Survey on the prevalence of parasites in ogol chicken of Chunbuk area (전북지방 오골계의 기생충감염 실태조사)

  • In order to monitor the parasites 417 fecal samples were taken from ogol chicken in Chonbuk area, The isolation and identification of the parasites were determined by the fecal examination using the floatation and microscopical examination, respectively. The detection rate of the eggs and unsporulated oocysts from the feces was 47.7%(199 heads), in the concerns of complicated infection with single, double and triple were 36.6%(153 heads), 9.5%(40 heads) and 1.4%(6 heads), respectively. The parasites isolated were identified as eimeria sup from 115 heads, dermanyssus spp from 103 heads, ascaridia galli from 26 heads, pterolichus obtusus from 13 heads, capillana spp from 6 heads and heterakiss gallirarum from 1 head, in order.

Prevalence and Identification of Cryptosporidium spp. from Swine Slurry

  • Cryptosporidium spp. were detected in 17 of 135 swine lagoon samples from five farms by 18S ribosomal DNA locus and PCR. Seventeen positive samples identified were included two distinctive genotypes C. suis and Cryptosporidium sp. based on a small-subunit rRNA gene-based PCR-restriction fragment length polymorphism analysis. Cryptosporidium spp. were detected out of farrowing, farrowing and nursery (mix), and finishing. Prevalence rate was 12.6% with infection rates between 3.7 and 18.5%. We concluded that Cryptosporidium oocysts can persist in treated lagoon and potentially contaminate surface water through improper discharge. This study was undertaken for the evaluation of the infection status of the genotypes of Cryptosporidium spp. in swine lagoon.

  • This study was performed to survey on the prevalence of swine internal parasites at 12 piggeries being operated by the sawdust fermentative pigsty in Kyongnam central area. To investigate the effects of viability of eggs, larva and cysts, temperatures at the underlayer of the sawdust floor and rooms of pens were taken four times each season for one year. In a pig farms which have been operated by the sawdust fermentative pigsty, eggs of Ascar is suum(8.8%), Trichr is suis(18.3%), Oesophagostomum sp(9.4%), Strongyloides ransomi(1.3%), Hyostrongylus sp(3.8%) and Metastronylus apri(2.3%) were detected from 480 heads of Pigs. Cysts and oocysts of Coccidia(12.7%), Balantidium coli(5.4%) were also detected form 480 heads of pigs. The change of the temperature in the pig room was variable between $13^{\circ}C\; to\; 27^{\circ}C$ accord-ing to outside condition, whereas that in underfloor was consistently stable between $28^{\circ}C \;to\; 31^{\circ}C$ which was strictly suitable for the development of eggs, larva and cysts.

Effects of the Feeds Supplemented with Korean Green Tea and Lactic Acid Bacteria on Infection of Eimeria maxima in Chickens (닭에서 녹차 및 유산균 혼합 사료 투여가 Eimeria maxima의 감염에 미치는 효과)

  • The chickens fed with the feeds supplemented with green tea(GT) and lactic acid bacteria(LB) were infected orally with 10,000 oocysts per chicken of E. maxima. The groups administered with the feeds supplemented with GT by 0.5% and 2.0% of feed showed the significant levels of decreasing in the number of oocysts shed for 5 days after E. maxima infection. The feeds supplemented with LB by 0.1% and 0.5% of feed were less effective in reducing the number of the fecal oocyst, compared with the groups administered with GT. To evaluate the immunoregulatory effects of the feed additives, the expression patterns of IL-2 and IFN-r in spleen cells were studied by RT-PCR and ELISA. The higher levels of IL-2 transcripts after E. maxima infection were observed in the groups with n, compared with the groups with LB and the mixture of GT and LB. The $IFN-\gamma$ mRNA bands were observed in the all of experimental groups except the uninfected control. The culture supernatants of Con A-stimulated spleen cells($5{\times}10^6cells/ml$) were measured for the concentration of IL-2 and $IFN-\gamma$ by ELISA. The levels of IL-2 and $IFN-\gamma$ on days 3 and 7 after E. maxima infection were significantly augmented in the groups with n. These results indicated that GT-supplemented feeds resulted in higher reduction of oocyst-shedding and more enhanced immune responses in the chicken infected with E. maxima, as compared with LB-supplemented feeds. According to the results, it was implicative that the supplements could be utilized for development of feed additives for anti-coccidiosis.

