Submerged Culture of Phellinus linteus for Mass Production of Polysaccharides

  • In order to increase the mycelial production of Phellinus linteus, which exhibits potent anticancer activity, some ingredients of the medium used to culture P. linteus were investigated. The optimal medium composition for the production of Phellinus linteus was determined to be as follows: fructose, 40 g/l; yeast extract, 20 g/l; $K_2HPO_4$, 0.46 g/l; $K_2HPO_4$, 1.00 g/l; M$MgSO_4\cdot7H_2SO$, 0.50 g/l; $FeCl_2\cdot6_2O$, 0.01 g/l; $MnCl_2\cdot4H_2O$, 0.036 g/l; $ZnCl_2$, 0.03 g/l; and $SuSO_4\cdot7H_2O$, 0.005 g/l. The optimal culture conditions were determined to be as follows: temperature, 28$^{\circ}C$; initial pH, 5.5; aeration, 0.6 vvm; and agitation, 100 rpm, respectively. Under optimal composition and conditions, the maximum mycelial biomass achieved in a 5 l jar fermentor was 29.9 g/l.

열처리를 통한 미세조류로부터 바이오수소 생산 향상 (Enhanced of Bio-Hydrogen Production from Microalgae by Thermal Pre-Treatment)

  • This study was conducted to increase the amount of bio-hydrogen production from microalgae(Chlorella vulgaris) in batch reactors by thermal pre-treatment. The optimization of thermal pre-treatment was conducted using statistic experimental design of response surface methodology. Two experimental parameters of temperature and reaction time were considered. The optimization condition was founded at the coded variables of <0.52, -0.07> corresponding to the experimental of heating temperature of $95.6^{\circ}C$ and reaction time of 57.9 min, respectively. Under the optimal condition, the maximum hydrogen production was predicted to 25.3mL $H_2/g$ dry cell weight (dcw), which was 9.1 times higher value of control(2.8mL $H_2/g$ dcw).

동물성 유지를 이용한 Biodiesel 생산기술에 관한 연구 (A Study on the Biodiesel Production Technology using Lard Oil)

  • Flesh wasters from tannery create major environmental problems. Despite their considerable fat content, these waster do not find important usage. Their disposal is also troublesome and costly. We have investigated the possible use of this fat as the production of biodiesel(fatty acid methyl esters) by transesterification using with fossil fuels. The fat released the waste by boiling water under the optimal condition (i.e., temperature, $120^{\circ}C;$ decompression, 200mbar) and used to dry without refining for the production of fatty acid methyl esters. Under the optimal condition, the experimental value of biodiesel yield was about 96%. The result of the chemical and GC analysis showed fatty acid composition and characteristics of biodiesel. Evaluation of the product indicated that it was suitable for use as a biodiesel fuel. In result of this experiment oil extract from fleshing process can be transformed into an environmentally affination fuel, to provide economical and ecological profits.

혈전 분해효소 생산균의 탐색 및 효소생산 최적조건의 조사 (Screening of Fibrinolytic Enzyme Producing from Microorganisms and Optimum Conditions of Enzyme Production)

  • 전통식품(고추장, 된장, 쌈장, 젓갈류, 김치)으로부터 혈전 용해능이 있는 미생물을 분리한 후 그 중 fibrin용해능이 가장 우수한 균주 KJ-23을 최종 선발하였다. 최종 선별한 균주 KJ-23을 Microbial identification system(MIS)의 Sherlock 95 program을 이용하여 동정한 결과 Bacillus brevis KJ-23으로 동정하였다. Bacillus brevis KJ-23균주의 최대혈전용해 생산을 위한 최적배지화 조건을 검토한 결과 nutrient broth, D-ribose 0.5%, malt extract 0.5%, $K_2$HPO$_4$ 0.3% 첨가시 최고의 활성을 보였으며, 금속염을 첨가했을 경우에는 오히려 활성이 감소하는 경향을 보여 금속원은 최적 배지화 조건에서 첨가하지 않았다. 또한 pH7.0, 3$0^{\circ}C$에서 진탕배양했을 경우 최대활성을 보였으며, 시간별 경시효과에서는 24시간에서 혈전용해능이 최대를 나타내었다.

Glutathione 생산균주의 분리 동정 및 생산조건 (Isolation, Identification and Culture Condition of Microorganism Producing Glutathione)

  • 춘천시 근교의 토양으로부터 glutathione 생산균주를 분리, 동정하고 glutathione 생산 최적조건을 검토하였다. 분리된 균주는 동정을 행한 결과 Candida sp.로 판단되었다. Glutathione 생산을 위한 Candida sp의 배지조성은 fructose 1.0, yeast extract 4.0, NaCl 0.04, thiamine-HCl5{\mu}g/ml$ 및 L-cysteine 0.04 이었으며 온도는 $25^{\circ}C$, pH 6.0에서 36시간 배양하였을 때 glutathione 생산이 가장 좋았다. Glutathion 생산을 위한 최적배지에서 Candida sp. 균주에의한 glutathione 생산량은 $92{\mu}g/ml$로 YM 배지보다 약 2.5배 정도 증가하였다.

뽕나무버섯(Armillaria mellea) 균사체 생산의 최적화 (Optimal Condition for Mycelial Production of Armillaria mellea)

  • 뽕나무 버섯균은 식용버섯으로써 노인병, 중풍, 현기증, 두통, 신경쇠약, 불면증, 사지마비, 소아마비, 간질 등에 효과가 있는 것으로 알려져 있으므로, 뽕나무 버섯 균사체의 고농도 배양을 위한 최적 조건확립 하고자 하였다. 뽕나무 버섯균사체 생산의 최적배지 선정을 위하여 7종의 배지를 대상으로 수행한 결과 맥아즙배지가 뽕나무 버섯 균사체 생산에 가장 우수하였다. 맥아즙 배지에서 뽕나무 버섯균 균사체 생산을 위한 적정조건은 맥아즙 농도 $15\;Brix^{\circ}$이었으며 온도, pH, 배양기간의 조건을 반응표면 분석법에 의해서 실행한 결과, $25.9^{\circ}C$, pH 5.72, 15.22일의 배양에서 3,40 g/50 ml의 최대 예측치를 나타내었다. 이는 실제 실험결과치와도 대체로 일치하는 경향이었다.

