Use of Lipid Extracts from Various Oil Grains to Supply Dietary Omega-3 Fatty Acids for Dairy Foods - A Preliminary Study

  • The objective of this research was to determine the grain oil most suitable for the production of omega-3 fatty acids (FAs) in China, in order to supply dietary omega-3 FAs. This study focused on the nutritional significance of omega-3 FAs and the wide search of sources of dietary omega-3 FA from oil grains produced in China. Five oil grains produced in the Yanbian region, namely sesame, flax, peanut, soybean, and corn, were selected and analyzed for their fat content and FA composition. Results showed that the lipid content in sesame, flax, and peanut, which was more than 30%, was higher than that in soybean and corn. The polyunsaturated FA content was found to be directly proportional to omega-3 FA content. Flax showed a considerably higher omega-3 FA content (53.43%) than other samples, suggesting its potential as a source of dietary omega-3 FA. The optimal daily intake of flax for males and females was determined to be 2.99 g (over one table spoon) and 2.06 g (nearly one table spoon), respectively. Flax showed the lowest ratio of omega-6 and omega-3 FA (0.32%), which can help reduce this ratio in the human body; the intake of equal amounts of flax and corn could achieve an optimum ratio of 1:1. These results can prove to be valuable in the preparation of various functional dairy foods in the future.

$\omega$6/$\omega$3계 불포화지방산을 투여한 후 혈장의 지질조성과 Tocopherol, Malondialdehyde 형성 및 적혈구의 Hemolysis 변화에 대한 Kinetic 연구 (Kinetic Study on Variations of Lipids, Tocopherol and Malondialdehyde Levels of Plasma and Red Blood Cell in Young Women fed Dietary $\omega$6/$\omega$3 Polyunsaturated Fatty Acids)

  • Ten college women were treated with either corn oil(CO< as a source of C18:2, $\omega$6) or fish oil concentrates(FO, as a source of C20:5+C22:6, $\omega$3) with experimental diet for 7 days and then returned to normal home made diet. Kinetic changes of plasma and RBC lipids, tocopherol, MDA levels and hemolysis were observed at day-0, -3, -7, -8, -10, -14 and -21 of experimental periods. Plasma and RBC tocopherol contents were significantly increased at day-3 and -7 by tocopherol tocopherol supplement in dietary oil, but there was no significant difference between CO and FO diet with large dose of tocopherol supplement. After stop experimental regimen at day-7, plasma tocopherol content was sustained at high level until day-10 but drastically decreased at day-14 and remained at low level at day-21. However, RBC tocopherol level was not greatly responded to the dietary intake of tocopherol and was varied in the very narrow range. MDa levels of plasma and RBC were not responded in the same way as tocopherol content of plasma. MDa content of RBC was very low compared to that of plasma. RBC hemolysis by incubation in hypotonic solution was negatively correlated to plasma tocopherol level and was not correlated to the level of MDa in either plasma or RBC. There were no significant change in the levels of plasma cholesterol, HDL-chol, triglyceride, phospholipid, and lipoprotein pattern throught experimental periods.

자기조립단분자막을 위한 보편적이고 기능화된 긴 사슬 알킬티올 연결자의 제조 (Preparations of Universal, Functionalized Long-Chain Alkylthiol Linkers for Self-assembled Monolayers)

  • 본 연구에서는 자기조립단분자막(SAM)의 연구에 유용한 일련의 $\omega$-mercapto alkyl amine 1 및 $\omega$-mercapto alkanoic acid 2를 만드는 제조공정을 서술하였다. 먼저 첫 번째 목표물질인 $\omega$-mercapto alkyl amine 1의 제조방법은 다음과 같다: 먼저 시중에서 시판되는 potassium phthalimide와 $\omega$-bromo alcohol을 $80{^{\circ}C}$, DMF 용매에서 치환반응으로 화합물 3을 합성하였다. 아민기와 알코올기를 포함하는 화합물 4을 합성하기 위하여 먼저 화합물 3를 hydrazine hydrate로 환류시킨 후, 이어서 c-HCl로 처리하여 부반응물 5를 수반하여 생성물 4를 76-98% 수율로 만들었다. $\omega$-aminoakanol 4의 hydroxyl기를 HBr로 브로민화반응을 하여 $\omega$-bromoamine 화합물 6을 34-97% 수율로 만들었다. 티올기를 도입하기 위하여 화합물 6을 95% 에탄올 속에서 thiourea와 치환 반응하여 $\omega$-aminoalkanthiuronium 7을 만든 다음, 이 화합물을 센 염기(KOH)와 센 산으로 처리하여 목표화합물, $\omega$-mercapto alkylamines 1을 총 5단계를 걸쳐서 제조하였다. 덧붙여 두 번째 목표물질인 $\omega$-mercapto alkanoic acid 2는 다음과 같이 2단계를 통하여 제조하였다: $\omega$-bromo alkanoic acid를 95% 에탄올 속에서 thiourea로 처리하여 화합물 7을 만든 다음, 센 염기(KOH)를 처리하여 thiuronium bromide 를 제거한 후, 다시 센 산(HCl) 수용액으로 처리하여 두 번째 목표화합물 $\omega$-mercapto alkanoic acid 2을 얻었다. 제조한 긴 사슬 알킬티올 1과 2 유도체들은 금속(Au, Pt, Ti)에 자기조립단분자 막을 형성함으로써 단백질, 효소 및 다양한 생체분자를 고정하는 연결자로 사용될 수 있으며, 그 밖에 금속의 표면개질을 이용하여 다양한 응용 연구를 위한 화학 도구로 사용될 수 있다.

