사립체 질환: 새로운 위대한 모방자가 될 것인가? (Mitochondrial Disease: Will it become a New Great Imitator?)

  • Mitochondrial disease is a group of disorders caused by dysfunctional mitochondria, the organelles that generate energy for the cell. Diagnosis of mitochondrial disease is difficult, subtle, and has many problems. It is more likely to miss the diagnosis of mitochondrial disease, especially in borderline cases where the symptoms of the disease are not severe. In this regard, urine organic acid analysis is noninvasive and can increase the sensitivity and specificity through repeated load test with few changes according to the specimen. And, It is considered to be suitable as a screening test for mitochondrial diseases because it has a great advantage of distinguishing from organic aciduria, urea cycle disorder and fatty acid oxidation disorder which may have similar symptoms. The purpose of this study was to investigate the clinical features and age distribution of mitochondrial diseases diagnosed by organic acid analysis and to establish the policy of diagnosis and treatment based on this study.

한국전통간장의 맛과 향에 관여하는 주요 향미인자의 분석(I) - 일반특성 및 당류와 유기산 분석 - (Analysis of Significant Factors in the Flavor of Traditional Korean Soy Sauce (I) - Analysis of General Characteristics, Sugars and Organic Acids Contents -)

  • This study was carried out in order to investigate general characteristics, sugars and organic acids contents of Korean traditional soy sauce, and to find out possibility of high quality soy sauce production in a short period through high concentration soy sauce making. In this study, we prepared three different types of soy sauce, low concentration soy sauce (Chungjang), high concentration soy sauce and Kyupjang, high quality traditional Korean soy sauce. pH of soy sauce were $4.46{\sim}4.90$ and did not show difference among three samples. Titratable acidity, buffering power and total free acid content were the highest value in Kyupjang. Kyupjang showed the highest contents of salt and pure extract. As the ripening period increased, the salt content increased in Chungjang samples, but decreased in Kyupjang. Reducing sugar contents of Kyupjang, high concentration soy sauce and low concentration soy sauce were 1.13%, 0.76% and 0.53%, respectively. Free sugar in soy sauce were analyzed maltose, glucose, galactose and fructose. Total free sugar content was highest in high concentration soy sauce, however, contents of glucose and fructose were higher in Kyupjang than in Chungjang samples. Galactose was the main free sugar in Chungjang, but glucose was in Kyupjang. Among identified volatile organic acid, acetic acid was present in the highest concentration, and volatile organic acid content was highest in the high concentration soy sauce at 150 days. 20 nonvolatile organic acids were detected in Korean traditional soy sauce. Succinic acid, lactic acid and 2,5-pyridine dicarboxylic acid were the main nonvolatile organic acid in soy sauce.

원각사지 십층석탑 오염물의 유기산 분석 (Analysis of the Organic Acid Contaminants on the surface of TEN-STORIED STONE PAGODA ON THE SITE OF WON-GAKSA)

  • TEN-STORIED STONE PAGODA ON THE SITE OFWON-GAKSA(Temple) which is one of three marble pagodas in South Korea, were dated from the thirteenth year of the reign of King Sejo(1467). On the roof and surface of each the stories, there were large amount of the contaminants such as pigeon′s excretions, dust and environmental elements for a long time. The pH of contaminants is not acid, but is 7.2, neutral. To find the species of organic acidscontained in the contaminants and the degree of damaging for a marble pagoda, we analyzed the contaminants using GC-MSD method by the following procedures. Organic acids were extracted by saponifying whole contaminants. After Saponification, the organic acids were mathylated to increase their volatility upon subsequent GC-MSD analysis. The mathyl esters of the organic acids are extracted from the acidified aqueous solution. And the organic extracts were washed with adilute base solution. The washed extract were analyzed by GC(Hewlett Packard 5890)with a nonpolar capillary column(Crosslinked 5% Ph Me Silicone, $50×0.2㎜×0.33\mum$film thickness, USA) and Mass Spectrometric Detector(Hewlett Packard 5970B).As the result, it was found that 12 organic acids were the main compound in pagoda′contaminants, and the amount of organic acid were negligible.

