Changes in the glucose and insulin responses according to high-protein snacks for diabetic patients

  • BACKGROUND/OBJECTIVES: This study aimed to develop healthy, appetizing high-protein snacks with enhanced isolated soy protein for diabetic patients and determine the blood glucose and insulin response after being consumed by these patients. MATERIALS/METHODS: Thirty adult patients aged between 30 and 75 years, with a ≤ 10-year history of type 2 diabetes and hemoglobin A1c of < 7.5%, were enrolled in this study. They made 3 clinical visits at one-week intervals. The control group consumed 50 g carbohydrates (white bread), whereas the test groups consumed high-protein grain (HP_G) or high-protein chocolate (HP_C) after an 8-hrs fast. Blood (2 ㎤) was drawn at 15, 30, 45, 60, 90, and 120 min before and after consumption to analyze the blood glucose and insulin concentrations. RESULTS: Compared to the commercial snacks, the developed high-protein snacks had below-average calorie, carbohydrate, and fat content and a 2.5-fold higher protein content. In diabetic patients who consumed these snacks, the postprandial blood glucose increased between 15 min and 2 h after consumption, which was significantly slower than the time taken for the blood glucose to increase in the patients who consumed the control food product (P < 0.001). Insulin secretion was significantly lower at 45 min after consumption (P < 0.05), showing that the high-protein snacks did not increase the blood glucose levels rapidly. The incremental area under the curve (iAUC), which indicated the degree of blood sugar and insulin elevation after food intake, was higher in the control group than the groups given the 2 developed snacks (P < 0.001), and there was no significant difference in insulin secretion. CONCLUSIONS: The results of the postprandial blood glucose and insulin response suggest that high-protein snacks are potential convenient sources of high-quality protein and serve as a healthier alternative for patients with type 2 diabetes, who may have limited snack product choices. Such snacks may also provide balanced nutrition to pre-diabetic and obese individuals.

  • Objectives: In this study, we investigated the antiobesity and antidiabetic effects of polyherbal extract, DM2 consisting of Atractylodis Rhizoma, Anemarrhenae Rhizoma, Cinnamomi Cortex, and Moutan Radicles Cortex in high fat diet-induced obesity mice. Methods: DM2 extract was prepared with a hot water. Six-week-old male C57BL/6N mice were fed a high-fat diet (HFD) for 8 weeks and then administrated with DM2 extract (500 mg/kg, p.o.) for 4 weeks. The changes of physiological markers, body weight (BW), food and water intakes, and the levels of fasting blood glucose (FBG) were measured once a week for 4 weeks in mice. The the serum levels of glucose, insulin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), total cholesterol (T-CHO), triglyceride, and low density lipoprotein cholesterol in sera were measured in mice using autometic chemical analyzer and enzyme linked immunosorbant assay. We also observed the histological changes of liver and pancreatic tissues with Hematoxylin & Eosin staining. Results: In physiological change, the increases of BW, calorie intake, and FBG in HFD-induced obese mice were significantly decreased after administration of DM2 extract for 4 weeks. The decrease of water intake was significantly increased in DM2 extract-administrated mice. In serological change, the administration of DM2 extract in obesity mice was significantly decreased the serum levels of glucose, insulin, T-CHO, AST, and ALT levels. We also found that DM2 extract inhibited the increase of lipid droplets in liver and the structural destruction of pancreatic tissues in obesity mice. Conclusion: Our study demonstrated that DM2 extract has antiobesity antidiabetic effects with body weight loss, decrease of glucose and insulin levels, and lipid accumulation on liver tissue.