Effects of Fatty Acids and Vitamin E Supplementation on Behavioral Development of the Second Generation Rat

  • In this study, we examined the effects of dietary fatty acids on the fatty acid composition of phospholipid fractions in regions of the brain and on behavioral development in rats. The Sprague Dawley rats were fed the experimental diets 3~4 wks prior to the conception. Experimental diets consisted of 10% fat(wt/wt) which were from either safflower oil (SO, poor in $\omega$3 fatty acids), mixed oil MO, P/M/S ratio : 1:1.4:1, $\omega$6/$\omega$3 ratio = 6.3), or mixed oil supplemented with vitamin E (+500 mg/kg diet). At 3 and 9 weeks of age, frontal cortex (FC), corpus striatum (CS), hippocampus (H), and cerebellum (CB) were dissected from the whole brain. The fatty acid content was determined in the different phospholipid fractions: phosphatidylcholine (PC), phosphatidyl-serine (PS), and phosphatidylethanolamine (PE) in the rat brain regions. In the visual discrimination test, the order of the cumulative errors made in Y-water maze test were SO > MO > ME. This suggested that the balanced diet supplemented with vitamin I had the most beneficial effect on learning ability. The overall characteristics of correlation between fatty acids and behavior development were that the frequency of cumulative errors were negatively correlated significantly with monounsaturated fatty acids (MUFAs), ie., 18:1 $\omega$9 and 22:1 $\omega$9. Docosa-hexaenoic acid (22:6 $\omega$3) of PS in frontal cortex (FC) was negatively correlated with the number of errors made in the Y-water maze test.22:5 $\omega$6 PS in hippocampus (H), PC and PE in corpus striatum (CS), PC in cerebellum (CB) were positively correlated with cumulative errors. And these errors were negatively correlated with 20:4 $\omega$ 6 of PE in corpus striatum (CS) and PC in cerebellum (CB). Especially, O1eic acid (18:1 u 9) in all phospholipid fractions (PC, PS, PE) of hippocampus was negatively correlated with the number of errors. These findings demonstrate that the MUFAs were might be essential for proper brain development, especially in hippocampus which is generally thought to be the regions of memory and learning.

  • ${\omega}3$계와 ${\omega}6$계 PUFA의 함유비율을 달리했을때 흰쥐장기의 지질대사 및 혈청 lipoprotein 조성에 미치는 영향을 알아보고자 실험한 결과를 요약하면 아래와 같다. 뇌 조직중의 총cholesterol농도는 전 식이군 간에 유의성은 없으나 ${\omega}6$계 linoleic acid 함유율이 높은 고추씨기름 10%단독 식이군에서 높은 함량을 보이는 반면 ${\omega}3$계 linolenic acid 함유율이 높은 들깨유 식이군 및 혼합 식이군에서 감소하는 경향을 보였다. 한편 중성지방 및 인지질 농도는 고추종자유 함유율이 높은 식이군에서 서서히 증가하지만 들깨유 함유량이 많아질수록 점차 감소하였다. 신장 조직중의 총cholesterol농도는 전 군간에 유의성이 없었으며, 중성지방 농도는 ${\omega}6$계 linoleic acid함유 비율이 높은 고추종자유 식이군이 가장 높았고 혼합 식이군에서 낮았다. 인지질 농도는 10% 고추종자유 식이군에서 현저히 낮았다. 고환 조직중의 총cholesterol농도는 혼합 식이군 및 들깨유 식이군에서 낮았으며, 중성지방 농도는 들깨유 식이군에서 상승하였고 혼합 식이군에서 감소하였다. 인지질 농도는 5군에서 낮았다. 혈청 lipoprotein pattern에서 HDL은 들깨유 함량이 많은 식이군(1,2,3군)에서 상승된 반면 고추씨기름 식이군에서는 가장 낮았으며, LDL은 들깨유 비율이 높아질수록 점차로 낮아졌다.

