Effects of Yunpaejaesueum extract on the contraction of isolated guinea pig trachea smooth muscle (윤폐제수음(潤肺除嗽飮)이 GUINEA PIG의 기관지(氣管支) 평활근(平滑筋)에 미치는 영향(影響))

  • In order to study the effects of Yunpaejaesueum known clinically for their effects of treatment for cough and asthma, the study was carried out to investigate the effect of Yunpaejaesueum extract on the contractile force of the isolated guinea pig trachea smooth muscle and elucidate its mechanism. The results were obtained as follows: 1. The isolated trachea smooth muscle of guinea pig was suspended in the organ bath with oxygenated Kreb's Henseleite bicarbonate buffer solution at $37^{\circ}C$, and the developed tension by the drug was recorded with isometric transducer(Nacro F-60). The resting tension was approximately 0.5g. 2. The isolated trachea smooth muscle of guinea pig was remakably relaxed by the administration of Yunpaejaesueum. 3. Yunpaejaesueum is significantly inhibited the contractile response of histimine 10-4 M in isolated guinea pig trachea smooth muscle. 4. Yunpaejaesueum is significantly inhibited the contractile response of acetylcholine 10-4 M in isolated guinea pig trachea smooth muscle. 5. Yunpaejaesueum is significantly inhibited the contractile response of 5-hydroxytryptamine 10-4 M in isolated guinea pig trachea smooth muscle. 6. Yunpaejaesueum is significantly inhibited the contractile response of prostaglandin F2a 10-7 M in isolated guinea pig trachea smooth muscle.

  • The purpose of this study is to investigate vasodilatation effect of complex saponin separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract on rabbit carotid artery. In this study, to determine vasodilatation effect of complex saponin separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract on rabbit carotid artery, arterial rings with intact or damaged endothelium were used for experiment using organ bath, and were contracted by endothelin. complex saponin, major active constituents of Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract, showed a moderate vasodilatation effect on the basilar arteries of rabbits. Therefore, treatment with complex saponin separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract may selectively accelerate cerebral blood flow through dilatation of the basilar artery. Theseis result suggest a potential role of complex saponin separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract as source of vasodilatation agent.

  • This study was carried out to investigate the effect of Jasoeumja extract on the contractile force of the isolated guinea pig trachea smooth muscle and elucidate its mechanism. The results were obtained as follows: 1.   • The purpose of this study was to determine the effects of oriental medical care on growth deficiency children. These cases were collected to investigate the effects of oriental medical care on growth deficiency children. So the cases were divided into 5 groups. The growth velocity was compared to a normal growth curve, and the results are as follows: 1. Oriental medical care was efficient when the epiphysial plates were not closed. 2. In female middle school beyond 1st grade students and in male middle school 3rd grade students who had almost closed epiphysial plates, the oriental medical care was not efficient and did not promote growth velocity. endothelium were used for the experiment using organ bath. 3. In the cases where the parent were very short, the effects of the oriental medical care to promote growth velocity was not efficient. 4. when we treated children of normal height, their growth velocity was over the normal growth curve. 5. In preschool aged children, oriental medical care was very efficient to promote children's growth. 6. The principle oriental medical care was to make balanced their body and digestive organs by using herb medicines that have the capability to promote the growth of the bones, and build strong muscles.

  • This study was undertaken to define the mechanism of Siegesbeckiae Herba-induced relaxation in rabbit common carotid artery contracted by agonists. In order to investigate the effect of Siegesbeckiae Herba on contracted rabbit carotid arterial strips, transverse strips with intact or damaged endothelium were used for the experiment using organ bath. To analyze the mechanism of Siegesbeckiae Herba-induced relaxation, Siegesbeckiae Herba extract infused into contracted arterial strips induced by agonists after treatment of lanthanum chloride, indomethacin, atropine, $N\omega-nitro-{_L}-arginine$, cobalt chloride or methylene blue. The relaxation effect of Siegesbeckiae Herba was dependent on the presence of endothelium, showing that Siegesbeckiae Herba-induced relaxation was not observed in the strips without endothelium. The endothelium-dependent relaxation induced by Siegesbeckiae Herba was suppressed by the pretreatment of lanthanum chloride, $N\omega-nitro-{_L}-arginine$, cobalt chloride or methylene blue, but it was not observed in the strips pretreated with indomethacin or atropine. These results demonstrated that Siegesbeckiae Herba may inhibit agonist-induced contraction through an increase in the cyclic GMP by the production of nitric oxide in the vascular endothelial cells.

  • This study was undertaken to define the effect of ChungSangSaWhaTang on the norepinephrine-induced arterial contraction and the mechanism of ChungSangSaWhaTang-induced relaxation. In order to investigate the effect of ChungSangSaWhaTang on contracted rabbit carotid arterial strips, transverse strips with intact or damaged endothelium were used for the experiment using organ bath. To analyze the mechanism of ChungSangSaWhaTang-induced relaxation, ChungSangSaWhaTang extract infused into contracted arterial strips induced by norepinephrine after treatment of indomethacin, $N_{\omega}$-nitro-L-arginine, or tetraethylammonium chloride. ChungSangSaWhaTang relax arterial strip with endothelium contracted by norepinephrine, but in the strips without endothelium, ChungSangSaWhaTang-induced relaxation was significantly inhibited. The endothelium-dependent relaxation induced by ChungSangSaWhaTang was decreased by the pretreatment of $N_{\omega}$-nitro-L-arginine, but it was not observed in the strips pretreated with indomethacin or tetraethylammonium chloride. We suggest that ChungSangSaWhaTang may inhibit agonist-induced contraction through the formation of nitric oxide in the vascular endothelial cells.

흰쥐 적출대동맥의 수축력에 미치는 열과 Nacl의 영향

  • In this study, in order to examine whether salt and heat shock stress would alter or not contraction and relaxation of isolated rat aorta. Under anesthesia with sodium pentobarbital(50 mg Kg$^{-1}$ i.p.), male Sprague Dawley rats weighing 300-330 g were subjected to 0, heat shock combined salt stress, where as the sham group was left at modified Krebs-bicarbonate solution. To measure contractile response of vascular ring preparation isolated from rat was determined in organ bath and was recorded on physiograph connected to isometric transducer. And the strip was checked for expression of heat shock protein(Hsps) by means of western blotting. The combination group of heat and 50 mM NaCl group increased vascular contractility, and the heat and 150 mM NaCl group decreased vascular contractility for 5 hours, and then recovered for 8 hours compared to that of control. Expressin of Hsp 70 of vascular muscle of rat aorta more increased by combination of heat and NaCl treatment than those of single treatment of heat or NaCl treatment, and vascular Hsp 70 showed a little decrease at 8 hours compared at 5 hours. These result indicate that mixed environmental stress either increased or decreased in vascular contractility by combination of heat and NaCl concentration.