A survey of cryptosporidiosis among 2,541 residents of 25 coastal islands in Jeollanam-do (Province), Republic of Korea

  • In order to determine the distribution and prevalence of human cryptosporidiosis on western and southern coastal islands of Jeollanam-do (Province), fecal samples were collected from 2,541 people residing on 25 islands, 13 in the western coasts and 12 in the southern coasts, during July and August 2000. Fecal smears were prepared following formalin-ether sedimentation of the samples and stained by a modified acid-fast procedure. The presence of Cyptosporidium oocysts was determined by light microscopy. Cyptosporidium oocysts were detected in 38 specimens (1.5%). The oocyst positive rate varied (0-6.0%) according to island; the highest was detected on Oenarodo (6.0%), followed by Naenarodo (5.6%) and Nakwoldo (5.4%). The majority (35 persons, 94.6%) of Cryptosporidium-infected individuals were older than 50 years of age. Men (22/1, 159; 1.9%) were infected at a higher rate than women (16/1, 382; 1.2%). The results of the present survey indicate that human Cyptosporidium infections (due to Cyptosporidium hominis and/or C. parvum) are maintained at a relatively low prevalence on coastal islands of Jeollanam-do, Republic of Korea.

  • Cryptosporidium, a coccidian protozoa, commonly causes a self-limiting diarrheal illness in humans and animals. Fecal samples from various animals in Chonbuk district were observed using Sheather's aoatation technique, Kinyoun's modified acid-fast staining, and osmic acid pre-fixed Giemsa staining. The oocysts were detected in 74 cages(29.6%) out of 250 cages of mature mice, 26 (13.3%) out of 195 mature house rats, 75 (15.0%) out of 4-week-old 500 fowls, 98(19.9%) out of 6 to 8-month-old 500 pigs, and 111(22,2%) out of 2 to 5-year-old 500 dairy cattle, respectively. The degree of prevalence was slight in general, but actual prevalence was higher than infection rate because the detection rates were higher in repeated-preparation examinations in comparison to the first examination. Meanwhile, large and small types of oocysts were detected from mice, house rats, pigs, and cattle, and midium type from fowls.

  • The responses of gastric mucosal mast cells (GMMCS) to infection with coccid- ian protozoa, Clptosporinium muris, in mice were examined during primary and challenge infections. Each of three-week-old ICR SPF mice was orally inoculated with a single dose of 2 × 106 oocysts of C. muffs (strain MCR). After oocyst shedding ceased, the mice were orally challenged with a single dose of 2×106 oocysts of the same species. GMMCS reached a peak on days 20-30 postinoculation (Pl) in number, and decreased thereafter. An increase on days 20-30 post-challenge-infection (PCI) was also observed. The mice showed, on the whole, normal profiles of oocyst shedding in droppings. The number of the cells of uninfected control mice remained constant. .Tudging from the above results, it is suggested that mastocytosis correlate with expulsion of C. mans in primary infection and the defense mechanism of challenge infection.

Toxoplasma gondii B1 Gene Detection in Feces of Stray Cats around Seoul, Korea and Genotype Analysis of Two Laboratory-Passaged Isolates

  • The increasing prevalence of Toxoplasma gondii infection in the human population in the Republic of Korea (= Korea) is due to various reasons such as an increase in meat consumption. However, the importance of cats in transmitting T. gondii infection through oocysts to humans has seldom been assessed. A total of 300 fecal samples of stray cats captured around Seoul from June to August 2013 were examined for T. gondii B1 gene (indicating the presence of oocysts) using nested-PCR. Fourteen (4.7%) of 300 cats examined were positive for B1 gene. Female cats (7.5%) showed a higher prevalence than male cats (1.4%). Cats younger than 3 months (5.5%) showed a higher prevalence than cats (1.5%) older than 3 months. For laboratory passage of the positive samples, the fecal suspension (0.2 ml) of B1 gene positive cats was orally inoculated into experimental mice. Brain tissues of the mice were obtained after 40 days and examined for the presence of tissue cysts. Two isolates were successfully passaged (designated KNIH-1 and KNIH-2) and were molecularly analyzed using the SAG5D and SAG5E gene sequences. The SAG5D and SAG5E gene sequences showed high homologies with the ME49 strain (less virulent strain). The results indicated the importance of stray cats in transmitting T. gondii to humans in Korea, as revealed by detection of B1 gene in fecal samples. T. gondii isolates from cats were successfully passaged in the laboratory for the first time in Korea.

