현곡(玄谷) 보비탕(補脾湯)의 구성한약과 그 기미배오(氣味配伍) 분석 (Hyeongok's Bobee-tang was Combined according to the Theory for Properties and Tastes of Herbal Medicines)

  • The objectives of this study lie in theoretical establishment of Bobee-tang for curing the asthenic syndrome of spleen through analyzing the component medicines and combination principles of Hyeongok's Bobee-tang, and furthermore, maximizing the clinical use of Bobee-tang. This study analyzed the component medicines and combination principles of Hyeongok's Bobee-tang based on the theory for properties and tastes of herbal medicines from the ${\ulcorner}$Yellow Emperor's Canon of Internal Medicine${\lrcorner}$, the theory for principal herbal medicine, assistant herbal medicine, adjuvant herbal medicine, dispatcher herbal medicine, and the five elements doctrine. Hyeongok's Bobee-tang is an odd prescription, composed of 7 kinds of ingredients : No.1 Radix Ginseng (2don;7.5 g), No.2 Rhizoma Atractylodis Macrocephalae (1don;3.75 g), No.3 Carapax Et Plastrumtestudinis (1don), No.4 Concha Ostreae (1don), No.5 Cortex Cinnamomi (1don), No.6 Rhizoma Zingiberis Praeparatae (1don), and No.7 Radix Glycyrrhizae (1don). First, to cure the asthenic syndrome of the spleen, the methods of invigorating the spleen and the heart, and purging the liver should be used according to the five elements doctrine. Secondly, herbal medicines appropriate for those treatment methods should be chosen according to the theory for properties and tastes of herbal medicine and thirdly, the combination of those herbal medicines should be carried out according to the theory for principal herbal medicine, assistant herbal medicine, adjuvant herbal medicine, dispatcher herbal medicine. As a good example, Hyeongok's Bobee-tang is combined according to the above theories. In conclusion, this formula was created by applying to the theory for properties and tastes of herbal medicines.

  • The inhibitory effects of 207 oriental medicines and 19 oriental prescriptions on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical were examined. The most effective oriental medicines were the fruit of Terminalia chebula, followed in order by the seed of Areca catechu, the heart wood of Caesalpinia sappan, the pericarp of Castanea crenata, the seed of Nelumbo nucifera, Rhus galls, the root of Sanguisorba officinalis, and the bark of Betula platyphylla at the concentration of 10 ${\mu}g$/mL. Of the 19 oriental prescriptions, Buyeonsan and Samsinhwan showed the potent scavenging activities of 88% and 71% on DPPH radical at the concentration of 5 ${\mu}g$/mL, respectively.

Anti-Inflammatory Effect of Ixeris dentata on Ultraviolet B-Induced HaCaT Keratinocytes

  • Human skin is the first line of defense for the protection of the internal organs of the body from different stimuli. Ultraviolet B (UVB) irradiation induces skin damage and inflammation through the secretion of various cytokines, which are immune regulators produced by cells. To prevent the initiation of skin inflammation, keratinocytes that have been irreversibly damaged by radiation must be removed through the apoptotic mechanism. Ixeris dentata (family: Asteraceae) is a perennial medicinal herb indigenous to Korea. It has been used in Korea, China, and Japan to treat in digestion, pneumonia, diabetes, hepatitis, and tumors. To gain insight into the anti-inflammatory effects of I. dentata, we examined its influence on UVB-induced pro-inflammatory cytokine production in human keratinocytes (HaCaT cells), by observing cells that were stimulated with UVB in the presence or absence of I. dentata. In the present study, pro-inflammatory cytokine production was determined by performing enzyme-linked immunosorbent assay, reverse transcription polymerase chain reaction, and western blot analysis to measure the activation of mitogen-activated protein kinase (MAPKs). I. dentata inhibited UVBinduced production of the pro-inflammatory cytokine interleukin (IL)-6 in a dose-dependent manner. Further, I. dentata inhibited the UVB-induced expression of cyclooxygenase (COX)-2. Furthermore, I. dentata inhibited the phosphorylation of c-Jun NH2-terminal kinase and p38 MAPKs, suggesting that it inhibits the secretion of the pro-inflammatory cytokines IL-6 and IL-8, and COX-2 expression, by blocking MAPK phosphorylation. These results suggest that I. dentate can potentially protect against UVB-induced skin inflammation.