오미자의 용출시간에 따른 풍미성분 변화에 관한 연구 (Changes in Flavor Component of Omija, Shizandra Chinensis Baillon, with Various Extraction times)

  • This study attempted to set up reasonable extraction time of Omija that was put in water for the various components to soak out. Changes of free sugars, organic acids, reducing sugar, total acid and tannin in Omija with various extraction times were investigated (together with the analysis of each components in Omija fruit). 1. High Performance Liquid Chromatography showed fructose, glucose, and sucrose to be the major free sugars of the Omija fruit. Free sugars and reducing sugar value in Omija beverage increased gradually in according with the extraction time, and marked 75.6% per total free sugars and 82.1% per total reducing sugar at 12 hours. 2. Gas Chromatography showed lactic acid, oxalic acid, fumaric acid, levulinic acid, succinic acid, malic acid, citric acid and pyroglutamic acid to be the major organic acids of the Omija fruit. Organic acids and total acids value in Omija beverage increased gradually on proportion to extraction time, and marked 97.0% per total organic acids at 9 hours and 79.0% per total acids at 12 hours. 3. Tannin content in Omija beverage was increased when extraction time was longer but it showed a low percentage as compared with the reducing sugar and total acid. Tannin content marked 48.8% per total tannin at 12 hours. 4. Sensory evaluation revealed that !1 hours of extraction produced the best quality products based in taste, flavor, color and over-all acceptability, considering the data, it seems possible to conclude that the optimum of time for extraction of Omija to water is 9 hours.

Surface modification of graphene oxide by citric acid and its application as a heterogeneous nanocatalyst in organic condensation reaction

  • A citric acid functionalized graphene oxide nanocomposite was successfully synthesized and the structure and morphology of the nanocatalyst were comprehensively characterized by Fourier transform infrared spectroscopy, energy-dispersive X-ray analysis, X-ray diffraction patterns, atomic force microscopy images, scanning electron microscopy images, transmission electron microscopy images, and thermogravimetric analysis. The application of this nanocatalyst was exemplified in an important condensation reaction to give imidazole derivatives in high yields and short reaction times at room temperature. The catalyst shows high catalytic activity and could be reused after simple work up and easy purification for at least six cycles without significant loss of activity, which indicates efficient immobilizing of citrate groups on the surface of graphene oxide sheets.

지방산을 이용한 Nuttallia olivacea 및 Nereidae의 먹이원에 관한 연구 (The Origin of Food Sources for Nuttallia olivacea and Nereidae by Fatty Acid Analysis)

  • The spatial variability in the food chain structure of an estuary environment (Nanakita estuary, Japan) was investigated using fatty acid. Potential organic matter sources (terrestiral plants, macroalgae, benthic microalgae, dinflagellates and bacteria), sedimentary organic matters and benthic invertebrates (Nuttallia olivacea and Nereidae) were sampled in four locations with different tidal flat type. The main objective of the present study was to determine the origin of sediment and the food sources of N. olivacea and Nereidae along with small-scale spatial variability. The origin of sedimentary organic matters were mainly the fatty acid of bacteria and benthic microalgae. Especially, The organic matter of terrestrial plant origin was found the highest in station C. The diets of N. olivacea and Nereidae were found to be dominated by diatoms and terrestrial plants. Whereas, macroalgae and dinoflagellates showed little influence to benthic invertebrates. Moreover, according to principal component analysis, it is showed that benthic invertebrates in the same region are using the same food without relation with species. On the other hand, the N. olivacea and Nereidae of station D clearly contrasts with station B in terms of main food sources. From these results, it is suggested that food competition of benthic invertebrates revealed high a connection between small-scale spatial variability and food source in estuary.

항암제 유발 말초신경병증환자와 유기산검사 마커와의 상관성 연구 (Correlation Analysis of Organic Acid Comprehensive Profile Markers with Chemotherapy Induced Peripheral Neuropathy in Cancer Patients)

  • Objectives: The purpose of this study is to evaluate the urinary organic acid comprehensive profile for chemotherapy induced peripheral neuropathy (CIPN). Methods: Participants are 66 patients with CIPN who had symptom (Visual analog scale ${\geq}30mm$, Eastern Cooperative Oncology Group ${\leq}2$). Participants were tested with organic acid comprehensive profile markers. Results: Positive Correlation was observed in the neurotransmitter metabolism markers, N-methyl-D-aspartate (NMDA) modulators markers, detoxification markers, energy production markers, amino acid metabolism markers, and intestinal dysbiosis markers. Especially, all the neurotransmitter metabolism markers were showed positive rate of 44%. In addition, neuro-endo-immune was associated with energy metabolism (mitochondrial dysfunction) in CIPN of cancer patient. especially detoxification, intestinal bacterial hyperplasia, vitamin deficiency (folate, complex B group, vitamin C). Conclusions: Significant urinary organic acid comprehensive profile results were obtained in cancer patients who induced peripheral neuropathy by chemotherapy.