Efficacy of Baycox(Bay Vi 9142) Suspension against Coccidium Isospora suis in piglets (신생자돈의 콕시듐증에 대한 Baycox(Bay Vi 9142) 현탁액의 효능)

  • I suis cocciosis in piglets seems to occur in the majority of Korean sow herds. Cocci야osis is characterized by a pasty diarrhea in piglets appearing at 5 to 11 days old. Morbidity is variable, Mortality is usually low but piglets growth is always retarded. And, the principal source of infection is the environment because of the high resistance of oocysts. Control of coccidiosis is usually frustrated since the majority of treatment have not constant results. Mundt et al(1990) developed a new strategy by administrating toltrazuril as an oral suspension. This new anticoccidial drug reduced clinical signs and mortality. Therefore, we will try to medicate piglets with Baycox(Bay Vi 9142) suspension in a sow herd, having a problem of I suis coccidiosis. The incidences of coccidiosis in suckling piglets in C and D bleeding stock farm were from 28.6 to 42.3 and 21.1~57.9 % during 3 to 21 day-old. The body weights of suckling piglets in treated groups were higher than those of control groups at 11 and 18 day-old. Also, those of C breeding stock firm were higher than those of D farm. The body weight gains of suckling piglet of treated groups were higher than those of control groups. Also, those of C breeding stock firm were higher than those of D farm. There was no dead piglet in all experimental groups. Some atrophic piglets were done away with. In C breeding stock farm, the incidences of diarrhea in treatment groups were significantly lower an those of control groups. Also, the degrees of diarrhea in treated groups were significantly lower than those in control groups. In D breeding stock farm, the incidences of diarrhea in treated groups were a little lower than those in control groups. Also the degrees of diarrhea in treated groups were a little lower than those in control groups. In the treated groups, the incidences and degrees of diarrhea were reduced at the 4th day and increased at the 13th day after treatment. The number of litters excreted oocysts in treated groups wert lower than those in control groups after treatment. Also, the OPG in treated groups were lower than those in control groups. According to above results, the effect of Baycox suspension against swine coccidiosis was very good. If we will treat with Barcox suspension swine coccidiosis in swine stock farms, we can get good effects of the improvement of body weight gains and diarrhea and e reduce of excreted costs in faeces.

  • Cryposporidium species have a wide host range. These coccidian parasites are found in close association with epithelial cells of many species of animals including mm. The gastrointestinal tract is most commonly affected in young ruminants and this parasite is thought to be considerable importance in calf diarrhea complex. Major outbreaks of cryptosporidiosis have been reported in calves, lambs, pigs and others including avian species. Cryptosporidiosis is transmitted by oocysts of Cryptosporidium species. Because cryptosporidiosis is common infection among animals, early literature considered it a zoonosis. Human infections contracted from calves, cats, and horse feces. However, the resrvoir host is longer considered the major source of infection. Mild cases of disease have been reported in farm workers. Immunosuppressed, very young and very old persons should avoid contact with this parasite because it may cause severe diarrhea. In order to detect of Crytosporidium sp infection from feces of cattle and pigs at Chonbuk Iksan and Kunsan area, sedimentation and modified acid fast stain were applied. The positive rate of Cryptosporidium sp infection from 1, 176 of cattle and 267 of pigs were 0.5 % and 16.8%, respectively. According to area in Iksan and Kunsan, the positive rates were 0.4% and 0.9% from cattle, and 18.9% and 12.1% from pigs, respectively. In cattle, positive detection rate was 0.6% in milking cows but not in Korean cattle.