Effect of Monounsaturated Fatty Acid-enriched Peanut Consumption on Serum Lipid in High Fat Diet-induced Mice (고지방 식이 유도 비만 마우스에서 고올레산 땅콩 섭취의 혈장 지질 개선 효과)

  • This study was conducted to evaluate the effect of high-oleate and normal-oleate peanut consumption on adipose mass and serum lipids in obese-induced C57BL/6J mice. After four weeks of the high-fat diet, mice were randomly divided into six groups: normal control (NC) diet, high-fat control (HFC) diet, high-oleate peanut-seed (HOPS) diet, normal-oleate peanut-seed (NOPS) diet, high-oleate peanut-oil (HOPO) diet, and olive-oil (OO) diet. After four weeks, all four experimental diet groups showed significantly lower body weight and epididymal fat weight than HFC group. In four experimental diet groups, serum triglycerides (TG), total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) were significantly lower, and high-density lipoprotein cholesterol (HDL-C) was significantly higher than HFC group. TG was significantly decreased in HOPS group (92.1±1.2 mg/dL) than NOPS group (101.7±5.3 mg/dL, p<0.05). Similarly, LDL-C was significantly lower in HOPS group (66.1±2.8 mg/dL) than NOPS (76.9±1.5 mg/dL, p<0.05), on the other hand, HDL-C indicated a significant elevation in HOPS (50.5±2.1 mg/dL) than NOPS group (45.2±1.6 mg/dL, p<0.05). This result suggests that the consumption of high-oleate peanut has a favorable effect on the plasma lipid profile.

Effects of Changbudodam-tang on Obesity-induced Rats (창부도담탕이 비만 유도 흰쥐에 미치는 영향)

  • Objectives: This study was designed to evaluate the efficacy of Changbudodam-tang on obesity by using high-fat diet rats. Methods: Rats were divided into five groups. Normal group: Normal diet, Control group: High-fat diet, Positive control group: High-fat diet+Dietamin 4 mg/kg/day, Changbudodam-tang-Low group: High-fat diet+Changbudodam-tang 250 mg/kg/day, Changbudodam-tang-High group: High-fat diet+Changbudodam-tang 500 mg/kg/day. Weight, food intake were measured every week. After 7 weeks, total cholesterol, high density lipoprotein-cholesterol, low density lipoprotein-cholesterol, Triglyceride, free fatty acid, aspartate aminotransferase, alanine aminotransferase, complete blood count were measured and messenger ribonucleic acid expression of adiponectin, peroxisome proliferator-activated receptor-γ, leptin were observed using Reverse transcription polymerase chain reaction of liver cells. Results: There was no difference in food intake between groups. Body weight tended to decrease compared with the Control group, but it wasn't statistically meaningfull. The total cholesterol, low density lipoprotein-cholesterol, Triglyceride, free fatty acid tended to decrease compared with the Control group. High density lipoprotein-cholesterol tended to decrease compared with the Control group, but it wasn't statistically meaningfull. White blood cell, red blood cell, hemoglobin, platelet, aspartate aminotransferase, alanine aminotransferase were not affected by Changbudodam-tang. The messenger ribonucleic acid expression of Adiponectin, peroxisome proliferator-activated receptor-γ, leptin, which are involved in the differentiation of adipocytes, was decreased compared with the Control group. Conclusions: Based on the results above, it is suggested that Changbudodam-tang can be applied to improving serum lipid levels in obese patients caused by high fat diets.

  • Obesity is closely associated with profound dyslipidemia, insulin resistance, and fatty liver disease. Recent reports have suggested that alterations in gut microbiota can be linked to diet-induced obesity. In this study, the anti-obesity effects of Weissella confusa WIKIM51 isolated from kimchi were investigated, as evidenced by: i) reduced lipid accumulation and downregulated adipogenesis-related genes in 3T3-L1 adipocytes; ii) suppressed gains in body weight and epididymal fat mass; iii) reduced serum lipid levels, for example, triglyceride and total cholesterol; iv) increased serum adiponectin levels and reduced serum leptin levels; v) downregulated lipogenesis and upregulated β-oxidation-related genes in the epididymal fat; and vi) altered microbial communities. The collective evidence indicate the potential value of W. confusa WIKIM51 as a functional food supplement for the prevention and amelioration of obesity.