  • Objectives : Ecklonia cava is brown alga(Laminariaceae) which grows is sea, it has antioxidant, diarrhea and anticoagulant effect. In this study, the effect of ethanol extract of Ecklonia cava (EC) on peptidoglycan(PGN) -induced NO production in RAW 264.7 cells was investigated. Methods : In the present study, IL-6 production was measured using the enzyme-linked immunosorbent assay(ELISA), prostaglandin $\E_2$($\PGE_2$) production was measured using the EIA kit, and inducible NO synthase(iNOS), cyclooxygenase-2(COX-2), and mitogen-activated protein kinase(MAPK) activation, as determined by western blot analysis and reverse transcription -polymerase chain reaction(RT-PCR). Results : EC inhibited PGN-induced NO and IL-6 production. Consistent with these observations, the protein expression of iNOS and COX-2 were inhibited by EC. Moreover, EC suppressed the phosphorylation of extracellular signal-regulated kinase(ERK) 1/2 in PGN-induced RAW 264.7. Conclusions : These results suggest that EC has inhibitory effects on PGN-induced $\PGE_2$, NO, and IL-6 production, as well as the expressions of iNOS and COX-2 in the murine macrophage. These inhibitory effects occur through blockades on the MAPKs phosphorylation.

경북북부지역 재배한약재와 토양의 중금속 함량연구 (Studies on the Heavy Metal Contents in Cultivated Herbal Medicines and Their Soils at North Kyungbuk Area)

  • The concentration of heavy metal were studied for the estimation of quality in herbal medicines which is Bupleuri Radix, Paeoniae Radix, Dioscoreae Rhizoma and Astragali Radix in products of north Kyungbuk area. And we compared with heavy metal concentration of herbal medicines in products of Kyungbuk and the other area of Korea. Also, we collected soils from several area in Kyungbuk, and determined heavy metal concentration of soils by ICP. We compared with herbal medicines in products of Kyungbuk and their soils. The results were as follow : Kyungbuk area appear to be more proper to cultivated land than other area. For the heavy metals contents of Kyungbuk area, in Gong Jung Ri, represented the higher level, it may be due to a near the road. Correlation coefficients of heavy metal contents in cultured herbal medicines and their soils did not give reproducible results.

중국, 베트남산 약용식물의 최종당화산물 생성저해활성 검색 (IV) (Screening of Herbal Medicines from China and Vietnam with Inhibitory Activity on Advanced Glycation End Products (AGEs) Formation (IV))

  • Advanced glycation end products (AGEs) have been implicated in the development of diabetic complications. The AGEs inhibitors or cross-link breakers attenuate various functional and structural manifestations of diabetic complications. In this study, 64 herbal medicines from China and Vietnam have been investigated with an in vitro evaluation system using AGEs inhibitory activity. Of these, eight herbal medicines ($IC_{50}$<50 ${\mu}g$/ml) were found to have strong AGEs inhibitory activity compared with aminoguanidine (14 days, $IC_{50}$=75.98 ${\mu}g$/ml; 28 days, $IC_{50}$=88.27 ${\mu}g$/ml). Particularly, four herbal medicines, Buddleja officinalis (whole plant), Syzygium cuminii (leaf), Eugenia caryophyllate (seed), and Paeonia suffruticosa (root) showed more potent inhibitory activity (approximately 5-6 fold) than the positive control aminoguanidine.

인터넷상 동의보감 방제의 약물구성 계보 표현 방법에 관한 연구 (Study on the Method of Expressing the Genealogy of the formation of herbal treatments from Treasure Mirror of Eastern Medicines)

  • As the IT technology develops there is a service on internet in which the herbal treatments from medicinal classics can be found in a form of a database. If an assay, and a verification on individual informations on materia medica and the herbal treatments from medicinal classics is established, it would be an enormously efficient asset for the formation of new treatment as well as interpreting the basic information on the efficacy of a treatment. If asp.net is used to make such a system, the user can have access to the informations on materia medica and its treatment without having to setting up a separate system, and especially the user can attain visual information on the genealogy of the herbal treatments in its formation. However, there is a need to develop a more efficient program in terms of organizing the informations on herbs and herbal treatments.