유기산혈증-유기산 분석 8년의 정리 (Organic acidemias in Korea - Eight years experience of organic acid analysis -)

  • 목 적 : 국내의 유기산 대사 이상 질환들의 상대적 발병 빈도와 임상양상을 알아보기 위하여 유기산 정량 분석이 가능하여진 1997년 7월부터 2005년 5월까지 8년간 전국에 있는 대학병원에서 의뢰된 1,787명의 환자의 검체를 분석한 결과를 3년 전 발표한 연구결과와 비교하여 정리하였다. 방 법 : 1997년 7월 1일부터 2005년 5월 31일까지 약 8년 동안 본원 임상유전학연구실로 분석이 의뢰되었던 1,787명 환자의 검체들의 분석결과를 신생아기(생후 2개월까지), 영아기(2개월 이후 2세까지), 유소아기(2세 이후 12 세까지) 및 청소년기 이후(12세 이후)로 분류하였고, 환자들의 주요증상 및 소견을 분류하였다. 유기산 분석은 83종의 유기산을 gas chromatography와 mass spectrometry를 이용하여 정량 분석하였으며, 특징적인 양상으로 진단하였다. 결 과 : 총 28가지 질환 470명의 환자를 이 기간 동안에 진단할 수 있었으며, 가장 빈도가 높았던 질환들은 cytosolic 3-ketothiolase 결핍증, 사립체의 호흡 연쇄 효소의 이상 질환군(mitochondrial respiratory chain disorders), PDHC 결핍증, mitochondrial 3-ketothiolase 결핍증, glutaric aciduria type II biotinidase 결핍증, 메틸말론산뇨증(methylmalonic aciduria), 프로피온산뇨증(propionic aciduria) 등이었다. 결 론 : 유기산 대사 장애 질환들은 각각의 발병 빈도는 낮으나 국내에서도 다양한 질환들이 발병되고 있으며, 이들 질환들은 대부분의 경우 신경학적 이상 소견을 보이고 있으므로 신경학적 이상 소견을 보이는 경우는 감별진단에 포함시켜야 된다. 3년전 Choi 등이 발표한 연구결과와 비교하여 새로운 질환의 출현은 없었고 빈도가 높은 질환의 발병빈도의 차이도 보이지 않았다. 3년 전의 연구당시 짧은 연구기간으로 인한 오차의 가능성을 염려하였으나 8년 동안의 긴 연구기간 동안 비슷한 상대적 발병빈도를 보여 이러한 질환들이 우리나라에서 다수 발병되는 것으로 판단되며 주의가 필요하다고 판단된다.

유기산 처리가 해도형 초극세 폴리에스터 섬유의 용출 특성에 미치는 영향 (1) (Accelerating Effect of Organic Acid Treatment on Weight Reduction Characteristics of Sea-Island Type PET Supermicrofiber (1))

  • In order to make a supermicrofiber fabric with PET/co-PET sea-island type filament fiber, the optimum conditions of dissolution of co-PET composite filament fiber was examined. The data set was made at various organic acid concentration and steam temperature with treated time as a main variable. At the same time, the microstructure changes by organic acid treatment of sea-island type PET supermicrofibers were monitored by thermal analysis morphology, DSC thermgrams and SEM images. Weight reduction behavior of supermicrofibers by caustic treatment was investigated in a comparative method. The termination of dissolution was also confirmed using DSC thermgrams and SEM images.

폴리락틱산/산화아연 기둥구조의 유기사포나이트 나노복합체의 특성, 구조 및 결정화 (Properties, Structure and Crystallization of Poly Lactic Acid/Zinc Oxide Pillared Organic Saponite Nanocomposites)

  • ZnO pillared saponite was synthesized via a microwave hydrolysis method. To enhance interfacial compatibility between zinc oxide (ZnO) pillared saponite and poly lactic acid (PLA), ZnO pillared organic saponite was prepared by intercalation modification of cetyltrimethylammonium bromide. Moreover, PLA/ZnO pillared organic saponite nanocomposites were prepared by melting processing. The microstructure analysis of PLA/ZnO pillared organic saponite nanocomposites showed that ZnO pillared organic saponite was exfoliated and homogeneouslydispersed in PLA matrix. The property results showed that ZnO pillared organic saponite improved the mechanical properties and thermal stabilities of PLA/ZnO pillared organic saponite nanocomposites. Differential scanning calorimetry (DSC) demonstrated that ZnO pillared organic saponite restrained the appearance of cold crystallization, lowered the glass transition temperature and melting temperature of PLA, and improved the crystallinity of PLA. The results demonstrated that ZnO pillared organic saponite had a good interfacial compatibility and heterogeneous nucleation effect in PLA matrix, and also played an active role in accelerating the crystallization process of PLA.