Protective Effect of Curcumin on Dextran Sulfate Sodium-Induced Ulcerative Colitis in Obese Mice (비만마우스에서 Dextran Sulfate Sodium 유도 궤양성 대장염에 대한 커큐민의 보호효과)

  • Ulcerative colitis is a disease that causes inflammation in the mucosal or submucosal layer of the colon. Previous studies have reported that obesity increases the prevalence of ulcerative colitis and aggravates the progression. This study was therefore undertaken to investigate whether curcumin inhibits the progression of ulcerative colitis caused by obesity. Mice were bred on a high-fat diet to induce obesity, and curcumin was administered with the high-fat diet to confirm the anti-inflammatory effect. To induce ulcerative colitis, dextran sulfate sodium (DSS) was administered orally, and clinical symptoms of colitis were subsequently observed. For histological evaluation of curcumin, the colon, liver and abdominal fat tissue samples were prepared and analyzed by hematoxylin and eosin (H&E) and Alcian blue-periodic acid-Schiff (PAS) staining. Our results confirm that consumption of curcumin resulted in decreasing the score of the disease activity index, and inhibited shortening of the colon length. In addition, inflammatory cell infiltration and mucosal damage were inhibited in the colon tissue of ulcerative colitis exacerbated by obesity. We further confirmed that exposure to curcumin significantly reduced the steatosis area of the liver and adipocytes of abdominal fat. In conclusion, we believe that curcumin can be applied as a therapeutic agent to treat ulcerative colitis, by inhibiting the progression of colitis caused by obesity.

Development of a Bodice Prototype Drafting Method for 20s Plus-size Women

  • This study attempted to develop a bodice prototype that is the basis for the production of upper body clothing for 20s plus-size women who have distinct characteristics from women with general body types. Through this, it was intended to provide basic data necessary for the development of plus-size women's clothing, which is revitalizing the market due to the increase in the obese population. Through the first evaluation of appearance and abdominal pressure, patterns such as moving the side neck, adding the amount of armhole dart, and adding the amount of sagging were modified. Through the second evaluation, corrections such as vertical side lines, reduction of the neck of the back center line, and adding the amount of armhole darts were performed. Through the third evaluation, the final pattern drafting method was developed by vertically modifying the side line and adding the amount of back armhole darts. In the case of 20s plus-size female body types, a drafting method distinguished from the general body type was required in the method of setting the side and hem due to the protrusion of the abdomen. This study can be said to be meaningful in that it proposed a bodice prototype drafting method suitable for the body type of 20s plus-size women. In the follow-up study, it is thought that the wearability should be evaluated through actual garment wearing.

  • In order to develop a bodice prototype suitable for obese women in their 20s, this study compared and analyzed a total of five pattern system methods, including prototype of France and Japan, one prototype for education in Korea and two prototype for industries in Korea. Through this, this study attempted to investigate the bodice pattern system suitable for the development of plus-size women's clothing. For the pattern drafting, pattern 1 was applied with the most body dimensions such as interscye fold front, interscye fold back, bust circumference, neck circumference, and waist back length, and pattern 2, pattern 4, and pattern 5 were made based on bust circumference and waist back length. As a result of the appearance evaluation, Pattern 3 was evaluated as the best pattern in all items except for the suitability of the center front length and the suitability of the side waist circumference position. However, it was evaluated as inappropriate in items such as the vertical side line, the suitability of the side waist circumference line position, the suitability of interscye fold back position and shape, and the suitability of the back shoulder dart position. Most of the pattern drafting methods are based on the size of the bust circumference, but other institutional methods are considered necessary when setting the neck circumference and shoulder length for 20s obesity women. In addition, it is also required to develop a method for setting the front center length due to abdominal protrusion.