INHIBITORY ACTION OF PROCESSED HERBAL MEDICINES ON THE PRODUCTION OF ADVANCED GLYCATION ENDPRODUCTS(AGEs)

  • Diabetic nephropathy is major chronic complication of diabetes mellitus. Advanced glycation endproducts(AGEs) are largely involved in the pathogenesis of diabetic nephropathy. The irreversibly formed AGEs do not return to normal even if hyperglycemia is corrected and continue to accumulate over the lifetime of protein. The AGEs inhibitor. aminoguanidine(AG), is the only protein glycation inhibitor currently under development. its safety however is desirable. To find possible AGEs inhibitor in herbal medicines, bovine serum albumin was added to a mixture of sugars and some of processed. unprocessed herbal medicines or AG. Cyperi rhizoma was processed in four different methods according to chinese pharmacopoeia and traditional literatures. In comparision to the negative control with no inhibitor and positive control with AG. alcoholic extracts of these processed cyperi rhizoma proved to have more potent inhibitory activities than that of unprocessed cyperi rhizoma. These results revealed that some processed herbal medicines have a more potent in vitro inhibitory action on AGEs formation than AG. suggesting the possible candidate for diabetic nephropathy from the processed herbal medicines.

  • Objectives : DojukSan is known to be effective for treating a urinary diseases and stomatitis. However, there has been a lack of studies regarding the effects of Dojuksan on the inflammatory activities and effector inflammatory disease mechanism about macrophage before is not known. To elucidate the molecular mechanisms of Dojuksan water extract (DJS) on pharmacological and biochemical actions in inflammation, we examined the effect of DJS on pro-inflammatory mediators in lipopolysaccharide (LPS)-stimulated macrophages. Methods : In the present study, pro-inflammatory cytokine production was determined by performing enzyme-linked immunosorbent assay, reverse transcription polymerase chain reaction, and western blot analysis to measure the activation of MAPKs. Cells were treated with 200 ng/mL of LPS 1 h prior to the addition of DJS. Cell viability was measured by MTS assay. The investigation focused on whether DJS inhibited nitric oxide (NO) and prostaglandin E2 ($PGE_2$) productions, as well as the expressions of inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), interleukin-6 (IL-6) and mitogen-activated protein kinases (MAPKs) in LPS-stimulated RAW 264.7 cells. Results : We found that DJS inhibited LPS-induced NO, $PGE_2$ and IL-6 productions as well as the expressions of iNOS and COX-2. Furthermore, DJS suppressed the LPS-induced phosphorylation of p38 MAPK and c-Jun NH2-protein kinase (JNK). Conclusions : These results suggest that DJS has inhibitory effects on LPS-induced $PGE_2$, NO, and IL-6 production, as well as the expressions of iNOS and COX-2 in the murine macrophage. 랫드에서 헛개나무와 천마 혼합 추출물의 에탄올 섭취에 의한 숙취 제거 효과 (Eliminatory Effect of Mixed Extract of Hovenia Dulcis Thunb and Gastrodia Elata on Ethanol-Induced Hangover in Rats)

  • To investigate an eliminatory effect of mixed extract of Hovenia dulcis Thunb, Gastrodia elata and Alnus japonica etc., on the ethanol-induced hangover, 12 hr-fasted male Sprague-Dawley rats weighing 220±20 g were given mixed extract (10 mL/kg, p.o.) and administered ethanol at a dose of 3 g/kg bw (25% in distilled water) orally 30 min postdosing. Blood was collected from caudal artery at 0.25, 0.5, 1, 2, 4, 6, 8, 10, and 12 hr and then the animals were sacrificed at 24 hr after the ethanol treatment. From 0 to 12 hr, the administration of mixed extract significantly decreased the area under the serum alcohol concentrations-vs.-time curves by 21 % compared with control group. In these experiments, liver function indices, such as alanine aminotransferase, aspartate aminotransferase and sorbitol dehydrogenase activities, showed unaltered results in all treated groups compared with the normal group. These results suggest that oral intake of the mixed extract may be effective on elimination of ethanol-induced hangover.