난분해성 우모분해 Pseudomonas geniculata에 의한 케라틴 분해효소 생산 및 식물성장 촉진 활성 (Keratinase Production by Recalcitrant Feather Degrading Pseudomonas Geniculata and Its Plant Growth Promoting Activity)

  • We investigated the optimal conditions for keratinase production by feather-degrading Pseudomonas geniculata H10 using one variable at a time (OVT) method. The optimal medium composition and cultural condition for keratinase production were determined to be glucose 0.15% (w/v), beef extract 0.08% (w/v), $KH_2PO_4$ 0.12% (w/v), $K_2HPO_4$ 0.02% (w/v), NaCl 0.07% (w/v), $MgSO_4{\cdot}7H_2O$ 0.03%, $MgCl_2{\cdot}6H_2O$ 0.04% along with initial pH 10 at 200 rpm and $25^{\circ}C$, respectively. The production yield of keratinase was 31.6 U/ml in an optimal condition, showing 4.6-fold higher than that in basal medium. The strain H10 also showed plant growth promoting activities. This strain had ammonification activity and produced indoleacetic acid (IAA), siderophore and a variety of hydrolytic enzymes such as protease, lipase and chitinase. Therefore, this study showed that P. geniculata H10 could be not only used to upgrade the nutritional value of feather wastes but also useful in situ biodegradation of feather wastes. Moreover, it is also a potential candidate for the development of biofertilizing agent applicable to crop plant soil.

Optimization of Culture Conditions for Erythritol Production by Torula sp.

  • The medium for erythritol production by Torula sp. in a 500-ml baffled flask was optimized to be 300 g/I sucrose, 10 g/I yeast extract, 3 g/I $KH_2PO_4$, and 10 mg/I $CuSO_4{\cdot}5H_2O{\;}at{\;}34^{\circ}C$ with initial pH of 5.5. Using this optimal medium, erythritol of 166 g/I was obtained after 140 h of cultivation, corresponding to 55.3% of the erythritol yield from sucrose with a productivity of 1.11 g/I/h. Optimal concentrations of carbbon and nitrogen sources in a fermentor were higher than that in a flask due to the higher oxygen supply of the fermentor. Employing the medium containing 300 g/I or 400 g/I sucrose for the determination of optimal C/N ratio, the C/N ratio was found to be more important than the nitrogen concentration for effective erythritol production, The optimal ratio of yeast extract to sucrose (g/g) was 20. The yield and productivity of erythritol were maximal in the medium containing 400 g/I sucrose and 20 g/I yeast extract. when dissolved oxygen in the culture was increased, the cell mass increased but the erythritol production was manimal in the range of 5 to 10% of dissolved oxygen. Under the optimal the rane of 5 to 10% of dissolved oxygen. Under the optimal culture condition of the fermentor, a final erythritol concentration of 200 gI was obtained after 120 h with a yield of 50% and the productivity was 1.67 g/I/h. 한천분해효소의 고생산성 변이주의 분리 및 최적배양조건 (Isolation and Its Optimal Culture Condition for High Agarase-Producing Mutant)

  • A marine bacterium Bacillus cereus ASK202, agarase producing strain, was treated with some mutagenic agents, ultraviolte(UV), 1-methyl-3-nitro-1-nitrosoguanidine(NTG), and ethyl methane sulfonate(EMS), several times for the increasing of the agarase production After mutagen treatment, we isolated one mutant strain treated with NTG showed the highest stability and agarase productivity and named as Bacillus cereus ASK202-N3. This Bacillus cereus ASK202-N3 strain was well grown in the modified marine medium containing 0.5%(w/v) agar, 0.3%(w/v) yeast extract, and 5.0%(w/v) NaCl, and the optimal initial pH, temperature and culture time were 7.8, $25^{\circ}C$ and 32h, respectively. In the optimal culture conditions, the agarase production was increased to 5.3 fold(850units/L) compared to that of the wild type.

Effect of Nutrients on the Production of Extracellular Enzymes for Decolorization of Reactive Blue 19 and Reactive Black 5

  • Several white-rot fungi are able to produce extracellular lignin-degrading enzymes such as manganese peroxidase (MnP), lignin peroxidase (LiP), and laccase. In order to enhance the production of laccase and MnP using Trametes versicolor KCTC 16781 in suspension culture, the effects of major medium ingredients, such as carbon and nitrogen sources, on the production of the enzymes were investigated. The decolorization mechanism in terms of biodegradation and biosorption was also investigated. Among the carbon sources used, glucose showed the highest potential for the production of laccase and MnP. Ammonium tartrate was a good nitrogen source for the enzyme production. No significant difference in the laccase production was observed, when glucose concentration was varied between 5 g/l and 30 g/l. As the concentration of nitrogen source increased, a lower MnP activity was observed. The optimal C/N ratio was 25 for the production of laccase and MnP. When the concentrations of glucose and ammonium tartrate were simultaneously increased, the laccase and MnP activities increased dramatically. The maximum laccase and MnP activities were 33.7 U/ml at 72 h and 475 U/ml at 96 h, respectively, in the optimal condition. In this condition, over 90% decolorization efficiency was observed.