EID3 Promotes Glioma Cell Proliferation and Survival by Inactivating AMPKα1

  • Objective : EID3 (EP300-interacting inhibitor of differentiation) was identified as a novel member of EID family and plays a pivotal role in colorectal cancer development. However, its role in glioma remained elusive. In current study, we identified EID3 as a novel oncogenic molecule in human glioma and is critical for glioma cell survival, proliferation and invasion. Methods : A total of five patients with glioma were recruited in present study and fresh glioma samples were removed from patients. Four weeks old male non-obese diabetic severe combined immune deficiency (NOD/SCID) mice were used as transplant recipient models. The subcutaneous tumor size was calculated and recorded every week with vernier caliper. EID3 and AMP-activated protein kinase α1 (AMPKα1) expression levels were confirmed by real-time polymerase chain reaction and Western blot assays. Colony formation assays were performed to evaluate cell proliferation. Methyl thiazolyl tetrazolium (MTT) assays were performed for cell viability assessment. Trypan blue staining approach was applied for cell death assessment. Cell Apoptosis DNA ELISA Detection Kit was used for apoptosis assessment. Results : EID3 was preferentially expressed in glioma tissues/cells, while undetectable in astrocytes, neuronal cells, or normal brain tissues. EID3 knocking down significantly hindered glioma cell proliferation and invasion, as well as induced reduction of cell viability, apoptosis and cell death. EID3 knocking down also greatly inhibited tumor growth in SCID mice. Knocking down of AMPKα1 could effectively rescue glioma cells from apoptosis and cell death caused by EID3 absence, indicating that AMPKα1 acted as a key downstream regulator of EID3 and mediated suppression effects caused by EID3 knocking down inhibition. These findings were confirmed in glioma cells generated patient-derived xenograft models. AMPKα1 protein levels were affected by MG132 treatment in glioma, which suggested EID3 might down regulate AMPKα1 through protein degradation. Conclusion : Collectively, our study demonstrated that EID3 promoted glioma cell proliferation and survival by inhibiting AMPKα1 expression. Targeting EID3 might represent a promising strategy for treating glioma.

Massa Medicata Fermentata improves fatty liver in high fat diet-fed nonalcoholic fatty liver disease's mouse model (고지방식이 유도 비알콜성지방간 마우스 모델에서 육신국(六神麯)의 지방간(脂肪肝) 개선효과)

  • Objectives : This study was undertaken to verify the effects of Massa Medicata Fermentata (MMF) on nonalcoholic fatty liver disease (NAFLD) using high fat diet-fed male mice. Methods : Fifty four male C57BL/6N mice (age matched) were used for all experiments. Nine standard chow diet-fed mice were used as normal group and forty five high fat diet-fed obese mice were randomly divided into 5 groups: control, atorvastatin-10mg/kg, MMF(1)-62.5mg/kg, MMF(2)-125mg/kg and MMF(3)-250mg/kg. After all groups were treated with several kinds of diets for 8 weeks, we measured body weight gain, adipose tissue weights, plasma lipid and glucose metabolism, visceral organ weights, histological analysis for liver on the mice. Results : MMF-treated mice had lower body weight gain compared with controls. Among MMF-treated mice, the effect was magnified in MMF(2). MMF(3)-treated mice had lower blood plasma total cholesterol (TC) and glucose level compared with controls. MMF decreased hepatic lipid accumulation, liver fibrosis and liver inflammation of mice compared with controls. The effects was maximized in MMF(2) and atorvastatin. Blood plasma aspartate aminotransferase (AST), alanine aminotransferase (ALT), ${\gamma}$-glutamyltransferase (${\gamma}$-GT) concentrations tends to be decreased by MMF compared with controls. Blood plasma AST, ALT, ${\gamma}$-GT concentrations and organ weights were not changed by MMF, indicating that all three kinds of MMF do not show any hepatotoxicity. Conclusions : These results suggest that MMF improves NAFLD by reducing body weight gain, hepatic lipid accumulation, liver fibrosis, liver inflammation.