어미 쥐의 $\omega$3계 및 $\omega$6계 지방산 식이가 제2세대 쥐의 뇌조직 지방산 성분에 미치는 영향 (Effect of Maternal Dietary $\omega$3 and $\omega$6 Polyunsaturated Fatty Acids on the Fatty Acid Composition of the Second Generation Rat Brain)

  • The change in fatty acid composition in brain tissue of the second generation rats(Sprague-Dawley strain) was studied using four different fat diets(Corn oil=CO, Soybean oil=SO, Perilla oil=PO, Fish oil=FO, 10% by Wt). The experimental diets were started from pregnancy in four different groups, each consisting of 9 rats. The seound generation rats were fed the same diet as their mothers. Animals were anesthetized with ether at 0, 3, 9 & 16 weeks of age. Whole brains were dissected out, brain tissues were, then, homogenized and lipids were extracted from brain tissues. The fatty acid compositions were measured after methylation by gas-liquid chromatography at 0, 3, 9 and 16 weeks of age of offspring. The changes in the relative concentrations of polyunsaturated fatty acids(PUFA) or more specifically docosahexaenoic acid(22 : 6, $\omega$3, DHA), the major $\omega$3 fatty acid component in rat brain at different age were similar to changes in the amount of DNA in brain tissue showing the maximum value during the lactation. The changes in saturated fatty acid(SFA) content showed a contrasting patten to those of PUFA, while monounsaturated fatty acid(MUFA) increased steadily throughout the experimental period. At birth, the relative concentrations of $\omega$3 series fatty acids the relative concentrations of PUFA, MUFA and SFA converged to very similar values respectively regardless of the dietary fatty acid compositions. In brain tissue, it is of value to note that while changes in relative concentrations of linoleic acid (18 : 2, $\omega$6, LA) and arachidonic acid(20 : 4, $\omega$6, AA) showed a precursor-product-like relationship, $\alpha$-linolenic acid(18 : 3, $\omega$3, $\alpha$-LnA) and DHA showed a different pattern. Even when the $\omega$3 fatty acid content in very low in maternal diet(CO), the second generation rat brain tissues appeared to secure DHA content, suggesting an essential role of this fatty acid in the brain. The fact that a large amount of $\alpha$-LnA in the maternal diet did not have a significant effect on the second generation rat brain $\alpha$-LnA content, indicated that DHA seemed essential component for the brain development in our experimental condition. In all groups, the relative content of $\alpha$-LnA in the brain tissues remained relatively constant throughout the experimental period at the very low level. The study of the specific concentrations and essential role(s) of DHA in each parts of brain tissue is needed in more details.

한국산 잣기름이 정상토끼 및 동맥경화증 유발 토기의 적혈구막 지방산조성에 미치는 영향 (Effect on Fatty Acid Composition of red Blood Cell Membranes of supplement of Korean Pinenut Oil, rich in 5-Olefinic acids, in Normocholesterolemic and Atherosclerotic New Zealand White Rabbits)

  • The present study was carried out in normal and atherosclerotic New Zealand white(NZW) rabbits, to evaluate the effect of dietary supplementation with Korean pinenut oil, on the total fatty acid composition of red blood cell membranes. Erythrocyte fatty acids of chain lengths ranging fro 12 : 0 to 22 : 6$\omega$3 were identified. In study I, palmitic acid(16 : 0) was found to be the main erythrocyte membranes fatty acid, followed in turn by stearic(18 : 0), oleic(18 : 1), and linoleic acids(18 : 2$\omega$6). There was a large increase in the proportion of 20 : 4$\omega$6 in 10% pinenut oil group on days 40 and 80(compared with control), although the increase was not statistically significant. Most erythrocyte fatty acids were less affected by the types or levels of oils supplemented for periods fo 40 and 80days. In study II,at the end of the dietary treatment, the levels of 12 : 0, 14 : 0, 15 : 0 and 18 : 0 were significantly decreased in the 5% soybean oil/cholesterol group, whereas those of 12 : 0, 14 : 0 and 18 : 0 were significantly decreased, but that of 18 : 2$\omega$6 was significantly increased in the 10% soybean oil/cholesterol group, compared to the cholesterol group. No significant differences in fatty acid composition were seen between the pinenut oil/cholesterol group, whereas the 10% soybean oil/cholesterol group had significantly elevated 18 : 2$\omega$6, compared to the cholesterol group. The proportion of 18 : 4$\omega$6 was significantly raised in the 5% soybean oil/cholesterol and 5% pinenut oil/cholesterol group, and the content of 16 : 1$\omega$7 was significantly lowered in the 5% and 10% soybean/cholesterol groups, compared to the cholesterol group at 10 weeks supplementation. In studies I,II and III, the total proportions of saturated fatty acids were not significantly affected by the types of levels of oils supplemented throughout the study period.

