Inhibitory effects of ethanol extract of Atractylodis Rhizoma on melanogenesis in B16/F10 melanoma cells

  • The inhibitory effect of Atractylodis Rhizoma alba extract(AM) on melanogenesis was studied by using B16/F10 melanoma in culture. Cells were cultured in the presence of various concentrations of AM for 48 hrs, and the experiment of total melanin content as a final product and activity of tyrosinase, a key enzyme, in melanogenesis. AM significantly inhibited tyrosinase activity, and melanin content in a dose-dependent manner. These results show that Atractylodes macrocephala extract could be developed as skin whitening components of cosmetics.

다형홍반 치험 1례 (A Case of Erythema Multiforme(EM))

  • Background and Objective: Erythema multiforme(EM) is related disorders of skin and mucous membranes which is typically associated with antecedent medications or infections. etc. EM is an erythematous maculopapular cutaneous eruption of variable form. However, the pathophysiology of the EM remains obscure. Treatment at present is symptomatic and supportive. There is no report on EM treated with oriental medical treatment. Methods: We conducted one patient with EM seen at Won-Kwang University Kwang-Ju Oriental Medical Hospital in 2004. We treated EM in a twenty-two years old man by a herbal medicine(Joa-Chang-Bang), a herbal medicine for external use, acupunture, moxa, COTDE-made cosmetics, and P-Tx(carbon). Conclusions. We had a significant results. So we report this case to estimate the efficiency of oriental medical treatment and management on EM.

HaCaT Cell에서 황기 에탄올 추출물의 Aquaporin-3 발현 효과 (Ethanol Extract of Astragalus membranaceus Bunge Induces Aquaporin-3 Expression in HaCaT Cells)

  • Astragalus membranaceus Bunge is used in herbal medicine in Eastern Asian countries including Korea. In this study, we assessed the effects of A. membranaceus extract (AM) on the aquaporin-3 (AQP3) protein expression in HaCaT cells. AM did not affect viability of HaCaT cells. AQP3 expression and cell migration seem to be maximal at $100{\mu}g/mL$ concentration. Epidermal growth factor receptor (EGFR) kinase inhibitor, PD153035, blocked AM-induced AQP3 expression and cell migration. In addition, an 80% ethanol extracts of herbal prescription, SinhyoTakleesan (ST), which is composed of A. membranaceus, Angelicae gigantis, Glycyrrhiza glabra Linne, and Lonicera japonica Flos also induced AQP3 expression at $20{\mu}g/mL$ in HaCaT cells. Collectively, these results suggest that AM induce AQP3 expression via EGFR pathway.

연교승마탕(連翹升麻湯)이 함유된 한방화장품이 아토피 피부염에 미치는 임상 연구 (A Clinical Research of Atopic Dermatitis treated by Yeongyuseungmatang in cosmetics)

  • Objective : This clinical research was conducted to test patients with Atopic Dermatitis by external application with Yeongyuseungmatang in cosmetics. Methods : We gave scores to 33 patients who visited the Dept. of Oriental Medical Opthalmology & Otolaryngology & Dermatology of Semyung Jecheon Oriental Medical Hospital from April 17th, 2008 to June 12th. Seventeen patients were treated with Yeongyuseungmatang(Experimental group) and sixteen patients were treated with Atopico skin care cream(Control group) for 4weeks. We observed change of total IgE, eosinophil count, Skin Temperature, Transepidermal Water Loss(TEWL), Skin Hydration and Skin pH. Also Clinic Index of Atopic Dermatitis(SCORAD Index) and global assesment of efficacy were used to evaluate the effects of Yeongyuseungmatang. Statistical analysis was performed by using frequency analysis and descriptive analysis. Statistical significance was achieved if the probability was less than 5%(p<0.05). Results : 1. After 4 weeks of external application treatment, SCORAD Index in Yeongyuseungmatang was significant statistically decreased compared with control group. 2. After 4 weeks of external application treatment, eosinophil count in Yeongyuseungmatang was significant statistically decreased compared with control group but total IgE was very increased both Yeongyuseungmatang and control group. 3. Statistically, Yeongyuseungmatang and control group didn't showed significant effect on Skin Temperature was measured by Thermometer SM815. 4. Transepidermal Water Loss(TEWL) in Yeongyuseungmatang was significant statistically decreased compared with control group measured by Tewameter TM300. 5. Skin Hydration in Yeongyuseungmatang was significant statistically increased compared with control group measured by Corneometer CM825. 6. Statistically, Yeongyuseungmatang and control group didn't showed significant effect on Skin pH was measured by Skin-pH-Meter PH905. 7. After 4 weeks of external application treatment, Yeongyuseungmatang showed significant effect on global assesment of efficacy. Conclusion : Considering the above results, we can speculate that cosmetics with Yeongyuseungmatang has some therapeutical effects in mitigating the symptoms of atopic dermatitis.

