasing pressure. This result shows that high pressure is of greater benefit for the ionization of Ba than for that of Ar. Oxygen depressed the intensity of the plasma more than Ar did. When the Ar/$O_2$ ratio decreased, the intensity of Ba decreased more sharply than that of Ti. This result indicates that the plasma composition strongly depends on the discharge gas atmosphere. When the oxygen increased, the Ba/Ti ratio and the thickness of the films decreased. The emission spectra showed consistent variation with applied power to the Ti target during co-sputtering of the $BaTiO_3$ and Ti targets. The co-sputtered films showed a Ba/Ti ratio of 1.05 to 0.73 with applied power to the Ti target. The films with different Ba/Ti ratios showed changes in grain size. Ti excess films annealed at $600^{\circ}C$ did not show the second phase such as $BaTi_2O_5$ and $TiO_2$.
Increased oxidative stress and changes in DNA methylation are frequently detected in bladder cancer patients. We previously demonstrated a relationship between increased oxidative stress and hypomethylation of the transposable long-interspersed nuclear element-1 (LINE-1). Promoter hypermethylation of a tumor suppressor gene, runt-related transcription factor 3 (RUNX3), may also be associated with bladder cancer genesis. In this study, we investigated changes of DNA methylation in LINE-1 and RUNX3 promoter in a bladder cancer cell (UM-UC-3) under oxidative stress conditions, stimulated by challenge with $H_2O_2$ for 72 h. Cells were pretreated with an antioxidant, tocopheryl acetate for 1 h to attenuate oxidative stress. Methylation levels of LINE-1 and RUNX3 promoter were measured by combined bisulfite restriction analysis PCR and methylation-specific PCR, respectively. Levels of LINE-1 methylation were significantly decreased in $H_2O_2$-treated cells, and reestablished after pretreated with tocopheryl acetate. Methylation of RUNX3 promoter was significantly increased in cells exposed to $H_2O_2$. In tocopheryl acetate pretreated cells, it was markedly decreased. In conclusion, hypomethylation of LINE-1 and hypermethylation of RUNX3 promoter in bladder cancer cell line was experimentally induced by reactive oxygen species (ROS). The present findings support the hypothesis that oxidative stress promotes urothelial cell carcinogenesis through modulation of DNA methylation. Our data also imply that mechanistic pathways of ROS-induced alteration of DNA methylation in a repetitive DNA element and a gene promoter might differ.
Growth kinetic parameters for mass cultivation of Chinese Hamster Ovary (CHO) cells are estimated by measuring oxygen uptake rates. It Is found that there is strong correlation between cell growth and oxygen consumption, showing that correlation factor is 0.83. Derived linear model predicts actual cell density very well. It tells that oxygen uptake rate can play important role in indirectly measuring cell density when conventional method of estimating cell density is no longer meaningful due to heavy cell clumpings. Cell yield per oxygen consumption, $Y_{\chi}o$ and mass transfer coefficient for oxygen, Ka are also estimated as 1.26$\times$10$^4$cells/mmole $O_2$ consumed and 1.01/h, respectively. Average specific growth rate over all runs is 2.891/day for CHO cells with producting 2 grams of tPA per day under continuous perfusion operations.
The protective effect of Acanthopanax senticosus (AS) extract on alloxan-induced pancreatic β-cell damage was investigated in HIT T-15 cells, a Syrian hamster pancreatic β-cell line. Alloxan caused the pancreatic β-cell damage through the generation of reactive oxygen free radicals, increased DNA fragmentation, and decreased cellular NAD/sup +/ levels. The β-cell damage was significantly prevented by the pretreatment with water soluble extract of AS roots. These results suggest that the protective effect of AS extract, on alloxan-induced β-cell damage, is primarily due to the inhibition of the generation of reactive oxygen free radical species (ROS) by alloxan.

We report a simple way to enhance the size gradient of an imprinted nanopattern through oxygen plasma etching under a nonuniform magnetic field. A sample substrate was placed next to a magnet, and then a nonuniform magnetic field condition was formed around the sample. Using oxygen plasma etching, a line pattern having an initial width of 273 nm was gradually modified from 248 nm at one end to 182 nm at the other end. Controlling the arrangement of the magnet and sample, we could induce a triangular shape size gradient. We verified that the gradually modified nanopatterns we produced are applicable to continual optical property control, showing a possibility to be utilized for optical components such as gratings and polarizers.

The optimal conditions for operating a moving-aeration bioreactor were determined as 30rpm and 150 (ml/min) of air flow rate, which can yield ca. 7.3 (l/h)of maximum mass transfer coefficient. It was also found that the agitation speed played much much important role than air input rate in oxgen transfer into the medium. $2.6{\times}10^6$ (cells/ml) and 0.6 (ml/l) of maximum cell denisty and IL-2 production were observed in batch cultivation of IL-2 producing BHK cell line. 0.53 (mM/l/h) of oxygen uptake rate was also estimated. The performance of a moving-aeration bioreactor (specific growth rate and oxygen uptake rate, etc.) was superior to other culture systems, such as cell-life and static membrane aeration bioreactors. Ii must be useful to apply this reactor to many culture processes by improving structural limitations in scaling-up the system.

Intake silencers of high performance and with less pressure drop are developed for a household oxygen generator. First, the acoustic power of the target oxygen generator are experimentally evaluated according to ISO code. Then, the transmission loss of and the flow characteristics inside the existing intake silencer are predicted and analyzed. On a basis of these results, two intake silencers are proposed, which are designed to induce the swirling flow inside the intake silencer and thus to reduce the flow loss. The predicted TL and the pressure drop for these two new silencers are compared to the existing one, which shows that the proposed ones have higher TLs as well as less pressure drop. The reason for these improvements are explained by investigating the flow characteristics of the new silencers in detail.

Abstract Oligotrophic microbial fuel cells (MFCs) were tested for the continuous monitoring of low biochemical oxygen demand (BOD) by using artificial wastewater, containing glucose and glutamate, as check solution. Ten times diluted trace mineral solution was used to minimize the background current level, which is generated from the oxidation of nitrilotriacetate used as a chelating agent. The feeding rate of 0.53 ml/min could increase the sensitivity from 0.16 to 0.43 ${\mu}$A/(mg BOD/l) at 0.15 ml/min. The dynamic linear range of the calibration curve was between 2.0 and 10.0 mg BOD/l, and the response time to the change of 2 mg BOD/l was about 60 min. The current signal from an oligotroph-type MFCs increased with the increase in salts concentration, and the salt effect could be eliminated by 50 mM phosphate buffer.