  • 본 연구에서는 세 모드 사이의 내부공진을 고려하여 강제진동 중인 보의 비선 형 해석을 다루고자 한다. 이 문제에 관심을 갖게 된 동기는 "연속계의 비선형해석 에서 더 많은 모드를 포함시키면 어떤 결과를 낳게 될 것인가\ulcorner" 라는 질문에서 생겨난 것이다.

  • 본 연구에서는 ${\omega}$-6 및 ${\omega}$-3계 지방산을 함유한 유지의 첨가비율을 달리한 실험사료를 어린 병아리에서 급여했을 때 면연 기관의 지방산조성에 미치는 영향을 조사하였다. 5개의 처리구에 각 처리당 3반복을 두고 총 75수의 1일령 수평아리를 공시하였다. 옥수수유(CO)와 들깨유(PO)를 중량비로 각각 8%+0%, , 6%+2%, 4%+4%, 2%+6%, 0%+8% 수준으로 첨가한 반정제 실험 사료를 7주간 급여하였다. 실험 종료시 증체량 및 사료섭취량은 처리 간에 큰 차이가 보이지 않았으며 체중 100g당 간장 중량 및 면역 기관 중량에서도 유의한 차이가 인정되지 않았다. 면역조직 내 지방산 조성은 EPA와 AA(arachidonic acid)를 포함하여 섭취한 사료 지방산의 조성을 잘 반영하는 경향을 보였는데, 면역기관 간에 그 정도에 차이가 있었다. 들깨유의 첨가수준이 증가함에 따라 linoleie acid LA)의 비율이 점진적으로 감소하였고 반면 a-linolenic acid(LNA) 비율은 현저하게 증가하는 경향이었다. 흉선에는 .LA와 LNA의 함량이 다른 조직보다 현저히 더 많아서 섭취하는 지방산의 조성이 가장 잘 반영되었다. 비장 조직에는 HPA 및 AA 농도와 EPA/LNA, AA/LA의 비율이 다른 조직에 비해 현저히 더 높았는데 이는 비장이 LA나 LNA로부터 AA나 EPA로 전환하는 능력이 높음을 의미한다. 실험사료 급여후 2, 4주후에 조사된 anti-BSA 항체가 유의한 차이는 인정되지 않았으나 CO 8%구에 비해 PO를 첨가한 모든 구에서 더 많이 생성되는 경향이 관찰되었고 특히 PO를 6% 및 8% 첨가한 실험구에서 20~30%나 더 높아졌다. 결론적으로 섭취하는 지방산 조성에 따라 면역기관내 모든 지방산 조성이 변하였으며, 면역기관에 따라 그 반응정도가 달라지는 것이 관찰되었다. 이는 eicosanoids 합성에도 영향을 미칠 수 있기 때문에 가금에서 질병에 대한 면역력 향상이라는 관점에서 앞으로 더 세밀한 연구가 필요하다.

과도전자탐사법에 의한 모르타르 중의 코팅 철근의 부식 측정 방법 개발 (The Development of Measuring Method of Coated Steel Corrosion in Mortar by Transient Electro-Magnetic(TEM) Method)

  • To study measurement of coated steel corrosion in mortar, a transient electro-magnetic(TEM) method was adapted. The sensors were made of enamelled wire with diameter of 0.25mm(transmitter), 0.1mm(receiver) and the secondary electro motive force(EMF) was measured with SIROTEMIII. The sensors configuration was used as in loop configuration. After coated steels were corroded by the salt spray during 3, 7, 15, 25days, they were embedded in mortar which were made from sand : cement : water ratio of 2 : 1 :0.5. To investigate coated steel corrosion in mortar, the sensors were used. ( sensorl - $T_x$ : $4{\Omega}$, $R_x$ : $10{\Omega}$, $3{times}3cm$, sensor2 - $T_x$ : $8{\Omega}$, $R_x$ : $10{\Omega}$, $3{times}3cm$, sensor3 - $T_x$ : $4{\Omega}$, $R_x$ : $10{\Omega}$, $6{times}6cm$, sensor4 - $T_x$ : $8{\Omega}$, $R_x$ : $10{\Omega}$, $6{times}6cm$). The obtained results showed that the secondary EMF was decreased with specimens of 3, 7days corroded coationg steel in mortar and then increased with specccimens of 15, 25days corroded one. And it was confirmed that measurement of coated steel corrosion in mortar by a transient electro-magnetic(TEM) method is possible.