  • Recently. the herbal industry has expanded into the herbal dietary supplements industry, the herbal cosmetics industry, etc. But still the main form of herbal drug usage is water extraction- packaging after herb preparation in private clinics. There was no report or data of investigations about industrial herbal preparations. To search for a better plan of herbal drug improvement and popularization. we surveyed the present state of Korean herbal preparation production. We made an industrial herbal preparation list, investigated the total amount of production, the chief items of manufacture. names of prescriptions each item is based on. scales of each production company, etc., and report on them on this article.

산수유 클로로포름 추출물과 분획물의 암세포주에 대한 세포독성 (Cytotoxicity Effects of Fraction and Chloroform Extracts from Corn is fructus on Cancer Cell Lines)

  • Cornis fructus were extracted by successive extractions and then fractionated with chloroform extract to get active fractions. This study was performed to determine the cytotoxic effect of chloroform extract from Corn is fructus on NIH 3T3 fibroblasts and cancer cell lines using MTT assay. All extracts did not exhibit cytotoxicity in NIH 3T3 fibroblasts. Chloroform extract exhibited antitumor activity in A549, MDA-MB-123, B16 melanoma and SNU-C4 cells. Futher fractionation with chloroform extract was performed to obtain effective fractions. 3 fraction showed the strongest cytotoxic effect against A549, MDA-MB-123, B16 melanoma and SNU-C4 cells. These results suggest that 3 fraction of the chloroform extract from Cornis fructus possessed bioactive material of antitumorous agents.

아토피 피부염 환자에서 황연해독탕(黃連解毒湯)이 함유된 한방화장품에 대한 임상 연구 (Clinical Research of Atopic Dermatitis Treated by Hwangryeonhaedok-Tang in Cosmetics)

  • This clinical research was conducted to test patients with Atopic Dermatitis by external application with Hwangryeonhaedok-Tang in cosmetics. We gave scores to 31 patients who visited the Dept. of Oriental Medical Opthalmology & Otolaryngology & Dermatology of Semyung University Jecheon Oriental Medical Hospital from May 15th, 2008 to June 26th. Fifteen patients were treated with the ointment that contain Hwangryeonhaedok-Tang (experimental group) and sixteen patients were treated with normal ointment that doesn't have Hwangryeonhaedok-Tang (control group) for 4 weeks. We observed change of total IgE, eosinophil count, Skin Temperature, Transepidermal Water Loss(TEWL), Skin Hydration and Skin pH. Also Clinical Index of Atopic Dermatitis(SCORAD Index) and global assesment of efficacy were used to evaluate the effects of Hwangryeonhaedok-Tang. Transepidermal Water Loss(TEWL) in both groups were increased but experimental group was significant decreased compared with control group. Skin Hydration in experimental group was significant statistically increased compared with control group. Statistically, both groups didn't showed significant effect on Skin pH. Only experimental group showed little increase. After 4 weeks of external application treatment, experimental group showed significant effect on global assesment of efficacy. Considering the above results, we can speculate that cosmetics with Hwangryeonhaedok-Tang has some therapeutical effects in mitigating the symptoms of Atopic Dermatitis.

  • Natural products have been used as drugs and cosmetics due to their bioactivity and their biochemical diversity. Natural products usually refer to secondary metabolites produced by various living organisms including marine animals, insects, microbes, amphibians, and plants. These secondary metabolites, which usually have molecular weights less than 2,000 amu, are unnecessary for survival, development, growth, and reproduction but play major roles in plant defense systems against other species. These secondary metabolites such as lignans, flavonoids, monoterpenes, and phenylethanoid glycosides showed various biological activities like anti-oxidant behavior, anti-cancer properties, neuroprotective properties, and so forth. Thus, isolation and elucidation of secondary metabolites from living organisms is of great significance to human life.

Capsosiphon Fulvescens Decreases Melanin Synthesis Via Downregulation of Tyrosinase and TRP-2 Expression

  • The green marine algae, Capsosiphon fulvescens (CF) is a food supplement cultivated in south coast of Southern Korea. The purpose of this study was to investigate the mechanism of CF-induced hypopigmentation. The present study was designed to determine the effect of CF extracton melanogenesis in B16 cells, particularly its specific effects on tyrosinase and tyrosinase-related protein 2 (TRP-2). We measured melanin contents and analyzed melanosome associated protein levels using Western blot and Reverse transcription-polymerase chian reaction (RT-PCR) analysis. CF extract markedly inhibited melanin synthesis and tyrosinase activity. In addition, cellular dendricity was slightly decreased by CF extract. In further experiments, CF extract significantly reduced the protein levels of tyrosinase and TRP-2 in B16 cells. RT-PCR analysis revealed that tyrosinase and TRP-2 mRNA levels were unaffected by CF treatment. Therefore, these results suggest that hypopigmentary effect of CF was due to post-translational degradationof tyrosinase and TRP-2.
